Infection during the perinatal period can adversely affect brain development, predispose infants to ischemic stroke and have lifelong consequences. We previously demonstrated that diet enriched in n-3 polyunsaturated fatty acids (n-3 PUFA) transforms brain lipid composition in the offspring and protects the neonatal brain from stroke, in part by blunting injurious immune responses. Critical to the interface between the brain and systemic circulation is the vasculature, endothelial cells in particular, that support brain homeostasis and provide a barrier to systemic infection. Here, we examined whether maternal PUFA-enriched diets exert reprograming of endothelial cell signalling in postnatal day 9 mice after modeling aspects of infection using LPS. Transcriptome analysis was performed on microvessels isolated from brains of pups from dams maintained on 3 different maternal diets from gestation day 1: standard, n-3 enriched or n-6 enriched diets. Depending on the diet, in endothelial cells LPS produced distinct regulation of pathways related to immune response, cell cycle, extracellular matrix, and angiogenesis. N-3 PUFA diet enabled higher immune reactivity in brain vasculature, while preventing imbalance of cell cycle regulation and extracellular matrix cascades that accompanied inflammatory response in standard diet. Cytokine analysis revealed a blunted LPS response in blood and brain of offspring from dams on n-3 enriched diet. Analysis of cerebral vasculature in offspring in vivo revealed no differences in vessel density. However, vessel complexity was decreased in response to LPS at 72 h in standard and n-6 diets. Thus, LPS modulates specific transcriptomic changes in brain vessels of offspring rather than major structural vessel characteristics during early life. N-3 PUFA-enriched maternal diet in part prevents an imbalance in homeostatic processes, alters inflammation and ultimately mitigates changes to the complexity of surface vessel networks that result from infection. Importantly, maternal diet may presage offspring neurovascular outcomes later in life.

The study focuses on developing a novel perilla seed oil (PeO)‐based polyunsaturated fatty acid–rich margarine fat analogue using enzymatic interesterification. PeO is a rich source of omega‐3 fatty acids, however, has limited application due to susceptibility to oxidative and thermal degradation. Moreover, low consumption of omega‐3 fatty acids in modern diets serves as a major cause for increased prevalence of cardiovascular diseases. The stability of such oils can be improved by techniques like blending and interesterification. However, blending lacks uniformity, which can be overcome by interesterification. The process conditions, namely, enzyme load, temperature, substrate molar ratio, and reaction time, were systematically optimized using response surface methodology. The optimized interesterified fat was characterized for fatty acid and triacylglyceride (TAG) composition. The physicochemical and functional properties, along with oxidative stability, were also evaluated to determine its overall quality. The structured lipid with zero trans and low saturated fat was developed with 50:50 substrate molar ratio of PeO and palm stearin at 54°C within 5.4 h at 6.2% w/w concentration of TLIM enzyme. It exhibited 23.8% degree of interesterification, 435.5 g hardness, and 33°C slip melting point. The TAG profile of the resulting fat was significantly modified with increased triunsaturated TAGs like LnLnLn and LnLnL but reduced trisaturated TAGs like PPP. The interesterification process lowered tocopherol composition by 3%. The acid, peroxide, IP (Induction Period), and p‐anisidine values of PeO significantly enhanced. The thermal behavior of the developed fat was also modified. Seabird reproductive success in northern latitudes is often linked with prey abundance, availability, size, or species. Causes are often broadly explained as changes in ocean currents/temperatures, or diets. Few studies trace diets down the food web to primary producers, thus missing what I propose as an underlying cause of seabird colony failure, dietary abundance of essential omega-3 polyunsaturated fatty acids (PUFA). PUFA differ from other nutrients because they are, in part, ligands—critical for reproduction and other physiological processes for the entire marine food web, not just for seabirds. Diatoms are one of the few life forms that can produce PUFA de novo , and a lack of PUFA in zooplankton and fish reduces their abundance and productivity, with consequences up the food web. Since the mid-1980s, historical prey of breeding seabirds has decreased, and frequency of colony failures has increased. In years of failure, prey often had less fat than historically preferred species. Likewise, proportions of alternate species' PUFA, from published papers, show lower values than in historical prey. Production of PUFA by diatoms is depressed in warmer and more acidic environments, and I hypothesize that these warmer and more acidic seas have affected production of PUFA over the past 3–4 decades. The present study investigated the impact of seasonal variation on biomass availability, proximate composition, fatty acid profile, minerals content, pigments, moisture content, and elemental composition of an edible green alga Gayralia brasiliensis collected from Shirgaon estuary Maharashtra, west coast of India. Additionally, the physico-chemical parameters of the Shirgaon estuary were analyzed and correlated with biomass abundance and biochemical composition of G. brasiliensis. The findings of the present study demonstrated a significant variation in the physico-chemical parameters of seawater throughout the seasons. During the summer season, the biomass abundance (258.23 ± 23.06 g m-2 FW), and the total protein content (11.12 ±1.37 % DW) were found highest while the total lipid content declined considerably. Nonetheless, the contents n-6 and n-3 PUFA increased significantly over the winter season. The concentration of micro-elements was highest during the winter season while the macro-elements found plenty during the monsoon season. The correlogram analysis revealed that biomass, total protein, total carbohydrate, chlorophyll pigments, and tissue C, H, N, and S were positively correlated with each other throughout all three seasons. Based on Pearson corelation analysis it is confirmed that among the environmental parameters, irradiance and temperature were found the most limiting factors for the growth of G. brasiliensis. Further, dissolved inorganic nitrate significantly influence the growth of G. brasiliensis negatively. Overall, the results of this study imply that G. brasiliensis should be harvested for desired metabolites during the respective seasons. Chronic prostatitis-induced excessive inflammation and oxidative stress (OS) damage substantially affect men's quality of life. However, its treatment remains a major clinical challenge. Therefore, the identification of drugs that can decrease chronic prostatitis and oxidative stress targets is urgent and essential. CXCR4 is a classic chemokine receptor that is crucially associated with the occurrence and development of inflammation. This investigation aimed to elucidate how CXCR4 affects prostatitis regression and progression. The effect of CXCR4 on chronic prostatitis was evaluated by HE staining, immunohistochemistry, immunofluorescence, PCR, and TUNEL analyses. Furthermore, CXCR4 influence on metabolism was also evaluated by monitoring body weight, body temperature, food intake, and LC/MS. Additionally, chromatin immunoprecipitation, Western blot, and double luciferase reporter gene assays were carried out to elucidate the mechanism by which CXCR4 modulates Fads2 transcription by PPARγ. Lastly, ROS, DHE, mito-tracker, and ATP were utilized to validate the α-linolenic acid's protective effect against OS in prostate epithelial cells. It was revealed that the inhibition of CXCR4 can effectively alleviate prostatitis in mice. Furthermore, downregulating CXCR4 expression can markedly reduce the inflammatory cell infiltration in mouse prostates, decrease the elevated levels of DNA damage markers,MDA and 4-HNE, and mitigate apoptosis of prostatic epithelial cells. Moreover, treatment of CXCR4 knockdown mice with a PPARγ inhibitor revealed different degrees of changes in the above phenotypes. Mechanistically, the PPARγ protein translocates to the nucleus and serves as a transcription factor to regulate Fads2 expression, thereby altering PUFA metabolism. Additionally, in vitro experiments indicated that α-linolenic acid can effectively alleviate OS damage and RWPE-1 cell apoptosis by protecting mitochondrial function and enhancing the antioxidant capacity of prostatic epithelial cells. Short-term (4 weeks) supplementation with n-3 polyunsaturated fatty acids eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) has recently been shown to improve protein metabolism in a dose dependent way in normal weight patients with Chronic Obstructive Pulmonary Disease (COPD). Furthermore, EPA/DHA supplementation was able to increase extremity lean soft tissue but not muscle function. No studies are available combining n-3 PUFAs and the leucine metabolite β-hydroxy-β-methylbutyrate (HMB) supplementation in chronic clinical conditions. Whether adding HMB to daily EPA/DHA supplementation for 10 weeks enhances muscle and brain health, daily functional performance, and quality of life of patients with COPD by further improving their protein and amino acid homeostasis remains unknown. Patients with COPD (GOLD: II-IV, n = 46) received daily for 10 weeks, according to a randomized double-blind placebo-controlled three-group design, EPA/DHA (n = 16), EPA/DHA to which HMB was added (n = 14), or placebo (n = 16). The daily dose of 2.0 g of EPA/DHA or soy + corn oil as the placebo was provided via gel capsules, and 3.0 g of Ca-HMB or maltodextrin as placebo as powders. At pre- and post-intervention, a pulse mixture of multiple amino acids was administered to measure postabsorptive net protein breakdown (netPB as primary endpoint) and whole body production (WBP) and conversion rates of the amino acids. As secondary endpoints, lean soft tissue and fat mass were assessed by dual-energy X-ray absorptiometry, upper and lower muscle function by handgrip and single leg isokinetic dynamometry, brain (cognitive, wellbeing) health by assessments, daily functional performance by measuring 6-min walk distance, 4-m gait speed, and postural balance, and quality of life by questionnaire. Plasma enrichments and concentrations were analyzed by LC-MS/MS, and systemic inflammatory profile and metabolic hormones by Luminex. HMB + EPA/DHA but not EPA/DHA supplementation increased postabsorptive netPB (p = 0.028), and WBPs of glutamine (p = 0.024), taurine (p = 0.039), and tyrosine (p = 0.036). Both EPA/DHA and HMB + EPA/DHA supplementation resulted in increased WBP of phenylalanine (p < 0.05). EPA/DHA but not HMB + EPA/DHA was able to increase WBP of arginine (p = 0.030), citrulline (p = 0.008), valine (p = 0.038), and conversion of citrulline to arginine (p = 0.009). Whole body and extremity fat mass were reduced after HMB + EPA/DHA supplementation only, whereas lean soft tissue was increased after EPA/DHA (p = 0.049) and HMB + EPA/DHA (p = 0.073). No other significant findings were observed. Reductions in several proinflammatory cytokines were observed in the HMB + EPA/DHA group including IL-2, IL-17, IL-6, IL-12P40, and TNF-β (p < 0.05). Ten weeks of supplementation with 2 g of EPA/DHA daily is sufficient to induce muscle gain in COPD but HMB is needed to induce fat loss. Whether HMB is solely responsible for the fat mass loss or has a synergistic effect with EPA/DHA remains unclear. The aim of this study was to investigate the effects of a supplement rich in ω-3 and ω-6 polyunsaturated fatty acids (PUFAs) and antioxidant vitamins on physical performance and body composition following a period of high-intensity functional training (HIFT). Nineteen healthy young adults (nine males, ten females) underwent an 8-week HIFT program (3 days·week−1) where they were randomized 1:1 into either the supplement group (SG)—n = 10, receiving a 20 mL daily dose of a dietary cocktail formula (Neuroaspis™ PLP10) containing a mixture of ω-3 and ω-6 PUFAs (12,150 mg), vitamin A (0.6 mg), vitamin E (22 mg), and γ-tocopherol (760 mg)—or the placebo group (PG)—n = 9, receiving a 20 mL daily dose of virgin olive oil. Body composition, cardiorespiratory fitness, muscle strength, and muscle endurance were assessed before and after the training period. Body mass did not change, but muscle mass increased by 1.7 ± 1.9% or 0.40 ± 0.53 kg in the SG (p = 0.021) and decreased by 1.2 ± 1.6% or 0.28 ± 0.43 kg (p = 0.097) in the PG, compared with baseline. VO2max, vertical jump, squat 1RM, bench press 1RM, and muscle endurance increased similarly in both groups. The effects of HIFT on physical performance parameters, muscle damage, and inflammation indices were not affected by the supplementation. In conclusion, HIFT combined with high doses of ω-3 and ω-6 PUFAs and antioxidant vitamins resulted in a small but significant increase in muscle mass and fat reduction compared with HIFT alone. Some health disorders, such as obesity and type 2 diabetes, are associated with a poor diet and low quality of the fat in it. The type and duration of the diet have an impact on the liver. This investigation uses the proteomic approach to identify changes in the mouse liver protein profile in adaptation to high-fat diets with different saturated fatty acid contents and linoleic acid (18:2n-6) to α-linolenic acid (18:3n-3) fatty acid ratios. Four groups of male mice were fed different diets: one standard diet and three high-fat diets were investigated. After six months on these diets, the animals were sacrificed for liver dissection. Two-dimensional electrophoresis was used to separate the complex liver protein mixture, which enabled the separation of proteins against a wide, 3–10 range of pH and molecular weights of 15–250 kDa. Protein profiles were analysed in the PDQuest Advanced 8.0.1 program. Differentially expressed spots were identified using matrix-assisted laser desorption/ionisation–time-of-flight tandem mass spectrometry and peptide mass fingerprinting. The levels of identified proteins were validated using Western blotting. Transcript levels were evaluated using a real-time quantitative PCR. The analysis of mouse liver protein profiles enabled the identification of 32 protein spots differing between nutritional groups. A diet high in polyunsaturated fatty acids modulated the levels of liver proteins involved in critical metabolic pathways, including amino acid metabolism, carbohydrate metabolism and cellular response to oxidative stress. Background and objective: Hemorrhagic stroke, characterized by acute bleeding due to cerebrovascular lesions, is associated with plasma lipids and endothelial damage. The causal relationship between genetic plasma lipid levels and hemorrhagic stroke remains unclear. This study employs a two-sample Mendelian randomization (MR) analysis to explore the causal relationship between plasma lipid profiles with different fatty acid chains and the risk of intracerebral and subarachnoid hemorrhage, the two main subtypes of hemorrhagic stroke. Methods: The datasets for exposure and outcome summary statistics were obtained from publicly available sources such as the GWAS Catalog, IEU OpenGWAS project, and FinnGen. The two-sample MR analysis was employed to initially assess the causal relationship between 179 plasma lipid species and the risk of intracerebral and subarachnoid hemorrhage in the Finnish population, leading to the identification of candidate lipids. The same methods were applied to reanalyze data from European populations and conduct a meta-analysis of the candidate lipids. The Inverse Variance Weighting (IVW) method served as the primary analysis for causal inference, with additional methods used for complementary analyses. Sensitivity analysis was conducted to clarify causal relationships and reduce biases. Results: Two analyses using Mendelian randomization were performed, followed by meta-analyses of the results. A causal relationship was established between 11 specific lipid species and the occurrence of intracerebral hemorrhage within the European population. Additionally, 5 distinct lipid species were associated with subarachnoid hemorrhage. Predominantly, lipids with linoleic acid and arachidonic acid side chains were identified. Notably, lipids containing arachidonic acid chains (C20:4) such as PC 18:1;0_20:4;0 consistently showed a decreased risk of both intracerebral hemorrhage [p < 0.001; OR(95% CI) = 0.892(0.835-0.954)] and subarachnoid hemorrhage [p = 0.002; OR(95% CI) = 0.794(0.689-0.916)]. Conversely, lipids with linoleic acid chains (C18:2) were associated with an increased risk of intracerebral hemorrhage. Conclusion: This study identifies a potential causal relationship between lipids with different fatty acid side chains and the risk of intracerebral and subarachnoid hemorrhagic stroke, improving the understanding of the mechanisms behind the onset and progression of hemorrhagic stroke. AbstractLong-chain polyunsaturated fatty acids (LC-PUFA), like 22:6n-3 (Docosahexaenoic acid; DHA) and 20:5n-3 (Eicosapentaenoic acid; EPA), are recognized for a range of important physiological roles in many aquaculture species. While the effects of EPA and DHA on a range of performance attributes and meat qualities are well recognized, an increasing awareness of their role in immune function, reproduction, bone formation and