Aggregation of protein-based therapeutics can occur during development, production, or storage and can lead to loss of efficacy and potential toxicity. Native mass spectrometry of a covalently linked pentameric monoclonal antibody complex with a mass of ∼800 kDa reveals several distinct conformations, smaller complexes, and abundant higher-order aggregates of the pentameric species. Charge detection mass spectrometry (CDMS) reveals individual oligomers up to the pentamer mAb trimer (15 individual mAb molecules; ∼2.4 MDa) whereas intermediate aggregates composed of 6-9 mAb molecules and aggregates larger than the pentameric dimer (1.6 MDa) were not detected/resolved by standard mass spectrometry, size exclusion chromatography (SEC), capillary electrophoresis (CE-SDS), or by mass photometry. Conventional quadrupole time-of-flight mass spectrometry (QTOF MS), mass photometry, SEC, and CE-SDS did not resolve partially or more fully unfolded conformations of each oligomer that were readily identified using CDMS by their significantly higher extents of charging. Trends in the charge-state distributions of individual oligomers provides detailed insight into how the structures of compact and elongated mAb aggregates change as a function of aggregate size. These results demonstrate the advantages of CDMS for obtaining accurate masses and information about the conformations of large antibody aggregates despite extensive overlapping m/z values. These results open up the ability to investigate structural changes that occur in small, soluble oligomers during the earliest stages of aggregation for antibodies or other proteins.

One of the primary hurdles in microdevice fabrication lies in ascertaining the most impactful tactics for adapting metal surfaces. Through a one-pot tackle and distinct mechanochemical reactions evoked by 15 min aqueous wet sand-milling (SM-15), we successfully grafted Mo-based metal–organic frameworks (Mo-MOFs) onto graphene oxides (GOs). Following this, a convenient and readily scalable methodology of electrophoretic deposition was implemented to create controllable thickness of SM-15 GOs@Mo-MOFs lubricating films, achieving considerable enhancements of 143% and 91% in hardness and Young's modulus, respectively, when compared to those of SM-15 Mo-MOFs. The successful synthesis of SM-15 GOs@Mo-MOFs was corroborated using strategies such as x-ray diffraction, Fourier transform infrared spectroscopy, and field emission scanning electron microscopy. Analyses using the micro-tribotester indicated that the new film exhibited a lowest friction coefficient of roughly 0.5 when imposed with a load of 5 N and sliding speed of 8 mm/s. In addition, the optical profiler nano-indentation in situ scanning probe microscope revealed that SM-15 GOs@Mo-MOFs films had smaller and shallower scratches and grooves compared to SM-15 Mo-MOFs ones. The calculated results of key descriptors (EHOMO, ELUMO, ΔE, etc.) in density functional theory quantitatively disclosed the interaction mechanisms between GOs@Mo-MOFs molecules and microdevices. We first scrutinized the innate properties of molecule adsorption energy and frictional mechanical behaviors using synergetic cross-scale simulations, such as Monte Carlo and finite element methods. Background: Plants that contain brewing microorganisms are used in traditional fermentation starters, which are an essential part of local diet, nutrition, life, and health. Regionally, the plant species used and the microorganisms included in traditional fermentation starters are diverse, endowing local fermented drinks with different flavors and health benefits. However, related traditional knowledge has been scarcely documented or revealed. Methods: An ethnobotanical survey was conducted in five towns of Nayong County in northwestern Guizhou, China. Snowball sampling, semi-structured interviews, free lists, and participatory observation were used to collect information on Jiuqu Plants (JPs) and jiuqu-making techniques. The PacBio platform was used to study the microbial community structure and diversity in the Chuanqing people's jiuqu. Results: In total, 225 informants were interviewed, including 116 who provided plants and technological processes for making Chinese baijiu jiuqu (CBJ) and 139 who provided information about making fermented glutinous rice jiuqu (FGRJ). This study found that older people have more abundant knowledge about CBJ plants. Poaceae was found to be the dominant family used in making CBJ and FGRJ (7 species each). Compared to individual plant parts, the whole plant is most commonly used in two kinds of jiuqu (19.5% in CBJ and 22.6% in FGRJ). The Chuanqing people's jiuqu is used to treat dietary stagnation and indigestion. The highest relative frequency of citation of the CBJ plant was Ficus tikoua Bureau, and the counterpart of the FGRJ plant was Buddleja macrostachya Benth. The dominant bacterial species in jiuqu were Gluconobacter japonicus (YQ1, YQ4) and Pediococcus pentosaceus (YQ2, YQ3), and the dominant fungal species was Rhizopus oryzae. Conclusion: For the first time, this study documents the unique traditional jiuqu knowledge and reveals the microbial mystery behind the FGRJ of the Chuanqing people. Therefore, this study encourages the use of online social media platforms in order to spread Jiuqu culture, the use of the new media wave in order to create multimedia databases, and also suggests that local communities should develop preservation intervention programs, in addition to nurturing the inheritors in order to prevent the disappearance of traditional Jiuqu knowledge. This research contributes to the conservation and demystification of the traditional jiuqu knowledge of the Chuanqing people and lays the foundation for further research on its microbiology, nutrition, and metabolomics. Background Hemoglobin (Hb) Hekinan II (A27; Glu-Asp) is an α-chain variant, and its interaction with the common Southeast Asian (--SEA/) α-thalassemia (α-thal) deletion is rarely reported. This study provides a clinical update of Hb Hekinan II associated with (--SEA/) α-thal. Methods A total of 11 simple heterozygotes and 20 composite heterozygotes for Hb Hekinan II and (--SEA/) α-thal were included based on molecular diagnosis. Results Hb Hekinan II exhibited a significant increase in hemoglobin, mean corpuscular volume, and mean corpuscular hemoglobin content, but a decrease in red blood cell level compared with α+ thalassemia deletion. Compared with (--SEA/) α-thal, composite heterozygotes for Hb Hekinan II and (--SEA/) α-thal showed similar erythrocyte parameters. Both heterozygotes with and without (--SEA/) α-thal showed low Hb A2 level. Hb Hekinan II showed abnormal performance in high-performance liquid chromatography but not in capillary electrophoresis. Conclusion Hb Hekinan II is a benign Hb variant. The heterozygotes exhibit clinically asymptomatic coinheritance with (--SEA/) α-thal having comparable hematological phenotype to simple (--SEA/) α-thal. Objectives Voxelotor can increase hemoglobin levels in patients living with sickle cell disease (SCD). A clinician who is monitoring voxelotor response may want to know whole-blood voxelotor concentration, but this cannot be measured in most clinical settings. However, voxelotor has been demonstrated to cause "peak splitting" in common methods of hemoglobin measurement such as capillary zone electrophoresis (CZE) and high-performance liquid chromatography (HPLC). We hypothesized that we could use the size of the peak split to estimate the whole-blood concentration. Methods Blood from people with SCD was dosed with known concentrations of voxelotor, and multiparameter regression was used to derive the relationship of voxelotor concentration to the degree of peak splitting observed. To validate these equations, 21 patients started on voxelotor at 1500 mg/d had blood samples drawn at days 0, 14, 30, and 60. Samples were sent out for gold standard voxelotor concentration testing. The derived equations were then used to calculate voxelotor concentration. Results Calculated concentrations correlated strongly with measured concentrations for both CZE (R 2 = 0.83, P <.001) and HPLC (R 2 = 0.76, P <.001). Voxelotor concentration also had a significant effect on increases in hemoglobin (R 2 = 0.40, P <.001). Conclusions Thus, peak splitting CZE and HPLC can be used to estimate voxelotor concentration. This work was devoted to a study of the composition of protein concentrates from amaranth grain (Amaranthus hypochondriacus L.) of variety Voronezh. Amaranth protein concentrates were obtained by alkaline extraction of proteins and neutralization of the solution followed by ultrafiltration, by separating the starch fraction with amylolytic enzymes, and by alkaline extraction of proteins and their precipitation at pH 4.5. Conditions for the extraction of proteins followed by chromatography–mass spectrometric