1. Autoregressive models for panel data causal inference with application to state-level opioid policies

Abstract: Motivated by the study of state opioid policies, we propose a novel approach that uses autoregressive models for causal effect estimation in settings with panel data and staggered treatment adoption. Specifically, we seek to estimate of the impact of key opioid-related policies by quantifying the effects of must access prescription drug monitoring programs (PDMPs), naloxone access laws (NALs), and medical marijuana laws on opioid prescribing. Existing methods, such as differences-in-differences and synthetic controls, are challenging to apply in these types of dynamic policy landscapes where multiple policies are implemented over time and sample sizes are small. Autoregressive models are an alternative strategy that have been used to estimate policy effects in similar settings, but until this paper have lacked formal justification. We outline a set of assumptions that tie these models to causal effects, and we study biases of estimates based on this approach when key causal assumptions are violated. In a set of simulation studies that mirror the structure of our application, we also show that our proposed estimators frequently outperform existing estimators. In short, we justify the use of autoregressive models to provide robust evidence on the effectiveness of four state policies in combating the opioid crisis.

2. A Creative Communication Partnership to Promote Curricula Dissemination on Social Media

Abstract: Genome Sciences Education Outreach (GSEO) has developed innovative programs that bring leading-edge science to teachers and students in K-12 schools. Disseminating educational materials equitably and accessibly to teacher stakeholders to maximize reach and impact is challenging for many programs. Traditionally, programs connect materials with teachers through local networks, in-person professional development sessions, and at regional and national conference presentations. The need for curricular changes in 2020 spurred the proliferation of online and digital educational materials and professional development opportunities. These digital materials--now available to a worldwide audience--require a shift in dissemination strategy to enhance the potential reach of these materials both locally and nationally. This manuscript reports a case study of a dissemination approach, to create a collaboration between GSEO and CommLead (the communications master's program at the University of Washington) to promote education materials developed by the publicly-funded Genes, Environment and Me Network (GEMNet) program. This manuscript describes the development and the ad hoc implementation and evaluation of a social media campaign to expand the reach of the GEMNet curricula. With a targeted social media campaign on Facebook, GSEO was able to dramatically and affordably increase the reach of the GEMNet curricula and expand the potential impact and utilization of educational materials to a nationwide teacher audience, highlighting the potential for other similar collaborations to efficiently enhance the dissemination strategy of other education outreach programs.

3. Evaluate the Safety Effects of Adopting a Stop-as-Yield Law for Cyclists in California

Abstract: The escalating number of injuries and fatalities among cyclists is a pressing safety concern. In the United States, communities are actively seeking strategies to boost cyclist safety, with some states implementing bike-specific policies, such as stop-as-yield laws, to support cyclists. Stop-as-yield laws allow cyclists to treat stop signs as yield signs. The laws are not yet widely implemented, and their potential safety impact is a subject of debate among transportation experts and advocates. This study investigates how stop-as-yield laws can positively or negatively affect safety and provides insights and guidelines for California policymakers and safety practitioners if the law passes in California. We collected cyclist data from five states that have enacted stop-as-yield laws—Idaho, Arkansas, Oregon, Washington and Delaware—and data from some of their contiguous states without such legislation. Using an observational before-after study with comparison groups at the state level, the research examined changes in cyclist crash frequencies after the laws were implemented. Additionally, a random-effects negative binomial regression model with panel data was employed to estimate a law’s overall impact. The results did not indicate a significant change in cyclist crashes among the states with stop-as-yield laws.

Abstract: Radium-225 (nuclear spin $I=1/2$) ions possess electronic hyperfine transitions that are first-order insensitive to magnetic field noise, which is advantageous for optical clocks and quantum information science. We report on laser cooling and trapping of radium-225 ions and hyperfine splitting measurements of the ion's $7s$ $^2S_{1/2}$, $7p$ $^2P_{1/2}$, and $6d$ $^2D_{3/2}$ states. We measured the ground state hyperfine constant, $A(^2S_{1/2}) = -27.684511056(9)\ \mathrm{GHz}$, and the quadratic Zeeman coefficient, $C_2 = 142.3(10)\ \mathrm{Hz\ G}^{-2}$, of the $^2S_{1/2} (F=0, m_F = 0) \leftrightarrow~^2S_{1/2} (F=1, m_{F} = 0)$ transition. We also measured the hyperfine constants of the $^2P_{1/2}$ state, $A(^2P_{1/2}) = -5.447(4)\ \mathrm{GHz}$, and the $^2D_{3/2}$ state, $A(^2D_{3/2}) = -619.7(11)\ \mathrm{MHz}$., Comment: 5 pages, 4 figures