stress response is also emerging. Against this background of demand, global supplies of LC-PUFA are dominated by fish oil production from a diversified range of sources, though news sources are emerging. Among those aquaculture sectors that are the largest users of LC-PUFA resources (salmonids, shrimp, and marine fish), there are varying degrees of capacity by each to endogenously synthesize LC-PUFA and this affects the degree to which they must be obtained via the diet. Salmonids, which are the largest user of these nutrients possess some capacity to make EPA and DHA de novo, although evidence supports that salmonids perform better when provided with them preformed. Requirements by shrimp for LC-PUFA are variable, with evidence indicating that some species have capacity to desaturate and elongate fatty acids, whereas others do not. This is consistent with the observation that some species can utilize short-chain polyunsaturated fatty acids, whereas others need pre-formed LC-PUFA in their diet. A third group, marine fish, have limited ability to desaturate and elongate precursor fatty acids and therefore have a critical requirement for LC-PUFA in their diet. Evidence across multiple species indicates that demands for these fatty acids are greater when the animals are young, and this demand decreases as they age. This study aimed to investigate the effect of different culture mediums based on the Walne medium on the growth rate, chemical composition, and fatty acid (FA) profile in I. galbana and N. oculata. This experiment was done in a factorial design with two culture media (Walne and modified Walne media) and two microalgae species, including I. galbana and N. oculata. The results showed that the modified culture medium increased total and daily fresh and dried biomass production of I. galbana and N. oculata. Modified culture media increased crude fat (CF) and crude protein (CP) content and total and daily lipid production in both the studied species. However, in both the studied culture mediums, N. oculata had higher growth and production performance compared to I. galbana. Modified growth media also affects the FA profiles of the studied microalgae species. Total saturated and unsaturated FA content was not influenced by the growth medium but modified media increased poly unsaturated FA (PUFA) at the expense of mono unsaturated FA (MUFA). Omega- 3 FA content (linolenic acid, Eicosapentaenoic acid (EPA), Docosahexaenoic acid (DHA)) was increased as a result of the medium modification in both species (P<0.05). However, linoleic acid content was affected differently in I. galbana and N. oculata. The linoleic acid concentration was reduced in modified medium grown N. oculata but increased in I. galbana (P<0.05). Palmitic acid and stearic acid contents were also decreased in both of the studied species in the modified medium (P<0.05). This study develops microalgal cultivation using a modified Walne medium for higher CP, CF, EPA, DHA contents, the ratio of omega3: omega6 FA, and biomass production in N. oculata and I. galbana microalgae. This study aimed to assess the impact of Chlorella vulgaris supplementation in broilers' diet, alone or in combination with vitamin E, on meat quality parameters, nutritional value, and oxidative stability during storage time. An experiment was conducted on 180 COBB 500 broiler chickens (14 days old), assigned into six treatments, following a 2 × 3 factorial arrangement. A corn–soybean meal diet was supplemented with three levels of C. vulgaris (0% in group C1, 1% in E1, 2% in E2), two levels of vitamin E (0% in C1, 250 ppm in C2), and a combination of them (1% C. vulgaris + 250 ppm vitamin (E3), 2% C. vulgaris + 250 ppm vitamin (E4)). Dietary incorporation of C. vulgaris, including those supplemented with vitamin E, resulted in a significant increase in meat protein content. DPA and DHA levels increased by 2.01-fold and 1.60-fold in the 2% C. vulgaris + vitamin E group. The PUFA/SFA ratio was increased across all dietary treatments (p < 0.0001). HPI and h/H registered the highest values as a result of 2% C. vulgaris supplementation, being linked with a positive effect in lowering cholesterol levels. Supplementation with 2% C. vulgaris and vitamin E exhibited a 1.45-fold increase in vitamin E concentration in thigh meat compared to the control group, being the highest level registered in thigh meat in this experiment. Metmyoglobin concentrations registered lower values in the thigh meat of the experimental groups, while deoxymyoglobin increased in the same groups when compared to the control group. The inclusion of C. vulgaris (1% and 2%) in combination with vitamin E (250 mg/kg) in broiler diets exhibited the best prevention of lipid oxidation after 7 days of refrigerated storage, defined by the highest efficiency factors assessed in terms of secondary oxidation products. Background: Metabolic syndrome is a significant risk factor for both type 2 diabetes mellitus and cardiovascular disease, with a high prevalence in Asia Pacific, particularly in Indonesia. To reduce its prevalence, several studies have recommended the use of tropical nuts, which can be developed as functional foods and complementary treatment. In this context, the bioactivities of tropical nuts can largely be attributed to their rich content of monounsaturated fatty acids, polyunsaturated fatty acids, fiber, minerals, vitamins, phytosterols, and polyphenols. Objectives: This literature review aims to evaluate the potential benefits and mechanism of action of tropical nuts against metabolic syndrome. Methods: The study design was a literature review of several articles from 3 online databases, including PubMed, Google Scholar, and ScienceDirect. Discussions: The results showed that tropical nuts (peanut, sacha inchi, cashew, tropical almond, and Brazil nut) had several biologically active components, such as arginine, fiber, fatty acid, mineral, vitamin, phenolic compounds, resveratrol, and phytosterol. The test samples were reported to have the ability to modulate Nrf2, SOD, MDA, GSH, GPx, and CAT due to their antioxidant activity. In inflammation, tropical nuts had a significant effect on NF-κB, NLRP3, TNF-ɑ, IL-8, IL-1ꞵ, IL-6, and IL-10. The results also showed their ability to enhance lipid synthesis, nitric oxide production, advanced glycation end-product, prostaglandin, SIRT3, homocysteine, protein kinase C, adhesion molecules, platelet aggregation, GLP-1, PYY, AGRP, PPARɑ/ꞵ/δ, GLUT4, and insulin receptor. Conclusions: Tropical nuts had beneficial effects on metabolic syndrome due to their bioactivities, including antioxidants, anti-inflammatory, anti-obesity, antidiabetic, antihypertensive, anti-dyslipidemia, and cardioprotective.

Thus, LPS modulates specific transcriptomic changes in brain vessels of offspring rather than major structural vessel characteristics during early life. N-3 PUFA-enriched maternal diet in part prevents an imbalance in homeostatic processes, alters inflammation and ultimately mitigates changes to the complexity of surface vessel networks that result from infection. Aim: The aim of the study was to assess the association of nutritional status and dental caries in children at an orphanage in Raichur city, Karnataka. Materials and Methods: A specially designed structured questionnaire was administered to a total of 101 children of the orphanage to gather demographic details, dietary habits, sugar exposure, and oral hygiene practices. The sweet score was recorded as given by Nizel and Papas, body mass index (BMI) was assessed using the Centers for Disease Control 2000, growth charts, dental caries, and odontogenic infections were recorded using decayed missing filled teeth in deciduous dentition (dmft)/Decayed Missing Filled Teeth in permanent dentition (DMFT) and pufa/PUFA (Pulpal involvement, Ulceration, Fistula, and Abscess) index (Monse et al., 2010), respectively. Results: A total of 101 participants were enrolled in the study including children aged 6–15 years old with a mean age of 10.67 ± 2.74 years. A statistically significant relation was found between the duration of stay in an orphanage and the prevalence of dental caries (P < 0.05). A statistically significant positive correlation was found between the duration of stay in the orphanage and BMI (0.382) dmft/DMFT (0.203). In addition, a statistically significant positive correlation was also observed between dmft/DMFT and pufa/PUFA. Conclusion: The study showed caries to be primarily associated with the sugar exposure and dietary habits, whereas untreated dental decay was the main cause of odontogenic infections. The untreated caries PUFA ratio was 13.98% with only one filled component, indicating the unmet needs for decayed teeth and lack of awareness and access to preventive and curative dental care facilities by the children of the orphanage.

