A Pd‐catalyzed asymmetric isomerization‐hydroamidocarbonylation of amide‐containing alkenes was developed, affording a variety of chiral α‐alkyl succinimides in moderate to good yields with high enantioselectivities. The key to success was introducing bulky 1‐adamentyl P‐substitution and 2,3,5,6‐tetramethoxyphenyl group into the rigid P‐chirogenic bisphosphine ligand to create stronger steric hinderance and deeper catalytic pocket. By this approach, regio‐ or stereo‐convergent synthesis of enantiomeric succinimides from the mixture of olefin isomers was achieved. [ABSTRACT FROM AUTHOR]

This article discusses the development of a practical method for Fischer indolization, a commonly used synthetic approach in chemistry. The researchers used bench-stable sodium arylhydrazine sulfonates as a substitute for arylhydrazine, which is typically used in Fischer indolization. The sodium arylhydrazine sulfonates were easily obtained from substituted anilines and could be used directly in the reaction without further purification. The method was successful in synthesizing various tetrahydrocarbazoles and tryptamine-based pharmaceutical molecules. A holistic model for predicting yield and linear selectivity for the hydroformylation of 1-octene was developed by machine learning using the experimental data collected from literatures. Physical organic chemistry (POC) parameter-based descriptors were adopted to represent pre-catalyst molecular features. Machine learning models trained respectively by Random Forests (RF) and Extreme Gradient Boost (XGBoost) algorithm showed remarkable performance on predicting linear selectivity. The method can also comprehensively map the correlation between reaction conditions and the results. The alkyne alkoxycarbonylation to α- or β-substituted acrylates was coupled with the electrocatalytic reduction of CO 2 to CO. The CO-enriched gaseous mixture produced from the electrocatalytic reduction of CO 2 was collected and directly used in the alkyne alkoxycarbonylation. The CO content was found to be critical to the process of carbonylation, and satisfying results were attained by using the gas mixture containing >15 vol % CO. This method offered an efficient but simple CO source from CO 2 electroreduction to the alkoxycarbonylation reaction in two-compartment manner.

The development of chiral hydrogen donor catalysts is fundamental in the expansion and innovation of asymmetric organocatalyzed reactions via an enantioselective hydrogen atom transfer (HAT) process. Herein, an unprecedented type of chiral C 2 -symmetric arylthiol catalysts derived from readily available enantiomeric lactate ester was developed. With these catalysts, an asymmetric anti-Markovnikov alkene hydroamination-cyclization reaction was established, affording a variety of pharmaceutically interesting 3-substituted piperidines with moderate to high enantioselectivity. Results of the designed control experiments and theoretical computation rationalized the origin of stereocontrol and disclosed the spatial effect of the moiety of chiral thiols on the enantioselectivity. We believed the facile synthesis, flexible tunability, and effective enantioselectivity-controlling capability of these catalysts would shed light on the development of versatile chiral HAT catalysts and related asymmetric reactions.

A convenient and efficient method for the synthesis of optically active difluoro-substituted indoline derivatives starting from the corresponding 3 H -indoles by chiral phosphoric acid-catalyzed transfer hydrogenation was developed.
