Abstract

Federated learning (FL) enables collaborative model training across institutions without sharing sensitive data, making it an attractive solution for medical imaging tasks. However, traditional FL methods, such as Federated Averaging (FedAvg), face difficulties in generalizing across domains due to variations in imaging protocols and patient demographics across institutions. This challenge is particularly evident in pancreas MRI segmentation, where anatomical variability and imaging artifacts significantly impact performance. In this paper, we conduct a comprehensive evaluation of FL algorithms for pancreas MRI segmentation and introduce a novel approach that incorporates adaptive aggregation weights. By dynamically adjusting the contribution of each client during model aggregation, our method accounts for domain-specific differences and improves generalization across heterogeneous datasets. Experimental results demonstrate that our approach enhances segmentation accuracy and reduces the impact of domain shift compared to conventional FL methods while maintaining privacy-preserving capabilities. Significant performance improvements are observed across multiple hospitals (centers).

Abstract

In this work, we have developed and characterized a position-sensitive CLYC detector that acts as the neutron imaging layer and $\gamma$-ray Compton scatterer of the novel dual \g-ray and neutron imaging system GN-Vision, which aims at simultaneously obtaining information about the spatial origin of \g-ray and neutron sources. We first investigated the performance of large 50$\times$50~mm$^{2}$ monolithic CLYC crystals coupled to a pixelated SiPM in terms of energy resolution and neutron-gamma discrimination. The response of two different 95\% $^{6}$Li-enriched CLYC detectors coupled to an array of 8$\times$8 SiPMs was studied in comparison to the results of a conventional photo-multiplier tube. Energy resolution ranging from 6-8\% for the $^{137}$Cs peak and a figure of merit of 3-4 for the neutron-gamma discrimination have been obtained. The spatial response of the CLYC-SiPM detector to $\gamma$-rays and neutrons has also been characterized using charge modulation-based multiplexing techniques based on a diode-coupled charge division circuit. Average resolutions close to 5~mm FWHM with good linearity are obtained in the transverse crystal plane. Abstract

Dosimetry in BNCT poses significant challenges due to the indirect effect of neutrons interacting with elements within the body and uncertainties associated with the uptake of boron compounds used in clinical practice. Current treatment planning relies on unconventional estimates of boron tumor uptake derived from prior PET scans and thus, an online boron-uptake monitor would be highly convenient. This work presents the first pilot experiments carried out at ILL-Grenoble with the high-efficiency Compton camera i-TED, hereby aiming at demonstrating its applicability for BNCT dosimetry by introducing real-time measurement of the boron concentration and imaging capabilities of spatial dose distribution. In this experiment, we measured the $^{10}$B uptake of different cancer cells of tongue squamous cell carcinoma, malignant melanoma and glioblastoma treated with BPA (80~ppm of $^{10}$B). The samples were irradiated with the thermal neutron spectrum of ILL-Grenoble and the 478keV $\gamma$-rays from the $^{7}$Li de-excitation after the neutron-boron reaction were registered both with the Compton imager and the high-sensitivity FIPPS HPGe array. These series of measurements allowed us to demonstrate the imaging capabilities of the Compton imaging device for this type of application, as well as to assess its sensitivity, which was found to be below 1 $\mu$g of $^{10}$B., Comment: 10 pages, 12 figures, submitted to the Special Issue of Applied Radiation and Isotopes associated to the 20th International Congress on Neutron Capture Therapy (20th ICNCT)