analysis and identification were selected. It was found that proteins from amaranth grain were more effectively extracted with a buffer with urea at protein concentrations of 1.7, 1.9, and 2.9 mg/cm3 in solution, respectively, while a buffer with detergents was more effective for the extraction of low-molecular-weight proteins at protein concentrations of 4.9, 2.9, and 9.0 mg/cm3 in solution, respectively. As a result of HPLC–MS/MS analysis followed by identification and search in the UNIPROT database, it was established that the main protein of amaranth grain is 11S-globulin, which acts as a reserve protein of amaranth seeds. In amaranth concentrates, 14 unique proteins belonging only to A. hipochondriacus L. were identified, and also proteins that did not belong to this species were reliably identified. Based on the results of semiquantitative analysis of the peptide profile of amaranth grain protein concentrates, a high frequency of occurrence of the main 11S-globulin proteins was established in all samples. The frequency of occurrence of other proteins in samples obtained by different methods differed significantly due to the peculiarities of protein isolation from amaranth grain. A general expression is derived for the Laplace transform of the time dependent transient electrophoretic mobility of spherical colloidal particles in a multi-particle suspension when a step external electric field is suddenly applied to the colloidal suspension. From the general mobility expression, we obtain a simple closed form expression for the Laplace transform of the time-dependent transient electrophoretic mobility of weakly charged spherical colloidal particles in an electrolyte solution. The obtained Laplace transform of the transient electrophoretic mobility of particles does not involve numerical integrations so that its inverse Laplace transform is easily obtained using calculation software. Looking at the development status of Nigeria and other developing nations, most low-income and rural households often use coal as a source of energy which necessitates its trade very close to the communities. Moreover, the effects of exposure to coal mining activities are rarely explored or yet to be studied, not to mention the numerous street coal vendors in Nigeria. This study investigated the oxidative stress levels in serum and urine through the biomarker 8-OHdG and DNA damage via single cell gel electrophoresis (alkaline comet assay). Blood and urine levels of 8-OHdG from 130 coal vendors and 130 population-based controls were determined by ELISA. Alkaline comet assay was also performed on white blood cells for DNA damage. The average values of 8-OHdG in serum and urine of coal vendors were 22.82 and 16.03 ng/ml respectively, which were significantly greater than those detected in controls (p < 0.001; 15.46 and 10.40 ng/ml of 8-OHdG in serum and urine respectively). The average tail length, % DNA in tail and olive tail moment were 25.06 μm, 18.71% and 4.42 respectively for coal vendors. However, for controls, the average values were 4.72 μm, 3.63% and 1.50 for tail length, % DNA in tail and olive tail moment respectively which were much lower than coal vendors (p < 0.001). Therefore, prolonged exposure to coal dusts could lead to higher serum and urinary 8-OHdG and significant DNA damage in coal vendors observed in tail length, % DNA in tail, and olive tail moment by single cell gel electrophoresis. It is therefore established that coal vendors exhibit a huge risk from oxidative stress and assessment of 8-OHdG with single cell gel electrophoresis has proven to be a feasible tool as biomarkers of DNA damage. Peripheral neuropathy is a prevalent complication in plasma cell disorders, posing significant diagnostic and therapeutic challenges. This study presents three cases initially diagnosed with chronic inflammatory demyelinating polyneuropathy (CIDP). Despite initial symptom regression post-immunomodulatory treatment, the patients exhibited progressive neurological deficits. Advanced laboratory evaluation confirmed monoclonal protein presence, yet traditional diagnostic methods, including bone marrow biopsy and flow cytometry, yielded normal results. Utilizing 18F-FDG PET/CT, we identified multiple hypermetabolic vertebral lesions, which upon biopsy, confirmed the diagnosis of plasmacytoma. Our findings underscore the utility of PET/CT as a reliable diagnostic tool for monoclonal gammopathy associated neuropathy, advocating for its consideration in cases with equivocal diagnosis. The red V2O5 sol was prepared by facile alternating stirring and ultrasonic dispersion route. A V2O5 film fabricated on ITO-PET by electrophoresis method was used to demonstrate a flexible electrochromic device. The effects of applied voltage and electrophoresis time for V2O5films were investigated in detail. When the electrophoresis time was kept 30 s, the applied voltage was above 3 V to deposit uniform V2O5-PET film. The electrophoresis time could more significantly regulate the deposition of V2O5 on ITO-PET film. A flexible electrochromic device with "sandwich" structure was fabricated using V2O5 film, and its electrochromic performance of the device was evaluated. The flexible V2O5 electrochromic devices show the multicolor electrochromic performance of yellow, yellow-green, green and orange-red. The coloration efficiency values of V2O5 devices were 21.6 cm2C − 1 (from initial yellow to green) and 26.9 cm2C − 1 (from green to orange-red), respectively, and the highest transmittance modulation range was 51% at 741 nm. The ion steric repulsion and ion-solvent interactions, due to the finite size of ions, are analyzed for electrophoresis of a droplet in a non-dilute electrolyte in which the ionic volume fraction is O (0.01). In this study, the viscosity of the suspension medium is considered to be dependent on the local ionic volume fraction. The electrochemical potential of the finite-sized ions is modified to take into account the steric interactions. The impact of the Marangoni stress due to the non-uniform distribution of the non-ionized surfactant is taken into account. Governing equations are solved under a weak applied field consideration. At a lower range of surface charge density, the mobility is found to increase with the droplet viscosity, in which the Marangoni stress is found to enhance the mobility. The counterion saturation created by the ion steric interaction attenuates the screening of the surface charge and hence, enhances the mobility. However, the enhanced viscosity of the medium for the non-dilute electrolyte reduces the mobility and this reduction is higher in a monovalent electrolyte as compared to a multivalent electrolyte. The standard model shows a singularity in mobility when the surface potential is varied. 1,2-propylene glycol and its 50-50 w/w mixture with water were used to prepare heat transfer fluids based on amino-functionalized silica. On top of pH-neutral dispersions (no reagents added except for the solvent and the particles), dispersions acidified with acetic acid and with HCl were used to enhance the positive electric charge of silica particles. The colloidal particles had a positive zeta potential >40 mV and showed apparent particle radii of 70 nm, and these properties remained unchanged on heating up to 80 °C for up to 28 days. The glycolate oxidase enzyme, a core photorespiratory enzyme in green plants, oxidizes glycolate to glyoxylate, playing an adaptive role in roots during hypoxia to normoxia transition. The present study aimed to purify glycolate oxidase enzymes from peas (Pisum sativumL.) and sorghum (Sorghum sudanense J.), and investigate their physicochemical and regulatory properties. The activity of one of the glycolate oxidase isoforms (LCO-like glycolate oxidase) was induced in pea and sorghum leaves. The enzymes were purified to an electrophoretically homogeneous condition. The purified enzymes' specific activity, molecular mass, and physicochemical characteristics were determined. The results showed that the LCO-like glycolate oxidase enzyme from pea leaves was 314.37E/mg proteins, the degree of purification was 207 times, and the yield was 11.5%. The sorghum leaf enzyme was 274.22E/mg proteins with the degree of purification and yield, estimated at 143 times and 13.8%, respectively. The molecular mass of the native enzymes was 182 and 168kDa for peas and sorghum, respectively. It was revealed that the enzyme consisted of four subunits with a molecular mass of 45 and 42kDa for glycolate oxidase from pea and sorghum leaves, respectively, indicating a homo-tetramer. The sorghum leaf enzyme exhibited a higher affinity for lactate substrate with an optimal temperature of 55°C and pH of 7.2. Meanwhile, the optimal temperature and pH of pea leaf enzyme were 50°C and 7.5, respectively. The availability of homogeneous specimens provides opportunities to investigate the role of LCO-like glycolate oxidase in the adaptive reactions of cellular plant metabolism. [ABSTRACT FROM AUTHOR]

