The bacterial stringent response is a global regulatory process in which polyphosphate kinase (Ppk) and lon protease are important players. Previous studies have shown that overexpression of the lon gene and deletion of the ppk gene significantly increased actinorhodin production in Streptomyces coelicolor (SCO). In this study, a recombinant SCOΔppk-lon cell, expressing the extra lon gene in Δppk cells, was simulated using a modified in silico (computational) model, ecSco-GEM, and the negative effect of Ppk on actinorhodin production was confirmed. In addition, we identified key enzymes that play a positive role in actinorhodin production. Of these, NADH dehydrogenase/complex-I, beta-ketoacyl-[acyl-carrier-protein] synthase III, glycine cleavage system, and superoxide dismutase were identified as the most significant. By confirming these results with experiments, we have shown that GEMs can be a reliable starting point for in vitro (lab-based) studies of Streptomyces.., (© 2024. Polyhydroxyalkanoates (PHAs) are important and completely biodegradable alternatives to regular plastics, and they can be produced by activated sludge systems during wastewater treatment. Wastewaters with high organic content are being used for PHA production, which is an important resource recovery option. In this context, the effect of sludge retention time and different carbon sources, such as acetate, peptone-mixture and industrial wastewater (containing acetic acid (AA), lactic acid (LA) and propionic acid (PA)), on PHA storage was investigated. Oxygen utilisation rate (OUR) profiles were generated in respirometric tests and were evaluated by activated sludge modelling. Results showed that high storage (AA: 70%; LA: 49%; PA: 60% and industrial wastewater: 52%) was achievable in the feast phase even when the biomass was fed with a high organic acid content substrate to which it is not acclimated.

Background: Digital technologies have penetrated most workplaces. However, it is unclear how such digital technologies affect the physical health of older workers., Objective: This scoping review aims to examine and summarize the evidence from scientific literature concerning the impact of digital technology on the physical health of older workers., Methods: This scoping review will be conducted following recommendations outlined by Levac et al and will adhere to the PRISMA-ScR (Preferred Reporting Items for Systematic Reviews and Meta-Analysis extension for Scoping Reviews) guidelines for reporting. Peer-reviewed papers written in English will be searched in the following databases: MEDLINE, Cochrane, ProQuest, Web of Science, Scopus, APA PsycInfo, and ERIH PLUS. The web-based systematic review platform Covidence will be used to create a data extraction template. It will cover the following items: study and participant characteristics, health measures, digital tool characteristics and usage, and research findings. Following the Population, Concept, and Context (PCC) framework, our review will focus on studies involving older workers aged 50 years or older, any form of digital technology (including teleworking and the use of digital tools at work), and how digital technologies affect physical health (such as vision loss, musculoskeletal disorders, and migraines). Studies that focus only on mental health will be excluded. Study selection based on title and abstract screening (first stage), full-text review (second stage), and data extraction (third stage) will be performed by a group of researchers, whereby each paper will be reviewed by at least 2 people. Any conflict regarding the inclusion or exclusion of a study and the data extraction will be resolved by discussion between the researchers who evaluated the papers; a third researcher will be involved if consensus is not reached., Results: A preliminary search of MEDLINE, Epistemonikos, Cochrane, PROSPERO, and JBI Evidence Synthesis was conducted, and no current or ongoing systematic reviews or scoping reviews on the topic were identified. In this study, thiazole derivatives containing sulphonamide, amide, and phenyl amino groups were synthesized to protect the free amino groups of 5-methyl-4-phenyl-2-aminothiazole and 4-phenyl-2-aminothiazole. Halogenated reactions of N-protected thiazole derivatives have been investigated. LCMS, FT-IR, 1 H NMR, and 13 C NMR spectroscopy techniques were used to elucidate the structures of the synthesized compounds. Inhibition effects of the N-protected thiazole derivatives against human carbonic anhydrase I, II (hCA I, hCA II), and acetylcholinesterase (AChE) were investigated. The best results among the synthesized N-protected thiazole derivatives showed K i values in the range of 46.85-587.53 nM against hCA I, 35.01-578.06 nM against hCA II, and in the range of 19.58-226.18 nM against AChE. Biothiols widely exist in living organisms and have a crucial function of maintaining redox balance in the human body. It is vital yet difficult to develop probes that can simultaneously detect and distinguish biothiols. In this study, a highly sensitive dual-modality nanosensor, NBD-Nap@NCC, was developed for the discrimination and determination of biothiols in real samples, and its practical application was elucidated based on RGB analysis using a smartphone. The sensitive nanosensor was successfully prepared through the surface modification of nanocrystalline cellulose (NCC), combining NBD and naphthalene fluorophores. Owing to the high electron-withdrawing behavior of the NBD group, which led to a PET mechanism between the fluorophores, the prepared NBD-Nap@NCC nanosensor had a very weak fluorescence response. However, after treatment with Hcy or Cys, NBD-Nap@NCC quickly provided remarkable and different rates of fluorescence "turn-on" responses in both blue and green channels, which was attributed to naphthalene and NBD fluorophores as a result of the inhibition of the PET mechanism. However, after treatment with GSH, only a significant blue-channel emission, which was attributed to the naphthalene fluorophore was obtained, indicating the inhibition of the PET mechanism. Furthermore, the NCC platform demonstrated improved sensitivity and selectivity because of the increased surface area and higher number of binding sites due to modification of the NBD group on the surface. The detection limit ranged from 0.910 to 1.150 μmol L -1 for biothiols with a large dynamic response range. The accuracy of the sensor in determining the concentrations of Hcy, Cys, and GSH in real samples was evaluated via HPLC and spike/recovery analysis. Additionally, paper-based analysis kits were fabricated for the practical detection of biothiols based on RGB changes using a smartphone application.

