2. Association between Chinese visceral adiposity index and lower urinary tract symptoms suggestive of benign prostatic hyperplasia (LUTS/BPH): a national cohort study

Abstract Background Benign prostatic hyperplasia (BPH) and its related lower urinary tract symptoms (LUTS) are commonly observed among aging males and have a substantial effect on quality of life. Metabolic syndrome, with a specific focus on obesity, is believed to play a role in the development of BPH. This study intends to explore the relationship between several obesity-related metrics, including the Chinese Visceral Adiposity Index (CVAI), and LUTS/BPH within a national cohort of Chinese men. Methods Data from the China Health and Retirement Longitudinal Study (CHARLS) 2015 were analyzed, encompassing a sample of 5,735 male participants aged 45 and older. Eight obesity-related indices—namely Body mass index (BMI), Waist-height ratio (WHtR), Triglyceride-glucose.BMI (TyG.BMI), TyG. Waist circumference (TyG.WC), TyG.WHtR, CVAI, Visceral adiposity index (VAI), and A body shape index (ABSI)—were examined. Logistic regression models, adjusted for potential confounders, were utilized to evaluate the associations between these indices and LUTS or BPH. The predictive capabilities of these indices were further assessed using receiver operating curves (ROC). Results Among the participants, 718 (12.5%) were diagnosed with LUTS/BPH. All obesity-related indices were significantly higher in the LUTS/BPH group compared to the healthy group. CVAI demonstrated the highest predictive ability for LUTS/BPH, with an area under the curve (AUC) of 0.58. The study highlighted a nonlinear relationship between LUTS/BPH and several obesity-related indices, including CVAI. Conclusions This study underscores the significant association between visceral fat, as measured by CVAI, and the risk of LUTS/BPH in Chinese men. CVAI emerged as the most effective predictor among the indices evaluated, suggesting its potential utility in identifying individuals at risk for LUTS/BPH. Further prospective studies are needed to confirm these findings and elucidate the underlying mechanisms.

High-entropy alloys (HEAs) are known for their distinctive microstructural features, outstanding performance, and potential applications, establishing them as novel metallic materials. Studies indicate that face-centered cubic HEAs generally offer increased strength and toughness at lower temperatures, suitable for cryogenic applications. This work on (FeCoNi)86Al7Ti7 HEA fabricated using powder bed fusion (PBF) studies the phase composition, microstructure, and mechanical properties at both room and cryogenic temperatures (298 K and 77 K). The PBF HEA exhibits a hierarchical microstructure with columnar grains, Ti-enriched cellular substructures entangled with high-density dislocations, and L21 nanoprecipitates, contributing to an excellent strength-ductility combination at room temperature. Notably, as the temperature decreases from 298 K to 77 K, both strength and ductility increase, with a higher yield strength of ∼1.0 GPa, ultimate tensile strength of ∼1.55 GPa, and ductility of ∼42%. Dislocation strengthening is dominant at both room and cryogenic temperatures, with dislocation slip as the primary deformation mechanism at 298 K and a combination of dislocation slips and stacking faults at 77 K.

Abstract Oxidative stress and neuronal apoptosis could be an important factor leading to post-hemorrhagic consequences after germinal matrix hemorrhage (GMH). Previously study have indicated that relaxin 2 receptor activation initiates anti-oxidative stress and anti-apoptosis in ischemia-reperfusion injury. However, whether relaxin 2 activation can attenuate oxidative stress and neuronal apoptosis after GMH remains unknown. To investigate the beneficial effect of relaxin 2 on oxidative stress injury and neuronal apoptosis by GMH, a total of 150 rat pups were subjected to GMH by an intraparenchymal injection of bacterial collagenase. Recombinant human relaxin-2 (rh-relaxin-2) was administered intraperitoneally injections at 1 h and 13 h after GMH. Lenti-virus with sgRXFP1 and sgCtrl was administered intracerebroventricular (i.c.v.) on the left side of the brain to inhibit the RXFP1 at 2d prior to GMH induction, and LY321499, ERK inhibitor, was administered by i.c.v. injection at 1 h on the left side of the brain prior to GMH induction, respectively. Co-immunoprecipitation, immunofluorescence, TUNEL, Fluoro-Jade C, DHE staining, western blot, Nitrix Oxide (NO) quantification and side effect experiments were performed to evaluate post-GMH. We found endogenous relaxin-2 interacts with RXFP1 and both protein colocalized in neurons on the first day after GMH. Additionally, RXFP1 activation with rh-relaxin-2 significantly inhibited oxidative stress and neuronal apoptosis in GMH + rh-relaxin-2 group compared with GMH + vehicle group. Moreover, rh-relaxin-2 treatment significantly inhibited the phosphorylation of ERK and nNOS, as well as upregulated expression of Bcl2 and NO and downregulated expression of Bax and Romo 1. The beneficial effects of rh-relaxin-2 were reversed by i.c.v. injection of lenti-virus with sgRXFP1 and LY321499, respectively. Furthermore, the side effect experiment showed rh-relaxin-2 did not affect neurological behavior and the function of liver and kidney. In conclusion, our finding showed that rh-relaxin-2 attenuated oxidative stress and neuronal apoptosis after GMH through RXFP1-ERK-nNOS-NO signaling pathway.