A general theory is developed for the time dependent transient electrophoretic mobility of spherical colloidal particles in a salt-free liquid medium containing only counterions when a step external electric field is suddenly applied to the colloidal suspension. It is found that as in the case of the steady electrophoretic mobility in a salt-free medium, there is a certain critical value of the particle surface charge separating two cases, that is, the low-surface-charge case and the high-surface-charge case. In the latter case the counterion condensation takes place near the particle surface. For the low-surface charge case, the transient electrophoretic mobility agrees with that of a sphere in an electrolyte solution in the limit of very low electrolyte concentrations. For the high-surface-charge case, however, the transient mobility becomes independent of the particle surface charge because of the counterion condensation effects. A simple expression is derived for the ratio of the transient electrophoretic mobility to the steady electrophoretic mobility, which is found to take the same form irrespective of the magnitude of the particle surface charge. AbstractValproic acid is an antiepileptic drug associated with skin-related issues like excessive hair growth, hair loss, and skin rashes. In contrast, Moringa oleifera, rich in nutrients and antioxidants, is gaining popularity worldwide for its medicinal properties. The protective properties of M. oleifera extract against skin-related side effects caused by valproic acid were investigated. Female rats were divided into control groups and experimental groups such as moringa, sodium valproate, and sodium valproate + moringa groups. A 70% ethanolic extract of moringa (0.3 g/kg/day) was given to moringa groups, and a single dose of sodium valproate (0.5 g/kg/day) was given to valproate groups for 15 days. In the skin samples, antioxidant parameters (such as glutathione, glutathione-S-transferase, superoxide dismutase, catalase, and total antioxidant capacity), as well as oxidant parameters representing oxidative stress (i.e. lipid peroxidation, sialic acid, nitric oxide, reactive oxygen species, and total oxidant capacity), were examined. Additionally, boron, hydroxyproline, sodium-potassium ATPase, and tissue factor values were determined. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis was also carried out for protein analysis in the skin samples. The results showed that moringa could increase glutathione, total antioxidant capacity, sodium-potassium ATPase, and boron levels, while decreasing lipid peroxidation, sialic acid, nitric oxide, total oxidant capacity, reactive oxygen species, hydroxyproline, and tissue factor levels. Introduction: Transient hypercalcaemia due to teriparatide occurs in up to 11% of patients though delayed hypercalcaemia (> 24 h post injection) is rare. We report the case of a female who developed significant delayed hypercalcaemia after teriparatide treatment for osteoporosis and review other cases in the literature to date. Case report: A 72-year-old female on teriparatide for the treatment of osteoporosis was found to have hypercalcaemia (3.30 mmol/l) on routine testing approximately 3 months after starting therapy. Serum calcium pretreatment was normal at 2.39 mmol/l. She was admitted to the hospital for investigations which identified a serum 25-hydroxyvitamin D of 94 nmol/l, a low parathyroid hormone of 6.0 pg/ml, and normal test results for 1,25 dihydroxyvitamin D (115 pmol/l), parathyroid hormone-related peptide (< 1.4 pmol/ml), serum electrophoresis and angiotensin-converting enzyme (39 IU/l). CT abdomen, pelvis, and thorax revealed no evidence of malignancy and an isotope bone scan ruled out skeletal metastases. Serum calcium normalised (2.34 mmol/l) several days after stopping teriparatide and calcium supplements and administering intravenous fluid. On restarting teriparatide, delayed hypercalcaemia reoccurred and treatment was switched to denosumab. Discussion: Delayed moderate to severe hypercalcaemia (serum calcium > 3.0 mmol/l) due to teriparatide is rare but may lead to therapy withdrawal. BACKGROUND: Adolescent idiopathic scoliosis is a complex condition whose pathogenesis may include inflammation and signs of joint and bone degeneration. OBJECTIVE: The main objective of this study is to evaluate the relationship between the severity of adolescent idiopathic scoliosis and inflammatory blood parameters. METHODS: The study recruited patients with adolescent idiopathic scoliosis who attended the Rehabilitation Center of the Apostolo Foundation in Merate (LC). The scoliosis curve (Cobb's angle) was used as a severity index to compare with inflammatory blood parameters (white blood cells subpopulations, immunoglobulins, protein electrophoresis). In addition, the study used an overall severity grading called "Scoliosis Score" which includes all spine angles and Risser's score (bone development index). RESULTS: Thirty-four subjects were recruited (mean age 14 years, 2 months), 30 females and 2 males. A significant correlation was found between Cobb's angle and the percentage values of beta-2 globulins in a directly proportional manner (r = 0.42, p = 0.01), and gamma globulins in an inversely proportional manner (r = - 0.366, p = 0.04). However, no significant correlation between Cobb's angle and the absolute values of white blood cells and percentage subpopulations was found (r = 0.0821 p = 0.655). A moderate, inverse correlation was found between the Scoliosis Score and the percentage of neutrophils (r = - 0.385, p = 0.02), a direct correlation was found between the Scoliosis Score and the percentage of lymphocytes (r = 0.404, p = 0.02). In addition, there was a strong correlation of the Scoliosis Score with alpha-2 globulin (r = 0.564, p = 0.0012), beta-1 globulin (r = 0.478, p = 0.0074), and beta-2 globulin (r = 0.370, p = 0.044) and an inverse relationship with gamma globulin (r = - 0.625, p = 0.0002). The main correlations were confirmed by regression analysis. CONCLUSION: The correlation between beta-2 globulins and gamma globulins with Cobb's angle and the Scoliosis Score suggests a link between spinal curvature and inflammation in scoliosis patients, This link may indicate the significance of these parameters for diagnosing, staging the disease, and monitoring therapies. Dental caries is a multifactorial chronic disease resulting from the intricate interplay among acid‐generating bacteria, fermentable carbohydrates, and several host factors such as saliva. Saliva comprises several proteins which could be utilized as biomarkers for caries prevention, diagnosis, and prognosis. Mass spectrometry‐based salivary proteomics approaches, owing to their sensitivity, provide the opportunity to investigate and unveil crucial cariogenic pathogen activity and host indicators and may demonstrate clinically relevant biomarkers to improve caries diagnosis and management. The present review outlines the published literature of human clinical proteomics investigations on caries and extensively elucidates frequently reported salivary proteins as biomarkers. This review also discusses important aspects while designing an experimental proteomics workflow. The protein–protein interactions and the clinical relevance of salivary proteins as biomarkers for caries, together with uninvestigated domains of the discipline are also discussed critically. Electronic paper displays rely on electrokinetic effects in nonpolar solvents to drive the displacement of colloidal particles within a fluidic cell. While Electrophoresis (EP) is a well-established and frequently employed phenomenon, electro-osmosis (EO), which drives fluid flow along charged solid surfaces, has not been studied as extensively. We hypothesize that by exploiting the interplay between these effects, an enhanced particle transport can be achieved. In this study, we experimentally investigate the combined effects of EP and EO for colloidal particles in non-polar solvents, driven by an electric field. We use astigmatism micro-particle tracking velocimetry (A- μ PTV) to measure the motion of charged particles within model fluidic cells. Using a simple approach that relies on basic fluid flow properties we extract the contributions due to EP and EO, finding that EO contributes significantly to particle transport. The validity of our approach is confirmed by measurements on particles with different magnitudes of charge, and by comparison to numerical simulations. We find that EO flows can play a dominant role in the transport of particles in electrokinetic display devices. This can be exploited to speed up particle transport, potentially yielding displays with significantly faster switching times. Chirality is a fundamental and ubiquitous property of nature