Abstract Background We aimed to explore the association and potential causality between polyunsaturated fatty acids concentrations and the risk of periodontal disease. Materials and methods Data were collected from the 2011–2014 National Health and Nutrition Examination Survey (NHANES). Weighted logistic regression analysis and restricted cubic spline (RCS) analysis were used to analyse the associations of the concentrations of omega-3 and omega-6 fatty acids and the omega-6/omega-3 fatty acids ratio with the risk of periodontitis. E-value and propensity score matching (PSM) analyses were used for sensitivity analyses. In addition, two-sample Mendelian randomisation (MR) analyses were performed to assess the potential causal impact of the concentrations of those fatty acids on periodontitis risk. Results A total of 2462 participants from the NHANES were included. Logistic regression analysis revealed that high omega-3 fatty acids levels were negatively associated with the risk of developing periodontitis (P 0.05). Conclusion Although omega-3 fatty acids and the omega-6/omega-3 fatty acids ratio were associated with the risk of periodontitis in cross-sectional studies, the MR results did not support a causal relationship between them. Therefore, there is no indication that an increase in the omega-3 fatty acids concentration or a decrease in the omega-6/omega-3 fatty acids ratio may be beneficial for preventing periodontitis.

Dietary intake of omega-3 polyunsaturated fatty acids (eicosapentaenoic acid, EPA) exerts antiarrhythmic effects, although the mechanisms are poorly understood. Here, we investigated the possible beneficial actions of EPA on saturated fatty acid-induced changes in the L-type Ca2+ channel in cardiomyocytes. Cardiomyocytes were cultured with an oleic acid/palmitic acid mixture (OAPA) in the presence or absence of EPA. Beating rate reduction in cardiomyocytes caused by OAPA were reversed by EPA. EPA also retrieved a reduction in Cav1.2 L-type Ca2+ current, mRNA, and protein caused by OAPA. Immunocytochemical analysis revealed a distinct downregulation of the Cav1.2 channel caused by OAPA with a concomitant decrease in the phosphorylated component of a transcription factor adenosine-3′,5′-cyclic monophosphate (cAMP) response element binding protein (CREB) in the nucleus, which were rescued by EPA. A free fatty acid receptor 4 (FFAR4) agonist TUG-891 reversed expression of Cav1.2 and CREB mRNA caused by OAPA, whereas an FFAR4 antagonist AH-7614 abolished the effects of EPA. Excessive reactive oxygen species (ROS) accumulation caused by OAPA decreased Cav1.2 and CREB mRNA expressions, which was reversed by an ROS scavenger. Our data suggest that EPA rescues cellular Cav1.2-Ca2+ channel decline caused by OAPA lipotoxicity and oxidative stresses via both free fatty acid receptor 4-dependent and -independent pathways. Athletes are characterized by special nutritional needs. Meeting their dietary needs associated with intensive exercise is a prerequisite for effective training and success in sports competitions. Hence, it is important to supply key performance nutrients, such as macronutrients, antioxidants, calcium, vitamin D, or iron, in adequate quantities. The increased need for these nutrients makes it necessary to look for food products on the market that more fully cover these needs. Such products may include organic foods. According to research, they have unique properties and are richer in selected nutrients, such as antioxidants. Hence, the aim of this review was to analyze the available literature as to whether organic foods have the potential to more fully cover the increased nutritional requirements of athletes for selected nutrients compared to conventionally produced foods. A narrative review of current literature was carried out. As the analysis showed, organic foods are characterized by a higher content of antioxidant bioactive compounds, a higher content of n-3 fatty acids, a better n:3/n:6 ratio, and a more optimal amino acid composition, which may result in an appropriate dietary ration design for athletes. Since time immemorial, molluscs have been a popular source of food in North East India. Given the historical importance of snail consumption in the region, the current investigation was carried out to assess the nutritional properties of mollusc meat/tissue and shell. Three species of freshwater snails (Cipangopaludina sp. Brotia costula, Filopaludina bengalensis) and the bivalve Lamellidens sp. were identified from five states of North East India. Proximate and elemental analyses of mollusc meat revealed high amounts of nutrients (more crude protein (range 26.75–22.83%) and low fat (3.82–2.70%)) and minerals (especially calcium). Breast cancer is the most common tumor in women. Chemotherapy is the gold standard for cancer treatment; however, severe side effects and tumor resistance are the major obstacles to chemotherapy success. Numerous dietary components and phytochemicals have been found to inhibit the molecular and signaling pathways associated with different stages of breast cancer development. In particular, this review is focused on the antitumor effects of PUFAs, dietary enzymes, and glucosinolates against breast cancer. The major databases were consulted to search in vitro and preclinical studies; only those with solid scientific evidence and reporting protective effects on breast cancer treatment were included. A consistent number of studies highlighted that dietary components and phytochemicals can have remarkable therapeutic effects as single agents or in combination with other anticancer agents, administered at different concentrations and via different routes of administration. These provide a natural strategy for chemoprevention, reduce the risk of breast cancer recurrence, impair cell proliferation and viability, and induce apoptosis. Some of these bioactive compounds of dietary origin, however, show poor solubility and low bioavailability; hence, encapsulation in nanoformulations are promising tools able to increase clinical efficiency. Psoriasis is a chronic systemic disease with a multifaceted pathomechanism and immunological basis, with the presence of inflammatory skin lesions and joint ailments. Diseases accompanying psoriasis include metabolic and cardiovascular disorders. It has been suggested that inflammation is involved in the development of each of these conditions. The main objective of this study was to analyse the fatty acid profile, including polyunsaturated fatty acids, in the erythrocyte membranes of patients suffering from psoriasis. A total of 58 adult patients of the Department of Skin and Venereal Diseases of the Pomeranian Medical University in Szczecin, suffering from psoriasis, were qualified for this study. The patients had undergone an interview and physical examination, during which the severity of psoriasis was assessed. All patients had their weight and height measured to assess their body mass index (BMI). After 3 months of treatment, biochemical parameters (ALT, AST, total cholesterol) and inflammatory markers (CRP) in the blood were assessed. In addition, the isolation of fatty acids (PUFAs, SFAs, MUFAs) from erythrocyte membranes and the qualitative and quantitative analysis of their profile using a gas chromatograph were carried out. In patients with severe psoriasis requiring systemic treatment, an altered profile of fatty acids in erythrocyte membranes was found, including a significantly lower concentration of polyunsaturated fatty acids (omega-3), which have an anti-inflammatory effect; a significantly higher concentration of saturated fatty acids; and a decreased concentration of oleic acid (omega-9), compared to the results obtained in patients with less severe psoriasis receiving topical treatment. In patients with psoriasis and BMI ≥ 25, significantly higher concentrations of AST and ALT in the blood and significantly higher concentrations of pro-inflammatory arachidonic acid in erythrocyte membranes were found. Heart failure (HF) with preserved ejection fraction (HFpEF) is the most common cause of pulmonary hypertension (PH) worldwide and is strongly associated with adverse clinical outcomes. The American Heart Association recently highlighted a call to action regarding the distinct lack of evidence-based treatments for PH due to poorly understood pathophysiology of PH attributable to HFpEF (PH-HFpEF). Prior studies have described cardiophysiological