Abstract The observation of high-T c superconductivity (HTSC) in concomitant with pressure-induced orthorhombic-tetragonal structural transition in bilayer La3Ni2O7 has sparked hopes of achieving HTSC by stabilizing the tetragonal phase at ambient pressure. Chemical pressure, introduced by replacing La3+ with smaller rare-earth R 3+ has been considered as a potential route. However, our experimental and theoretical investigation reveals that such substitutions, despite causing lattice contraction, actually produce stronger orthorhombic distortions, requiring higher pressures for the structural transition. A linear extrapolation of P c versus the average size of A-site cations (), yields a putative critical value of c ≈ 1.23 Å for P c ≈ 1 bar. The negative correlation between P c and indicates that replacing La3+ with smaller R 3+ ions is unlikely to reduce P c to ambient pressure. Instead, substituting La3+ with larger cations like Sr2+ or Ba2+ might be a feasible approach. Our results provide guidance for realizing ambient-pressure HTSC in bilayer nickelates.

Abstract Purpose To evaluate the efficacy of an enhanced recovery after surgery (ERAS) strategy for upper tract urothelial carcinoma (UTUC) patients undergoing laparoscopic radical nephroureterectomy (LRNU). Methods 90 patients who received LRNU at Zhongnan Hospital of Wuhan University between January 2018 and July 2022 were retrospectively analyzed, including 43 in the ERAS group and 47 in the pre-ERAS group. Abstract It is significant to study the stability of surrounding rock in soft rock tunnels to ensure construction safety and improve efficiency. Through triaxial shear tests on soft rock at various confining pressures, we observed the failure characteristics transitioning from strain softening to strain hardening as confining pressure increases. An improved Hoek-Brown strength criterion has been proposed to characterize the critical confining pressure effect of soft rock, with tensile strength in the tensile zone aligning with experimental results, showing an error of less than 5%. By assuming that the micro-unit strength of soft rock follows a two-parameter Weibull distribution, we constructed a statistical damage constitutive model for soft rock in mountain tunnels. This model provides a more realistic depiction of damage change, failure processes, and stress-strain evolution in soft rock, thereby validating its scientific and rational foundation. The research findings can serve as a valuable reference for mountain tunnel construction.