Abstract

The $^{246}$Cm(n,$\gamma$) and $^{248}$Cm(n,$\gamma$) cross-sections have been measured at the Experimental Area 2 (EAR2) of the n_TOF facility at CERN with three C$_6$D$_6$ detectors. This measurement is part of a collective effort to improve the capture cross-section data for Minor Actinides (MAs), which are required to estimate the production and transmutation rates of these isotopes in light water reactors and innovative reactor systems. In particular, the neutron capture in $^{246}$Cm and $^{248}$Cm open the path for the formation of other Cm isotopes and heavier elements such as Bk and Cf and the knowledge of (n,$\gamma$) cross-sections of these Cm isotopes plays an important role in the transport, transmutation and storage of the spent nuclear fuel. The reactions $^{246}$Cm(n,$\gamma$) and $^{248}$Cm(n,$\gamma$) have been the two first capture measurements analyzed at n_TOF EAR2. Until this experiment and two recent measurements performed at J-PARC, there was only one set of data of the capture cross-sections of $^{246}$Cm and $^{248}$Cm, that was obtained in 1969 in an underground nuclear explosion experiment. In the measurement at n_TOF a total of 13 resonances of $^{246}$Cm between 4 and 400 eV and 5 of $^{248}$Cm between 7 and 100 eV have been identified and fitted. The radiative kernels obtained for $^{246}$Cm are compatible with JENDL-5, but some of them are not with JENDL-4, which has been adopted by JEFF-3.3 and ENDF/B-VIII.0. The radiative kernels obtained for the first three $^{248}$Cm resonances are compatible with JENDL-5, however, the other two are not compatible with any other evaluation and are 20% and 60% larger than JENDL-5.

Abstract

Automated volumetric segmentation of the pancreas on cross-sectional imaging is needed for diagnosis and follow-up of pancreatic diseases. While CT-based pancreatic segmentation is more established, MRI-based segmentation methods are understudied, largely due to a lack of publicly available datasets, benchmarking research efforts, and domain-specific deep learning methods. In this retrospective study, we collected a large dataset (767 scans from 499 participants) of T1-weighted (T1W) and T2-weighted (T2W) abdominal MRI series from five centers between March 2004 and November 2022. We also collected CT scans of 1,350 patients from publicly available sources for benchmarking purposes. We developed a new pancreas segmentation method, called PanSegNet, combining the strengths of nnUNet and a Transformer network with a new linear attention module enabling volumetric computation. We tested PanSegNet's accuracy in cross-modality (a total of 2,117 scans) and cross-center settings with Dice and Hausdorff distance (HD95) evaluation metrics. We used Cohen's kappa statistics for intra and inter-rater agreement evaluation and paired t-tests for volume and Dice comparisons, respectively. For segmentation accuracy, we achieved Dice coefficients of 88.3% (std: 7.2%, at case level) with CT, 85.0% (std: 7.9%) with T1W MRI, and 86.3% (std: 6.4%) with T2W MRI. There was a high correlation for pancreas volume prediction with R^2 of 0.91, 0.84, and 0.85 for CT, T1W, and T2W, respectively. We found moderate inter-observer (0.624 and 0.638 for T1W and T2W MRI, respectively) and high intra-observer agreement scores. All MRI data is made available at https://osf.io/kysnj/. Abstract

The neutron time-of-flight facility n_TOF at CERN is a spallation source dedicated to measurements of neutron-induced reaction cross-sections of interest in nuclear technologies, astrophysics, and other applications. Since 2014, Experimental ARea 2 (EAR2) is operational and delivers a neutron fluence of $4\times 10^7$ neutrons per nominal proton pulse, which is 50 times higher than the one of Experimental ARea 1 (EAR1) of $8\times10^5$ neutrons per pulse. The high neutron flux at EAR2 results in high counting rates in the detectors that challenged the previously existing capture detection systems. For this reason, a Segmented Total Energy Detector (sTED) has been developed to overcome the limitations in the detectors response, by reducing the active volume per module and by using a photomultiplier (PMT) optimized for high counting rates. This paper presents the main characteristics of the sTED, including energy and time resolution, response to $\gamma$-rays, and provides as well details of the use of the Pulse Height Weighting Technique (PHWT) with this detector. The sTED has been validated to perform neutron-capture cross-section measurements in EAR2 in the neutron energy range from thermal up to at least 400 keV. The detector has already been successfully used in several measurements at n_TOF EAR2.

