Abstract The chloroplast (cp) genome sequence of Mussaenda pubescens, a promising resource that is used as a traditional medicine and drink, is important for understanding the phylogenetic relationships among the Mussaenda family and genetic improvement and reservation. This research represented the first comprehensive description of the morphological characteristics of M. pubescens, as well as an analysis of the complete cp genome and phylogenetic relationship. The results indicated a close relationship between M. pubescens and M. hirsutula based on the morphological characteristics of the flower and leaves. The cp was sequenced using the Illumina NovaSeq 6000 platform. The results indicated the cp genome of M. pubescens spanned a total length of 155,122 bp, including a pair of inverted repeats (IRA and IRB) with a length of 25,871 bp for each region, as well as a large single-copy (LSC) region and a small single-copy (SSC) region with lengths of 85,370 bp and 18,010 bp, respectively. The results of phylogenetic analyses demonstrated that species within the same genus displayed a tendency to group closely together. It was suggested that Antirhea, Cinchona, Mitragyna, Neolamarckia, and Uncaria might have experienced an early divergence. Furthermore, M. hirsutula showed a close genetic connection to M. pubescens, with the two species having partially overlapping distributions in China. This study presents crucial findings regarding the identification, evolution, and phylogenetic research on Mussaenda plants, specifically targeting M. pubescens.

Background: In recent years, numerous studies have suggested that the gut microbiota and its metabolites are closely related to myocardial infarction. Utilizing insights from these research findings may be advantageous in the prevention, treatment, and prognosis of myocardial infarction. We have employed bibliometric methodology to summarize the progress made in this research area over the past 20 years, identify the hotspots, and highlight the developmental tendencies, providing a reference for future research in this field. Methods: We searched the content related to this field in the Web of Science Core Collection database, with a time range from 2001 to 2023. We used VOSviewer, CiteSpace, and Scimago Graphica software to visualize the search results. Results: We included 889 reports in this study. The country with the most publications was China, while the country with the greatest influence was the United States. An analysis of institutions showed that the Chinese Academy of Medical Sciences had the largest volume of publications, whereas the Cleveland Clinic had the most influential ones. An author analysis showed Stanley L Hazen to have published the most and to also have been the most influential researcher. An analysis of all the journals publishing articles related to the search terms showed that PLoS One journal had the highest number of publications (18 articles), while Atherosclerosis journal had the most influential articles. The results of our reference analysis showed a strong association between Trimethylamine N-oxide and myocardial infarction. We found that increased intestinal permeability may be related to the progression of cardiovascular diseases, a high-fiber diet may help in the prevention of diseases such as myocardial infarction, and populations with a high intake of red meat may have an increased risk of myocardial infarction. Keyword analysis suggested that ‘cardiac fibrosis’ and ‘major bleeding’ were promising research directions in the future, and supplementing food intake with short-chain fatty acids was looked upon as a promising approach to treating coronary heart disease. Conclusion: The gut microbiota are closely related to myocardial infarction, and investigating this relationship is crucial for the prevention and treatment of myocardial infarction, where interdisciplinary research and international cooperation are indispensable.

Catechins, a class of polyphenolic compounds found in tea, have attracted significant attention due to their numerous health benefits, particularly for the treatment and protection of hypertension. However, the potential targets and mechanisms of action of catechins in combating hypertension remain unclear. This study systematically investigates the anti-hypertensive mechanisms of tea catechins using network pharmacology, molecular docking, and molecular dynamics simulation techniques. The results indicate that 23 potential anti-hypertensive targets for eight catechin components were predicted through public databases. The analysis of protein–protein interaction (PPI) identified three key targets (MMP9, BCL2, and HIF1A). KEGG pathway and GO enrichment analyses revealed that these key targets play significant roles in regulating vascular smooth muscle contraction, promoting angiogenesis, and mediating vascular endothelial growth factor receptor signaling. The molecular docking results demonstrate that the key targets (MMP9, BCL2, and HIF1A) effectively bind with catechin components (CG, GCG, ECG, and EGCG) through hydrogen bonds and hydrophobic interactions. Molecular dynamics simulations further confirmed the stability of the binding between catechins and the targets. This study systematically elucidates the potential mechanisms by which tea catechins treat anti-hypertension and provides a theoretical basis for the development and application of tea catechins as functional additives for the prevention of hypertension.