mechanisms to explain the development of isolated postcapillary PH (ipc-PH); however, the consequent increase in pulmonary vascular (PV) resistance (PVR) may lead to the less understood and more fatal combined pre- and postcapillary PH (cpc-PH). Metabolic disease and inflammatory dysregulation have been suggested to predispose PH, yet the molecular mechanisms are unknown. Although PH-HFpEF has been studied to partly share vasoactive neurohormonal mediators with primary pulmonary arterial hypertension (PAH), clinical trials that have targeted these pathways have been unsuccessful. The increased mortality of patients with PH-HFpEF necessitates further study into viable mechanistic targets involved in disease progression. We aim to summarize the current pathophysiological and clinical understanding of PH-HFpEF, highlight the role of known molecular mechanisms in the progression of PV disease, and introduce a novel concept that lipid metabolism may be attenuating and propagating PH-HFpEF. Leukocyte telomere length (LTL) is a biomarker of aging that may be influenced by dietary factors. Omega-3 fatty acids (n-3 FA) have been suggested to affect LTL. However, research on this effect has been inconclusive. The aim of the study was to test the hypothesis about the positive effect of n-3 FA on LTL. Fat-1 transgenic mice, which can convert omega-6 fatty acids (n-6 FA) to n-3 FA and have elevated levels of endogenous n-3 FA in their tissues, were used to study the effects of n-3 FA on LTL at different ages. Blood samples from 10-month-old wild-type (WT) mice (n = 10) and fat-1 mice (n = 10) and 3-month-old WT mice (n = 5) and fat-1 mice (n = 5) were used to measure relative and absolute LTL. The levels of proteins critical for telomere maintenance were examined by Western blot analysis. Fat-1 transgenic mice had longer leukocyte telomeres than their WT siblings, suggesting a slower rate of age-related telomere shortening in fat-1 mice. In animals aged 10 months, the LTL was significantly longer in fat-1 than in WT mice (mean ± SEM; relative LTL: WT = 1.00 ± 0.09 vs. fat-1: 1.25 ± 0.05, P = 0.031; absolute LTL: WT = 64.41 ± 6.50 vs. fat-1: 78.53 ± 3.86, P = 0.048). The difference in LTL observed in three-month-old mice was insignificant, however the mean LTL was still longer in fat-1 mice than in the WT mice. Fat-1 mice also had abundant levels of two shelterin proteins: TRF1 (27%, P = 0.028) and TRF2 (47%, P = 0.040) (telomeric repeat binding factor 1 and 2) compared to WT animals. This study, for the first time in a unique animal model free of dietary confounders, has demonstrated that increased levels of n-3 FA in tissues can reduce telomere attrition. The data presented indicate the possibility of using omega-3 fatty acids to reduce accelerated telomere attrition and, consequently, counteract premature aging and reduce the risk of age-related diseases. Background. Lipoprotein-associated phospholipase A2 (Lp-PLA2) may play an important role in the development of atherosclerotic cardiovascular disease (ASCVD). Increased plasma levels of Lp-PLA2 may predict future cardiovascular (CV) events in type 2 diabetes (T2D). The potential beneficial effects of polyunsaturated fatty acids (PUFA) on ASCVD have been widely investigated. However, the impact of different PUFA concentrations on Lp-PLA2 remains uncertain. Objectives. We sought to determine the intergender differences in a population of patients with both T2D and ASCVD regarding Lp-PLA2 mass and the association between Lp-PLA2 mass and plasma levels of PUFA. Materials and methods: In this cross-sectional study, we measured the Lp-PLA2 mass, PUFA concentrations and inflammatory markers in 74 patients (49 males and 25 females) with T2D and ASCVD. Results: In this very high-risk population, males had, on average, 33.6% higher levels of Lp-PLA2 than females. The Lp-PLA2 mass was positively associated with interleukin 6 (IL-6) (r = 0.27, p = 0.019), creatinine (r = 0.29, p = 0.03) and triglyceride levels (r = 0.41, p = 0.002). Additionally, male gender and higher levels of triglycerides, leptin, oxidized low-density lipoprotein (oxLDL), and intercellular adhesion molecule 1 (ICAM-1) were independent predictors for an increased Lp-PLA2. Moreover, arachidonic acid (AA) negatively correlated with Lp-PLA2 (r = -0.26, p = 0.024), which was especially apparent in the female subgroup. Conclusions. In the population of patients with ASCVD and T2D, males present with higher plasma levels of Lp-PLA2 than females. Additionally, higher plasma levels of AA were associated with lower Lp-PLA2 levels. Coronary artery disease is one of the significant causes of death and is still a health problem for developed and developing countries. Increased cholesterol in the blood is caused by heredity and high-fat consumption. The effect of dietary fat on artery disease is related to the impact of fatty acid components and cholesterol on blood cholesterol. This study aims to determine the relationship between consumption of Saturated Fatty Acids (SFA), Monounsaturated Fatty Acids (MUFA), and Polyunsaturated Fatty Acids (PUFA) with total cholesterol levels in patients with coronary artery disease. The research design used was descriptive-analytic in clinical nutrition with a cross-sectional approach. The population in this study were 405 patients with coronary artery disease at the artery clinic of RSUD Dr. M. Yunus Bengkulu. The sample was 32 patients collected using a purposive sampling technique. Using the Chi-Square test, data analysis was used to determine the relationship between the consumption of SFA, MUFA, and PUFA with total cholesterol levels in patients with coronary artery disease. The study’s results found a significant relationship between SFA consumption and coronary artery disease. Still, conversely, there was no significant relationship between MUFA and PUFA consumption with total cholesterol level in coronary artery disease patients, namely that SFA consumption was inadequate (p-value = 0.043, OR = 0.407), inadequate MUFA consumption (p-value = 0.710), and inadequate of PUFA consumption (p-value= 0.465). The conclusion is that saturated fatty acids are related to total cholesterol in coronary artery disease while conversely to monounsaturated fatty acids and polyunsaturated fatty acids. The microalgae Chlorella vulgaris and Tetraselmis chui are valued for their nutrient-rich content, including lipids and polyunsaturated fatty acids (PUFA). However, little is known about how storage and processing affect their lipid quality. This study aimed to assess the impact of domestic storage and cooking practices in dried biomass of C. vulgaris and T. chui. Four conditions were tested: control (newly opened package), light (storage at room temperature and daily light regimen for three weeks), frozen (storage in the freezer at −20 °C for three weeks), and heated (three cycles of 90 min at 100 °C). Lipid extracts were analyzed by GC-MS and LC-MS, and antioxidant activity through DPPH and ABTS radical scavenging assays. Tested storage conditions promoted a decrease in fatty acid content and in diacyl/lyso lipid species ratios of phospholipid (PC/LPC, PE/LPE) and betaine lipids (DGTS/MGTS). Lipid extracts from light treatment showed the lowest antioxidant activity in C. vulgaris (ABTS, IC40: 104.9; DPPH, IC20: 187.9 ± 15.0), while heat affected the antioxidant activity of T. chui (ABTS, IC40: 88.5 ± 2.8; DPPH, IC20 209.4 ± 10.9). These findings underscore the impact of managing storage and processing conditions to optimize the nutritional and functional benefits of C. vulgaris and T. chui in food and feed applications. Introduction: In aquatic animals, colors not only reflect their sensory qualities but also affect their nutritional components and market price. Noble scallop Chlamys nobilis is an importantly economical bivalve cultured in the south sea of China and renowned for its various shell colors. Methods: In this study, the nutrient compositions were compared among golden, yellow, and brown shell colour noble scallops originating from the same population for breeding new varieties. Results: Among three colour scallops, there is no significant difference in the content of moisture, ash, and total protein content (TPC) but significant differences in total carotenoid content (TCC), total lipid content (TLC), and amino acids (AAs) (P < 0.05), with the TCC of the brown scallop being about 2 times lower. The yellow scallop contained the