involved in multiple fields of science. In particular, the possible resolution of the enantiomeric forms of a molecule is crucial in the pharmaceutical, food, and agrochemical industries. The search for efficient, broad-spectrum, and yet simple methods for obtaining enantiomerically pure substances is a current challenge. Enantioselective resolution methods rely on an asymmetric environment that allows the two antipodes of a chiral molecule to be distinguished. In addition to enantiomeric separation techniques, such as chromatography and electrophoresis, new promising approaches involving out-of-the-scheme synergistic effects between chiral selectors (CS) and external stimuli are emerging. Pregnancy has its own protein dynamics, reflecting the hormonal profile. Quantitative and qualitative changes in plasma protein profile may provide useful information about this condition. Any alterations may be a signal heralding clinical or subclinical pathology. The objective of our study was to compare the plasma protein profile between selected months of pregnancy in cows for a better understanding gestation course. For this purpose, we collected blood from healthy pregnant (n = 30; n = 6 for each pregnancy stage) and non‐pregnant (C; n = 6) Holstein‐Friesian cows during a routine veterinary examination. Collected samples were selected according to pregnancy month (first, second, third, sixth, and ninth), prepared, and separated by two‐dimensional electrophoresis. The Delta‐2D program compared and statistically evaluated scanned gel images from the appropriate months. The mean volume of the spots was considered. The MALDI TOF/TOF spectrometer was used to identify statistically significant proteins. There were 11 distinct proteins found, including peptidyl‐prolyl cis‐trans isomerase F, oligoribonuclease, and PRELI domain‐containing protein 3B (all of them have the lowest abundance in the C group), alpha‐1B‐glycoprotein, L‐gulonolactone oxidase, hemopexin (first month with higher abundance than control), alpha‐2‐HS‐glycoprotein (significantly higher abundance in the first month than in remaining groups), ermin (absent in the first month and lower abundance in the third and sixth months than in the remaining groups and control), endophilin‐A2 (significant differences between the control and the second, third, sixth, and ninth months), apolipoprotein A‐I (significant difference between control and the first and sixth months), alpha‐1‐antiproteinase (significant difference between control and the ninth month). The fractions obtained from low temperature-induced crystallisation of ethanolic extracts of green-and red-oak leaf and okra seed were accounted for total polyphenol content and the DPPH antioxidant activity, resulting in 5.99, 4.44, and 12.35 mg gallic acid equivalent per g sample, and 0.45, 0.35, and 0.99 mg Trolox equivalent per g sample, respectively. Insulin resistance was the result after incubating C2C12 skeletal muscle cells in high glucose DMEM for 20 h. Oxidative stress and inflammation were triggered by re-incubating these resistant cells with insulin. The expression of p-p38 MAPK and NF-κB proteins and the NF-B p65 transcription factor activity were enhanced. Such inflammatory protein markers were reduced when the crystalised extracts replaced insulin. Increased catalase activity and NO production were also determined for the incubations using the crystalised extracts. It was suitable to include these vegetables in the daily diets of prediabetic individuals for better management of the patient’s health status by increasing insulin sensitivity and decreasing inflammation. Background: Isatuximab, an IgG-kappa (IgGκ) anti-cluster of differentiation 38 (CD38) monoclonal antibody approved for use in patients with relapsed or refractory multiple myeloma (MM), can potentially interfere with the visualization of endogenous monoclonal protein (M-protein) on standard immunofixation electrophoresis (IFE) and lead to inaccurate classification of a patient's response to therapy. The Hydrashift 2/4 isatuximab IFE assay (Hydrashift isatuximab assay) removes isatuximab interference from IFE. Using samples from patients enrolled in clinical trials of isatuximab-based therapy for MM, we demonstrate how the Hydrashift isatuximab assay improves the ability to detect residual M-protein and offer recommendations for when the assay is most useful. Methods: Samples from 141 patients with a variety of known M-protein isotypes were selected and analyzed by standard IFE and the Hydrashift isatuximab assay. A positive control containing isatuximab was run on every standard IFE and Hydrashift gel. Results: The Hydrashift isatuximab assay reliably shifted the migration of isatuximab in patient samples. Standard IFE was adequate for determining 104 patients' M-protein status, and the Hydrashift isatuximab assay confirmed these results. In samples from 37 patients with a history of IgGκ MM and a single IgGκ band visible on standard IFE near the isatuximab migration site, the Hydrashift isatuximab assay was able to separate isatuximab from endogenous M-protein, identifying residual M-protein in 17 samples and preventing false-positive interpretations of standard IFE in 20 samples. Conclusions: The Hydrashift isatuximab assay is most useful in patients with known IgGκ MM when a single IgGκ band appears near the isatuximab migration site on standard IFE during isatuximab-based therapy. Introduction: Since its discovery in 1961, methicillin-resistant Staphylococcus aureus (MRSA) has been recognized as a significant healthcare-associated pathogen (HA-MRSA) and a notorious 'superbug'. Typing is crucial for surveillance, epidemiology analysis, infection control of MRSA and sequencing of the spa gene is one of the most common methods used for determining the origin of this bacterium in humans and animals. This research aimed to determine the antibiotic resistance and spa type of S. aureus strains collected from outpatients in two hospitals in the Wasit province of Iraq. Material & Methods: The study analyzed 200 outpatient MRSA isolates by collecting nasal and sputum samples from patients. Standard biochemical and molecular methods based on the nuc gene were used to identify S. aureus bacteria and amplify the mecA and spa genes. The Kirby-Bauer disc diffusion method was employed to determine the antibiotic sensitivity of the isolates using penicillin, cefoxitin, vancomycin, gentamicin, erythromycin, tetracycline, imipenem, clindamycin, chloramphenicol and rifampicin. Results: Methods. The prevalence of MRSA was more common in women than in men. Antibiogram results showed that most of the isolates were resistant to penicillin (94.2%) and sensitive to imipenem (100%), clindamycin (100%), and chloramphenicol (100%). Of these 35 isolates, 30 (87.5%) and 26 strains (74.3%) were positive for the mecA and spa genes. Typing based on spa gene sequencing revealed four different patterns: t386, t3579, U0002 and U0234. Conclusion: Variations in the spa gene among different S. aureus isolates may he of clinical importance when treating staphylococcal infections. Electric film heaters (EFHs) applied on flat steel substrates show promising potential in the production of heating devices. The basis for manufacturing EHFs is heat-resistant steel, the surface of which is coated with a heat-resistant electrical insulation coating. These coatings are glass crystalline and are applied mainly by screen printing. Compared to screen printing, the electrophoretic method of coating is more productive. In this work, glass in the MgO-CaO-B2O3-SiO2 oxide system was chosen for obtaining glass crystalline coatings. The paper provides a description of the material composition and the method used to prepare a suspension for epy electrophoretic deposition of coating. The main technological parameters for forming electrophoretic coatings with a specific thickness are determined based on experimental results. Utilizing calculated data on the properties of the selected glass, the study substantiates the most rational temperature and time regime for coating firing. These conditions promote crystallization and result in the production of heat-resistant coatings with sufficient electrical insulating properties for steel substrates of EFHs. [ABSTRACT FROM AUTHOR]

The objective of this research was to identify the morphology and molecular profile of crab caught from the coast of Tambala Village, Tombariri District, Minahasa Regency, North Sulawesi. Crabs were collected from the intertidal zone at night during the lowest tide. Crabs were morphological identified by description of shape, color and size of the carapace. Molecular identification was done through DNA barcoding including DNA extraction, amplification of the cytochrome c oxidase subunit I (COI) gene and electrophoresis. Morphological and genetic analysis identified the crab species as Metopograpsus oceanicus.