Abstract: Active Galactic Nuclei (AGN) are prime candidate sources of the high-energy, astrophysical neutrinos detected by IceCube. This is demonstrated by the real-time multi-messenger detection of the blazar TXS 0506+056 and the recent evidence of neutrino emission from NGC 1068 from a separate time-averaged study. However, the production mechanism of the astrophysical neutrinos in AGN is not well established which can be resolved via correlation studies with photon observations. For neutrinos produced due to photohadronic interactions in AGN, in addition to a correlation of neutrinos with high-energy photons, there would also be a correlation of neutrinos with photons emitted at radio wavelengths. In this work, we perform an in-depth stacking study of the correlation between 15 GHz radio observations of AGN reported in the MOJAVE XV catalog, and ten years of neutrino data from IceCube. We also use a time-dependent approach which improves the statistical power of the stacking analysis. No significant correlation was found for both analyses and upper limits are reported. When compared to the IceCube diffuse flux, at 100 TeV and for a spectral index of 2.5, the upper limits derived are $\sim3\%$ and $\sim9\%$ for the time-averaged and time-dependent case, respectively., Comment: 14 Pages 7 Figures

Abstract: We present a search for an eV-scale sterile neutrino using 7.5 years of data from the IceCube DeepCore detector. The analysis uses a sample of 21,914 events with energies between 5 and 150 GeV to search for sterile neutrinos through atmospheric muon neutrino disappearance. Improvements in event selection and treatment of systematic uncertainties provide greater statistical power compared to previous DeepCore sterile neutrino searches. Our results are compatible with the absence of mixing between active and sterile neutrino states, and we place constraints on the mixing matrix elements $|U_{\mu 4}|^2 < 0.0534$ and $|U_{\tau 4}|^2 < 0.0574$ at 90% CL under the assumption that $\Delta m^2_{41}\geq 1\;\mathrm{eV^2}$. These null results add to the growing tension between anomalous appearance results and constraints from disappearance searches in the 3+1 sterile neutrino landscape., Comment: 11 pages, 5 figures. Abstract: The recent IceCube detection of TeV neutrino emission from the nearby active galaxy NGC 1068 suggests that active galactic nuclei (AGN) could make a sizable contribution to the diffuse flux of astrophysical neutrinos. The absence of TeV $\gamma$-rays from NGC 1068 indicates neutrino production in the vicinity of the supermassive black hole, where the high radiation density leads to $\gamma$-ray attenuation. Therefore, any potential neutrino emission from similar sources is not expected to correlate with high-energy $\gamma$-rays. Disk-corona models predict neutrino emission from Seyfert galaxies to correlate with keV X-rays, as they are tracers of coronal activity. Using through-going track events from the Northern Sky recorded by IceCube between 2011 and 2021, we report results from a search for individual and aggregated neutrino signals from 27 additional Seyfert galaxies that are contained in the BAT AGN Spectroscopic Survey (BASS). Besides the generic single power-law, we evaluate the spectra predicted by the disk-corona model. Assuming all sources to be intrinsically similar to NGC 1068, our findings constrain the collective neutrino emission from X-ray bright Seyfert galaxies in the Northern Hemisphere, but, at the same time, show excesses of neutrinos that could be associated with the objects NGC 4151 and CGCG 420-015. Abstract: Active Galactic Nuclei (AGN) are promising candidate sources of high-energy astrophysical neutrinos since they provide environments rich in matter and photon targets where cosmic ray interactions may lead to the production of gamma rays and neutrinos. We searched for high-energy neutrino emission from AGN using the $\textit{Swift}$-BAT Spectroscopic Survey (BASS) catalog of hard X-ray sources and 12 years of IceCube muon track data. First, upon performing a stacked search, no significant emission was found. Second, we searched for neutrinos from a list of 43 candidate sources and found an excess from the direction of two sources, Seyfert galaxies NGC 1068 and NGC 4151. We observed NGC 1068 at flux $\phi_{\nu_{\mu}+\bar{\nu}_{\mu}}$ = $4.02_{-1.52}^{+1.58} \times 10^{-11}$ TeV$^{-1}$ cm$^{-2}$ s$^{-1}$ normalized at 1 TeV, with power-law spectral index, $\gamma$ = 3.10$^{+0.26}_{-0.22}$, consistent with previous IceCube results. The observation of a neutrino excess from the direction of NGC 4151 is at a post-trial significance of 2.9$\sigma$. If interpreted as an astrophysical signal, the excess observed from NGC 4151 corresponds to a flux $\phi_{\nu_{\mu}+\bar{\nu}_{\mu}}$ = $1.51_{-0.81}^{+0.99} \times 10^{-11}$ TeV$^{-1}$ cm$^{-2}$ s$^{-1}$ normalized at 1 TeV and $\gamma$ = 2.83$^{+0.35}_{-0.28}$.