same nutrients as TCC, TLC, ΣPUFAs (including EPA and DHA), and n-3/n-6 as the golden scallop, and both were significantly higher than the brown scallop (P < 0.05), but they had significantly lower ΣSFAs than the latter (P < 0.05). The correlation between the nutrient indices and the shell color of scallops was 67.5%. There is a significantly positive correlation between TCC and ΣPUFAs (P < 0.05), while a significantly negative correlation exists between TCC and ΣSFAs (P < 0.05). More importantly, the yellow scallop had significantly higher EAAs and FAAs than the golden scallop (P < 0.05). Discussion: This study showed that the yellow noble scallop not only enriched in carotenoids and PUFAs but also enriched in amino acids. In this brief, non-exhaustive essay, we review major adaptive changes in reproductive and developmental traits of decapod crustaceans, which have passed through evolutionary transitions from ancestral life in coastal marine environments to brackish, limnic, polar, or deep-sea habitats. Two major approaches can be recognised in studies of life-history evolution, focussing on: (1) developmental changes in adaptive physiological or biochemical traits of successive ontogenetic stages, comparing, for instance, osmoregulatory capabilities in larvae, juveniles, and adults ("ontogeny of adaptations" perspective); (2) adaptive changes in the modes of reproduction and development, e.g., enhanced maternal energy investment in egg production, abbreviated, non-feeding larval development ("adaptations in ontogeny"). Using both scientific approaches, Christoph D. Schubart has significantly contributed to our understanding of evolutionary transitions in crustaceans. In our essay, we pay particular attention to the significance of nutritional selection factors in relation to presumably adaptive developmental, physiological, and biochemical traits in different life-history stages. In this context, we highlight the key roles of lipids and carotenoid pigments, in particular of essential polyunsaturated fatty acids (PUFAs) and astaxanthin. While PUFAs and other lipid compounds have received wide scientific attention, we propose that future studies should further analyse also the potential role of astaxanthin and other essential carotenoids. Background: LC-MS is an ever-increasingly used methodology for clinical applications. Due to the superior selectivity and sensitivity, in certain situations, it can offer an advantage or be the only option for diagnostics and biomonitoring applications. Methods: A high selectivity sensitive LC-MS/MS method was developed for direct quantification of free plasma polyunsaturated fatty acids as well as conjugated membrane polyunsaturated fatty acids, using isocratic reverse phase elution. A quick and simple sample purification method was used in order to ensure high-throughput analysis of biological samples. The method was validated with regard to selectivity, sensitivity, linearity, accuracy, precision, carryover, and recovery, as well as other relevant parameters. Results and Conclusions: The method was developed and validated with respect to all relevant parameters and was successfully used in a number of clinical diagnostics and biomonitoring applications. The simple sample purification process allowed for an easy learning curve for analysts and other users, while ensuring a low chance of systematic or random errors and thus reliable results usable in a clinical setting. In this study, fatty acid compounds in the muscle tissues of the economically important Mesopotamian catfish (Silurus triostegus, Heckel, 1843) and Shabut (Arabibarbus grypus, (Heckel, 1843)), which live in the Ilısu Dam Lake on the Tigris River, were investigated. For fatty acid analysis, muscle tissues were taken from the samples obtained from local fishermen, placed in tubes and lipids were obtained by applying chloroform/methanol method. The methylation steps of fatty acids in the samples were performed in accordance with TS EN ISO 12966:2 (TS EN ISO 12966-2., 2017) method. The total fatty acid content of saturated fatty acids (SFA) value was detected as 27,97 ± 0,26 in A. grypus, 32.46 ± 0.37 in S. triostegus, The value of Monounsaturated Fatty Acids (MUFA) varied between 42.63 ± 0.6% in A. grypus and 41.5 ± 0.51% in S. triostegus. Total polyunsaturated polyunsaturated acids (PUFA) ranged between 21.83 ± 1.49 for A. grypus and 25.82 ± 1.94 for S. triostegus. MUFA values in all samples were higher than SFA and PUFA values for both fish species. Docosa Hexaenoic Acid (DHA) and Eicosa Pentaenoic Acid (EPA) are commonly consumed omega-3 types. A. grypus and S. triostegus are within the recommended limits for EPA (C20:5n3) and DHA (C22:6n3). For A. grypus, ω3/ω6 values were (1,06); for S. triostegus ω3/ω6 values were (1.38). When the ω3/ω6 values of both fish species were analysed for human health, it was determined that the ω3/ω6 values of omega fatty acids A. grypus and S. triostegus were above 1. Numerous publications have demonstrated that polymorphisms in FADS1 and FADS2 gene cluster influence the levels of long chain polyunsaturated fatty acids (LC PUFAs) synthesized in the body from essential precursors. They can be consumed through the diet as well. These LC PUFAs perform important plastic and regulatory functions in the body. The associations between the polymorphisms of FADS genes and the fatty acid (FA) composition of the body with some inflammatory and chronic diseases were shown. To date, very few studies in the Russian Federation researched the distribution of polymorphic variants in FADS1 and FADS2 genes and no studies examined the effects of these polymorphisms on the level of FAs in tissues.So, the purpose of this paper was to study the frequency of genotypes and alleles of the rs174544 polymorphism of the FADS1 gene and the rs174583 polymorphism of the FADS2 gene and their influence on the FA composition of erythrocyte membranes in residents of the Central region of the Russian Federation (RF). Additionally, the effect of ω-3 PUFAs fortified food products on this composition was examined. Methods: The study included 70 volunteers aged 23–69 years from Moscow region who provided informed consent. They were genotyped for rs174544 polymorphism of FADS1 gene and rs174583 polymorphism of FADS2 gene by real-time PCR. The participants of the study consumed fruit or vegetable smoothies enriched with either vegetable (chia seeds flour) or animal (fish oil flour) sources of ω-3 PUFAs for 1 month. The fatty acid composition of erythrocyte membranes in some of the volunteers was analysed using gas-chromatography before and after fortification of their diet with ω-3 PUFAs. Statistical analysis was performed using SPSS 23. Results: The frequency of genotypes and alleles of polymorphisms studied were similar to those published for the European region. For rs174544, the genotype frequencies were CC (61%), CA (29%), and AA (10%), frequency of allele A was 24%. For rs174583, the genotype frequencies were CC (51%), CT (39%), and TT (10%), frequency of allele T was 29%. Carriers of minor alleles of both polymorphisms studied had significantly higher levels of ω-6 and ω-3 LC PUFAs in erythrocyte membranes compared to those carrying other genotypes. This differs from the results of other studies where carriers of minor alleles as were shown to be slow converters of precursors to LC PUFAs. The reason of the discrepancy was most likely associated with the obesity and dietary preferences of our study participants who carry the minor alleles. Over the period of one-month consumption of food sources providing an additional intake of either 2 g/day ω-3 PUFAs from fish oil or 6.5 g/day of ω-3 PUFAs from chia seed flour markedly reduced the incorporation of ω-6 PUFAs into erythrocyte membranes in carriers of minor alleles. Additionally, there was an increase in the content of saturated palmitic acid in the membranes that noticeably changed the fatty acid composition of erythrocyte membranes. Conclusion: Our results confirm the observation that blood fatty acid composition related to polymorphisms of FADS gene is highly influenced by BMI. The incorporation of ω-6 PUFAs into erythrocyte membranes decreases when the diet is fortified with ω-3 PUFAs. This decrease can help reduce the formation of pro-inflammatory eicosanoids and the risk of various inflammatory and chronic diseases, especially in carriers of minor alleles of the genetic polymorphisms studied.