Drug molecules exist as complexed with serum proteins such as human serum albumin (HSA) and/or unbound free form in the blood circulation. Drugs can be effective only when they are free. Thus, it is important to understand aspects that are important for interaction between drugs and interacting proteins. In this study, interactions among 2990 FDA approved drugs and HSA were computational analyzed to unravel principles that are critical for drug-HSA interactions. Docking results showed that drugs have higher affinity toward cavity-1 (C1) than cavity-2 (C2). A total of 1131 drug molecules have docking score greater than 60 while 768 molecules have docking score greater than 60 when they are docked in C2. In addition, three solvent channels have potential to direct solvent to C1 cavity while C2 does not have any effective channel. The post MD analyses demonstrated that drugs are making polar interactions with basic amino acids in the binding cavities. Verbscoside and ceftazidime both have stable low RMSD values throughout MD simulation with 2 Å on average in C1 cavity. The ligand RMSD shows less stability for verbscoside, which is around 4 Å when it is in complex with HSA in C1. The individual contribution of the residues K192, K196, R215, and R254 to ceftazidime are -1.92 ± 0.18, -3.09 ± 0.09, -2.17 ± 0.17, and - 2.32 ± 0.098, respectively. These residues contribute the binding energy of the verbscoside by -6.06 ± 0.08, -2.10 ± 0.06, and - 1.57 ± 0.03 kcal/mol individually in C1 cavity. C2 is making polar interactions with drug via R469, K472, and K488 residues and their contribution to the two drugs are -3.13 ± 0.21 kcal/mol for R469, -1.94 ± 0.18 kcal/mol for K472, and -1.96 ± 0.11 kcal/mol for K488 to total binding energy of ceftazidime. The binding energy of verbscoside is 57.17 ± 7.00 kcal/mol and Arg-407 has the highest contribution this bind energy individually with -4.29 ± 0.12 kcal/mol. Drugs with hydrogen bond donor/acceptor chemical adducts such as verbscoside involve higher hydrogen bond formation in C1 pocket. Neurodegenerative diseases are characterized by progressive deterioration of the nervous system. Alzheimer's disease (AD), Parkinson's disease (PD), amyotrophic lateral sclerosis (ALS), and Huntington's disease (HD) are prominently life-threatening examples of neurodegenerative diseases. The complexity of the pathophysiology in neurodegenerative diseases causes difficulties in diagnosing. Although the drugs temporarily help to correct specific symptoms including memory loss and degeneration, a complete treatment has not been found yet. New therapeutic approaches have been developed to understand and treat the underlying pathogenesis of neurodegenerative diseases. With this purpose, clustered-regularly interspaced short palindromic repeats/CRISPR-associated protein (CRISPR/Cas) technology has recently suggested a new treatment option. Editing of the genome is carried out by insertion and deletion processes on DNA. Safe delivery of the CRISPR/Cas system to the targeted cells without affecting surrounding cells is frequently investigated. Extracellular vesicles (EVs), that is exosomes, have recently been used in CRISPR/Cas studies. In this review, CRISPR/Cas and EV approaches used for diagnosis and/or treatment in AD, PD, ALS, and HD are reviewed. In this study, new peripherally substituted symmetric zinc and magnesium phthalocyanines (4 and 5) were successfully prepared by cyclotetramerization of the tetrahydropyrimidone (THPM)-linked phthalonitrile 3. The identity of the compounds were confirmed primarily through spectroscopic analysis including NMR, FT-IR, UV-Vis and MALDI-TOF mass spectroscopy. The photophysical and photochemical properties of the synthesized phthalocyanines (Pcs) were examined using UV-Vis absorption and fluorescence emission spectroscopy techniques. The quantum yields of singlet oxygen were found to be 0.50 and 0.33 for compounds 4 and 5 in DMSO, respectively. In addition to photo-physicochemical properties, the enhanced biological activities of compounds 4 and 5 were investigated using a range of biological assays, namely, antibiofilm, microbial cell viability, antioxidant, DNA cleavage, antimicrobial and photodynamic antimicrobial assays. The maximum DPPH inhibition of 4 and 5 was detected as 40.46% and 25.76% at 100 mg L -1 , respectively. Fragmentation of the DNA molecule was observed at concentrations of 25 mg L -1 , 50 mg L -1 and 100 mg L -1 for 4 and 5. Additionally, effective inhibition of microbial cell viability was observed with the targeted Pcs. The antibiofilm properties of these compounds were found to be concentration-dependent. The biofilm inhibition activities of 4 and 5 were found to be 96.01% and 92.04% for S. aureus , while they were 95.42% and 91.27%, for P. aeruginosa , respectively. The antimicrobial activities of 4 and 5 on different microorganisms were evaluated using the microdilution assay. In the case of photodynamic antimicrobial treatment, the newly synthesized Pcs showed more effective antimicrobial inhibition compared to the control. These findings suggest that compounds 4 and 5 can be used as promising photodynamic antimicrobial agents for the treatment of many diseases, particularly infectious diseases.

The neuronal etiology of obesity is centered around a diet-induced inflammatory state in the arcuate nucleus of the hypothalamus, which impairs the functionality of pro-opiomelanocortine neurons (POMCs) responsible for whole-body energy homeostasis and feeding behavior. Intriguingly, systemic salt inducible kinase 2 (SIK2) knockout mice demonstrated reduced food intake and energy expenditure along with modestly dysregulated metabolic parameters, suggesting a causal link between the absence of SIK2 activity in POMCs and the observed phenotype. To test this hypothesis, we conducted a comparative secretomics study from POMC neurons following pharmacologically induced endoplasmic reticulum (ER) stress induction, a hallmark of metabolic inflammation and POMC dysregulation in diet-induced obese (DIO) mice. Our data provide significant in vitro evidence for the POMC-specific SIK2 activity in controlling energy metabolism and feeding in DIO mice by regulating the nature of the related POMC secretome. Our data also suggest that under physiological stress conditions, SIK2 may act as a gatekeeper for the secreted inflammatory factors and signaling molecules critical for cellular survival and energy homeostasis. Upadacitinib (UPA) is a selective and reversible oral Janus kinase (JAK) 1 inhibitor and is of great importance in treating inflammatory bowel disease (Zheng et al., Int Immunopharmacol 126:111229, 2024; Foy et al., JAAD Case Rep 42:20-22, 2023). Although there are limitations to the effectiveness of UPA, it has received positive responses in clinical trials and is approved for the treatment of atopy dermatitis (AD) (Li et al., Int Immunopharmacol 125:111193, 2023). In this study, a nanoparticle-doped molecularly imprinted polymer (MIP)-based electrochemical sensor was developed for sensitive and selective detection of UPA. The developed sensor was designed as a thin film layer using the photopolymerization method on the surface of the prepared nanoparticle-doped polymerization solution glassy carbon electrode (GCE). Various nanoparticles, such as multi-walled carbon nanotube, titanium dioxide, oxide, and zinc oxide (ZnO) nanoparticles, were the most suitable for UPA. Surface characterization of the developed sensor was done by scanning electron microscopy (SEM), and electrochemical characterization was done by cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The quantitative analysis of UPA was performed in 5.0 mM [Fe (CN) 6 ] 3-/4- solution using the differential pulse voltammetry (DPV) technique. Under optimum conditions, the calibration range was between 0.1 and 1 pM. The limit of detection (LOD) and limit of quantification (LOQ) were calculated as 0.005 pM and 0.017 pM, respectively. The sensor's accuracy was proven by performing a recovery study in serum. The sensor's selectivity was also evaluated using common interfering substances such as KNO 3 , CaCl 2 , Na 2 SO 4 , uric acid, ascorbic acid, dopamine, and paracetamol. According to the results obtained, the performance of the designed sensor was found to be quite sensitive and selective in determining UPA. The developed UPA-ZnO/3-APBA@MIP-GCE sensor showed high sensitivity and selectivity towards UPA. In addition, the selectivity, the most important feature of the MIP-based sensor, was confirmed by imprinting factor (IF) calculations using tofacitinib (TOF) and ruxolitinib (RUX). In this study, the performance of a novel organic tea compost developed for the first time in the world from raw tea waste from tea processing factories and enriched with worms, beneficial microorganisms, and enzymes was tested in comparison to chemical fertilizers in tea plantations in Rize and Artvin provinces, where the most