In the field of autonomous driving, the accuracy and real-time requirements for 3D object detection technology continue to improve, which is directly related to the commercialization process and market popularity of autonomous vehicles. Despite the efficiency of pillar-based coding for onboard systems, it falls short in terms of accuracy and the reduction of incorrect positives. In this paper, we will examine how to solve the problem of high incorrect rate and low accuracy of existing methods. Firstly, a MAP coding module is introduced to optimize previous point cloud feature coding modules, allowing for the efficient extraction of fine-grained features from point cloud data. Then, we introduce an innovative sparse dual attention (SDA) to efficiently filter out irrelevant details in feature extraction, thereby improving the pertinence and efficiency of information extraction. Finally, to address the potential loss of information from single local feature extraction, a local and global fusion module (CTGC) is introduced. Our method has proactively demonstrated its efficiency and accuracy through rigorous experimentation across diverse datasets. BackgroundDe novo neuroendocrine prostate cancer (NEPC) is a rare subtype of prostate cancer (PCa) and few markers are available for screening and monitoring. Potential circulating or fluid markers might facilitate early diagnosis thus improving prognosis of NEPC, especially for de novo NEPC.Case presentationA man of 71-year was presented with elevated level of serum carcinoembryonic antigen (CEA) (1296.5 ng/ml) and normal PSA (0.47ng/ml). Gastrointestinal endoscopy showed no signs of gastric or colorectal cancer. Fluorodeoxyglucose positron emission tomography-computed tomography (FDG PET-CT) and prostate-specific membrane antigen PET-CT (PSMA PET-CT) indicated prostate cancer with metastases including pelvic lymph nodes, bone as well as lung metastases. Biopsy of prostate revealed mixed carcinoma including small cell neuroendocrine carcinoma (SCNEC) and adenocarcinoma (Gleason score of 4 + 5). Immunohistochemistry (IHC) staining and next generation sequencing demonstrated a strong expression of chromogranin A (CgA), synaptophysin (SYN) and CEA, and a germline mutation in BRCA2, respectively. After a prostatic massage, an increased level of CEA (137 ng/ml vs 5 ng/ml) was detected in urine. Olaparib, a Poly ADP-ribose polymerase inhibitor (PARPi), combined with androgen deprivation therapy (ADT) were administrated. FDG PET-CT indicated tumor regression in both quantity and size three months later, and CEA levels of serum and urine decreased to 23 ng/ml and 2.4 ng/ml 4 months later, respectively.ConclusionThis is the first report of a de novo NEPC presented with an elevated level of CEA, in which CEA was also detected in urine specimen post a prostatic massage. After a combination treatment of ADT for 3 months, levels of CEA in both serum and urine decreased sharply when tumor regressed radiologically. CEA might be a marker of screening and monitoring of NEPC.

Traditional object detection methods usually underperform when locating tiny or small drones against complex backgrounds, since the appearance features of the targets and the backgrounds are highly similar. To address this, inspired by the magnocellular motion processing mechanisms, we proposed to utilize the spatial–temporal characteristics of the flying drones based on spiking neural networks, thereby developing the Magno-Spiking Neural Network (MG-SNN) for drone detection. The MG-SNN can learn to identify potential regions of moving targets through motion saliency estimation and subsequently integrates the information into the popular object detection algorithms to design the retinal-inspired spiking neural network module for drone motion extraction and object detection architecture, which integrates motion and spatial features before object detection to enhance detection accuracy. To design and train the MG-SNN, we propose a new backpropagation method called Dynamic Threshold Multi-frame Spike Time Sequence (DT-MSTS), and establish a dataset for the training and validation of MG-SNN, effectively extracting and updating visual motion features. Experimental results in terms of drone detection performance indicate that the incorporation of MG-SNN significantly improves the accuracy of low-altitude drone detection tasks compared to popular small object detection algorithms, acting as a cheap plug-and-play module in detecting small flying targets against complex backgrounds.

Excessive utilization of chemical fertilizers degrades the quality of medicinal plants and soil. Bio-organic fertilizers (BOFs) including microbial inoculants and microalgae have garnered considerable attention as potential substitutes for chemical fertilizer to enhance yield. In this study, a field experiment was conducted to investigate the effects of BOF partially substituting chemical fertilizer on the growth and quality of medicinal plant Polygala tenuifolia. The growth parameters, bioactive component contents, soil properties and composition of rhizosphere microorganisms were measured. The results indicated that substituting 40% of chemical fertilizer with microalgae showed the most pronounced growth-promoting effect, leading to a 29.30% increase in underground biomass and a 19.72% increase in 3,6’-disinapoylsucrose (DISS) content. BackgroundThe Oxidative Balance Score (OBS) quantifies the overall oxidative stress burden, with higher scores indicating greater antioxidant (relative to prooxidant) activity. This study aimed to examine the association between peripheral arterial disease (PAD) and OBS.Methods and materialsData from the National Health and Nutrition Examination Survey (NHANES, 1999–2004) were analyzed for participants with ankle-brachial index (ABI) measurements. The total Oxidative Balance Score (OBS) comprised a lifestyle OBS (four lifestyle categories) and a dietary OBS (16 dietary factors). Logistic regression analyses evaluated associations between PAD and total OBS, lifestyle OBS, and dietary OBS. Restricted cubic spline (RCS) analyses assessed dose–response relationships between ABI, PAD, and OBS. Mediation analyses investigated the roles of glucolipid metabolism and renal function in the OBS-PAD association. 15. Quantum and complex-valued hybrid networks for multi-principal element alloys phase prediction

