Pygopus (Pygo) has been identified as a specific nuclear co-activator of the canonical Wingless (Wg)/Wnt signaling pathway in Drosophila melanogaster. Pygo proteins consist of two conserved domains: an N-terminal homologous domain (NHD) and a C-terminal plant homologous domain (PHD). The PHD's ability to bind to di- and trimethylated lysine 4 of histone H3 (H3K4me2/3) appears to be independent of Wnt signaling. There is ongoing debate regarding the significance of Pygo's histone-binding capacity. Drosophila Pygo orthologs have a tryptophan (W) > phenylalanine (F) substitution in their histone pocket-divider compared to vertebrates, leading to reduced histone affinity. In this research, we utilized CRISPR/Cas9 technology to introduce the Pygo-F773W point mutation in Drosophila, successfully establishing a viable homozygous Pygo mutant line for the first time. Adult mutant flies displayed noticeable abnormalities in reproduction, locomotion, heart function, and lifespan. RNA-seq and cluster analysis indicated that the mutation primarily affected pathways related to immunity, metabolism, and posttranslational modification in adult flies rather than the Wnt signaling pathway. Additionally, a reduction in H3K9 acetylation levels during the embryonic stage was observed in the mutant strains. These findings support the notion that Pygo plays a wider role in chromatin remodeling, with its involvement in Wnt signaling representing only a specific aspect of its chromatin-related functions. The effective identification of remanufacturing design requirements is crucial to ensure that remanufactured products meet the required standards and demands. However, in addition to considering the performance demands of customers for products, remanufactured products should also take into account the various types and degrees of failure across diverse parts. To this end, under the dual demands, a knowledge graph – based requirement identification model is proposed to intelligently transform complex remanufacturing demands into standardised design requirements. Firstly, excavation of design information from representative cases is conducted, including failure characteristics, customer demands and design requirements (FCDR). A FCDR ontology model is proposed by establishing the mapping relationship between demands and design requirements. Secondly, the triplet of design information is integrated into the BERT-BiLSTM-CRF model to obtain the accurate entity, and the ALBERT-BiLSTM-Attention model is used to extract the FCDR relationship, so that a knowledge graph can be constructed. Thirdly, by using coupling weighting technique to consider the relevance of the dual demands, normalised failure characteristics and customer demands are accurately extracted and mapped to similar design requirement nodes in knowledge graph. For the catalyst for water splitting, low overpotential and longer stability is very important. Here, we developed a simple strategy to prepare an efficient catalyst for oxygen evolution by partially phosphating NiFe-LDH and then etching and growing Prussian blue analogues on it. Benefiting from its unique hierarchical structure, the synthesized NiFeCo PBA-NiFeP catalyst showed excellent electrocatalytic performance in 1 M KOH. At 100 mA/cm2, the overpotential of the oxygen evolution reaction (OER) is only 245 mV and exhibits stability for over 50 h. This is superior to most of the OER catalysts reported so far. Its excellent performance is due to its unique hierarchical structure and the synergistic effect between polymetals, which enables it to have abundant active sites and efficient electron transfer. This controllable and universal strategy will provide a new idea for developing interfacial composites and preparing high efficiency electrocatalysts. • A new NiFeCo-PBA-NiFeP composite was prepared by hydrothermal-phosphating-etching process. • The unique hierarchical structure facilitates gas release and contact between the active site and the solution. • The synergistic effect between multiple metals improves the OER properties of the materials. • The combination of NiFeP and NiFeCo PBA improves the OER performance of the material. Due to the intricate mechanism of Abbe error's impact on spatial accuracy, the accumulated Abbe error in the traditional spatial accuracy model is difficult to identify and eliminate, resulting in reduced modeling accuracy and limiting accuracy improvement. This paper proposes a data-driven spatial accuracy modeling approach for machine tools under the influence of Abbe error, using a three-axis coupling measurement optical path to directly measure the comprehensive spatial accuracy data of the machine tool containing Abbe error. Firstly, to effectively identify Abbe error in the comprehensive spatial accuracy, an Abbe error quantization function is established, which analyzes its formation mechanism in the measurement process to eliminate Abbe error in the machine tool's spatial accuracy data. Furthermore, to address the issue of limited data samples after eliminating Abbe error, data samples are extended based on the degradation mechanism of machine tool spatial accuracy at different coordinate positions, resulting in a high-precision spatial error model for machine tools. Finally, the experiment is conducted on a three-axis CNC machine tool, with a model accuracy of over 95%. The integration of green scheduling and lot splitting scheduling is indispensable for ensuring the coordinated optimization