intensive tea cultivation is carried out in Turkey. In the field trials, the developed organic tea vermicompost was incorporated into the root zones of the plants in the tea plantations in amounts of 1000 (OVT1), 2000 (OVT2) and 4000 (OVT4) (kg ha -1 ). The experimental design included a control group without OVT applications and positive controls with chemical fertilizers (N: P: K 25:5:10, (CF) 1200 kg ha -1 ) commonly used by local growers. The evaluation included field trials over two years. The average yields obtained in two-year field trials in five different areas were: Control (6326), OVT1 (7082), OVT2 (7408), OVT4 (7910), and CF (8028) kg ha -1 . Notably, there was no significant statistical difference in yields between the organic (at 4000 kg ha -1 ) and chemical fertilizers (at 1200 kg ha -1 ). The highest nutrient contents were obtained when CF and OVT4 were applied. According to the average values across all regions, the application of OVT4 increased the uptake of 63% N, 18% K, 75% P, 21% Mg, 19% Na, 29% Ca, 28% Zn, 11% Cu and 24% Mn compared to the control group. The application of chemical fertilizers increased the uptake of 75% N, 21% K, 75% P, 21% Mg, 28% Na, 27% Ca, 30% Zn, 18% Cu and 31% Mn compared to the control group. The organic fertilizer treatment had the lowest levels of antioxidants compared to the control groups and the chemical fertilizers. It was also found that the organic fertilizer increased the levels of amino acids, organic acids and chlorophyll in the tea plant. Its low antioxidant activity and proline content prepared them for or protected them from stress conditions. This study hypothesizes that advanced machine learning (ML) models can more accurately predict certain critical water quality parameters in marine environments compared to conventional regression techniques. We specifically evaluated the spatio-temporal distribution of Chlorophyll-a (Chl-a) and Secchi Disk Depth (SDD) in the Gulf of Izmit using in-situ measurements and Sentinel-2 satellite imagery from October 2021 and 2022. Among the models tested, the Support Vector Regression (SVR) model showed better predictive performance, achieving the lowest RMSE for SDD (1.11-1.70 m) and Chl-a (1.16-4.97 mg/m 3 ) and the lowest MAE for SDD (0.86-1.43 m) and Chl-a (1.03-3.17 mg/m 3 ). Additionally, the study observed a shift from hypertrophic to eutrophic Chl-a conditions and from mesotrophic-eutrophic to oligotrophic SDD conditions between 2021 and 2022, aligning with SVR model predictions and in-situ observations. ETS domain transcription factor superfamily is highly conserved throughout metazoa and is involved in many aspects of development and tissue morphogenesis, and as such, the deregulation of ETS proteins is quite common in many diseases, including cancer. The PEA3 subfamily in particular has been extensively studied with respect to tumorigenesis and metastasis; however, they are also involved in the development of many tissues with branching morphogenesis, such as lung or kidney development. In this review, we aim to summarize findings from various studies on the role of Pea3 subfamily members in nervous system development in the embryo, as well as their functions in the adult neurons. We further discuss the different signals that were shown to regulate the function of the Pea3 family and indicate how this signal-dependent regulation of Pea3 proteins can generate neuronal circuit specificity through unique gene regulation. Finally, we discuss how these developmental roles of Pea3 proteins relate to their role in tumorigenesis., (© 2024. A wide range of cells respond to fibroblast growth factor 2 (FGF2) by proliferation via activation of the Ras/ERK1/2 pathway. In this study, the potential involvement of salt inducible kinase SIK2) in this cascade within retinal Müller glia is explored. It is found that SIK2 phosphorylation status and activity are modulated in an FGF2-dependent manner, possibly via ERK1/2. With SIK2 downregulation, enhanced ERK1/2 activation with delayed attenuation and increased cell proliferation is observed, while SIK2 overexpression hampers FGF2-dependent ERK1/2 activation. In vitro kinase and site-directed mutagenesis studies indicate that SIK2 targets the pathway element GRB2-associated-binding protein 1 (Gab1) on Ser266. This phosphorylation event weakens Gab1 interactions with its partners growth factor receptor-bound protein 2 (Grb2) and Src homology region 2 domain containing phosphatase 2 (Shp2). The determination of urea concentration is essential for human health owing to its crucial role in the ability to metabolize nitrogen-containing substances. This study developed new electrochemical enzymatic detection systems via the synergistic effect of the superior features of novel electropolymerizable pyranine-aniline (PA, 4), polyaniline (PANI) compounds, graphene quantum dots (GQDs) and zeolitic imidazolate framework-8 (ZIF8). The novel compound 4 was characterized via 1 H-NMR, 13 C-NMR, FTIR, and MALDI-TOF mass spectroscopies. Furthermore, Cu-GQD@ZIF8 hybrid materials containing GQD and integrated electroactive Cu metal were prepared in this study. The surface morphology of the prepared Cu-GQD@ZIF8 hybrid material was investigated through microscopic methods such as SEM and TEM, and chemical characterizations were performed using FTIR, XPS, XRD, and TGA analyses. After the characterization of the novel materials, the urease (Urs) enzyme was bound to the new modified electrode surface. Next, the enzymatic biosensor properties of the Urs/Cu-GQD@ZIF8/PANI/PA/GCE sensor electrode for urea detection via reduction of PANI were investigated by DPV and CV techniques. The LOD and LOQ values of the presented sensor were calculated to be 0.77 μM and 2.31 μM, respectively, in the linear range of 1.0-80.0 μM, based on DPV measurements. The presented biosensor system determined the amount of urea in an artificial serum sample, and its accuracy was confirmed via the recovery test and GC-MS analysis.

Herein, we report polyphosphonate covalent organic frameworks (COFs) constructed via P-O-P linkages. The materials are synthesized via a single-step condensation reaction of the charge-assisted hydrogen-bonded organic framework, which is constructed from phenylphosphonic acid and 5,10,15,20-tetrakis[p-phenylphosphonic acid]porphyrin and is formed by simply heating its hydrogen-bonded precursor without using chemical reagents. Above 210 °C, it becomes an amorphous microporous polymeric structure due to the oligomerization of P-O-P bonds, which could be shown by constant-time solid-state double-quantum 31 P nuclear magnetic resonance experiments. The polyphosphonate COF exhibits good water and water vapor stability during the gas sorption measurements, and electrochemical stability in 0.5 M Na 2 SO 4 electrolyte in water. The reported family of COFs fills a significant gap in the literature by providing stable microporous COFs suitable for use in water and electrolytes. New solvated Mo(VI) complexes were isolated from the reaction of [MoO 2 (acac) 2 ] with asymmetric isatin bisthiocarbohydrazone ligands. The ligands were obtained from the reaction of isatin monothiocarbohydrazone with 3,5-dibromo salicylaldehyde (L1), 3,5-dichloro salicylaldehyde (L2) and 3-chloro-5-bromo salicylaldehyde (L3), respectively. In the complexes, the ligands serve as ONS donors and coordinate to the [MoO 2 ] 2+ nucleus. The bonding sites are azomethine nitrogen atom, phenolic oxygen atom and thiol sulfur atom. The sixth coordination site is completed by an oxygen atom from an ethanol solvent. The ethanol-coordinated Mo(VI) complexes, C1-C3, [MoO 2 L(EtOH)] (L: L1-L3), were characterized using elemental analysis, IR and 1 H NMR spectroscopies, and conductivity measurements. By crystallizing ethanol-solvated solid complexes from an EtOH/DMSO mixture, DMSO-solvated complexes (C4-C6) suitable for X-ray crystallography were obtained. Crystal structure analysis supports the proposed complex structures and geometries, but the ethanol in the sixth coordination site has been replaced by DMSO. When the anticarcinogenic effects of the ligands and complexes (C1-C3) on the C6 cell line were examined, it was found that the complexes showed higher activity than the ligands. The C3 complex appears to have the best anti-cancer activity compared to doxorubicin. Additionally, all compounds were determined to have high total antioxidant capacity. In this study, spatiotemporal analysis of forest fires in Turkiye was undertaken, with a specific focus on the large-scale atmospheric systems responsible for causing these fires. For this purpose, long-term variations in forest fires were classified based on the occurrence types (i.e. natural/lightning, negligence/inattention, arson, accident, unknown). The role of large-scale atmospheric circulations causing natural originated forest fires was investigated using NCEP/NCAR Reanalysis sea level pressure, and surface wind products for the