Background: Deoxyribonucleic acid (DNA) molecules can be effectively visualized when stained and observed under ultraviolet light during the electrophoresis process. The commonly used dye Ethidium bromide (EtBr) is considered hazardous due to its potential to cause mutations, cancer, and congenital disabilities. Various alternative dyes have been reported, one of which is hematoxylin. Hematoxylin compounds do not have mutagenic potential and are easier to apply than EtBr. However, there is no optimal variation in concentration and duration of staining for clear and effective visualization of DNA bands. Purpose: To find the concentration and staining time of harris hematoxylin staining for DNA bands from agarose gel electrophoresis. Method: An experimental method with a group comparison statistical design. The amplified DNA I6S rRNA gene from Escherichia coli (584 bp), which had undergone electrophoresis, was stained using harris hematoxylin dye at 0.01%; 0.02%; 0.03%; 0.04%; and 0.05% concentrations and immersion times soaking times of 10, 15, 20, and 30 minutes variations. The intensity of the DNA bands was analyzed using ImageJ. The staining power of the experimental groups was compared to the intensity of control dye and given a grading score of 1 - 4. The experiment was repeated twice, and the mean grading score was calculated. The highest mean value was considered the most optimal value. Result: A concentration of 0.02% showed relatively constant staining intensity for each soaking time. A mean value of 3.5 was obtained for a 0.01% concentration for 15 minutes. A 0.03% and 0.04% concentrations for 20 minutes. Conclusion: The highest mean value of 4 was obtained for Harris hematoxylin at 0.05% for 15 minutes.

The effect of four meat thawing techniques including chill temperature, room temperature, tap water and microwave on quality characteristics, lipid oxidation, proteins degradation, microbial spoilage and the microstructure of beef longissimus lumborum muscle were compared. A fresh portion of longissimus lumborum muscle was obtained from eight beef carcasses after 24 h postmortem. The findings indicated that all of the thawed samples, particularly the microwave beef samples, had a markedly worse quality than those of fresh meat. Among the thawing methods, changes in the beef quality of the chill temperature samples were significantly less. The increases in malondialdehyde value and decreases in free thiol content indicated that proteins and lipids in the beef samples subjected to room temperature, tap water, and microwave thawing methods were oxidized while thawed beef at a chill temperature did not significantly affect the lipid-protein oxidation compared to fresh beef. The greater myosin degradation was observed in room temperature, tap water and microwave thawing treatments, while actin degradation was not affected. Higher growth of total bacterial counts was presented by samples subjected to tap water and at room temperature thawing compared to microwave and chill temperature thawing. The sensory traits of the chiller thawing method were better than those of other methods. Images from scanning electron microscopy demonstrated that the myofibril configurations in the thawing samples were more loose than in the fresh beef samples, which had a compact and organized structure. This was demonstrated by the thawing samples' evidently higher myofibril gap value.

Abstract Introduction Patients with psoriatic arthritis have some lipid metabolism changes and higher risk of metabolic syndrome (MetS) and cardiovascular diseases, regardless of traditional risk factors, suggesting that chronic inflammation itself plays a central role concerning the atherosclerosis. However, there is a lack of information regarding atherogenic pattern and lipoprotein subfractions burden in these individuals. Aim To evaluate the HDL and LDL-cholesterol plasmatic levels and their subfractions after a nutritional intervention in patients with psoriatic arthritis (PsA). Methods This was a randomized, placebo-controlled clinical trial of a 12-week nutritional intervention. PsA patients were randomly assigned to 1-Placebo: 1 g of soybean oil daily, no dietetic intervention; 2-Diet + Supplementation: an individualized diet, supplemented with 604 mg of omega-3 fatty acids, three times a day; and 3-Diet + Placebo: individualized diet + 1 g of soybean oil. The LDL subfractions were classified as non-atherogenic (NAth), atherogenic (Ath) or highly atherogenic (HAth), whereas the HDL subfractions were classified as small, medium, or large particles, according to the current recommendation based on lipoproteins electrophoresis. Results A total of 91 patients were included in the study. About 62% of patients (n = 56) had an Ath or HAth profile and the main risk factors associated were male gender, longer skin disease duration and higher BMI. Thirty-two patients (35%) had a high-risk lipoprotein profile despite having LDL plasmatic levels below 100 mg/dL. The 12-week nutritional intervention did not alter the LDL subfractions. However, there were significant improvement of HDL subfractions. Conclusion Recognizing the pro-atherogenic subfractions LDL pattern could be a relevant strategy for identifying PsA patients with higher cardiovascular risk, regardless total LDL plasmatic levels and disease activity. In addition, a short-term nutritional intervention based on supervised and individualized diet added to omega-3 fatty acids changed positively the HDLLARGE subfractions, while LDLLARGE subfraction was improved in hypercholesterolemic individuals. The study of genetic resources using prolamin polymorphism in wheat cultivars from countries with different climatic conditions makes it possible to identify and trace the preference for the selection of the alleles of gliadine-coding loci characteristic of specific conditions. The aim of the study was to determine the “gliadin profile” of the collection of common wheat (Triticum aestivum L.) from breeding centers in Russia and Kazakhstan by studying the genetic diversity of allelic variants of gliadin-coding loci. Intrapopulation (μ ± Sμ) and genetic (H) diversity, the proportion of rare alleles (h ± Sh), identity criterion (I) and genetic similarity (r) of common wheat from eight breeding centers in Russia and Kazakhstan have been calculated. It has been ascertained that the samples of common wheat bred in Kostanay region (Karabalyk Agricultural Experimental Station, Kazakhstan) and Chelyabinsk region (Chelyabinsk Research Institute of Agriculture, Russia) had the highest intrapopulation diversity of gliadin alleles. The proportion of rare alleles (h) at Gli-B1 and Gli-D1 loci was the highest in the wheat cultivars bred by the Federal Center of Agriculture Research of the South-East Region (Saratov region, Russia), which is explained by a high frequency of occurrence of Gli-B1e (86 %) and Gli-D1a (89.9 %) alleles. Based on identity criterion (I), the studied samples of common wheat from different regions of Kazakhstan and Russia have differences in gliadincoding loci. The highest value of I = 619.0 was found when comparing wheat samples originated from Kostanay and Saratov regions, and the lowest I = 114.4, for wheat cultivars from Tyumen and Chelyabinsk regions. Some region-specific gliadin alleles in wheat samples have been identified. A combination of Gli-A1f, Gli-B1e and Gli-Da alleles has been identified in the majority of wheat samples from Kazakhstan and Russia. Alleles (Gli-A1f, Gli-A1i, Gli-A1m, Gli-A1o, Gli-B1e, Gli-D1a, Gli-D1f, Gli-A2q, Gli-B2o, and Gli-D2a) turned out to be characteristic and were found with varying frequency in wheat cultivars in eight regions of Russia and Kazakhstan. The highest intravarietal polymorphism (51.1 %) was observed in wheat cultivars bred in Omsk region (Russia) and the lowest (16.6 %), in Pavlodar region (Kazakhstan). On the basis of the allele frequencies, a “gliadin profile” of wheat from various regions and breeding institutions of Russia and Kazakhstan was compiled, which can be used for the selection of parent pairs in the breeding process, the control of cultivars during reproduction, as well as for assessing varietal purity.

