Multiple myeloma (MM) is characterized by clonal plasma cell proliferation in the bone marrow, challenging prognosis prediction. We developed a gene-pairing prognostic risk model using m6A regulatory genes and a nested LASSO method. A cutoff of − 0.133 categorized MM samples into high-risk and low-risk groups. The model showed strong prognostic performance in 2088 newly diagnosed MM samples and predicted response to combination therapy (daratumumab, carfilzomib, lenalidomide, and dexamethasone) in patients who failed or relapsed from bortezomib-containing regimens, with an AUC of 0.9. It distinguished between smoldering MM and MM (cutoff: − 0.45) and between MM and plasma cell leukemia (cutoff: 0.0857). Single-cell analysis revealed higher risk scores at relapse. Combining MM cell lines and sample data, we identified potential drugs and targets (ADAT2 and NUP153) effective against high-risk MM. Integrating the m6A risk model with the International Staging System (ISS) enhanced stratification accuracy. These insights support precision treatment of MM. [ABSTRACT FROM AUTHOR]

This paper developed an efficient microbial activator formula and conducted an in-depth study on its efficacy and mechanism in promoting the degradation of petroleum hydrocarbons in oil-contaminated soil. A 60-day microbial remediation experiment conducted on oily soil revealed that the microbial activators significantly boosted the activities of dehydrogenase and catalase, subsequently speeding up the degradation of petroleum hydrocarbons in the soil. The overall degradation rate reached as high as 71.23%, with the most significant degradation effect observed in asphaltenes, achieving a degradation rate of 93.98%. This was followed by aromatic hydrocarbons (90.45%), saturated hydrocarbons (84.39%), and asphaltenes (65%). Compared to traditional microbial stimulation methods, this activator demonstrated significant superiority. Microbial diversity analysis reveals that microbial activators can effectively activate microbial activity in soil targeting refractory petroleum hydrocarbon components. By comparing the changes in microbial community structure before and after the addition of microbial activators, we found that the activators promoted an increase in the abundance of microorganisms belonging to the Bacillota, Pseudomonadota, and Bacteroidetes, which have petroleum hydrocarbon degradation functions, and facilitated the evolution of microbial community structure towards a direction more conducive to petroleum hydrocarbon degradation. KEGG metabolic pathway analysis revealed that the degradation pathways for alkanes, aromatic hydrocarbons, and PAHs are primarily present in these bacterial phylum. Ultrahigh-voltage potassium-ion batteries (PIBs) with cost competitiveness represent a viable route towards high energy battery systems. Nevertheless, rapid capacity decay with poor Coulombic efficiencies remains intractable, mainly attributed to interfacial instability from aggressive potassium metal anodes and cathodes. Additionally, high reactivity of K metal and flammable electrolytes pose severe safety hazards. Herein, a weakly solvating fluorinated electrolyte with intrinsically nonflammable feature is successfully developed to enable an ultrahigh-voltage (up to 5.5 V) operation. Through breaking the anionic solvation barrier, synergistic interfacial modulation can be achieved by the formation of robust anion-derived inorganic-rich electrode-electrolyte interfaces on both the cathode and anode. As proof of concept, a representative KVPO4F cathode can sustain 1600 cycles with 84.4% of capacity retention at a high cutoff voltage of 4.95 V. Meanwhile, K plating/stripping process in our designed electrolyte also demonstrates optimized electrochemical reversibility and stability with effectively inhibited potassium dendrites. These findings underscore the critical impact of anion-dominated solvation configuration on synergistic interfacial modulation and electrochemical properties.