selected episodes. According to the main results, Mediterranean (MeR), Aegean (AR), and Marmara (MR) regions of Turkiye are highly susceptible to forest fires. Statistically significant number of forest fires in the MeR and MR regions are associated with global warming trend of the Eastern Mediterranean Basin. In monthly distribution, forest fires frequently occur in the MeR part of Turkiye during September, August, and June months, respectively, and heat waves are responsible for forest fires in 2021. As a consequence of the extending summer Asiatic monsoon to the inner parts of Turkiye and the location of Azores surface high over Balkan Peninsula result in atmospheric blocking and associated calm weather conditions in the MeR (e.g. Mugla and Antalya provinces). When this blocking continues for a long time, southerly winds on the back slopes of the Taurus Mountains create a foehn effect, calm weather conditions and lack of moisture in the soil of Antalya and Mugla settlements trigger the formation of forest fires., (© 2024. Worldwide, fermented foods (FF) are recognized as healthy and safe. Despite the rapid increase of research papers, there is a lack of systematic evaluation of the health benefits and risks of FF. The COST Action CA20128 "Promoting innovation of fermented foods" (PIMENTO) aims to provide a comprehensive assessment on the available evidence by compiling a set of 16 reviews. Seven reviews will cover clinical and biological endpoints associated with major health indicators across several organ systems, including the cardiovascular, gastrointestinal, neurological, immune, and skeletal systems. Nine reviews will address broader biological questions associated with FF including bioactive compounds and vitamin production, nutrient bioavailability and bioaccessibility, the role of FF in healthy diets and personalized nutrition, food safety, regulatory practices, and finally, the health properties of novel and ethnic FF. For each outcome assessed in the reviews, an innovative approach will be adopted based on EFSA's published guidance for health claim submissions. In particular, each review will be composed of three parts: (1) a systematic review of available human studies; (2) a non-systematic review of the mechanism of action related to the clinical endpoints measured by the human studies identified in part 1; and (3) a non-systematic review of the characterization of the FF investigated in the human studies identified in part 1. Electrochemical sensors have a broad range of industrial applications due to their sensitivity, speed, and cost-effectiveness. These sensors enable the continuous monitoring and control of critical parameters in various industrial processes. For instance, they are essential in food safety, environmental monitoring, biomedical applications, and pharmaceutical production. In the food industry, electrochemical sensors facilitate the rapid and reliable detection of contaminants and pathogens in food products, thus enhancing product quality and consumer safety. An electrochemical sensor was developed with the molecularly imprinted polymer (MIP) technique to detect deltamethrin with high sensitivity and selectivity. The sensor was fabricated by electrodeposition of Co 3 O 4 on indium tin oxide (ITO), followed by electropolymerization of o-phenylenediamine with deltamethrin as a template molecule. The template molecules were then removed from the modified electrode by a methanol. The MIP-based electrochemical sensor exhibited high sensitivity and selectivity towards deltamethrin. Under the optimized conditions, the LOD values for the MIP/Co 3 O 4 /ITO electrode in the first and second linear regressions were 1.53 nM for linear range of 2.82 nM to 56.5 nM and 0.34 μM for linear range of 0.25 μM to 3.98 μM. Moreover, the LOD values for the NIP/Co 3 O 4 /ITO electrode in the first and second regressions were 2.43 nM for the linear range of 3.91 nM to 65.0 nM and 726.0 nM for the linear range of 0.023 μM to 4.5 μM. The developed electrochromic pesticide sensor, being an electrochemical-based molecularly imprinted polymer (MIP) sensor incorporating electrochromic materials, enables both target-specific pesticide detection and visual pesticide identification based on color changes dependent on pesticide concentration. Consequently, this system is more advantageous compared to electrochemical-based MIP sensors, as it provides both qualitative and quantitative determinations. The aim of this study was to determine the antiviral activity of cannabidiol (CBD) against SARS-CoV-2 infection. CBD is the second most studied cannabinoid obtained from Cannabis plants. We investigated the potential use of CBD, which has so far proven to have a positive effect on different diseases, in the SARS-CoV-2 infection. To test this, in vivo studies were carried out using K18-hACE2 transgenic mice. To reveal the potential therapeutic effect of the CBD at the histopathological and molecular level challenge experiments were performed. The study was designed with two groups (n = 10) and in the treatment group animals were infected with SARS-CoV-2 virus strain B.1.1.7 alpha before the administration of CBD. While the disease progressed and resulted in death in the control group that was infected by the virus alone, it was observed that the infection slowed down and the survival rate increased in the mice treated with CBD along with the virus. In this study, we investigate the intricate regulatory mechanisms underlying the circadian clock in Drosophila, focusing on the light-induced conformational changes in the cryptochrome (DmCry). Upon light exposure, DmCry undergoes conformational changes that prompt its binding to Timeless and Jetlag proteins, initiating a cascade crucial for the starting of a new circadian cycle. DmCry is subsequently degraded, contributing to the desensitization of the resetting mechanism. The transient and short-lived nature of DmCry protein-protein interactions (PPIs), leading to DmCry degradation within an hour of light exposure, presents a challenge for comprehensive exploration. To address this, we employed proximity-dependent biotinylation techniques, combining engineered BioID (TurboID) and APEX (APEX2) enzymes with mass spectrometry. This approach enabled the identification of the in vitro DmCry interactome in Drosophila S2 cells, uncovering several novel PPIs associated with DmCry. Validation of these interactions through a novel co-immunoprecipitation technique enhances the reliability of our findings. Importantly, our study suggests the potential of this method to reveal additional circadian clock- or magnetic field-dependent PPIs involving DmCry. Both clinical diagnosis and neuropathological diagnosis are commonly used in literature to categorize individuals as Alzheimer's disease (AD) or non-AD in omics analyses. Whether these diagnostic strategies result in distinct profiles of molecular abnormalities is poorly understood. Here, we analysed one of the most commonly used AD omics datasets in the literature from the Religious Orders Study and Memory and Aging Project (ROSMAP) cohort and compared the two diagnosis strategies using brain transcriptome and metabolome by grouping individuals as non-AD and AD according to clinical or neuropathological diagnosis separately. Differentially expressed genes, associated pathways related with AD hallmarks and AD-related genes showed that the categorization based on neuropathological diagnosis more accurately reflects the disease state at the molecular level than the categorization based on clinical diagnosis. We further identified consensus biomarker candidates between the two diagnosis strategies such as 5-hydroxylysine, sphingomyelin and 1-myristoyl-2-palmitoyl-GPC as metabolite biomarkers and sphingolipid metabolism as a pathway biomarker, which could be robust AD biomarkers since they are independent of diagnosis strategies. We also used consensus AD and consensus non-AD individuals between the two diagnostic strategies to train a machine-learning based model, which we used to classify the individuals who were cognitively normal but diagnosed as AD based on neuropathological diagnosis (asymptomatic AD individuals). The majority of these individuals were classified as consensus AD patients for both omics data types. ICSI is one of the most commonly used techniques to treat infertility. The sperm selection for the procedure is done 'randomly' by the embryologist according to the motility and morphology parameters which is known not to reflect the potential of a sperm for fertilization, pregnancy and a healthy childbearing. Since the apoptosis rate is higher in sperm cells of infertile patients, it is more likely to choose an apoptotic sperm by the 'random selection method'. We recently introduced a novel sperm selection technique namely 'Annexin-V coated polystrene bead technique'(APB-Tech), for the selection of non-apoptotic sperm cells. The principal of the