Abstract Background Electrotransfection is based on application of high-voltage pulses that transiently increase membrane permeability, which enables delivery of DNA and RNA in vitro and in vivo. Its advantage in applications such as gene therapy and vaccination is that it does not use viral vectors. Skeletal muscles are among the most commonly used target tissues. While siRNA delivery into undifferentiated myoblasts is very efficient, electrotransfection of siRNA into differentiated myotubes presents a challenge. Our aim was to develop efficient protocol for electroporation-based siRNA delivery in cultured primary human myotubes and to identify crucial mechanisms and parameters that would enable faster optimization of electrotransfection in various cell lines. Results We established optimal electroporation parameters for efficient siRNA delivery in cultured myotubes and achieved efficient knock-down of HIF-1α while preserving cells viability. The results show that electropermeabilization is a crucial step for siRNA electrotransfection in myotubes. Decrease in viability was observed for higher electric energy of the pulses, conversely lower pulse energy enabled higher electrotransfection silencing yield. Experimental data together with the theoretical analysis demonstrate that siRNA electrotransfer is a complex process where electropermeabilization, electrophoresis, siRNA translocation, and viability are all functions of pulsing parameters. However, despite this complexity, we demonstrated that pulse parameters for efficient delivery of small molecule such as PI, can be used as a starting point for optimization of electroporation parameters for siRNA delivery into cells in vitro if viability is preserved. Conclusions The optimized experimental protocol provides the basis for application of electrotransfer for silencing of various target genes in cultured human myotubes and more broadly for electrotransfection of various primary cell and cell lines. Together with the theoretical analysis our data offer new insights into mechanisms that underlie electroporation-based delivery of short RNA molecules, which can aid to faster optimisation of the pulse parameters in vitro and in vivo.

Abstract Copper-doped ZnO nanoparticles with the formula Zn1−x(Cu)O, where x = 0.0, 0.03, 0.05, and 0.07 were produced using the co-precipitation process. Physical, chemical, and structural properties were properly examined. Powdered X-ray diffraction (P-XRD) patterns revealed the formation of hexagonal wurtzite crystal structure in all samples, through atomic substitutional incorporation in the Cu-doped ZnO lattice. The presence of Cu ions and their dissolution in the host ZnO crystal structure was supported by FT-IR spectra. HR-TEM images were used to assess the average size, morphology, and shape regularity of the synthesized samples. The form and homogeneity of the ZnO changed when Cu ions were substituted, as evidenced by FE-SEM/EDX analysis. The presence of copper signals in the Cu-doped samples indicates that the doping was successful. The decrease in zeta potential with an increased copper doping percentage designates that the nanoparticles (NPs) are more stable, which could be attributed to an increase in the ionic strength of the aqueous solution. The synthesized NPs were evaluated for their substantial in vitro antioxidant properties. In addition, the antimicrobial efficacy of the materials was tested against pathogenic microorganisms. Regarding the anti-diabetic activity, the 7Cu ZnO sample showed the highest inhibitory effect on the α-amylase enzyme. No variations were observed in the activities of the acetylcholinesterase enzyme (AChE) and proteinase enzymes with ZnO and samples doped with different concentrations of Cu. Therefore, further studies are recommended to reveal the in-vitro anti-diabetic activity of the studied doped samples. Finally, molecular docking provided valuable insights into the potential binding interactions of Cu-doped ZnO with α-amylase, FabH of E. coli, and Penicillin-binding proteins of S. aureus. These outcomes suggest that the prepared materials may have an inhibitory effect on enzymes and hold promise in the battle against microbial infections and diabetes.

Dermatophytes are the keratinophilic fungi which infect humans and is the most recurring type of disease. The high level of transmissibility creates an epidemiological risk and emphasises the significance of these illnesses. However, a growing number of reports describing dermatophytes can cause deep infections in diabetic and immunocompromised patients, by invading deep layers like the dermis and hypodermis. Despite the prevalence and significance of dermatophytes in clinical mycology, it is not always possible to accurately diagnose this specific infection due to its overlapping structures among species of dermatophytes. Since it is difficult to identify species that exhibit weak characteristics in the morphological highlights, identification of the dermatophyte is often relied on its morphological analysis, which is a laborious process and demands skill. The massive shift in genetic variation, the source of infection, and epidemiological research can be discovered using molecular approaches. Therefore, the development of an accurate laboratory test for dermatophyte species identification is essential for the prevention and efficient management of dermatophytoses. One such methodology allows use of PCR technology which has many methods for molecular level characterization which is rapid, efficient, and capable of producing DNA polymorphisms specific to various dermatophyte species based on distinctive band patterns seen by agarose gel electrophoresis. The RAPD-PCR approach will be used in this study protocol to molecularly characterize the dermatophytes for precise speciation of the sample. In addition to improving knowledge of fungal biology and pathology with a focus on adaptive mechanisms to combat difficult conditions from host counteractions, there is a need to improve awareness of the importance of these diseases through accurate epidemiological data. The advantages of molecular approaches for characterizing objects over traditional methods are their sensitivity and specificity.