Abstract: We present the first three-parameter fit to a 3+1 sterile neutrino model using 7.634 years of data from the IceCube Neutrino Observatory on $\nu_\mu+\overline{\nu}_\mu$ charged-current interactions in the energy range 500-9976 GeV. Our analysis is sensitive to the mass-squared splitting between the heaviest and lightest mass state ($\Delta m_{41}^2$), the mixing matrix element connecting muon flavor to the fourth mass state ($|U_{\mu4}|^2$), and the element connecting tau flavor to the fourth mass state ($|U_{\tau4}|^2$). Predicted propagation effects in matter enhance the signature through a resonance as atmospheric neutrinos from the Northern Hemisphere traverse the Earth to the IceCube detector at the South Pole. The result is consistent with the no-sterile neutrino hypothesis with a probability of 4.3 %. Profiling the likelihood of each parameter yields the 90 % confidence levels: $ 2.4\,\mathrm{eV}^{2} < \Delta m_{41}^2 <9.6\,\mathrm{eV}^{2} $ , $0.0081 < |U_{\mu4}|^2 < 0.10$ , and $|U_{\tau4}|^2< 0.035$, which narrows the allowed parameter-space for $|U_{\tau4}|^2$. However, the primary result of this analysis is the first map of the 3+1 parameter space exploring the interdependence of $\Delta m_{41}^2$, $|U_{\mu4}|^2$, and $|U_{\tau4}|^2$.

Abstract: We provide supporting details for the search for a 3+1 sterile neutrino using data collected over eleven years at the IceCube Neutrino Observatory. The analysis uses atmospheric muon-flavored neutrinos from 0.5 to 100\, TeV that traverse the Earth to reach the IceCube detector, and finds a best-fit point at $\sin^2(2\theta_{24}) = 0.16$ and $\Delta m^{2}_{41} = 3.5$ eV$^2$ with a goodness-of-fit p-value of 12\% and consistency with the null hypothesis of no oscillations to sterile neutrinos with a p-value of 3.1\%. Several improvements were made over past analyses, which are reviewed in this article, including upgrades to the reconstruction and the study of sources of systematic uncertainty. We provide details of the fit quality and discuss stability tests that split the data for separate samples, comparing results. Abstract: This Letter presents the result of a 3+1 sterile neutrino search using 10.7 years of IceCube data. We analyze atmospheric muon neutrinos that traverse the Earth with energies ranging from 0.5 to 100 TeV, incorporating significant improvements in modeling neutrino flux and detector response compared to earlier studies. Notably, for the first time, we categorize data into starting and through-going events, distinguishing neutrino interactions with vertices inside or outside the instrumented volume, to improve energy resolution. The best-fit point for a 3+1 model is found to be at $\sin^2(2\theta_{24}) = 0.16$ and $\Delta m^{2}_{41} = 3.5$ eV$^2$, which agrees with previous iterations of this study. The result is consistent with the null hypothesis of no sterile neutrinos with a p-value of 3.1\%., Comment: 9 pages, 3 figures. Abstract: Galactic PeVatrons are sources that can accelerate cosmic rays to PeV energies. The high-energy cosmic rays are expected to interact with the surrounding ambient material or radiation, resulting in the production of gamma rays and neutrinos. To optimize for the detection of such associated production of gamma rays and neutrinos for a given source morphology and spectrum, a multi-messenger analysis that combines gamma rays and neutrinos is required. In this study, we use the Multi-Mission Maximum Likelihood framework (3ML) with IceCube Maximum Likelihood Analysis software (i3mla) and HAWC Accelerated Likelihood (HAL) to search for a correlation between 22 known gamma-ray sources from the third HAWC gamma-ray catalog and 14 years of IceCube track-like data. No significant neutrino emission from the direction of the HAWC sources was found. We report the best-fit gamma-ray model and 90% CL neutrino flux limit from the 22 sources. From the neutrino flux limit, we conclude that the gamma-ray emission from five of the sources can not be produced purely from hadronic interactions. We report the limit for the fraction of gamma rays produced by hadronic interactions for these five sources.