technique is based on the binding affinity of an apoptotic sperm to 'Annexin-V covered beads' enabling to distinguish a viable and a healthy sperm by light microscopy. The aim of the present study was to observe the effects of this technique on ICSI outcomes in mice. Sibling-oocyte trial was conducted and the outcome measures were compared with the results of traditional sperm selection method. Embryo and blastocyst qualities and blastocyst development rates were significantly increased in APB-Tech group, while the other parameters were not affected. This study was designed to address the question: does antioxidant-containing embryo culture media affect DNA methyltransferases, global DNA methylation, inner cell mass/trophoblast differentiation, intracellular reactive oxygen species (ROS) levels, and apoptosis? Mouse zygotes were cultured in embryo culture media containing MitoQ, N-acetyl-L-cysteine (NAC), acetyl-L-carnitine (ALC), α-lipoic acid (ALA), or the mixture of NAC + ALC + ALA (AO) until the blastocyst stage, whereas in vivo-developed blastocysts were used as control. Protein expression levels of Dnmt1, 3a, 3b, and 3l enzymes were analyzed by immunofluorescence and western blot, while global DNA methylation, apoptosis, and ROS levels were evaluated by immunofluorescence. NAC, ALC, and MitoQ significantly increased the levels of all Dnmts and global methylation. ALA significantly induced all Dnmts, whereas global methylation did not show any difference. NAC and mixture AO applications significantly induced Nanog levels, ALA and MitoQ increased Cdx2 levels, while the other groups were similar. ALA and MitoQ decreased while ALC increased the levels of intracellular ROS. This study illustrates that antioxidants, operating through distinct pathways, have varying impacts on DNA methylation levels and cell differentiation in mouse embryos. Background: This study investigates the discourse on anxiety management strategies within the TikTok platform, analyzing a substantial dataset of 45,639 comments collected over a year., Aims: The primary aim is to explore the various strategies users employ to manage anxiety, focusing on how these strategies are discussed and shared on TikTok., Methods: A mixed-method approach was utilized, combining Latent Dirichlet Allocation (LDA) for topic modeling with qualitative analysis. This methodology allowed for the identification of nine distinct topics, which were further grouped into three main categories: Self-Empowerment and Coping Strategies, Community Support and Social Connectivity, and Recognizing and Navigating Triggers., Results: The analysis revealed a diverse range of strategies users employ to manage anxiety, spanning personal coping mechanisms, social support networks, and the recognition and mitigation of triggers. These findings underscore TikTok's role as a dynamic space for sharing, exploring, and validating experiences related to anxiety management., Conclusions: TikTok offers unique opportunities for identity construction and community support amidst the challenges of anxiety. However, the study acknowledges limitations, such as potential biases in keyword-based data collection and the complexity of capturing multimodal discourse on the platform. The conclusion emphasizes the need for further refinement of digital mental health platforms, calling for sophisticated algorithmic solutions to enhance user support and content relevance.

This paper discusses service times based on vehicle class at barrier-operated toll booths. Service times were measured using four vehicle classifications: car (C), medium goods vehicle (MGV), trucks & bus (TB), and articulated truck (AT) with two payment options: electronic (ETC) and manual toll collection (MTC). Each toll booth has both payment systems and resulting in mixed toll booth utilization in terms of payment and vehicles. The collected data was utilized to estimate logistic regression, and non-parametric statistical tests were performed. Findings indicate, when vehicle size increases, so does service time for even ETC. Furthermore, logistic regression used to compute threshold service times shifting from ETC to MTC for C, MGV, TB, and AT, which were 5.92, 7.51, 10.00, and 12.80 s, respectively. The transition time for a vehicle switch from ETC to MTC based on the logistic curve is denoted by threshold service time. Below the threshold, ETC is more favorable option and affected by barrier and vehicle class. Overall, the results indicate that operators can benefit from more dynamic operational conditions facilitated by threshold service times, enabling adaptive and efficient toll booth operations. A panorama ensures a stunning wide-angle field of view up to 360° representation of a scene, exceeding the limits of a normal photograph. Panoramic cameras satisfy the single-viewpoint characteristic. There are several types of panoramic cameras for 360-degree imaging. Multi-camera panoramic imaging systems pose a difficulty in obtaining a single projection center for the cameras. In a variety of practical implementations of panoramic cameras, it is possible to calculate three-dimensional coordinates from a panoramic image, especially using the Direct Linear Transformation (DLT) method. In this study, not only a defining method of the non-uniform image coordinate system is presented by utilizing the C-Means algorithm for a single panoramic image, captured with a Ladybug2 panoramic camera in a panoramic calibration room but also the use of an elliptical panoramic projection coordinate system is defined by the Singular Value Decomposition method in a panoramic view. The results of the suggested method have been compared with the DLT algorithm for a single panoramic image which defined a conventional photogrammetric image coordinate system. Coiled-coil domain-containing 124 protein is a multifunctional RNA-binding factor, and it was previously reported to interact with various biomolecular complexes localized at diverse subcellular locations, such as the ribosome, centrosome, midbody, and nucleoli. We aimed to better characterize the subcellular CCDC124 translocation by labelling this protein with a fluorescent tag, followed by laser scanning confocal microscopy methods. As traditional GFP-tagging of small proteins such as CCDC124 often faces limitations like potential structural perturbations of labeled proteins, and interference of the fluorescent-tag with their endogenous cellular functions, we aimed to label CCDC124 with the smallest possible split-GFP associated protein-tagging system (GFP11/GFP1-10) for better characterization of its subcellular localizations and its translocation dynamics. By recombinant DNA techniques we generated CCDC124-constructs labelled with either single of four tandem copies of GFP11 (GFP11 ×  1 ::CCDC124, GFP11 ×  4 ::CCDC124, or CCDC124::GFP11 ×  4 ). We then cotransfected U2OS cells with these split-GFP constructs (GFP11 ×  1(or X4) ::CCDC124/GFP1-10) and analyzed subcellular localization of CCDC124 protein by laser scanning confocal microscopy. Tagging CCDC124 with four tandem copies of a 16-amino acid short GFP-derived peptide-tag (GFP11  × 4 ::CCDC124) allowed better characterization of the subcellular localization of CCDC124 protein in our model human bone osteosarcoma (U2OS) cells. Thus, by this novel methodology we successfully identified GFP11 ×  4 ::CCDC124 molecules in G3BP1-overexpression induced stress-granules by live cell protein imaging for the first time. Indole-based agents are frequently used in targeted or supportive therapy of several cancers. In this study, we investigated the anticancer properties of originally synthesized novel indolin-2-one derivatives (6a-d) against Malignant Mesothelioma, Breast cancer, and Colon Cancer cells. Our results revealed that all derivatives were effectively delayed cell proliferation by inhibiting the ERK1/2, AKT, and STAT3 signaling pathways in a concentration-dependent manner. Additionally, these variants induced cell cycle arrest in the S phase, accompanied by elevated levels of p21 and p27 expressions. Derivatives also initiated mitochondrial apoptosis through the upregulation of Bax and downregulation of Bcl-2 proteins, leading to the activation of caspase 3 and PARP cleavage in exposed cells. Remarkably, three of the indolin-2-one derivatives displayed significant selectivity towards Breast and Colon Cancer cells, with compound 6d promising as the most potent and wide spectral one for all cancer cell lines., (© 2024 The Author(s). In the current investigation, total phenolics and flavonoids of the methanolic extract obtained from the trunk bark of Acacia cyanophylla Lindl. were quantified by LC-HRMS technique. DPPH and ABTS reagents were employed to assay the antioxidant potential. The anti-tyrosinase and anti-α-amylase potentials were also assayed. The findings revealed that thirteen polyphenolic compounds were detected in the methanolic extract with trans-taxifolin (23.2 