The object of research is the technique for obtaining glued sausage casings, in which adhesive related structures and electrophoresis for tanning are used to glue the layers of intestinal raw materials. Devising a technique for gluing sausage casings from intestinal raw materials with the use of adhesive related structures by stitching by tanning, via intensified electrophoresis, has been substantiated. The technology of glued sausage casings from intestinal raw materials, which are classified as unclaimed residues and waste, has been proposed. The technology differs from known similar technologies by applying a technique for gluing intestinal membranes using an adhesive related construct and electrophoresis during tanning. The devised gluing technique and the device for its implementation make it possible to avoid violation of the integrity of the raw material, are universal in relation to it, and are easy to operate. The rational parameters of electrophoresis, which is used to accelerate the tanning process of the place of gluing of the layers of the intestinal membranes using an adhesive related construct, have been determined. The potential difference between the electrodes and the duration of exposure of the gluing site for gluing the original intestinal raw materials: pork belly, lamb belly, beef belly were determined. At a potential difference of 100 V, the duration of exposure of the gluing site for pork and lamb bellies is 17 minutes, and for beef bellies − 20 minutes. It is noted that the results differ in the duration of exposure of the gluing site, which is probably due to the thickness of the layer of the raw material. The proposed technology would contribute to improving the efficiency of the technology of glued sausage casings from intestinal raw materials and could be used at enterprises for the processing of agricultural products and the meat processing industry. The analysis of dietary supplements is far less regulated than pharmaceuticals, leading to potential quality issues. Considering their positive effect, many athletes consume supplements containing l-histidine and β-alanine. A new microfluidic method for the determination of l-histidine and β-alanine in dietary supplement formulations has been developed. For the first time, capacitively coupled contactless conductivity detection was employed for the microchip electrophoresis of amino acids in real samples. A linear relationship between detector response and concentration was observed in the range of 10–100 µmol L–1 for l-histidine (R2 = 0.9968) and β-alanine (R2 = 0.9954), while achieved limits of detection (3 × S/N ratio) were 4.2 µmol L–1 and 5.2 µmol L–1, respectively. The accuracy of the method was confirmed using recovery experiments as well as CE-UV-VIS and HPLC-UV-VIS techniques. The developed method allows unambiguous identification of amino acids in native form without chemical derivatization and with the possibility of simultaneous analysis of amino acids with metal cations. We developed a simple, paper‐based device that enables sensitive detection by mass spectrometry (MS) without solid phase extraction or other sample preparation. Using glass fiber filter papers within a 3D printed holder, the device employs electrokinetic manipulations to stack, separate, and desalt charged molecules on paper prior to spray into the MS. Due to counter‐balanced electroosmotic flow and electrophoresis, charged analytes stack on the paper and desalting occurs in minutes. One end of the paper strip was cut into a sharp point and positioned near the inlet of a MS. The stacked analyte bands move toward the paper tip with the EOF where they are ionized by paper spray. The device was applied to analysis of PFAS in tap water with sub part‐per‐trillion detection limits in less than ten minutes with no sample pretreatment. Analysis of opioids in urine also occurs in minutes. The crucial parameters to enable stacking, separation, and MS ionization of both positively and negatively charged analytes were determined and optimized. Experimental and computational modeling studies confirm the electrokinetic stacking and analyte transport mechanisms. On‐paper separations were carried out by stacking analyte bands at different locations depending on their electrophoretic mobility, achieving baseline separation in some cases. Serum protein electrophoresis (SPE) in combination with immunotyping (IMT) is the diagnostic standard for detecting monoclonal proteins (M-proteins). However, interpretation of SPE and IMT is weakly standardized, time consuming and investigator dependent. Here, we present five machine learning (ML) approaches for automated detection of M-proteins on SPE on an unprecedented large and well-curated data set and compare the performance with that of laboratory experts.SPE and IMT were performed in serum samples from 69,722 individuals from Norway. IMT results were used to label the samples as M-protein present (positive, n=4,273) or absent (negative n=65,449). Four feature-based ML algorithms and one convolutional neural network (CNN) were trained on 68,722 randomly selected SPE patterns to detect M-proteins. Algorithm performance was compared to that of an expert group of clinical pathologists and laboratory technicians (n=10) on a test set of 1,000 samples.The random forest classifier showed the best performance (F1-Score 93.2 %, accuracy 99.1 %, sensitivity 89.9 %, specificity 99.8 %, positive predictive value 96.9 %, negative predictive value 99.3 %) and outperformed the experts (F1-Score 61.2 ± 16.0 %, accuracy 89.2 ± 10.2 %, sensitivity 94.3 ± 2.8 %, specificity 88.9 ± 10.9 %, positive predictive value 47.3 ± 16.2 %, negative predictive value 99.5 ± 0.2 %) on the test set. Background: Accelerated ageing is a method to assess seed storage performance by exposing the seeds to higher temperature and relative humidity to make seeds lose its vigour and viability quickly. The seed physiological parameters are then compared with the natural ageing to derive certain conclusions. But, the molecular pattern of seed deterioration under accelerated ageing varies compared to that of natural ageing. Methods: Chickpea, seed lots of variety JG-11 and Annigeri-1 were subjected to natural ageing in cloth bag with nine per cent initial seed moisture under ambient conditions. Simultaneously, representative sample of same seed lot were also aged at 41.C and 95±2% humidity up to 120 hours. Result: Both the ageing methods had significant negative effect on seed physiological and biochemical quality parameters. The germination potential of seeds tested after 48 hours of accelerated ageing (83.5%) was equal 10 months of natural ageing (85%) in JG-11, while also in Annigeri-1, 48 hours of accelerated ageing (78%) was nearly equal to 10 months of natural ageing (80%). The protein expression analyzed through 2D-PAGE at similar germination potential brought out by two independent ageing methods showed higher protein down regulation ratio (3.4) in accelerated ageing than in natural ageing (3.2) unveiling its rapidity in the seed deterioration process. Fungal diseases are crucial factors in reducing chickpea production. Ascochyta Blight is caused by the necrotrophic fungi Didymella rabiei and is one of the most destructive diseases in most world areas. Therefore, a completely randomized factorial design with five replications was applied to evaluate the effects of Ascochyta Blight disease fungi on the chickpea plant. The chlorophyll index, the activities of superoxide dismutase isoforms, and evolutionary analyses were performed to get further insights. Also, 2D electrophoresis of chickpea leaf proteins, gene ontology, and protein-protein interactions analysis was performed. The results did not show any significant effect of A. rabiei infection on the wet weight of chickpea seedlings. Chlorophyll index levels significantly decreased with A. rabiei infection in both chickpea lines. Electrophoretic analysis of superoxide dismutase on 8% polyacrylamide gel revealed three isoforms. The activities of superoxide dismutase isoforms significantly increased with A. rabiei disease. Identification of proteins was performed according to their isoelectric points and approximate molecular weights. Leaf proteome analysis of chickpea lines showed that the expression of eight reproducible spots changed significantly under A. rabiei disease condition. Candidate proteins were components of defense and regulation systems. High expression of Dual specificity protein, Peptide methionine sulfoxide reductase B2, and chloroplastic phosphatase 1B (the proteins involved in the defense system) reveals their essential functions under A. rabiei infection. Background: Upon infection activated plasma cells produce large quantities of antibodies which can lead to the emergence of a monoclonal component (MC), detectable by serum protein electrophoresis (SPEP). This study aims to investigate any correlation between SARS‐CoV‐2 infection and MC development and, if identified, whether it persists during follow‐up. Methods: SPEPs of 786 patients admitted to hospitals between March 01 2020 and March 31 2022 were evaluated. Positive (SARS‐CoV‐2+) and negative (SARS‐CoV‐2−) patients to nasopharyngeal swab for SARS‐CoV‐2 by RT‐PCR were included. The persistence/new occurrence of MC was investigated for all patients during follow‐up. Patient groups were compared by chi‐square analysis. Results: MC was identified in 12% of all patients admitted to hospital, of which 28.7% were SARS‐CoV‐2+. The most common immunoglobulin isotype in both groups was IgG‐k. There was no correlation between MC development and SARS‐CoV‐2 infection (p = 0.173). Furthermore, the risk of MC persistence in