Summary: This study introduces a hybrid network model for phase classification, integrating quantum networks and complex-valued neural networks. This architecture uses elemental composition as its only input, eliminating complex feature engineering. Parameterized quantum networks handle sparse elemental data and convert data from real to complex domains, increasing information dimensionality. Complex-valued neural networks process data in the complex domain, significantly reducing information loss during transitions. The experimental results show that the hybrid model achieves a phase classification accuracy of 94.93%, outperforming the best machine learning model by 2.27% and the quantum model by 8.67%. Precision, recall, and F1-score are also excellent at 0.9494, 0.9493, and 0.9500, respectively. Additional tests on phase transitions in AlxCoCrFeNi alloys confirm the model’s robust generalization, identifying transition thresholds at 0.46 and 0.88, closely matching the 0.45 and 0.88 reported in related studies.

16. A homophilic and dynamic influence maximization strategy based on independent cascade model in social networks

Influence maximization (IM) is crucial for recommendation systems and social networks. Previous research primarily focused on static networks, neglecting the homophily and dynamics inherent in real-world networks. This has led to inaccurate simulations of information spread and influence propagation between nodes, with traditional IM algorithms’ selected seed node sets failing to adapt to network evolution. To address this issue, this paper proposes a homophilic and dynamic influence maximization strategy based on independent cascade model (HDIM). Specifically, HDIM consists of two components: the seed node selection strategy that accounts for both homophily and dynamics (SSHD), and the independent cascade model based on influence homophily and dynamics (ICIHD). SSHD strictly constrains the proportions of different node types in the seed node set and can flexibly update the seed node set when the network structure changes. ICIHD redefines the propagation probabilities between nodes, adjusting them in response to changes in the network structure. Experimental results demonstrate HDIM’s excellent performance. Specifically, the influence range of HDIM exceeds that of state-of-the-art methods. Furthermore, the proportions of various activated nodes are closer to those in the original network.

BackgroundsCollagen type I alpha 1 chain (COL1A1) is a key protein encoding fibrillar collagen, playing a crucial role in the tumor microenvironment (TME) due to its complex functions and close association with tumor invasiveness. This has made COL1A1 a focal point in cancer biology research. However, studies investigating the relationship between COL1A1 expression levels and clinical characteristics of ovarian cancer (OC) remain limited.MethodsThis study integrated resources from publicly available online databases and immunohistochemistry (IHC) techniques to analyze and validate COL1A1 expression in OC tissues, and evaluated its potential association with clinical features in OC patients. The prognostic value of COL1A1 was assessed using Kaplan-Meier (KM) survival curve analysis. The TIMER and TISIDB databases to explore the potential relationship between COL1A1 expression and immune microenvironment in OC tissues. The LinkedOmics and INPUT2 databases were used to analyze differential gene expression in OC, This was followed by enrichment analysis using the Kyoto Encyclopedia of Genes and Genomes (KEGG) and Gene Ontology (GO) annotations to identify and predict potential signaling pathways associated with COL1A1.ResultsOur study demonstrated that COL1A1 expression was significantly elevated in OC tissues compared to normal ovarian tissues. This elevated expression was closely associated with tumor metastasis, poor prognosis, and advanced pathological stages in OC patients. Moreover, COL1A1 expression showed a significant correlation with immune cell infiltration and the expression of immune-related genes within the TME.Further analyses revealed that COL1A1 and its co-expressed genes were primarily enriched in key signaling pathways involved in OC invasion, metastasis, and angiogenesis, indicating its potential role in driving OC progression.ConclusionsOur study found that upregulation of COL1A1 expression is significantly associated with lymph node metastasis of OC and can affect the immune microenvironment. Based on this, COL1A1 could serve as a promising biomarker for OC prognosis and provide a new perspective for the development of potential immunotherapies for patients with OC.

