Abstract Effective antibody responses are essential to generate protective humoral immunity. Different inflammatory signals polarize T cells towards appropriate effector phenotypes during an infection or immunization. Th1 and Th2 cells have been associated with the polarization of humoral responses. However, T follicular helper cells (Tfh) have a unique ability to access the B cell follicle and support the germinal center (GC) responses by providing B cell help. We investigated the specialization of Tfh cells induced under type-1 and type-2 conditions. We first studied homogenous Tfh cell populations generated by adoptively transferred TCR-transgenic T cells in mice immunized with type-1 and type-2 adjuvants. Using a machine learning approach, we established a gene expression signature that discriminates Tfh cells polarized towards type-1 and type-2 response, defined as Tfh1 and Tfh2 cells. The distinct signatures of Tfh1 and Tfh2 cells were validated against datasets of Tfh cells induced following lymphocytic choriomeningitis virus (LCMV) or helminth infection. We generated single-cell and spatial transcriptomics datasets to dissect the heterogeneity of Tfh cells and their localization under the two immunizing conditions. Besides a distinct specialization of GC Tfh cells under the two immunizations and in different regions of the lymph nodes, we found a population of Gzmk+ Tfh cells specific for type-1 conditions. In human individuals, we could equally identify CMV-specific Tfh cells that expressed Gzmk. Our results show that Tfh cells acquire a specialized function under distinct types of immune responses and with particular properties within the B cell follicle and the GC.

Resumo Objetivo Verificar e correlacionar a resiliência e o significado do trabalho em enfermeiros hospitalares no contexto da pandemia de COVID-19. Método Estudo descritivo e correlacional, realizado entre junho e novembro de 2021, com 164 enfermeiros de dois hospitais de São Paulo com os instrumentos a Escala de Resiliência Connor-Davidson e o Inventário do Significado do trabalho. Os dados foram analisados por meio da estatística descritiva e inferencial sendo adotados os seguintes testes: Teste de correlação de Pearson; Teste de Kruskal-Wallis; Teste t de Student; Teste de Dunn; Teste de correlação de Spearman; Análise de Variância Univariada; Teste de normalidade de Shapiro-Wilk; Teste de Levene. O valor de significância estatística adotado foi de 5% (p ≤ 0,05). Resultados Média de idade 37,54 anos (DP = 8,52), sendo 81,1% (133) do sexo feminino, média da resiliência 76,98 (DP=11,32). Correlações significativas positivas foram encontradas entre: escolaridade, idade e a resiliência com os atributos “Fonte de realização e independência econômica”, “Expressão de respeito e de acolhimento”, “Fonte de desafio, responsabilidade e sustento”, “Desafiar-se”, “Crescer economicamente”, “Sentir prazer e proteção”, “Contribuir socialmente e ser assistido” e “Ser reconhecido”; Conclusão Houve correlação da resiliência com o significado do trabalho, colaborando com o enfrentamento das adversidades encontradas na pandemia de COVID-19.

Objective: To verify and correlate resilience and meaning of work in hospital nurses during the COVID-19 pandemic. Method: This is a descriptive and correlational study, carried out between June and November 2021, with 164 nurses from two hospitals in São Paulo using the Connor-Davidson Resilience Scale and the Inventário do Significado do Trabalho (Meaning of Work Inventory). The data were analyzed using descriptive and inferential statistics, Pearson's correlation test, Kruskal-Wallis test, Student's t test, Dunn's test, Spearman's correlation test, Univariate Analysis of Variance, Shapiro-Wilk normality test, Levene's test. The statistical significance value adopted was 5% (p ≤ 0.05). Results: Mean age 37.54 years (SD = 8.52), with 81.1% (133) female and mean resilience of 76.98 (SD = 11.32). Significant positive correlations were found between education, age and resilience with the attributes "Source of achievement and economic independence", "Expression of respect and acceptance", "Source of challenge, responsibility and livelihood", "Challenging", "Economic growth", "Feeling pleasure and protection", "Contributing socially and be assisted" and "Being recognized"; Conclusion: There was a correlation between resilience and the meaning of work, helping to face the adversities encountered in the COVID-19 pandemic. Recent investigations into the microbiota and fermentation of Coffea canephora fruits and beans have yielded limited advancements globally, highlighting a developing field of study. Consequently, this review seeks to consolidate existing literature through an analysis focusing on the microbiota and fermentation processes inherent in the postharvest processing of Canephora coffee. To this end, a comprehensive examination of the principal microorganisms inherent to this species, the application of starter cultures in fermentation, and the repercussions of fermentation on the chemical and sensory attributes of the beverage will be expounded. These investigations underscore the influence exerted by the fermentation process and the introduction of microorganism inoculation on Canephora coffees’ chemical composition and sensory characteristics. Fermentation emerges as a mechanism facilitating the modification of coffee flavor and aroma, thereby presenting avenues for innovative enhancements in producing distinct Canephora coffee beverages. Research to assess the microbiota of Canephora coffees from various origins has the potential to advance our understanding of the microbial ecology specific to this species. Such studies will play a crucial role in identifying pertinent starter cultures that could be used to produce high-quality coffees.

Abstract: Yacon (Smallanthus sonchifolius) is a tuberous root with a high phenolic compounds and fibers content which has a prebiotic effect, both important for preventing and/or promoting the reduction of non-communicable chronic diseases risks. However, this root’s shelf life is highly reduced and culminates the reduction of the beneficial to health compounds, which can be avoided by some strategies. In this sense, this work aimed to produce yacon beverages added with anti-browning agents (cysteine or citric acid) and evaluated the spontaneous fermentation during storage. Three yacon beverages have been produced with cysteine or citric acid (0.05% w/w) and a control beverage. Beverages were analyzed by microbiological counts, nutritional composition, physical-chemical characteristics, total phenolic compounds and phenolic acids, for 60 day storage. The data were evaluated by ANOVA and compared using the Duncan test (p≤0.05) or regression models were adjusted. Regarding microorganisms, all beverages have reached 109 CFU.ml-1 after 30 day storage. To the anti-browning agents use, the differences were occasional; however, the citric acid beverage had a desirable pH (< 4.5) for a fermented beverage, which allows inhibition of undesirable bacteria and a higher proportional initial FOS amount and less variation with 30 and 60 day storage. However, the citric acid beverage have presented smaller amount of phenolic compounds like chlorogenic and gallic acids (230.37 ± 168.63 and 40.87 ± 1.32, respectively). So, we conclued that spontaneous fermentation was a preparation technique which has added nutritional and functionals values to the yacon beverage, in special to beverage added citric acid, with a higher proportion of FOS.