13. 3D MR Fingerprinting for Dynamic Contrast-Enhanced Imaging of Whole Mouse Brain

Abstract: Quantitative MRI enables direct quantification of contrast agent concentrations in contrast-enhanced scans. However, the lengthy scan times required by conventional methods are inadequate for tracking contrast agent transport dynamically in mouse brain. We developed a 3D MR fingerprinting (MRF) method for simultaneous T1 and T2 mapping across the whole mouse brain with 4.3-min temporal resolution. We designed a 3D MRF sequence with variable acquisition segment lengths and magnetization preparations on a 9.4T preclinical MRI scanner. Model-based reconstruction approaches were employed to improve the accuracy and speed of MRF acquisition. The method's accuracy for T1 and T2 measurements was validated in vitro, while its repeatability of T1 and T2 measurements was evaluated in vivo (n=3). The utility of the 3D MRF sequence for dynamic tracking of intracisternally infused Gd-DTPA in the whole mouse brain was demonstrated (n=5). Phantom studies confirmed accurate T1 and T2 measurements by 3D MRF with an undersampling factor up to 48. Dynamic contrast-enhanced (DCE) MRF scans achieved a spatial resolution of 192 x 192 x 500 um3 and a temporal resolution of 4.3 min, allowing for the analysis and comparison of dynamic changes in concentration and transport kinetics of intracisternally infused Gd-DTPA across brain regions. The sequence also enabled highly repeatable, high-resolution T1 and T2 mapping of the whole mouse brain (192 x 192 x 250 um3) in 30 min. We present the first dynamic and multi-parametric approach for quantitatively tracking contrast agent transport in the mouse brain using 3D MRF.

Abstract: BACKGROUND: Neurotrophic tyrosine receptor kinase (NTRK) gene fusions are rare oncogenic drivers prevalent in 0.3% of solid tumors. They are most common in salivary gland cancer (2.6%), thyroid cancer (1.6%), and soft-tissue sarcoma (1.5%). Currently, there are 2 US Food and Drug Administration-approved targeted therapies for NTRK gene fusions: larotrectinib, approved in 2018, and entrectinib, approved in 2019. To date, the real-world uptake of tyrosine receptor kinase inhibitor (TRKi) use for NTRK-positive solid tumors in academic cancer centers remains largely unknown. OBJECTIVE: To describe the demographics, clinical and genomic characteristics, and testing and treatment patterns of patients with NTRK-positive solid tumors treated at US academic cancer centers. METHODS: This was a retrospective chart review study conducted in academic cancer centers in the United States. All patients diagnosed with an NTRK fusion-positive (NTRK1, NTRK2, NTRK3) solid tumor (any stage) and who received cancer treatment at participating sites between January 1, 2012, and July 1, 2023, were included in this study. Patient demographics, clinical characteristics, genomic characteristics, NTRK testing data, and treatment patterns were collected from electronic medical records and analyzed using descriptive statistics as appropriate. RESULTS: In total, 6 centers contributed data for 55 patients with NTRK-positive tumors. The mean age was 49.3 (SD = 20.5) years, 51% patients were female, and the majority were White (78%). The median duration of time from cancer diagnosis to NTRK testing was 85 days (IQR = 44-978). At the time of NTRK testing, 64% of patients had stage IV disease, compared with 33% at cancer diagnosis. Prevalent cancer types in the overall cohort included head and neck (15%), thyroid (15%), brain (13%), lung (13%), and colorectal (11%). NTRK1 fusions were most common (45%), followed by NTRK3 (40%) and NTRK2 (15%). Across all lines of therapy, 51% of patients (n = 28) received a TRKi. Among TRKi-treated patients, 71% had stage IV disease at TRKi initiation. The median time from positive NTRK test to initiation of TRKi was 48 days (IQR = 9-207). TRKis were commonly given as first-line (30%) or second-line (48%) therapies. Median duration of therapy was 610 (IQR = 182-764) days for TRKi use and 207.5 (IQR = 42-539) days for all other first-line therapies. CONCLUSIONS: This study reports on contemporary real-world NTRK testing patterns and use of TRKis in solid tumors, including time between NTRK testing and initiation of TRKi therapy and duration of TRKi therapy.