of economic and environmental benefits in flexible job-shop scheduling (FJS). However, this integration involves not only the indicator of greenness and economy but also the process of production planning and scheduling, which is substantially complicated. To this end, a hierarchical integrated scheduling method is proposed by comprehensively considering the multilevel organizational structure and task configuration characteristics of flexible job-shop, as well as the differences in objectives on different scheduling levels: workshop level, process unit level, machine tool level. On the workshop level, a lot splitting model is presented to obtain the optimal processing task set for each production cycle with the minimum expected cost (startup cost, tardiness cost, and holding cost). On the process unit level, a task allocation model is given to allocate the optimal workload for each machine tool with the minimum processing energy consumption and maximum machine load. On the machine tool level, an operation sequencing model is established to obtain the optimal processing sequence for each machine tool with the minimum standby energy consumption and makespan. According to the solving characteristics of the hierarchical models, a multi-objective algorithm is applied. Finally, a case study is demonstrated to validate the proposed method. Infection with bovine leukemia virus (BLV) leads to enzootic bovine leukosis, the most prevalent neoplastic disease in cattle. Due to the lack of commercially available vaccines, reliable eradication of the disease can be achieved through the testing and elimination of BLV antibody-positive animals. In this study, we developed a novel competitive ELISA (cELISA) to detect antibodies against BLV capsid protein p24. Recombinant p24 protein expressed by Escherichia coli , in combination with the monoclonal antibody 2G11 exhibiting exceptional performance, was used for the establishment of the cELISA. Receiver-operating characteristic curve analysis showed that the sensitivity and specificity of the assay were 98.85 % and 98.13 %, respectively. Furthermore, the established cELISA was specific for detecting BLV-specific antibodies, without cross-reactivity to antisera for six other bovine viruses. Significantly, experimental infection of cattle and sheep with BLV revealed that the cELISA accurately monitors seroconversion. In a performance evaluation, the established cELISA displayed a high agreement with Western blotting and the commercial BLV gp51 cELISA kit in the detection of 242 clinical samples, respectively. In conclusion, the novel p24 cELISA exhibited the potential to be a reliable and efficient diagnostic tool for BLV serological detection with a broad application prospect. [ABSTRACT FROM AUTHOR]

To promote energy saving, high efficiency and quality production in the mechanical manufacturing industry, a large number of studies have been conducted on modelling and evaluation. However, most of the approaches have focused on single objective modelling, evaluation and conventional machining method like unidirectional turning (UDT). This paper presents a multi-objective modelling and evaluation method for energy, efficiency, economy and quality using a new process of forward-and-reverse multidirectional turning (MDT). Firstly, the multi-objective influential factors of cutting parameters for energy consumption, cutting time, production cost and surface roughness of MDT are analyzed to establish the corresponding models, respectively. Then taking the energy consumption, cutting time, production cost and surface roughness as the objectives, the Technique for Order Preference by Similarity to Ideal Solution (TOPSIS) is applied for the multi-objective evaluation through different processes. The reprogramming of lipid metabolism serves an important role in occurrence and development of liver cancer. Fatty acid hydroxylase domain containing 2 (FAXDC2) is a hydroxylase involved in the synthesis of cholesterol and sphingomyelin and downregulated in various types of cancer. There are no reports on the relationship between FAXDC2 and liver carcinogenesis. The present study used multiple portals and publicly available tools to explore its correlation with liver cancer. The results showed that the expression of FAXDC2 decreased in liver cancer and the methylation level near the promoter increased. Patients with liver cancer and with low expression of FAXDC2 had a poor prognosis. Gain of function and loss of function strategies were performed to evaluate its roles in liver cancer cells. CCK-8 assay showed that overexpression of FAXDC2 inhibited the viability of liver cancer cells (HepG2). Flow cytometry analysis indicated that HepG2 cells with overexpressing FAXDC2 showed an S phase arrest, associated with cyclin-dependent kinase 2 decreased. Transwell experiments showed that increasing FAXDC2 inhibited HepG2 cell invasion ability, accompanied by the upregulation of E-cadherin. Notably, knockdown of FAXDC2 had no significant effect on cell cycle and invasion functions. Based on the cBioPortal platform, FAXDC2 was predicted to closely correlate to the ERK signal in tumorigenesis. Western blotting results showed that overexpression of FAXDC2 decreased the phosphorylation level of ERK in liver cancer cells. The present study first identified FAXDC2 as a liver cancer suppressor, which might inhibit the proliferation and invasion of liver cancer cells through the mechanism associated with ERK signaling.