g/kg), as the major constituent. A. cyanophylla extract displayed a higher activity with DPPH test (IC 50 =10.14±1.00 μg/mL) than with ABTS (IC 50 =15.27±2.09 μg/mL). The same extract also exhibited interesting α-amylase inhibitory action (IC 50 value of 4.00±0.17 μg/mL). Moreover, methanolic trunk bark extract exerted strong anti-tyrosinase capacity with an IC 50 of 5.12±0.41 μg/mL in comparison to kojic acid (IC50=10.22±0.85 μg/mL) used as positive control. Background: Evaluating the potential of using both synthetic and biological products as targeting agents for the diagnosis, imaging, and treatment of infections due to particularly antibiotic-resistant pathogens is important for controlling infections. This study examined the interaction between Gp45, a receptor-binding protein of the ϕ11 lysogenic phage, and its host Staphylococcus aureus (S. aureus), a common cause of nosocomial infections., Methods: Using molecular dynamics and docking simulations, this study identified the peptides that bind to S. aureus wall teichoic acids via Gp45. It compared the binding affinity of Gp45 and the two highest-scoring peptide sequences (P1 and P3) and their scrambled forms using microscopy, spectroscopy, and ELISA., Results: It was found that rGp45 (recombinant Gp45) and chemically synthesised P1 had a higher binding affinity for S. aureus compared with all other peptides, except for Escherichia coli. Furthermore, rGp45 had a capture efficiency of > 86%; P1 had a capture efficiency of > 64%., Conclusion: These findings suggest that receptor-binding proteins such as rGp45, which provide a critical initiation of the phage life cycle for host adsorption, might play an important role in the diagnosis, imaging, and targeting of bacterial infections. Stroke is the second result for death and ischemic stroke constitutes most of all stroke cases. Ischemic stroke takes place when blood clot or embolus blocks cerebral vessel and interrupts blood flow, which often leads to brain damage, permanent disability, or death. There is a 4.5-h (golden hour) treatment window to restore blood flow prior to permanent neurological impairment results. Current stroke treatments consist mechanical system or thrombolytic drug therapy to disrupt or dissolve thrombus. Promising method for stroke treatment is mechanical retrieving of thrombi employing device deployed endovascularly. Advent of smart materials has led to research fabrication of several minimally invasive endovascular devices that take advantage of new materials capabilities. One of these capabilities is shape memory, is capability of material to store temporary form, then activate to primary shape as subjected to stimuli. Shape memory polymers (SMPs) are employed as good materials for thrombectomy device fabrication. Therefore, current review presents thrombectomy device development and fabrication with SMPs. Design, performance, limitations, and in vitro or in vivo clinical results of SMP-based thrombectomy devices are identified. Dual functional materials can be beneficial for simultaneous application in different fields. Herein, tubular graphitic carbon nitride (TCN) was anchored on natural diatomite (DT) by performing a simple hydrothermal-calcination method and the as-obtained composite (TCN/DT) was utilized in both photocatalytic remediation and thermal energy storage. The optimal sample, TCN/DT/3, could degrade 88.9 % of tetracycline, which was about 2.87 times than that of the pristine TCN. This could be due to extended light absorption ability, altered band structure and enhanced separation rate of photoinduced carrier. The photocatalytic efficiency remained 78.0% after fifth cycle, indicating its reusability feature. The reaction was mainly driven by superoxide radicals as well as holes and hydroxyl radicals mediated the reaction. The TCN/DT/3/Vis system showed good performance at near-neutral pH, also the system could be efficiently performed under tap water and drinking water. On the other hand, the usage of TCN/DT/3 catalyst as a framework for shape-stabilized stearic acid (SA) based composite phase change materials (PCMs) was explored. The composite PCM exhibited higher thermal energy storage capacity accompanied with improved thermal conductivity in comparison with DT/PCM composite. The COVID-19 pandemic has caused havoc around the globe since 2019 and is considered the largest global epidemic of the twentieth century. Although the first antiviral drug, Remdesivir, was initially introduced against COVID‑19, virtually no tangible therapeutic drugs exist to treat SARS-CoV-2 infection. FDA-approved Paxlovid (Nirmatrelvir supplemented by Ritonavir) was recently announced as a promising drug against the SARS-CoV-2 major protease (M pro ). Here we report for the first time the remarkable inhibitory potentials of lead epigenetic-targeting drugs (epi-drugs) against SARS-CoV-2 M pro . Epi-drugs are promising compounds to be used in combination with cancer chemotherapeutics to regulate gene expression. The search for all known epi-drugs for the specific inhibition of SARS-CoV-2 M pro was performed for the first time by consensus (three high-order program) molecular docking studies and end-state free energy calculations. Several epi-drugs were identified with highly comparable binding affinity to SARS-CoV-2 M pro compared to Nirmatrelvir. In particular, potent histone methyltransferase inhibitor EPZ005687 and DNA methyltransferase inhibitor Guadecitabine were prominent as the most promising epi-drug inhibitors for SARS-CoV-2 M pro . Long Molecular dynamics (MD) simulations (200 ns each) and corresponding MM-GBSA calculations confirmed the stability of the EPZ005687-M pro complex with MM-GBSA binding free energy (ΔG bind ) -48.2 kcal/mol (EPZ005687) compared to Nirmatrelvir (-44.7 kcal/mol). Taken together, the antiviral activities of the highlighted epi-drugs are reported beyond widespread use in combination with anti-cancer agents. The current findings therefore highlight as yet unexplored antiviral potential of epi-drugs suitable for use in patients struggling with chronic immunosuppressive disorders.Communicated by Ramaswamy H. The study examines the immobilization of the urease enzyme on a range of High Internal Phase Emulsion (polyHIPE) materials, assessing characteristics, efficiency, and performance. It also investigates the impact of polyHIPE type, quantity, incubation time, and various parameters on the process and enzyme activity. Surface morphology and functional groups of polyHIPE materials were determined through scanning electron microscopy (SEM) and fourier transform infrared spectroscopy (FT-IR) analyses, revealing significant alterations after modification with polyglutaraldehyde (PGA). The maximum immobilization efficiency of 95% was achieved by adding PGA to polyHIPE materials with an incubation period of 15 h. The optimized conditions for immobilized enzyme using a Box-Behnken design (BBD) of response surface methodology (RSM) were as follows: temperature (40.8 °C), pH (7.1) and NaCl concentration (0.007 g/L). Furthermore, the immobilized enzyme demonstrated remarkable reusability, retaining 75% of its initial activity after six cycles, and sustained shelf-life stability, retaining over 40% activity after 10 days at room temperature. Kinetic analyses revealed that immobilized urease exhibited higher affinity for the substrate, but lower rate of substrate conversion compared to the free enzyme. This study focused on an innovative practical method using computer vision for particle size measurement, which serves as a key precursor for predicting the elastic modulus of polymer nanocomposites. This approach involved the morphological segmentation of the nanodispersed phase. It aimed, for the first time, to address the impractical conditions resulting from the assumption of idealized single-particle sizes in a monodispersed system during modeling. Subsequently, a micromechanical finite element framework was employed to determine the interphase thickness and modulus in ultrahigh molecular weight polyethylene/nanozeolite composites, following the quantification of nanoparticle sizes. The size measurement approach relied on morphological images extracted from scanning electron microscopy micrographs of impact-fractured surfaces. To compute the interphase thickness, experimental data was fitted to an interphase-inclusive upper-bound Hashin-Shtrikman model, with the measured average particle size per composition serving as a crucial input. Subsequently, the interphase elastic modulus was computed based on its thickness, employing a hybrid modified-Hashin-Hansen and Maxwell model. These estimated interfacial variables were then utilized as inputs for the finite element model to determine the tensile modulus. A comparison between the model results and measured data revealed a maximum discrepancy of 3.29%, indicating the effectiveness of the methodology employed in quantifying interfacial properties.

