The morphological development of rice (Oryza sativa L.) leaves is closely related to plant architecture, physiological activities, and resistance. However, it is unclear whether there is a co-regulatory relationship between the morphological development of leaves and adaptation to drought environment. In this study, a drought-sensitive, roll-enhanced, and narrow-leaf mutant (renl1) was induced from a semi-rolled leaf mutant (srl1) by ethyl methane sulfonate (EMS), which was obtained from Nipponbare (NPB) through EMS. Map-based cloning and functional validation showed that RENL1 encodes a cellulose synthase, allelic to NRL1/OsCLSD4. The RENL1 mutation resulted in reduced vascular bundles, vesicular cells, cellulose, and hemicellulose contents in cell walls, diminishing the water-holding capacity of leaves. In addition, the root system of the renl1 mutant was poorly developed and its ability to scavenge reactive oxygen species (ROS) was decreased, leading to an increase in ROS after drought stress. Meanwhile, genetic results showed that RENL1 and SRL1 synergistically regulated cell wall components. Our results revealed a theoretical basis for further elucidating the molecular regulation mechanism of cellulose on rice drought tolerance, and provided a new genetic resource for enhancing the synergistic regulation network of plant type and stress resistance, thereby realizing simultaneous improvement of multiple traits in rice.

An excellent Fe-3.3 wt% Si steel was fabricated by double cold rolling and final annealing. The evolution of the microstructure and texture was studied by optical microscope (OM), X-ray diffraction (XRD), ex situ, and quasi-in situ electron backscattered diffraction (EBSD) to investigate the recrystallization behavior. Double cold rolling significantly reduced the adverse γ texture in the final annealed sheets, and a stronger η texture was observed. With a reduction ratio of 50% and 65% during double cold rolling, the γ texture almost disappeared, whereas the η texture was obviously improved. Consequently, the texture factor reached its peak, leading to a reduction in iron loss and an enhancement of magnetic induction. By combining texture regulation with dislocation strengthening, the magnetic properties of Fe-3.3 wt% Si steel were improved, and the yield strength also increased. The final sheet exhibiting exceptional magnetic characteristics and enhanced strength attained a reduction in iron loss (P10/400 = 21.84 W/kg) of 6.43 W/kg, along with an enhancement of magnetic induction (B50 = 1.698 T) of 0.038 T and yield strength (Rp0.2 = 578 MPa) of 37 MPa compared to a single-stage cold rolling process. The heading date, a critical trait influencing the rice yield and quality, has always been a hot topic in breeding research. Appropriately delaying the flowering time of excellent northern rice varieties is of great significance for improving yields and enhancing regional adaptability during the process for introducing varieties from north to south. In this study, genes influencing the heading date were identified through genome-wide association studies (GWAS). Using KenDao 12 (K12), an excellent cultivar from northern China, as the material, the specific flowering activator, OsMADS50, was edited using the genome-editing method to regulate the heading date to adapt to the southern planting environment. The results indicated that the osmads50 mutant line of K12 flowered about a week later, with a slight increase in the yield and good adaptability in the southern region in China. Additionally, the expressions of key flowering regulatory genes, such as Hd1, Ghd7, Ehd1, Hd3a, and RFT1, were reduced in the mutant plants, corroborating the delayed flowering phenotype. Yield trait analysis revealed that the primary factor for improved yield was an increase in the number of effective tillers, although there is potential for further enhancements in the seed-setting rate and grain plumpness. Furthermore, there were significant increases in the length-to-width ratio of the rice grains, fat content, and seed transparency, all contributing to an overall improvement in the rice quality. In summary, this study successfully obtained a rice variety with a delayed growth period through OsMADS50 gene editing, effectively implementing the strategy for adapting northern rice varieties to southern climates. This paper investigates the fault-tolerant control problem for vehicular platoons with time-varying actuator fault directions and distance constraints. A bias constraint function is introduced to convert the asymmetric constraints into symmetric ones, based on which a unified barrier Lyapunov function (BLF) method is proposed to ensure distance constraints. Further, an adaptive fixed-time fault-tolerant controller in the context of a sliding mode control technique is proposed, wherein a new Nussbaum function is adopted to address the effects of unknown time-varying actuator fault directions. It is proved that both individual vehicle stability and string stability can all be guaranteed, and the effectiveness of the proposed algorithm is verified through numerical simulations. Summary: Grain weight is an important determinant of grain yield. However, the underlying regulatory mechanisms for grain size remain to be fully elucidated. Here, we identify a rice mutant grain weight 9 (gw9), which exhibits larger and heavier grains due to excessive cell proliferation and expansion in spikelet hull. GW9 encodes a nucleus‐localized protein containing both C2H2 zinc finger (C2H2‐ZnF) and VRN2‐EMF2‐FIS2‐SUZ12 (VEFS) domains, serving