Summary: Polyunsaturated fatty acids (PUFAs), which are abundant in fish oil, easily oxidise. Therefore, stabilisation is necessary, preferably through encapsulation and the addition of antioxidants. This research looked on multiple emulsification for PUFA‐rich fish oil by utilising a wall material (whey protein and chitosan) and an antioxidant (betalain) combination. Spray‐dried, microparticles filled with PUFA were successfully created (80% yield) and characterised. It was determined that the emulsion droplet size was 0.917 μm. Good oxidative stability was observed after 4 weeks of storage, as evidenced by a moderate (4%) drop in EPA and DHA content and a low peroxide value (PV) of 4.8 ± 0.23 meq/kg oil in microparticles. Microencapsulation of PUFA‐rich fish oil was successfully accomplished using multiple emulsion technology, as evidenced by encapsulation efficiency values of 87.73 ± 0.21% and loading efficiency values of 67 ± 0.11%. These results suggest that structurally stabilised healthful lipids could be added to foods using these innovative emulsions, which provide greater stability and versatility for a wide variety of uses. Crypthecodinium cohnii is a marine microalga that can accumulate high amounts of polyunsaturated fatty acids (PUFAs) and thus replace conventional routes of fish oil production. They are associated with the destruction of marine resources and multiple downstream/purification complications. The major drawbacks of using C. cohnii for industrial-scale production are associated with low PUFA productivity. One of the means of increasing the PUFA synthesis rate is to maintain the medium component concentrations at optimal values throughout cultivation, thus increasing PUFA production efficiency, which can result in the successful transfer of the process to pilot and/or industrial scale. The goal of the present research was to develop techniques for increasing the efficiency of PUFA production via C. cohnii cultivation. Multiple experiments were carried out to test and fine-tune the cultivation medium composition and oxygen transfer factors. The biomass yields from individual components, yeast extract, sea salts, and glucose amounted to 5.5, 0.65, and 0.61 g·g−1, respectively. C. cohnii cell susceptibility to mechanical damage was experimentally evaluated. Power inputs of <276.5 W/m3 did not seem to promote cell destruction when Pitched-blade impellers were used. The obtained cultivation conditions were shown to be efficient in terms of increasing the biomass productivity and the omega-3 fatty acid content in C. cohnii. By using the applied methods, the maximal biomass productivity reached 8.0 g·L−1·day−1, while the highest obtained biomass concentration reached 110 g·L−1. Dietary supplements are used for potential health benefits and are mainly intended to provide those nutrients that may be insufficiently consumed through regular dietary intake. They are mostly made from natural sources and are readily available in the form of capsules, tablets, or liquid form. Nowadays, omega-3 fatty acids (n-3 FAs) supplements are in high demand and have gained noteworthy popularity as the human body cannot produce them, and need to be administrated externally. They are polyunsaturated fatty acids (PUFA) characterized by at least one cis double bond at the third and fourth omega-end carbons, with primary examples being α-linolenic acid (ALA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA) and are classified as functional foods. The natural sources of these supplements include aquatic organisms like freshwater and marine fish, microalgae, seaweeds; nuts and seeds plant oils, and fortified foods. n-3 FAs, being biologically active molecules, are highly susceptible to oxidation due to double bonds in their long chains, leading to degradation over time, during storage. Various encapsulation systems, such as gels, emulsions, and powders, have been developed to effectively encapsulate PUFAs to enhance their chemical stability, dispersibility, and bioavailability. These quality control measures are essential to guarantee the effectiveness and safety of products containing n-3 FAs. This paper explores various sources of omega-3 fatty acid supplements and summarizes the multiple manufacturing techniques used for the production of these products. Additionally, the article tried to correlate quality aspects with the regulations for such products in an attempt to prevent exposure of consumers to harmful ingredients contained within these products. Several meta-analyses investigating the efficacy of n -3 PUFA in alleviating depression symptoms have reported conflicting findings. In the present study, we aimed to perform an umbrella meta-analysis to provide a definite conclusion. A comprehensive systematic search of PubMed, Scopus, Embase, Web of Science and Cochrane Central Library was performed up to June 2021. Meta-analysis studies evaluating the effects of n -3 PUFA on depression symptoms were included. The quality of the included meta-analyses was assessed using AMSTAR questionnaire. Out of 101 studies, twenty-two studies with twenty-six effect sizes (ES) were eligible for inclusion. Sixteen ES showed significant improving effect of n -3 supplementation on depression symptoms among which eleven ES had small ES. The other studies observed no significant effect. Available evidence suggests that n -3 PUFA (EPA, DHA) supplementation could be considered as an effective add-on therapeutic approach in relieving depression symptoms. [ABSTRACT FROM AUTHOR]

In this study, the fatty acid composition of smoked salmon fillets sold in grocery stores in Türkiye was investigated. Gas chromatography (GC) was used to determine fatty acid composition from extracted lipid. The main saturated fatty acids observed in smoked salmon samples were myristic acid (C14:0), palmitic acid (C16:0) and stearic acid (C18:0); monounsaturated fatty acids were palmitoleic acid (C16:1), oleic acid (C18:1n9), and vaccenic acid (C18:1n7); polyunsaturated fatty acids (PUFA) were linoleic acid (C18:2n6), eicosapentaenoic acid (EPA, C20:5n3) and decosahexaenoic acid (DHA, C22:6n3). Among the monounsaturated fatty acids, oleic acid was found to be the fatty acid with the highest value. Oleic acid amounts were found to vary between 27.22% and 35.52%. PUFA values in smoked salmon fillet groups were determined as 27.77%, 27.49%, 32.94% and 30.62%. The highest EPA value was determined in F1 group with 4.29% and the lowest value was determined in F2 group with 2.07%. DHA amounts were between 11.74% and 6.22%. The ratio of Σn6/Σn3 was between 0.80 (F4 group) and 1.16 (F2 group). As a result, it was concluded that the smoked fish fillets examined had high nutritional quality in terms of fatty acids. Among the groups, especially F3 and F4 groups were found to have rich content in terms of PUFA and Σn3 values. [ABSTRACT FROM AUTHOR]

The present study evaluated the effects of heavy metals, viz., lead, mercury, and cadmium, on growth, chlorophyll a, b, c, carotenoids, and PUFA content of marine microalgae Chlorella sp. and Cylindrotheca fusiformis. At 96-h exposure, the IC50 values for Hg2+, Pb2+, and Cd2+ were 0.85 mg/L, 2.4 mg/L, and 5.3 mg/L respectively, in Chlorella sp. In C. fusiformis, IC50 values for Hg2+, Pb2+, and Cd2+ were 0.5 mg/L, 1.2 mg/L, and 3 mg/L respectively. The pigment contents of both microalgae were significantly affected upon heavy metal exposure. In Chlorella sp. and C. fusiformis, the exposed concentrations of Hg2+ averagely decreased the PUFA content by 76.34% and 78.68%, respectively. Similarly, Pb2+-exposed concentrations resulted in 54.50% and 82.64% average reductions in PUFA content of Chlorella sp. and C. fusiformis, respectively. Cd2+-exposed concentrations showed 32.58% and 40.54% average reduction in PUFA content of Chlorella sp. and C. fusiformis, respectively. Among the environmental stress conditions, the dark treatment has increased total PUFA content by 6.63% in Chlorella sp. and 3.92% in C. fusiformis. It was observed that the 50% nitrogen starvation (two-stage) significantly improved the PUFA production from 26.47 ± 6.55% to 40.92 ± 10.74% in Chlorella sp. and from 11.23 ± 5.01 to 32.8 ± 14.17% in C. fusiformis. The toxicity for both microalgae was followed