Summary: The incidence of premature myocardial infarction (PMI) has been rising and acute kidney injury (AKI) occurring in PMI patients severely impacts prognosis. This study aimed to develop and validate a prediction model for AKI specific to PMI patients. The MIMIC-Ⅲ-CV and MIMIC-Ⅳ databases were utilized for model derivation of PMI patients. Single-center data served for external validation. There were 571 and 182 AKI patients in the training set (n = 937) and external validation set (n = 292) cohorts, respectively. Finally, a 7-variable model consisting of: Sequential Organ Failure Assessment (SOFA) score, coronary artery bypass grafting (CABG), ICU stay time, loop diuretics, estimated glomerular filtration rate (eGFR) HCO3- and Albumin was developed, achieving an AUC of 0.85 (95% CI: 0.83–0.88) in the training set. External validation also confirmed model robustness. This model may assist clinicians in the early identification of patients at elevated risk for PMI. Further validation is warranted before clinical application.

Aspergillus flavus is notorious for contaminating food with its secondary metabolite—highly carcinogenic aflatoxins. In this study, we found that exogenous nitric oxide (NO) donor could influence aflatoxin production in A. flavus. Flavohemoglobins (FHbs) are vital functional units in maintaining nitric oxide (NO) homeostasis and are crucial for normal cell function. To investigate whether endogenous NO changes affect aflatoxin biosynthesis, two FHbs, FHbA and FHbB, were identified in this study. FHbA was confirmed as the main protein to maintain NO homeostasis, as its absence led to a significant increase in intracellular NO levels and heightened sensitivity to SNP stress. Dramatically, FHbA deletion retarded aflatoxin production. In addition, FHbA played important roles in mycelial growth, conidial germination, and sclerotial development, and response to oxidative stress and high-temperature stress. Although FHbB did not significantly impact the cellular NO level, it was also involved in sclerotial development, aflatoxin synthesis, and stress response. Our findings provide a new perspective for studying the regulatory mechanism of the development and secondary mechanism in A. flavus.

Mapping neuronal networks is a central focus in neuroscience. While volume electron microscopy (vEM) can reveal the fine structure of neuronal networks (connectomics), it does not provide molecular information to identify cell types or functions. We developed an approach that uses fluorescent single-chain variable fragments (scFvs) to perform multiplexed detergent-free immunolabeling and volumetric-correlated-light-and-electron-microscopy on the same sample. We generated eight fluorescent scFvs targeting brain markers. Six fluorescent probes were imaged in the cerebellum of a female mouse, using confocal microscopy with spectral unmixing, followed by vEM of the same sample. The results provide excellent ultrastructure superimposed with multiple fluorescence channels. Using this approach, we documented a poorly described cell type, two types of mossy fiber terminals, and the subcellular localization of one type of ion channel. Because scFvs can be derived from existing monoclonal antibodies, hundreds of such probes can be generated to enable molecular overlays for connectomic studies.

Aim: Estuarine vegetation routinely experiences natural tidal fluctuations and is highly vulnerable to extreme events such as heavy rainfall, leading to changes in plant population structure and adaptability. Genetic and epigenetic modifications are widely considered to be mechanisms of phenotypic variation, triggered in plants responding to extreme changing environments. However, understanding on correlations among genetic, epigenetic, and phenotypic variation of wild plant populations is still limited. Location: China. Methods: In this study, populations of the typical wetland clonal species Phragmites australis were selected from four estuaries along various latitudes in China. Genetic and epigenetic diversity and phenotypic variation of these populations were analysed. Results: Phenotypic variation of P. australis populations was the highest at Yellow River Estuary and the lowest at Min River Estuary. Across all estuaries, Genetic and epigenetic diversity was strongly linear‐correlated. Genetic diversity had significant correlations with variation in reproductive traits, whereas epigenetic diversity had significant correlations with variation in growth traits. Climatic factors of mean annual temperature and precipitation, as well as soil nitrogen and phosphorus, were negatively correlated with variation in genetic diversity, epigenetic diversity, and variation in reproductive traits of P. australis populations along latitudes. Variation in growth traits was negatively correlated with soil salinity, reflecting the limiting effect of salinity on plant growth. Main Conclusion: Our findings found that genetic and epigenetic variations may play different roles in phenotypic variation of P. australis populations along latitudes, the variation becomes greater when the climatic and edaphic conditions deteriorate. The findings shed new light on the adaptation and evolution of wetland plant populations along a large latitudinal scale, and may contribute to the revegetation of estuary wetlands. In order to solve the problem of image quality and morphological characteristics of primary underglaze brown decorative pattern extraction, this paper proposes a method of primary underglaze brown decorative pattern extraction based on the coupling of single scale gamma correction and gray sharpening. The single-scale gamma correction is combined with the gray sharpening method. The single-scale gamma correction improves the contrast and brightness of the image by nonlinear transformation, but may lead to the loss of image detail. Gray sharpening can enhance the high frequency component and improve the clarity of the image, but it will introduce noise. Combining these two technologies can compensate for their shortcomings. The experimental results show that this method can improve the efficiency of last element underglaze brown decorative pattern extraction by enhancing the image retention detail and reducing the influence of noise. The experimental results showed that F1Score, Miou(%), Recall, Precision and Accuracy(%) were 0.92745, 0.82253, 0.97942, 0.92458 and 0.92745, respectively. [ABSTRACT FROM AUTHOR]