SARS‐CoV‐2‐negative patients was revealed to be higher than in the SARS‐CoV‐2+ at follow‐up (HR = 0.591, p = 0.05). Conclusions: Our study suggests that the detection of MC during SARS‐CoV‐2 infection is most likely due to the hyperstimulation of the humoral immune system, as also occurs in other viral infections. Sitophilus oryzae and Sitophilus zeamais are among the main pests of stored grains worldwide, they are very similar species and have great destructive potential, causing numerous damages to grain production. The control of these insects occurs, in general, through chemical insecticides such as the agrochemicals pirimiphos-methyl, an organophosphate, and bifenthrin, a pyrethroid, which are widely used to prevent infestation. Four strains of these two species were exposed to the two classes of agrochemicals mentioned. The polyacrylamide gel electrophoresis technique was used to identify the esterase patterns of the species and eight esterases were identified for S. zeamais, of which four cholinesterases, two carboxylesterases, one acetylesterase, and one arylesterase; and seven esterases for S. oryzae, being four cholinesterases, two carboxylesterases, and one acetylesterase. After exposure to agrochemicals, through dose-response bioassays, the LC50 was determined for each strain, with the SoB strain of S. oryzae being the most resistant to bifenthrin, the SzB strain of S. zeamais being the most resistant to pirimiphos-methyl and the SoA strain of S. oryzae was the most sensitive to both compounds. Analysis of the esterase patterns of insects exposed to these agrochemicals revealed the inhibition and probable involvement of carboxylesterases and cholinesterases in the detoxification processes of organophosphates and pyrethroids. Summary: Foodomics is an emerging probing method of phenotype investigation of the different milk proteins and their subtypes. The polymorphic nature of the β‐casein (β‐Cn) protein has shown fourteen different protein variants to date in bovines. The analysis of the β‐Cn genetic polymorphism from the milk of the crossbred dairy animals is crucial for the quality assurance of the consumers from the various health concerns, especially those linked with the A1 phenotype which yields β‐casomorphin‐7 on in vivo digestion. Jersey‐crossed Indian cattle have been widely utilised in dairy because of their better milk production and survival performance trait. In this investigation, an SDS‐PAGE coupled with a high‐resolution accurate mass spectrometry‐based proteomics approach has been applied to identify the presence of specific phenotype of the β‐Cn protein in the milk of the 24 Indian crossbred (Jersey crossed) animals. Amino acid sequential analysis has been done using different search modules, as MS Amanda and Sequest HT showed 17 cows are producing A2 β‐Cn (Pro~67) while only seven animals yielded the A1 variant (His~67). The maximum number of Indian Jersey‐crossed animals are lactating milk having A2 β‐Cn. The A2 milk from the crossbred animals is free from the negative impact on health caused by β‐casomorphin‐7 (BCM‐7) released during digestion of the A1 phenotype. The review provides an overview of recent developments and applications of capillary electromigration (CE) methods for the determination of important physicochemical parameters of various (bio)molecules and (bio)particles. These parameters include actual and limiting (absolute) ionic mobilities, effective electrophoretic mobilities, effective charges, isoelectric points, electrokinetic potentials, hydrodynamic radii, diffusion coefficients, relative molecular masses, acidity (ionization) constants, binding constants and stoichiometry of (bio)molecular complexes, changes of Gibbs free energy, enthalpy and entropy and rate constants of chemical reactions and interactions, retention factors and partition and distribution coefficients. For the determination of these parameters, the following CE methods are employed: zone electrophoresis in a free solution or in sieving media, isotachophoresis, isoelectric focusing, affinity electrophoresis, electrokinetic chromatography, and electrochromatography. The present article deals with the theoretical development of the electrophoresis of core-shell structured soft particles in which the inner rigid core is decorated with a fluid and ion-permeable polymeric shell layer. Note that such a particle resembles several biocolloids (e.g., bacteria, virus, humic acid), functionalized nanoparticles, and environmental entities, to name a few. For such a structured particle, the conventional ζ -potential concept loses its meaning, and an extensive theory is required to analyze the electrohydrodynamics of the particle considering the penetration of ionized liquid across the shell layer. Note that the dielectric permittivity of the shell layer is often lower than that of the bulk aqueous medium, which induces the ion partitioning effect. Besides, in several practical situations, the hydrodynamic slipping may occur along the slipping plane. In addition, the slipping plane may not always be located along the surface of the inner core due to grafting of a polymeric shell layer along its surface, and thus, it may be assumed to be located somewhere within the surface polymeric layer. The slipping plane separates two regions with different Brinkman parameters. The region outside the slipping plane the Brinkmann screening length takes a finite value, which allows fluid flow across this region. In the region inside the slipping plane, the Brinkman parameter may practically be equal to infinity, but electrolyte ions still can penetrate this region. Considering all the physical aspects indicated above, we have proposed a simple model to study the electrophoresis of soft particles within the flat-plate regime. Based on the weak charge limit, we adopt the Debye-Hückel linearization to simplify the governing equations, and the explicit form of electrophoretic mobility is derived. Several closed-form analytic expressions are further deduced from the general mobility expressions valid under various limiting situations. We have illustrated our findings graphically to highlight the impact of pertinent parameters on the electrophoretic mobility of such a particle. In addition, we have further provided an estimate of the parametric range in which the particle may attain a zero mobility. Background: Beta‐2 transferrin (B2‐Tf) gel electrophoresis (GE) is the preferred non‐invasive diagnostic modality for confirming cerebrospinal fluid (CSF) in body fluids. While B2‐Tf GE testing is highly sensitive and specific for CSF, false‐positive (FP) and false‐negative (FN) results can lead to diagnostic and therapeutic dilemmas. Several series have demonstrated potential causes of false B2‐Tf GE results, but few studies have reported reasons for these errors. The purpose of this systematic review was to describe sources of B2‐Tf GE errors. Methods: A systematic review was performed by searching OVID, EMBASE, and Web of Science databases for B2‐Tf GE studies. After applying exclusion criteria, original research studies directly addressing erroneous B2‐Tf GE results underwent qualitative analysis. Results: Of the 243 abstracts screened, 71 underwent full‐text review and 18 studies reporting B2‐Tf GE errors were included for analysis. There were 15 potential FPs, 12 actual FPs, 12 potential FNs, 19 actual FNs, and 14 indeterminate results. There were also 246 potentially indeterminate results from in vitro studies. Reasons for B2‐Tf GE errors included serum transferrin alterations (n = 17; all potential), infection related (n = 13; 9 potential), orbital or salivary contamination (n = 2; 1 potential), and collection related (n = 255; 246 potential). There were 31 false or indeterminate results with unspecified reasons. There were no reported errors due to laboratory processing. Conclusions: Multiple potential or actual reasons for false or indeterminate results have been reported for B2‐Tf GE testing of rhinorrhea and otorrhea. Background: Cervical cancer is one of the most common gynecological malignancies. Previous studies have shown that the ethanol extract of Sophora moorcroftiana seeds (EESMS) possesses an antiproliferative effect on several tumors in vitro. Therefore, in this study, we assessed the impact of EESMS on human cervical carcinoma (HeLa) cell proliferation. Methods: The proliferation and apoptotic effects of HeLa cells treated with EESMS were evaluated using 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide assay, dual acridine orange/ethidium bromide double staining, flow cytometry, and western blotting. Single-cell level atomic force microscopy (AFM) was conducted to detect the mechanical properties of HeLa cells, and proteomics and bioinformatics methods were used to elucidate the molecular mechanisms of EESMS. Results: EESMS treatment inhibited HeLa cell proliferation by blocking the G0/G1 phase, increasing the expression of Caspase-3 and affecting its mechanical properties, and the EESMS indicated no significant inhibitory effect on mouse fibroblasts L929 cell line. In total, 218 differentially expressed proteins were identified using two-dimensional electrophoresis, and eight differentially expressed proteins were successfully identified using matrix-assisted laser desorption/ionization-time-of-flight mass spectrometry. The differentially expressed proteins were involved in various cellular and biological processes. Conclusion: This study provides a perspective on how cells change through biomechanics and a further theoretical foundation for the future application of Sophora moorcroftiana as a novel low-toxicity chemotherapy medication for treating human cervical cancer.