Abstract

Abstract MYCN amplification predicts poor prognosis in childhood neuroblastoma. To identify MYCN oncogenic signal dependencies we performed N-ethyl-N-nitrosourea (ENU) mutagenesis on the germline of neuroblastoma-prone TH-MYCN transgenic mice to generate founders which had lost tumorigenesis. Sequencing of the mutant mouse genomes identified the Ring Finger Protein 121 (RNF121 WT ) gene mutated to RNFM158R associated with heritable loss of tumorigenicity. While the RNF121WT protein localised predominantly to the cis-Golgi Complex, the RNF121 M158R mutation in Helix 4 of its transmembrane domain caused reduced RNF121 protein stability and absent Golgi localisation. RNF121WT expression markedly increased during TH-MYCN tumorigenesis, whereas hemizygous RNF121 WT gene deletion reduced TH-MYCN tumorigenicity. The RNF121WT-enhanced growth of MYCN-amplified neuroblastoma cells depended on RNF121WT transmembrane Helix 5. RNF121WT directly bound MYCN protein and enhanced its stability. High RNF121 mRNA expression associated with poor prognosis in human neuroblastoma tissues and another MYC-driven malignancy, laryngeal cancer. RNF121 is thus an essential oncogenic cofactor for MYCN and a target for drug development.

Abstract

The global rise in plastic production has led to significant plastic deposition in aquatic ecosystems, releasing chemical compounds as plastics degrade. Among these, bisphenol A (BPA) is a major global concern due to its endocrine-disrupting effects and widespread presence in aquatic environments. Furthermore, the toxicity of BPA on aquatic organisms can be modulated by global change stressors such as temperature, which plays an essential role in the metabolism of organisms, including the degradation and accumulation of toxic compounds. In this study, we aimed to understand how temperature can modulate the toxic effect of BPA on a phytoplankton species (Cryptomonas ovata) and how this effect can be transferred to its herbivorous consumer (Daphnia magna). To do this, we first determined the sensitivity of C. ovata over a BPA gradient (0–10 mg L−1). Subsequently, we experimentally determined how the increase in temperature (+5ºC) could modify the toxic effect of BPA on the physiology, metabolism and growth of the phytoplankton. Finally, we investigated how this effect transferred to the growth rate of D. magna through food. Our results show a negative effect of BPA on C. ovata from 5 mg BPA L−1, affecting its photosynthetic yield of photosystem II, net primary production, respiration, and growth. This effect was accelerated when the temperature was higher. Additionally, the growth rate of D. magna also decreased when fed on C. ovata grown in the presence of BPA and high temperature. Our results indicate that high temperature can accelerate the toxic effects of BPA on organisms located at the base of the food web and this effect could be transferred to higher levels through food.

Abstract

Community engagement and local governance are important components of health interventions aiming to empower local populations. Yet, there is limited evidence on how to effectively engage with communities and codevelop interventions, especially in Southeast Asian contexts. Despite rapid progress, the Lao People’s Democratic Republic (Lao PDR) still has high maternal and child mortality, with essential service coverage showing significant disparities across socioeconomic strata. Long-standing challenges in community health were exacerbated by the COVID-19 pandemic and reinforced by poor trust between users and health providers. However, the pandemic also provided an opportunity to develop approaches for enhanced community engagement and local governance capacity to tackle health inequities. The Community Network Engagement for Essential Healthcare and COVID-19 Responses through Trust (CONNECT) Initiative, developed by the Lao PDR government, WHO and partners, has resulted in initial positive outcomes in community health such as increased vaccination uptake, facility births and trust in health providers. This case study describes the iterative, adaptive process by which the CONNECT Initiative was developed, and how the core components, key stakeholders, theory of change and evaluation framework evolved from grounded observations and hypotheses. Lessons learnt include (1) awareness of entry points and existing structures to strengthen local governance for health through mutually beneficial intersectoral collaboration; (2) building relationships and trust with an adaptive, grounds-up approach for sustainability and scalability. As a model which can be adapted to other settings, this case study provides evidence on how to engage with communities, strengthen local governance and codevelop interventions towards greater health equity.

Abstract

Heterochromatin is a pivotal element in the functional organization of genomes. In our study, we delve into the heterochromatin pattern of association by the PML (promyelocytic leukemia) protein. By using PML chromatin immunoprecipitation and sequencing data and comparing computational methodologies to depict PML chromatin association, we describe PML-associated domains or PADs as large heterochromatic regions that exhibit similar genomic features across cancer cell lines. We show that PADs are specifically enriched in non-coding genes, duplicated gene clusters, and repetitive DNA elements. Moreover, we find enriched binding motifs of KZFPs, which are involved in orchestrating epigenetic repression at repetitive DNA elements. Hence, our findings suggest that PML conservatively associates to heterochromatic domains enriched in repetitive DNA elements and duplicated gene clusters in cancer. These findings contribute to a broader understanding of the complex regulatory framework of genome organization by heterochromatin in cancer.