Higher heating value (HHV) is one of the most important parameters in determining the quality of the fuels. In this study, comparatively large datasets of ultimate and proximate analysis are constructed to be used in HHV estimation of several classes of fuels, including char & fossil fuels, agricultural wastes, manure (chicken, cow, horse, sheep, llama, and pig), sludge (like paper, paper-mil, sewage, and pulp), micro/macro-algae's, wastes (RDF and MSW), treated woods, untreated woods, and others (non-fossil pyrolysis oils) between the HHV range of 4.22-55.55 MJ/kg. The relationships of carbon, hydrogen, and oxygen atomic ratios for fuel classes are illustrated by using ternary plots, and the effects of elemental composition on HHV was analyzed with the extensive dataset. Then, the ultimate (U) and ultimate & proximate (UP) datasets were utilized separately to estimate the HHV by using artificial neural networks (ANN). Hyperparameter optimization was carried out and the best performing ANNs were determined for each dataset, which yielded R 2 values of 0.9719 and 0.9715, respectively. The results indicated that while ANNs trained by both datasets perform remarkably well, utilization of U dataset is sufficient for HHV estimation. The development of multitargeted drugs represents an innovative approach to cancer treatment, aiming to enhance drug effectiveness while minimizing side effects. Herein, we sought to elucidate the inhibitory effect of selagibenzophenone B derivatives on the survival of cancer cells and dual topoisomerase I/II enzyme activity. Results demonstrated that among the compounds, SelB-1 selectively inhibited the proliferation and migration of prostate cancer cells while exhibiting minimal effects on healthy cells. Furthermore, SelB-1 showed a dual inhibitory effect on topoisomerases. Computational analyses mirrored the results from enzyme inhibition assays, demonstrating the compound's strong binding affinity to the catalytic sites of the topoisomerases. To our surprise, SelB-1 did not induce apoptosis in prostate cancer cells; instead, it induced autophagic gene expression and lipid peroxidation while reducing GSH levels, which might be associated with ferroptotic death mechanisms. Pentachlorophenol is a very toxic chemical that is used as a pesticide, fungicide, herbicide, wood preservative, etc., and it should be monitored in terms of human health and environmental production. Another environmental problem is the increase in the use of facemasks, especially during the COVID-19 pandemic. This study provides a value added chemicals to sustainability of recycling process. Fluorescent carbon dots (CDs) were synthesized from waste facemasks and investigated their fluorescence sensor performances. UV-Vis and fluorescence spectra of the synthesized carbon dots were recorded in different organic solvents. The sensor properties of these carbon dots against pesticides were investigated, and a 'turn-off' response was observed toward pentachlorophenol. The limit of detection was found 8.5 µM in the linear range from 43.3 µM to 375 µM. This study showed that waste plastics such as facemasks can be recycled to obtain carbon dots, which are used in different technological areas such as photocatalysis, bioimaging, etc., as well as in sensors., (© 2024. We developed an efficient method that enables selective photodimerization of 5-arylpenta-2,4-dienoic acids (i.e., vinylogous cinnamic acids). The use of 1,8-dihydroxynaphthalene as a template ensures proximity of the two reacting olefins so that irradiation of template-bound dienoic acids gives mono [2 + 2] cycloaddition products in good to excellent yields (up to 99%), as single regioisomers, and with high diastereoselectivities (dr = 3:1 to 13:1). The geometrical and stereochemical features of compounds 12a , 16a , and 22a were analyzed by X-ray crystallography.

Requiring no fuel for generation and negligible material/energy for operation and maintenance, photovoltaic (PV) systems have environmental impacts mostly due to the production of modules and the commissioning of power plants. Thus, extending the service lifetime of these systems from 30 to 40 years through an enhanced lamination process for module production potentially reduces environmental impacts per unit energy generated. Life cycle assessment is employed to evaluate the environmental impacts under scenarios for resource utilizations for the new lamination process, operation and maintenance requirements in the extended service lifetime, and degradation rates of the devised modules. Extending the service lifetime significantly reduces environmental impacts across categories, with a 21-27% reduction in global warming potential on the pessimistic and optimistic ends. At least 20% impact reduction is achieved in most impact categories, even under a pessimistic scenario. Considering uncertainty models in the life cycle inventories, samples are generated for scenarios via Monte Carlo simulation, and with significant improvements with large effects in most environmental impact categories, deterministic impact comparisons are supported by ANOVA and Tukey tests. The microstrip of modulated width is a realization of a one-dimensional photonic crystal operating in the microwave regime. Like any photonic crystal, the periodic microstrip is characterised by the presence of frequency bands and band gaps that enable and prohibit wave propagation, respectively. The frequency bands for microstrip of the symmetric unit cell can be distinguished by 0 or π Zak phase. The sum of these topological parameters for all bands below a given frequency gap determines the value of the surface impedance at the end of the microstrip. We demonstrate that edge modes are absent in a finite microstrip terminated at both ends in the centres of unit cells, but they can be induced by adding the defected cells. Edge modes present at both ends of the microstrip enable microwave tunneling with high transitivity in the frequency gap with or without a change in phase. This has been demonstrated experimentally and developed in detail using numerical simulations and model calculations. Background: With recent advances in artificial intelligence, the use of this technology has begun to facilitate comprehensive tissue evaluation and planning of interventions. This study aimed to assess different convolutional neural networks (CNN) in deep learning algorithms to detect keratinized gingiva based on intraoral photos and evaluate the ability of networks to measure keratinized gingiva width., Methods: Six hundred of 1200 photographs taken before and after applying a disclosing agent were used to compare the neural networks in segmenting the keratinized gingiva. Segmentation performances of networks were evaluated using accuracy, intersection over union, and F1 score. Keratinized gingiva width from a reference point was measured from ground truth images and compared with the measurements of clinicians and the DeepLab image that was generated from the ResNet50 model. The effect of measurement operators, phenotype, and jaw on differences in measurements was evaluated by three-factor mixed-design analysis of variance (ANOVA)., Results: Among the compared networks, ResNet50 distinguished keratinized gingiva at the highest accuracy rate of 91.4%. The measurements between deep learning and clinicians were in excellent agreement according to jaw and phenotype. When analyzing the influence of the measurement operators, phenotype, and jaw on the measurements performed according to the ground truth, there were statistically significant differences in measurement operators and jaw (p < 0.05)., Conclusions: Automated keratinized gingiva segmentation with the ResNet50 model might be a feasible method for assisting professionals. This study focused on testing different AI models, specifically CNN, to identify and measure a specific type of gum tissue called keratinized gingiva using photos taken inside the mouth. Out of 1200 photos, 600 were used in the study to compare the performance of different CNN in identifying gingival tissue. The accuracy and effectiveness