Abstract: More than 10,000 photomultiplier tubes (PMTs) with a diameter of 80 mm will be installed in multi-PMT Digital Optical Modules (mDOMs) of the IceCube Upgrade. These have been tested and pre-calibrated at two sites. A throughput of more than 1000 PMTs per week with both sites was achieved with a modular design of the testing facilities and highly automated testing procedures. The testing facilities can easily be adapted to other PMTs, such that they can, e.g., be re-used for testing the PMTs for IceCube-Gen2. Single photoelectron response, high voltage dependence, time resolution, prepulse, late pulse, afterpulse probabilities, and dark rates were measured for each PMT. Abstract: We report on a measurement of astrophysical tau neutrinos with 9.7 years of IceCube data. Using convolutional neural networks trained on images derived from simulated events, seven candidate $\nu_\tau$ events were found with visible energies ranging from roughly 20 TeV to 1 PeV and a median expected parent $\nu_\tau$ energy of about 200 TeV. Considering backgrounds from astrophysical and atmospheric neutrinos, and muons from $\pi^\pm/K^\pm$ decays in atmospheric air showers, we obtain a total estimated background of about 0.5 events, dominated by non-$\nu_\tau$ astrophysical neutrinos. Thus, we rule out the absence of astrophysical $\nu_\tau$ at the $5\sigma$ level. The measured astrophysical $\nu_\tau$ flux is consistent with expectations based on previously published IceCube astrophysical neutrino flux measurements and neutrino oscillations., Comment: Accepted for publication in Physical Review Letters. Abstract: The IceCube Neutrino Observatory relies on an array of photomultiplier tubes to detect Cherenkov light produced by charged particles in the South Pole ice. IceCube data analyses depend on an in-depth characterization of the glacial ice, and on novel approaches in event reconstruction that utilize fast approximations of photoelectron yields. Here, a more accurate model is derived for event reconstruction that better captures our current knowledge of ice optical properties. When evaluated on a Monte Carlo simulation set, the median angular resolution for in-ice particle showers improves by over a factor of three compared to a reconstruction based on a simplified model of the ice. The most substantial improvement is obtained when including effects of birefringence due to the polycrystalline structure of the ice. Abstract: A measurement of the diffuse astrophysical neutrino spectrum is presented using IceCube data collected from 2011-2022 (10.3 years). We developed novel detection techniques to search for events with a contained vertex and exiting track induced by muon neutrinos undergoing a charged-current interaction. Searching for these starting track events allows us to not only more effectively reject atmospheric muons but also atmospheric neutrino backgrounds in the southern sky, opening a new window to the sub-100 TeV astrophysical neutrino sky. The event selection is constructed using a dynamic starting track veto and machine learning algorithms. We use this data to measure the astrophysical diffuse flux as a single power law flux (SPL) with a best-fit spectral index of $\gamma = 2.58 ^{+0.10}_{-0.09}$ and per-flavor normalization of $\phi^{\mathrm{Astro}}_{\mathrm{per-flavor}} = 1.68 ^{+0.19}_{-0.22} \times 10^{-18} \times \mathrm{GeV}^{-1} \mathrm{cm}^{-2} \mathrm{s}^{-1} \mathrm{sr}^{-1}$ (at 100 TeV). The sensitive energy range for this dataset is 3 - 550 TeV under the SPL assumption. Nos\'e's papers follow the former approach. Because the latter approach is not only constructive and simple, but also can be generalized to other forms of the equations of motion, we illustrate it here. Abstract: Background: Introduced in 2010, the sub-discipline of gerontologic biostatistics (GBS) was conceptualized to address the specific challenges in analyzing data from research studies involving older adults. However, the evolving technological landscape has catalyzed data science and statistical advancements since the original GBS publication, greatly expanding the scope of gerontologic research. There is a need to describe how these advancements enhance the analysis of multi-modal data and complex phenotypes that are hallmarks of gerontologic research. Methods: This paper introduces GBS 2.0, an updated and expanded set of analytical methods reflective of the practice of gerontologic biostatistics in contemporary and future research. Results: GBS 2.0 topics and relevant software resources include cutting-edge methods in experimental design; analytical techniques that include adaptations of machine learning, quantifying deep phenotypic measurements, high-dimensional -omics analysis; the integration of information from multiple studies, and strategies to foster reproducibility, replicability, and open science. Abstract: Name that Neutrino is a citizen science project where volunteers aid in classification of events for the IceCube Neutrino Observatory, an immense particle detector at the geographic South Pole. From March 2023 to September 2023, volunteers did classifications of videos produced from simulated data of both neutrino signal and background interactions. Name that Neutrino obtained more than 128,000 classifications by over 1,800 registered volunteers that were compared to results obtained by a deep neural network machine-learning algorithm. Possible improvements for both Name that Neutrino and the deep neural network are discussed.