High-efficiency electrocatalytic hydrogen evolution reaction (HER) is among the promising approaches for future low-cost hydrogen production. The cost and effectiveness challenges of electrocatalysts have significantly limited the development of electrochemical HER. Metal–organic framework (MOF) derivatives with tunable conformations and remarkable activity have shown unique capabilities in electrocatalytic hydrogen precipitation. Herein, we report a simple method for the synthesis of MOF-derived MoO2/NC with decorated Pt nanostructures. Thus, the synthesized MoO2/NC/Pt-20 nanostructures with a rod-like structure exhibit better pH-universal HER performance with overpotentials of 32, 53, and 70 mV in alkaline, acidic, and neutral conditions (10 mA·cm–2), respectively. Also, the electrocatalysts show low cell voltages of 1.58, 1.59, and 1.70 V at 10 mA·cm–2 for overall water splitting in alkaline, acidic, and neutral conditions, respectively. Characterized by long spans, low aspect ratios, and intricate construction sequences, super-large cross-section tunnels present substantial construction risks. Therefore, the selection of the optimal excavation method and construction sequence is crucial for ensuring the safety of tunnel construction and minimizing project costs. This paper takes a super large transverse-section highway tunnel as a case study, employing field monitoring data combined with ABAQUS software to analyze the stress and deformation of surrounding rock and support structures under different excavation methods. The findings reveal that the deformation of surrounding rock and support structures excavated by the Double-Side Drift Method is smaller than those caused by the three-benching seven-step method and the CRD excavation method. Nevertheless, the significant stresses of surrounding rock and support structures are released by the Double-Side Drift Method, leading to potential stress concentrations. Thus, it is necessary to ensure the rapid completion of early support and quick sealing of the tunnel. Furthermore, the sixth process achieves smaller deformation (including arch displacement and surface settlement) of the tunnel, a shorter construction period, and lower economic costs when compared to other construction processes. Consequently, it can obviously be concluded that both the Double-Side Drift Method and the sixth construction process stand out as the most appropriate choices for excavating super large cross-section tunnels. The zoning excavation method is fully employed to control the deformation of foundation pits constructed in urban soft soil areas. However, the similarities and differences in forces and deformations between foundation pits excavated by the zonal method and those excavated by the conventional method still need to be further explored. In this study, the deformation was monitored and analyzed by taking the zonal excavation of a foundation pit of the 'New World' project in Hangzhou City as the research object. The measured results showed that the pre-built diaphragm wall for the first excavated foundation pit restricted the deformation of the first excavated diaphragm wall. The presence of extensive construction and unloading activities also changed the deformation pattern of the soil. Further, finite element simulations were carried out. The simulation results revealed that excavating the foundation pit first caused displacements in the pre-built diaphragm wall. The displacements transmitted by non-adjacent pits through the pre-built diaphragm wall were small and were concentrated at the junction of the two sub-pits. Adjacent foundation pits caused large displacements of the pre-built diaphragm wall with similar displacement patterns. Phthalates are common endocrine disruptors. The placental barrier can be crossed by phthalates and may have a negative impact on the health of the fetus. However, the association between prenatal exposure to phthalates and birth size is still debatable. Here, we performed this meta-analysis to assess the relationship between prenatal phthalates exposure and birth size. Eighteen studies were finally included by searching PubMed, Embase, Scopus, Ovid, and Web of Science databases and standardized regression coefficients and standard errors were used to pool effect size. Our results showed that prenatal exposure to MMP (=-0.04, 95%CI: −0.08, −0.01) and MEP (=-0.01, 95%CI: −0.01, −0.002) was significantly associated with birth weight. However, no significant associations were identified for phthalate exposure with birth length, head circumference and chest circumference. Because the limiting of studies, more high-quality case–control studies or cohort studies are urgently needed to draw the best conclusions. [ABSTRACT FROM AUTHOR]