Abstract Graphene films have been applied in the thermal management of electronic devices due to their high thermal conductivity. However, the ever‐increasing power and local heat flux density of electronic chips require graphene films with excellent heat flux carrying capacity. Enhancing the heat flux carrying capacity is highly challenging, and the key is to maintain high thermal conductivity while increasing film thickness. Gases released during film assembly and the resulting catastrophic structural destruction should be responsible for the trade‐off between film thickness and thermal conductivity. Herein, the evolution of the pore structure is investigated during the assembly of graphene films and propose the construction of gas escape channels for the preparation of thick graphene films. The process involves using humidification treatment and freeze‐drying GO films to pre‐construct the ordered flat pore structure. The microstructure optimization of graphene films with more order, fewer wrinkles and defects, and larger grain size is achieved. After optimization, graphene films with ultra‐high thermal conductivity (1781 W m−1 K−1) and a thickness over 100 µm are realized. These films exhibit exceptional heat dissipation and cooling capabilities in high heat flux density (≈2000 W cm−2). This finding holds significant potential for guiding the thermal management of high‐power devices.

Sensitivity to motion direction is a feature of visual neurons that is essential for motion perception. Recent studies have suggested that direction selectivity is re-established at multiple stages throughout the visual hierarchy, which contradicts the traditional assumption that direction selectivity in later stages largely derives from that in earlier stages. By recording laminar responses in areas 17 and 18 of anesthetized cats of both sexes, we aimed to understand how direction selectivity is processed and relayed across 2 successive stages: the input layers and the output layers within the early visual cortices. We found a strong relationship between the strength of direction selectivity in the output layers and the input layers, as well as the preservation of preferred directions across the input and output layers. Moreover, direction selectivity was enhanced in the output layers compared to the input layers, with the response strength maintained in the preferred direction but reduced in other directions and under blank stimuli. We identified a direction-tuned gain mechanism for interlaminar signal transmission, which likely originated from both feedforward connections across the input and output layers and recurrent connections within the output layers. This direction-tuned gain, coupled with nonlinearity, contributed to the enhanced direction selectivity in the output layers. Our findings suggest that direction selectivity in later cortical stages partially inherits characteristics from earlier cortical stages and is further refined by intracortical connections.

The monosynaptic cortico-motoneuronal connections suggest the possibility of individual motor units (MUs) receiving independent commands from motor cortex. However, previous studies that used corticomuscular coherence (CMC) between electroencephalogram (EEG) signals and electromyogram (EMG) signals have not directly explored the corticospinal functionality at the single motoneuron level. The objective of this study is to find out whether synchronous activities exist between the motor cortex and individual MUs. Corticomuscular coherence was calculated between the EEG signals and the MU firing event trains which were extracted using the EMG decomposition technique. The results showed that some but not all MUs indeed had significant coherent activities with the contralateral motor cortex, which we named the cortico-motoneuronal coherence (CMnC). In contrast to the CMC only occurring in β and γ bands, CMnC occurred across the four common EEG frequency bands (θ, α, β and γ). Further, we identified individual MUs that showed significant interactions with the motor cortex. These coherent MUs (CohMU) could still be found even when the EMG signals were not coupled with the cortical activities. Compared with conventional CMC, our preliminary results indicated that the CMnC could potentially help to investigate the complex coupling between cortical and muscular activities due to its ability to separate different correlated components. This study proves that corticomuscular coherence exists at a single MU level, which provides a new perspective for the research on corticomuscular coupling. Further study on the CMnC could help deepen our understanding of the neural control of movement.

21. Contact Load and Elastohydrodynamic Lubrication Analysis of Eccentric Bearings in RV Reducer Considering the Effects of Roller Profile Modification

The mechanical and tribological behavior of eccentric bearings is crucial for the performance of a RV reducer. By combining the finite element model (FEM) and the elastohydrodynamic lubrication (EHL) method, a comprehensive model for the cylindrical roller bearings applied in the RV reducer is developed in this study. During the modeling phase, FEM is utilized to determine the bearing load, taking into account the structural flexibility. The FEM result demonstrates that a 15% increase in the maximum bearing’s load is detected by the FEM compared to the analytical model. After the simulations, the effects of the roller profile modification, the bearing load and the rolling speed on the bearing performance are revealed. The numerical results indicate that the combined generatrix shape roller results in weaker edge effects and stress concentration compared with that of the straight generatrix shape roller and the arc generatrix shape roller. The optimal values of modification length and modification quantity under various load and rolling speed conditions are provided. Furthermore, durability tests on RV reducers equipped with the three types of rollers were conducted. Experimental outcomes demonstrate that the combined generatrix shape roller significantly improves the reliability and fatigue life of the RV reducer, corroborating the numerical analyses.