24. 'One Person Cannot Change It; It's Going to Take a Community': Addressing Inequity through Community Environmental Education

Abstract: Environmental injustice is often an intersection of economic, social, and environmental disparities. Addressing the inequities borne by communities overburdened with such disparities requires local learning opportunities. Exploring how and what participants learn during community education projects can help inform and improve practice, which was the focus of this study. This study reports on a larger community environmental education project involving participatory action research, which involved community residents in Chicago learning to monitor local air quality using low-cost air sensors. The experiences of 14 volunteer air monitors were collected using focus group interviews and analyzed using Braun and Clarke's (2006) approach to thematic analysis. Participant learning focused on new and existing skills related to science/technology, interpersonal communication, and local environment. Volunteers built skills in using low-cost air sensors, taught other community members about air monitoring and local air quality, and devised strategies for improving community air quality and health. This exploration of the experiences of community residents learning to use low-cost air monitors has three applications to community education practice related to addressing inequity: utilizing community members as educators, developing community capacity to engage with science, and normalizing equitable processes. The study's findings mark a contribution by the field of adult and community education to both Critical Science Agency and low-cost air monitoring literature, in addition to the Education for Sustainability literature by addressing the lack of focus on sustainability and equity by highlighting a community-based PAR project focused on developing local capacity of marginalized communities to address air quality issues.

25. CONFINTEA VII and the Call to Climate Action: Clarifying Roles for Adult Learning and Education

Abstract: Climate change and climate education concern more than climate and the environment, as our social and economic systems both rely upon and are impacted by climate and environment. We cannot address the human impact on our global climate and local environments without several nations reconstructing their social and economic systems to be equitable and inclusive. According to several key global guidance documents related to adult learning and education (ALE) and climate education, adult educators have significant roles in developing learners capable of engaging in climate and environmental action. However, defining climate education and roles for adult educators is not well-defined or explicit. To gain clarity in how ALE providers can facilitate learners' understanding of how social/political, economic, and natural systems intersect to create and perpetuate the oppression, marginalization, and destruction of humans and the world upon which we depend, a close review of global guidance documents is warranted. This article analyzes key documents, such as the Marrakech Framework for Action (MFA), Education for Sustainable Development: A roadmap (ESD for 2030), and the International Council for Adult Education Spotlight Report for CONFINTEA VII. This process resulted in clarification of the definition of climate education from an ALE perspective and suggestions of explicit roles for ALE providers in the realm of climate education.

Abstract: Background: Intramedullary nail fixation for proximal humerus fractures has been shown to provide satisfactory results. The quality of reduction correlates with clinical outcomes, the rate of complications, avascular necrosis, and postoperative loss of fixation. The purpose of this study was to evaluate the clinical outcomes and complications of 2-part proximal humerus fractures compared to 3- or 4-part proximal humerus fractures. Methods: A single-center retrospective review was carried out of patients who underwent an intramedullary nail for a proximal humerus fracture by one of three surgeons between the years of 2009 and 2022, and who had a minimum of 12-months follow-up. Abstract: IntroductionHispanic/Latino populations are underrepresented in Alzheimer Disease (AD) genetic studies. Puerto Ricans (PR), a three-way admixed (European, African, and Amerindian) population is the second-largest Hispanic group in the continental US. We aimed to conduct a genome-wide association study (GWAS) and comprehensive analyses to identify novel AD susceptibility loci and characterize known AD genetic risk loci in the PR population.Materials and methodsOur study included Whole Genome Sequencing (WGS) and phenotype data from 648 PR individuals (345 AD, 303 cognitively unimpaired). We used a generalized linear-mixed model adjusting for sex, age, population substructure, and genetic relationship matrix. To infer local ancestry, we merged the dataset with the HGDP/1000G reference panel. Subsequently, we conducted univariate admixture mapping (AM) analysis.ResultsWe identified suggestive signals within the SLC38A1 and SCN8A genes on chromosome 12q13. This region overlaps with an area of linkage of AD in previous studies (12q13) in independent data sets further supporting. Univariate African AM analysis identified one suggestive ancestral block (p = 7.2×10−6) located in the same region. The ancestry-aware approach showed that this region has both European and African ancestral backgrounds and both contributing to the risk in this region. We also replicated 11 different known AD loci -including APOE- identified in mostly European studies, which is likely due to the high European background of the PR population.ConclusionPR GWAS and AM analysis identified a suggestive AD risk locus on chromosome 12, which includes the SLC38A1 and SCN8A genes. Our findings demonstrate the importance of designing GWAS and ancestry-aware approaches and including underrepresented populations in genetic studies of AD.