of these models were measured and compared to human clinician ratings. The study found that the ResNet50 model was the most accurate, correctly identifying gingival tissue 91.4% of the time. When the AI model and clinicians' measurements of gum tissue width were compared, the results were very similar, especially when accounting for different jaws and gum structures. The study also analyzed the effect of various factors on the measurements and found significant differences based on who took the measurements and jaw type. Enterotypes have been shown to be an important factor for population stratification based on gut microbiota composition, leading to a better understanding of human health and disease states. Classifications based on compositional patterns will have implications for personalized microbiota-based solutions. There have been limited enterotype based studies on colorectal adenoma and cancer. Here, an enterotype-based meta-analysis of fecal shotgun metagenomic studies was performed, including 1579 samples of healthy controls (CTR), colorectal adenoma (ADN) and colorectal cancer (CRC) in total. Gut microbiota of healthy people were clustered into three enterotypes ( Ruminococcus -, Bacteroides - and Prevotella -dominated enterotypes). Reference-based enterotype assignments were performed for CRC and ADN samples, using the supervised machine learning algorithm, K-nearest neighbors. Differential abundance analyses and random forest classification were conducted on each enterotype between healthy controls and CRC-ADN groups, revealing novel enterotype-specific microbial markers for non-invasive CRC screening strategies. Furthermore, we identified microbial species unique to each enterotype that play a role in the production of secondary bile acids and short-chain fatty acids, unveiling the correlation between cancer-associated gut microbes and dietary patterns. The enterotype-based approach in this study is promising in elucidating the mechanisms of differential gut microbiome profiles, thereby improving the efficacy of personalized microbiota-based solutions.

Background: Today, synthetic chemicals are used in vitrification solutions for cryopreservation studies to mimic natural cryoprotectants that supply tolerance to organisms in nature against freezing stress. In the case of plants, PVS2, containing glycerol, dimethyl sulfoxide (Me2SO), ethylene glycol and sucrose, is considered as the golden standard for successful cryopreservation. However, Me2SO can generally cause toxicity to certain plant cells, adversely affecting viability after freezing and/or thawing. Hence, the replacement (or substantial reduction) of Me2SO by cheap, non-toxic and natural cryoprotectants became a matter of high priority to vitrification solutions or reducing their content gained escalating importance for the cryopreservation of plants. Fructans, sucrose derivatives mainly consisting of fructose residues, are candidate cryoprotectants., Objective: Inspired by their protective role in nature, we here explored, for the first time, the potential of an array of 8 structurally different fructans as cryoprotectants in plant cryopreservation., Materials and Methods: Arabidopsis thaliana L. seedlings were used as a model system with a one-step vitrification method. PVS2 solutions with different Me2SO and fructan contents were evaluated., Results: It was found that branched low DP graminan, extracted from milky stage wheat kernels, led to the highest recovery (85%) among tested fructans with 12.5% Me2SO after cryopreservation, which was remarkably close to the viability (90%) observed with the original PVS2 containing 15% Me2SO. Moreover, its protective efficacy could be further optimized by addition of vitamin C acting as an antioxidant., Conclusion: Such novel formulations offer great perspectives for cryopreservation of various crop species. Worldwide, where the demand for novel and greener solutions for sustainable agricultural production is increasing, the use of eco-friendly products such as seaweed-derived biostimulants as pre-sowing treatment represent a promising and important approach for the future. Cystoseira barbata, a brown seaweed species abundant in the Mediterranean Region, was collected from the Marmara Sea and subjected to water, alkali, and acidic extractions, and the biostimulant activity of these extracts was tested on wheat (Triticum durum cv. Saricanak-98) using different rates through application to the seeds or germination medium (substrate) applications. The different extracts were characterized by mineral, total phenolic, free amino acid, mannitol, polysaccharide, antioxidant concentrations and hormone-like activity. The effects of the extracts on growth parameters, root morphology, esterase activity, and mineral nutrient concentrations of wheat seedlings were investigated. Our results suggest that the substrate application was more effective in enhancing the seedling performance compared to the seed treatment. High rates of seaweed extracts applied to substrates increased the shoot length and fresh weight of wheat seedlings by up to 20 and 25%, respectively. The substrate applications enhanced the root fresh weights of wheat seedlings by up to 25% when compared to control plants. Among the biostimulant extract applications, the water extract at the highest rate yielded the most promising results in terms of the measured parameters. Cystoseira barbata extracts with different compositions can be used as effective biostimulants to boost seedling growth. The aim of this study was to analyze the phytochemical profile of Acacia cyclops trunk bark ethyl acetate extract using LC-tandem mass spectrometry for the first time, along with evaluating its antioxidant and anti-tyrosinase properties. Consequently, we determined the total phenolic and flavonoid contents of the extract under investigation and identified and quantified 19 compounds, including phenolic acids and flavonoids. In addition to assessing their antioxidant potential against DPPH (2,2-diphenyl-1-picrylhydrazyl) and ABTS (2,2'-azino-bis-[3-ethylbenzothiazoline-6-sulfonic] acid) assays, in vitro and in silico studies were conducted to evaluate the tyrosinase inhibitory properties of the A. cyclops extract. The ethyl acetate trunk bark extract exhibited a substantial total phenolic content and demonstrated significant antioxidant activity in terms of free radical scavenging, as well as notable tyrosinase inhibitory action (half-maximal inhibitory concentration [IC 50 ] = 14.08 ± 1.10 μg/mL). The substantial anti-tyrosinase activity of the examined extract was revealed through molecular docking analysis and druglikeness prediction of the main selected compounds. Ionic liquids (ILs) are interesting chemical compounds that have a wide range of industrial and scientific applications. They have extraordinary properties, such as the tunability of many of their physical properties and, accordingly, their activities; and the ease of synthesis methods. Hence, they became important building blocks in catalysis, extraction, electrochemistry, analytics, biotechnology, etc. This study determined antifungal activities of various imidazolium-based ionic liquids against yeast Saccharomyces cerevisiae via minimum inhibitory concentration (MIC) estimation method. Increasing the length of the alkyl group attached to the imidazolium cation, enhanced the antifungal activity of the ILs, as well as their ability of the disruption of the cell membrane integrity. FTIR studies performed on the S. cerevisiae cells treated with the ILs revealed alterations in the biochemical composition of these cells. Interestingly, the alterations in fatty acid content occurred in parallel with the increase in the activity of the molecules upon the increase in the length of the attached alkyl group. This trend was confirmed by statistical analysis and machine learning methodology. The classification of antifungal activities based on FTIR spectra of S. cerevisiae cells yielded a prediction accuracy of 83%, indicating the pharmacy and medicine industries could benefit from machine learning methodology.