These robots represent a current area of significant research interest within the field of robotics. Snake robots have a wide range of applications in many fields, advancing the integration of bionics, robotics, and cybernetics, while playing a crucial role in performing survey and rescue missions. This survey presents the latest technological advancements in modeling, motion control, and guidance laws for planar snake robots, and provides a unified perspective based on the existing results. To achieve target-tracking control of robots in complex environments, we present a feasible approach that integrates guided vector field technology and transforms the target-tracking and obstacle avoidance problem into a reference angle tracking issue. Finally, this paper analyzes and summarizes the development process and key technologies of snake robot control and provides an outlook on future development trends.

23. Characterization of the Phosphotransferase from Bacillus subtilis 1101 That Is Responsible for the Biotransformation of Zearalenone

Bacillus microorganisms play an important role in the zearalenone (ZEA) biotransformation process in natural environments. The phosphotransferase pathway in Bacillus is both widespread and relatively well conserved. However, the reaction kinetics of these phosphotransferases remain poorly understood, and their catalytic activities are suboptimal. In this study, a ZEA phosphotransferase, ZPH1101, was identified from Bacillus subtilis 1101 using genome sequencing. The product transformed by ZPH1101 was identified as phosphorylated ZEA (ZEA-P) through LC-TOF-MS/MS analysis. The experiments conducted on MCF-7 cells demonstrated that ZEA-P exhibited a lower level of estrogenic toxicity than ZEA. The optimal reaction conditions for ZPH1101 were determined to be 45 °C and pH 8.0. The maximum velocity (Vmax), Michaelis constant (Km), and catalytic constant (kcat) were calculated through fitting to be 16.40 μM·s−1·mg−1, 18.18 μM, and 54.69 s−1, respectively. Furthermore, adding 1 mmol/L Fe2+ or Fe3+ to the reaction system increased the efficiency of ZPH1101 in converting ZEA by 100% relative to the system containing solely 1 mmol/L ATP and 1 mmol/L Mg2+, suggesting that low concentrations of Fe2+ or Fe3+ can improve the ZPH1101-mediated transformation of ZEA. This study contributes to the enzymatic removal of ZEA and broadens the spectrum of strain and enzyme options available to researchers for ZEA detoxification efforts.

Sugarcane (Saccharum spp.), a critical crop for sugar and bioenergy production, faces challenges in genetic improvement due to limited genetic diversity from selective breeding. Expanding genetic resources through intergeneric hybridization, particularly with Narenga porphyrocoma, offers a promising avenue to introduce traits like stress resistance and high biomass productivity. However, verifying true hybrids remains challenging with traditional morphological methods. This study employed tetra-primer ARMS-PCR and genomic in situ hybridization (GISH) to accurately identify intergeneric hybrids between S. officinarum and N. porphyrocoma. Species-specific primers were designed based on SNPs in the nrDNA-ITS region for ARMS-PCR, enabling effective differentiation of parental and hybrid genotypes, while GISH confirmed the chromosomal composition of hybrids, revealing an n + n inheritance pattern. The results demonstrated the potential of N. porphyrocoma to improve sugarcane’s tillering and leaf length, although sucrose content was lower in hybrids, suggesting the need for further breeding efforts. This study uniquely contributes to sugarcane breeding by providing an effective method for hybrid verification and laying a foundation for incorporating beneficial N. porphyrocoma genes into sugarcane cultivars.