Abstract: Abstract Background Naloxone is an effective and safe opioid reversal medication now approved by the U.S. Food and Drug Administration (FDA) for use with or without a prescription. Despite this, naloxone dissemination lags at a time when U.S. opioid-related mortality expands. The authors proposed distributing naloxone to all U.S. medical students using established statewide standing prescription orders for naloxone, eliminating the financial burden of over-the-counter costs on students and streamlining workflow for the pharmacy. By focusing naloxone distribution on medical students, we are able to capitalize on a group that is already primed on healthcare intervention, while also working to combat stigma in the emerging physician workforce. Methods Beginning August 2022, the authors established a partnership between Harvard Medical School (HMS) and the outpatient pharmacy at Brigham and Women’s Hospital (BWH) to facilitate access to naloxone for HMS medical students. BWH developed a HIPAA-secure electronic form to collect individual prescription information. BWH pharmacists processed submissions daily, integrating the naloxone prescription requests into their workflow for in-person pick-up or mail-order delivery. The electronic form was disseminated to medical students through a required longitudinal addiction medicine curriculum, listserv messaging, and an extracurricular harm reduction workshop. Results Over the 2022–2023 academic year, 63 medical students obtained naloxone kits (two doses per kit) through this collaboration. Conclusions We propose that medical schools advocate for a hospital pharmacy-initiated workflow focused on convenience and accessibility to expand naloxone access to medical students as a strategy to strengthen the U.S. emergency response and prevention efforts aimed at reducing opioid-related morbidity and mortality. Expansion of our program to BWH internal medicine residents increased our distribution to over 110 healthcare workers, and efforts to expand the program to other BWH training programs and clinical sites such as the emergency department and outpatient infectious disease clinics are underway. With more than 90,000 medical students in the U.S., we believe that widespread implementation of targeted naloxone training and distribution to this population is an accessible approach to combating the public health crisis of opioid-related overdoses.

Abstract: Abstract Species distribution models (SDMs) are often built upon the “niche conservatism” assumption, such that they ignore the possibility of “evolutionary rescue” and may underestimate species' future range limits under climate change. We select aphids and ladybirds as model species and develop an eco‐evolutionary model to explore evolutionary rescue in a predator–prey system under climate change. We model the adaptive change of species' thermal performances, accounting for biotic interactions. Our study suggests that, without considering evolutionary adaptation, the warming climate will result in a reduction in aphid populations and the extinction of ladybirds in large parts of the United States. However, when incorporating evolutionary adaptation into the model, aphids can adapt to climate change, whereas ladybirds demonstrate geographic variation in their evolutionary rescue potential. Specifically, ladybirds in southern regions are more likely to be rescued than those in the north. In certain northern regions, ladybirds do not avoid extinction due to severe warming trends and seasonality of the climate. While higher warming trends do prompt stronger evolutionary changes in phenotype, they also lead to reduced aphid population abundance such that ecology constrains ladybird population growth. Higher seasonality induces an ecological effect by limiting the length of reproductive season, thereby reducing the capacity for evolutionary rescue. Together, these findings reveal the complex interplay between ecological and evolutionary dynamics in the context of evolutionary adaptation to climate change.