as a negative regulator of grain size and weight. Interestingly, the non‐frameshift mutations in C2H2‐ZnF domain result in increased plant height and larger grain size, whereas frameshift mutations in both C2H2‐ZnF and VEFS domains lead to dwarf and malformed spikelet. These observations indicated the dual functions of GW9 in regulating grain size and floral organ identity through the C2H2‐ZnF and VEFS domains, respectively. Further investigation revealed the interaction between GW9 and the E3 ubiquitin ligase protein GW2, with GW9 being the target of ubiquitination by GW2. Genetic analyses suggest that GW9 and GW2 function in a coordinated pathway controlling grain size and weight. Our findings provide a novel insight into the functional role of GW9 in the regulation of grain size and weight, offering potential molecular strategies for improving rice yield. The immune system requires a high energy expenditure to resist pathogen invasion. Macrophages undergo metabolic reprogramming to meet these energy requirements and immunologic activity and polarize to M1-type macrophages. Understanding the metabolic pathway switching in large yellow croaker ( Larimichthys crocea ) macrophages in response to lipopolysaccharide (LPS) stimulation and whether this switching affects immunity is helpful in explaining the stronger immunity of hypoxia-tolerant L. crocea . In this study, transcript levels of glycolytic pathway genes ( Glut1 and Pdk1 ), mRNA levels or enzyme activities of glycolytic enzymes [hexokinase (HK), phosphofructokinase (PFK), pyruvate kinase (PK), and lactate dehydrogenase A (LDHA)], aerobic respiratory enzymes [pyruvate dehydrogenase (PDH), isocitrate dehydrogenase (IDH), and succinate dehydrogenase (SDH)], metabolites [lactic acid (LA) and adenosine triphosphate (ATP)], levels of bactericidal products [reactive oxygen species (ROS) and nitric oxide (NO)], and transcripts and level changes of inflammatory factors [IL1β, TNFα, and interferon (IFN) γ] were detected in LPS-stimulated L. crocea head kidney macrophages. We showed that glycolysis was significantly induced, the tricarboxylic acid (TCA) cycle was inhibited, and metabolic reprogramming occurred, showing the Warburg effect when immune cells were activated. To determine the potential regulatory mechanism behind these changes, Lc HIF-1α was detected and found to be significantly induced and transferred to the nucleus after LPS stimulation. LcHif-1α interference led to a significant reduction in glycolytic pathway gene transcript expression, enzyme activity, metabolites, bactericidal substances, and inflammatory factor levels; a significant increase in the aerobic respiration enzymes; and decreased migration, invasion, and phagocytosis. Further ultrastructural observation by electron microscopy showed that fewer microspheres contained phagocytes and that more cells were damaged after LcHif-1α interference. LcHif-1α overexpression L. crocea head kidney macrophages showed the opposite trend, and promoter activities of Ldha and Il1β were significantly enhanced after LcHif-1α overexpression in HEK293T cells. Our data showed that Lc HIF-1α acted as a metabolic switch in L. crocea macrophages and was important in polarization. Hypoxia-tolerant L. crocea head kidney showed a stronger Warburg effect and inhibited the TCA cycle, higher metabolites, and bactericidal substance levels. Seed endophytic microbiomes are shaped by host and environmental factors and play a crucial role in their host growth and health. Studies have demonstrated that host genotype, including hybridization, affects seed microbiomes. Heterosis features are also observed in root-associated microbiomes. It remains unclear, however, whether heterosis exists in seed endophytic microbiomes and whether hybrid microbiota provide noticeable advantages to host plant growth, especially to seed germination. Here, we investigated the structure of seed endophytic bacterial and fungal communities from three hybrid rice varieties and their respective parents using amplicon sequencing targeting 16S rRNA and ITS2 genes. Heterosis was found in diversity and composition of seed endophytic microbiomes in hybrids, which hosted more diverse communities and significantly higher abundances of plant growth-promoting taxa, such as Pseudomonas and Rhizobium genera compared with their parental lines. Co-occurrence network analysis revealed that there are potentially tighter microbial interactions in the hybrid seeds compared with their parent seeds. Finally, inoculation of seed-cultivable endophytes, isolated from hybrids, resulted in a greater promotion of seed germination compared with those isolated from parent lines. These findings suggest that heterosis exists not only in plant traits but also in seed endophytic microbiota, the latter in turn promotes seed germination, which offers valuable guidance for microbiome-assisted rice breeding.IMPORTANCEGenetic and physiological changes associated with plant hybridization have been studied for many crop species. Still, little is known about the impact of hybridization on the seed microbiota. In this study, we indicate that hybridization has a significant impact on the endophytic bacterial and fungal communities in rice seeds. The seed endophytic microbiomes of hybrids displayed distinct characteristics from those of their parental lines and exhibited potential heterosis features. Furthermore, the inoculation of seed-cultivable endophytes isolated from hybrids exhibited a greater promotion effect on seed germination compared with those isolated from the parents.