Aiming to address the problem of low recognition accuracy in the current cleaning loss detection system of rapeseed harvesters, a rapeseed cleaning loss detection system was developed using the energy distinction method based on the principle of impact piezoelectricity. A signal processing circuit, centered around a hardware integral circuit and a triple voltage comparison circuit, was designed. The energy of the impact signals generated by rapeseed kernels and impurities was calculated through hardware integration. The distinction threshold for the energy of the impact signals generated by kernels and impurities under the operating wind speed of the cleaning system was found through experiments, and a fitting model relating the fan speed to the distinction threshold was constructed. A loss detection and counting system for rapeseed kernels was designed to realize the statistics and real-time display of rapeseed kernels regarding cleaning loss. Performance verification tests were conducted on mixtures of rapeseed kernels and impurities with different mixing ratios, and a field test was carried out on the platform of a 4LZY-5.0Z rapeseed combine harvester. The test results showed that the accuracy of the designed loss detection system for kernel identification was more than 91.6%. Under operating conditions of 700, 900 and 1200 r/min fan speeds in the combine harvester cleaning system, the relative errors of the loss detection system compared to manual detection were 5%, 4.7% and 3.8%, respectively. The developed loss detection system for rapeseed kernels has high detection accuracy and good overall performance, which means it can provide feedback information for the control of the harvester.

Abstract The association of necrosis in tumors with poor prognosis implies a potential tumor-promoting role. However, the mechanisms underlying cell death in this context and how damaged tissue contributes to tumor progression remain unclear. Here, we identified p38 mitogen-activated protein kinases (p38 MAPK, a.k.a. p38) as a key player in promoting cell death and the inflammatory response to ischemic stress associated with necrotic tumors. We found that glioblastoma (GBM) cells expressing patient-derived Kirsten rat sarcoma (KRAS) or phosphoinositide-3-kinase (PI3K) active mutants showed enhanced cell death under ischemia-mimetic conditions in vitro and were more likely to develop into necrotic tumors in vivo. Cell death in both settings depended on p38, which is also required for tumor progression driven by KRAS or PI3K. Under ischemia-mimetic conditions, GBM cells undergo reactive oxygen species (ROS)-dependent cell death. Gene expression in these cells recapitulated multiple features observed in peri-necrotic tumors from patient GBM. Further studies showed the involvement of a positive feedback loop between the p38-MAPK-activated protein kinase 2 (MAPKAPK2, a.k.a. MK2) signaling axis and the unfolded protein response signaling components activating transcription factor 4 (ATF4) and inositol-requiring enzyme 1 (IRE1α) in driving ischemic tumor cell death. This signaling cascade was further potentiated by RAS or PI3K activation under ischemic conditions, contributing to the inflammatory gene expression response. Therefore, our study suggests that p38 could be targeted to relieve the inflammatory response in necrotic tumors and inhibit GBM progression.

Abstract Backgrounds Adapter proteins (APs) complex is a class of heterotetrameric complexes comprising of 4-subunits with important regulatory functions in eukaryotic cell membrane vesicle trafficking. Foxtail millet (Setaria italica L.) is a significant C4 model plant for monocotyledon studies, and vesicle trafficking may plays a crucial role in various life activities related to growth and development. Despite this importance, studies on AP complexes in foxtail millet have been lacking. Results This research conducted genome-wide identification and systematical analysis of AP complexes in foxtail millet. 33 SiAP complex genes were identified and classified into 7 groups, distributed unevenly across 9 chromosomes in foxtail millet. Among these genes, 11 segmental duplication pairs were found. Out of the 33 SiAP complex genes, 24 exhibited collinear relationships with Setaria viridis, while only one showed relationship with Arabidopsis thaliana. Gene structure and motif composition were investigated to understand the function and evolution of these SiAP complex genes. Furthermore, these promoter region of the SiAP complex genes contains 49 cis-elements that are associated with responses to light, hormones, abiotic stress, growth and development. The interaction network between the SiAP complexes was analyzed, and there were strong interactions among the SiAP complex proteins. Expression patterns of SiAP complex genes in different organs and developmental stages of foxtail millet were investigated. The majority of the SiAP complex genes exhibited expressed in multiple tissues, with some genes being predominantly expressed in specific tissues. Subsequently, we selected SiAP4M and SiAP2M for validation of subcellular localization. The signal of 35 S:: SiAP4M: GFP (Long) and 35 S:: SiAP4M: GFP (Short) fused proteins were primarily observed in the nucleus, while the signal of 35 S:: SiAP2M: GFP fused proteins was widely distributed on the cell membrane and vesicles. Conclusions Overall, this study presents a comprehensive map of the SiAP complexes in foxtail millet. These findings not only administer to understanding the biological functions of AP complexes in foxtail millet growth and development but also offer insights for enhancing genetic breeding in this crop.