39. Oncogenic and immunological targets for matched therapy of pediatric blood cancer patients: Dutch iTHER study experience

Abstract

Abstract Over the past 10 years, institutional and national molecular tumor boards have been implemented for relapsed or refractory pediatric cancer to prioritize targeted drugs for individualized treatment based on actionable oncogenic lesions, including the Dutch iTHER platform. Hematological malignancies form a minority in precision medicine studies. Here, we report on 56 iTHER leukemia/lymphoma patients for which we considered cell surface markers and oncogenic aberrations as actionable events, supplemented with ex vivo drug sensitivity for six patients. Prior to iTHER registration, 34% of the patients had received allogeneic hematopoietic cell transplantation (HCT) and 18% CAR‐T therapy. For 51 patients (91%), a sample with sufficient tumor percentage (≥20%) required for comprehensive diagnostic testing was obtained. Up to 10 oncogenic actionable events were prioritized in 49/51 patients, and immunotherapy targets were identified in all profiled patients. Targeted treatment(s) based on the iTHER advice was given to 24 of 51 patients (47%), including immunotherapy in 17 patients, a targeted drug matching an oncogenic aberration in 12 patients, and a drug based on ex vivo drug sensitivity in one patient, resulting in objective responses and a bridge to HCT in the majority of the patients. In conclusion, comprehensive profiling of relapsed/refractory hematological malignancies showed multiple oncogenic and immunotherapy targets for a precision medicine approach, which requires multidisciplinary expertise to prioritize the best treatment options for this rare, heavily pretreated pediatric population.

Abstract

Abstract The dystrophin protein has well‐characterized roles in force transmission and maintaining membrane integrity during muscle contraction. Studies have reported decreased expression of dystrophin in atrophying muscles during wasting conditions, and that restoration of dystrophin can attenuate atrophy, suggesting a role in maintaining muscle mass. Phosphorylation of S3059 within the cysteine‐rich region of dystrophin enhances binding between dystrophin and β‐dystroglycan, and mimicking phosphorylation at this site by site‐directed mutagenesis attenuates myotube atrophy in vitro. To determine whether dystrophin phosphorylation can attenuate muscle wasting in vivo, CRISPR‐Cas9 was used to generate mice with whole body mutations of S3059 to either alanine (DmdS3059A) or glutamate (DmdS3059E), to mimic a loss of, or constitutive phosphorylation of S3059, on all endogenous dystrophin isoforms, respectively. Sciatic nerve transection was performed on these mice to determine whether phosphorylation of dystrophin S3059 could attenuate denervation atrophy. At 14 days post denervation, atrophy of tibialis anterior (TA) but not gastrocnemius or soleus muscles, was partially attenuated in DmdS3059E mice relative to WT mice. Attenuation of atrophy was associated with increased expression of β‐dystroglycan in TA muscles of DmdS3059E mice. Dystrophin S3059 phosphorylation can partially attenuate denervation‐induced atrophy, but may have more significant impact in less severe modes of muscle wasting.

Abstract

Checkers is a strategy game for two players on an $8\times 8$ square board. This document outlines the creation of a checkers game, employing alpha-beta pruning as a search algorithm and transposition tables to enable the game to learn from past play sessions. The utilization of these techniques enhances the game’s strategic decision-making process, contributing to an improved overall gaming experience. The execution time of the machine’s moves in games with and without using transposition tables showed that it is possible to make some games faster by using the data stored in the tables. However, under some conditions this can be more time-consuming than the search performed by the alpha-beta algorithm. A user survey resulted in positive outcomes, indicating a favorable user experience when using the implemented game with the transposition tables. Comparative analysis against other algorithms demonstrated the successful implementation of the approach. This research contributes to the field of game AI optimization by integrating alpha-beta pruning and transposition tables into a web-based checkers game. The implementation offers a balance between computational efficiency and user engagement in the web-based checkers game.