in the order Hg2+ > Pb2+ > Cd2+. Natural-based chemicals from microalgae such as lipids and pigments are the interests in industries and the bioeconomy. Cold-adapted Ankistrodesmus sp. OR119838, an isolated strain from Cheshmeh–Sabz Lake in northeastern Iran, was cultivated using a two-stage culture strategy under different environmental conditions. With doubling the nitrate concentration at the vegetative stage (170 mg/L) and increasing the light intensity (180 µmol photons/m2/s) the highest specific growth rate (0.61 ± 0.02 per day) and biomass productivity (121.1 ± 7.2 mg/L/day) were observed at 25 °C. In the optimal growth condition Chl a and Chl b contents of Ankistrodesmus sp. OR119838 reached the highest amount (11.07 ± 0.14 and 11.23 ± 0.29 µg/mL, respectively) at 25 °C. While carotenoid content correlated negatively with optimum biomass productivity (− 0.708) and had the best value (12.23 ± 0.29 µg/mL) in nitrogen deficiency (42 mg/L) and intense light conditions (180 µmol photons/m2/s) at 15 °C. Lipid content was increased with declined nitrate concentration (42 mg/L), high light intensity, and 180 µmol photons/m2/s at 25 °C. The highest percentage of polyunsaturated fatty acids (71.94%) and α-linolenic acid (57.73 ± 6.63%) was observed in conditions with 170 mg/L nitrate concentration and low light intensity (40 µmol photons/m2/ s) at the low temperature (15 °C). While saturated fatty acids content (43.27%) and palmitic acid reached the highest amount under 40 µmol photons/m2/s, 42 mg/L nitrate at 25 °C (35.02 ± 5.33%). Biomass productivity of Ankistrodesmus sp. OR119838, as a cold-adapted strain, decreased by only 8.2% with a 10-degree decline in temperature. Eutrophication, climate-induced warming, and salinity fluctuations are altering the fatty acid profiles and the availability of essential polyunsaturated fatty acids (PUFAs) in marine zooplankton communities. Limnocalanus macrurus Sars G.O., 1863 is a large calanoid copepod inhabiting the low-salinity areas in the Baltic Sea, where it is a major source of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) to commercially important fish. L. macrurus is sensitive to warming, eutrophication and hypoxia. As an opportunistic feeder, it is capable of dietary shifts, which affects its fatty acid profiles. Although much studied in boreal lakes, there are only a few studies on the fatty acid profiles of the Baltic Sea populations. This study aimed to compare the fatty acid profiles of L. macrurus in three basins of the Baltic Sea, in relation to the community fatty acids and environmental variables. We collected samples of L. macrurus and filtered plankton community for gas chromatographic fatty acid analyses in August 2021 on R/V Aranda. The nutritional quality of L. macrurus to consumers was lower in the Gulf of Finland (GoF) compared to the Gulf of Bothnia, indicated by the low levels of DHA and EPA, as well as the low n-3/n-6 ratio of PUFAs. The lower ratio of 18:1n-7 to 18:1n-9 implied higher degree of omnivory in GoF. In contrast, a diatom marker 16:1n-7 had high proportion in the Bothnian Bay. High temperatures in GoF may have restricted feeding in the upper water column, possibly forcing a shift towards cyanobacteria or seston-based diet, as interpreted from a high proportion of 18:2n-6 and 18:3n-3. We conclude that the ability of L. macrurus to utilize multiple food sources increases its resilience to environmental change, while the consequences on the nutritional quality may have further cascading effects on the food webs. Epilepsy often occurs with other neurological disorders, such as autism, affective disorders, and cognitive impairment. Research indicates that many neurological disorders share a common pathophysiology of dysfunctional energy metabolism, neuroinflammation, oxidative stress, and gut dysbiosis. The past decade has witnessed a growing interest in the use of metabolic therapies for these disorders with or without the context of epilepsy. Over one hundred years ago, the high-fat, low-carbohydrate ketogenic diet (KD) was formulated as a treatment for epilepsy. For those who cannot tolerate the KD, other diets have been developed to provide similar seizure control, presumably through similar mechanisms. These include, but are not limited to, the medium-chain triglyceride diet, low glycemic index diet, and calorie restriction. In addition, dietary supplementation with ketone bodies, polyunsaturated fatty acids, or triheptanoin may also be beneficial. The proposed mechanisms through which these diets and supplements work to reduce neuronal hyperexcitability involve normalization of aberrant energy metabolism, dampening of inflammation, promotion of endogenous antioxidants, and reduction of gut dysbiosis. This raises the possibility that these dietary and metabolic therapies may not only exert anti-seizure effects, but also reduce comorbid disorders in people with epilepsy. In temperate lakes, eutrophication and warm temperatures can promote cyanobacteria blooms that reduce water quality and impair food-chain support. Although parasitic chytrids of phytoplankton might compete with zooplankton, they also indirectly support zooplankton populations through the “mycoloop”, which helps move energy and essential dietary molecules from inedible phytoplankton to zooplankton. Here, we consider how the mycoloop might fit into the biodiversity–ecosystem functioning (BEF) framework. BEF considers how more diverse communities can benefit ecosystem functions like zooplankton production. Chytrids are themselves part of pelagic food webs and they directly contribute to zooplankton diets through spore production and by increasing host edibility. The additional way that chytrids might support BEF is if they engage in “kill-the-winner” dynamics. In contrast to grazers, which result in “eat-the-edible” dynamics, kill-the-winner dynamics can occur for host-specific infectious diseases that control the abundance of dominant (in this case inedible) hosts and thus limit the competitive exclusion of poorer (in this case edible) competitors. Thus, if phytoplankton diversity provides functions, and chytrids support algal diversity, chytrids could indirectly favour edible phytoplankton. All three mechanisms are linked to diversity and therefore provide some “insurance” for zooplankton production against the impacts of eutrophication and warming. Aims To investigate fatty acid, including polyunsaturated fatty acids (PUFA), and cerebroside production of a large diversity of fungi from the Ascomycota, Basidiomycota , and Mucoromycota phyla. Methods and results Seventy-nine fungal strains were grown in Kavadia medium using a microcultivation system, i.e. Duetz microtiter plates. Following cultivation, fatty acid and cerebroside contents were analyzed by gas chromatography-flame ionization detection (GC-FID) and high performance thin-layer chromatography (HPTLC), respectively. Mucoromycota fungi appeared as the most promising candidates for omega-6 PUFA production. The best omega-6 producer, including γ-linolenic acid (GLA, 18:3n-6), was Mucor fragilis UBOCC-A109196 with a concentration of 647 mg L−1 total omega-6 PUFA (representing 35% of total fatty acids) and 225 mg L−1 GLA (representing 12% of total fatty acids). Arachidonic acid concentration (20:4n-6) was the highest in Mortierella alpina UBOCC-A-112046, reaching 255 mg L−1 and 18.56% of total fatty acids. Interestingly, several fungal strains were shown to produce omega-7 monounsaturated fatty acids. Indeed, Torulaspora delbrueckii strains accumulated palmitoleic acid (16:1n-7) up to 20% of total fatty acids, reaching 114 mg L−1 in T. delbrueckii UBOCC-A-214128, while C. elegans UBOCC-A-102008 produced mainly paullinic acid (20:1n-7) with concentrations up to 100 mg L−1. Concerning cerebroside production, HPTLC appeared as a relevant approach for their detection and quantification. Promising candidates belonging to the Mucoromycota phylum were found, especially in the Absidia genus with A. spinosa UBOCC-A-101332 as the best producer (12.7 mg L−1). Conclusions The present study highlighted PUFA and cerebroside production in a large diversity of fungi and the fact that members of the Mucoromycota phylum are good producers of PUFA as well as cerebrosides.