Abstract: This is a case report of a developmentally healthy 14-month-old female with partial thickness scald burns at multiple sites (the face, trunk, and bilateral upper extremities) that covered 14% of her total body surface area. Novel procedural pain therapy using intravenous lidocaine and oral ketamine was administered to perform daily wound care with minimal discomfort. Following this daily analgesic protocol there was sufficient analgesia and sedation with no adverse side effects. Currently, there are no studies where this analgesic combination was used during burn wound care and dressing changes in pediatric cases. The success of this protocol in a pediatric case highlights the safety and efficacy of oral ketamine when used in conjunction with intravenous lidocaine which potentially allows for a lower dose per procedure and a reduced risk of adverse effects.

Abstract: Abstract Background Uncovering the functional relevance underlying verbal declarative memory (VDM) genome-wide association study (GWAS) results may facilitate the development of interventions to reduce age-related memory decline and dementia. Methods We performed multi-omics and pathway enrichment analyses of paragraph (PAR-dr) and word list (WL-dr) delayed recall GWAS from 29,076 older non-demented individuals of European descent. We assessed the relationship between single-variant associations and expression quantitative trait loci (eQTLs) in 44 tissues and methylation quantitative trait loci (meQTLs) in the hippocampus. We determined the relationship between gene associations and transcript levels in 53 tissues, annotation as immune genes, and regulation by transcription factors (TFs) and microRNAs. To identify significant pathways, gene set enrichment was tested in each cohort and meta-analyzed across cohorts. Analyses of differential expression in brain tissues were conducted for pathway component genes. Results The single-variant associations of VDM showed significant linkage disequilibrium (LD) with eQTLs across all tissues and meQTLs within the hippocampus. Stronger WL-dr gene associations correlated with reduced expression in four brain tissues, including the hippocampus. More robust PAR-dr and/or WL-dr gene associations were intricately linked with immunity and were influenced by 31 TFs and 2 microRNAs. Six pathways, including type I diabetes, exhibited significant associations with both PAR-dr and WL-dr. These pathways included fifteen MHC genes intricately linked to VDM performance, showing diverse expression patterns based on cognitive status in brain tissues. Conclusions VDM genetic associations influence expression regulation via eQTLs and meQTLs. The involvement of TFs, microRNAs, MHC genes, and immune-related pathways contributes to VDM performance in older individuals.

Abstract: Abstract GATAD2B (GATA zinc finger domain containing 2B) variants are associated with the neurodevelopmental syndrome GAND, characterized by intellectual disability (ID), infantile hypotonia, apraxia of speech, epilepsy, macrocephaly and distinct facial features. GATAD2B encodes for a subunit of the Nucleosome Remodeling and Histone Deacetylase (NuRD) complex. NuRD controls transcriptional programs critical for proper neurodevelopment by coupling histone deacetylase with ATP-dependent chromatin remodeling activity. To study mechanisms of pathogenesis for GAND, we characterized a mouse model harboring an inactivating mutation in Gatad2b. Homozygous Gatad2b mutants die perinatally, while haploinsufficient Gatad2b mice exhibit behavioral abnormalities resembling the clinical features of GAND patients. We also observed abnormal cortical patterning, and cellular proportions and cell-specific alterations in the developmental transcriptome in these mice. scRNAseq of embryonic cortex indicated misexpression of genes key for corticogenesis and associated with neurodevelopmental syndromes such as Bcl11b, Nfia and H3f3b and Sox5. These data suggest a crucial role for Gatad2b in brain development.

Abstract: The IceCube Neutrino Observatory instruments about 1 km3 of deep, glacial ice at the geographic South Pole. It uses 5160 photomultipliers to detect Cherenkov light emitted by charged relativistic particles. An unexpected light propagation effect observed by the experiment is an anisotropic attenuation, which is aligned with the local flow direction of the ice. We examine birefringent light propagation through the polycrystalline ice microstructure as a possible explanation for this effect. The predictions of a first-principles model developed for this purpose, in particular curved light trajectories resulting from asymmetric diffusion, provide a qualitatively good match to the main features of the data. This in turn allows us to deduce ice crystal properties. Since the wavelength of the detected light is short compared to the crystal size, these crystal properties include not only the crystal orientation fabric, but also the average crystal size and shape, as a function of depth. By adding small empirical corrections to this first-principles model, a quantitatively accurate description of the optical properties of the IceCube glacial ice is obtained. In this paper, we present the experimental signature of ice optical anisotropy observed in IceCube light-emitting diode (LED) calibration data, the theory and parameterization of the birefringence effect, the fitting procedures of these parameterizations to experimental data, and the inferred crystal properties.
