Background/Objectives: The primary method used to pharmacologically arrest cancer development and its metastasis is to disrupt the cell division process. There are a few approaches that may be used to meet this objective, mainly through inhibiting DNA replication or mitosis. Despite intensive studies on new chemotherapeutics, the biggest problem remains the side effects associated with the inhibition of cell division in non-tumoural host cells. Methods: The efficacy and selectivity of the kojic acid derivative (L1) was studied in vitro with the use of tumoural (Caco2, SW480, HT29, T98G) and non- tumoural (HEK293T, RAW) cell lines. Light and electron microscopy observations were supported by the next generation sequencing (NGS), cytoflow, and spectroscopy analysis of mRNA and biomolecules, respectively. Results: The light and electron microscopy observations showed that L1 treatment leads to significant morphological changes in Caco2 cells, which are characteristic of mitosis arrest. Moreover, the fluorescent tubulin staining revealed the formation of tubulin ring structure associated with the apoptotic stage. Mitotic exit into apoptosis was further conformed by the cytoflow of early/late apoptosis stages and caspase-3 analysis. NGS investigation showed differentiated expressions of genes involved in mitosis and apoptosis processes. The observed IC50 in tumoural cell lines were as follows: Caco2 (IC50 = 68.2 mM), SW480 (IC50 = 15.5 mM), and HT29 (IC50 = 4.7 mM). Conclusions: The findings presented here suggest that L1 could be a valid candidate for oral prevention and/or chemotherapy in colorectal cancer. Considering high selectivity of L1 versus tumoural cell lines, more in-depth mechanistic studies could reveal unknown stages in carcinogenesis. [ABSTRACT FROM AUTHOR]

Environmental stressors such as micro‐ and nanosized plastic particles (MNPs) or crude oil have a detrimental effect on aquatic animals; however, the impact upon the cardiovascular system of fish remains relatively under‐researched. This study presents a novel approach for investigating the effect of crude oil and MNPs on the cardiac system of fish. We used salmonid larvae and cardiac cell cultures derived from hearts of salmonid fish and exposed them to environmental stressors. Following exposure to plastic particles or crude oil, the larvae exhibited some variation in contraction rate. In contrast, significant alterations in the contraction rate were observed in all cardiac cell cultures. The greatest differences between the control and treatment groups were observed in cardiac cell cultures derived from older brown trout. Following 7 days of exposure to MNPs or crude oil in Atlantic salmon larval hearts or cardiac cell cultures, there were only minor responses noted in mRNA expression of the selected marker genes. These findings show the use of a novel in vitro technique contributing to the existing body of knowledge on the impact of MNPs and crude oil on the cardiovascular system of salmonids and the associated risk. Alzheimer's disease (AD) is a major contributor to dementia and the most common neurodegenerative disorder. In AD pathophysiology, matrix metalloproteinases (MMPs)—proteolytic enzymes, best known to be responsible for remodeling and degradation of the extracellular matrix—were suggested to play an important role. Due to the diverse nature of the published data and frequent inconsistent results presented in available papers, it was considered essential to analyze all aspects of MMP literature with respect to AD pathophysiology and attempt to outline a unifying concept for understanding their role in AD. Thus, the main contribution of this review article is to summarize the most recent research on the participation of MMP in AD pathophysiology obtained using the cell cultures to understand the molecular principles of their action. Furthermore, an updated comprehensive view regarding this topic based exclusively on papers from human studies is provided as well. It can be concluded that determining the exact role of any particular MMPs in the AD pathophysiology holds promise for establishing their role as potential biomarkers reflecting the severity or progression of this disease or for developing new therapeutic agents targeting the processes that lead to AD. The increasing evolution of pathogen resistance is a global problem that requires novel solutions. Recently, an increased interest in ethnomedicinal sources can be observed in the derivation of new medicines. The return to traditional medicinal formulations handed down for generations is being followed, but it is necessary to revise them again, taking into account the generally accepted research protocol. We aimed to evaluate the antimicrobial potential of historical deposits of Silesian healing clay (SHC), used in ethnomedicine against Gram-positive bacteria and to assess their biological activity using a primary dermal fibroblast line (NHDF) and a model monocyte line (THP1). Information on medicinal clay deposits that occur in Silesia and are traditionally used in ethnomedicine or ancient medicine and known as terra sigillata Silesiaca or SHC, was selected on available source materials and old prints and maps from the archives of the Polish Geological Institute (Wrocław, Poland). Subsequently, their places of occurrence were identified and traced in the field by taking three deposits from the Silesia territory: Upper Silesia (D1), Opole Silesia (D2), and Lower Silesian (D3) Voivodeships for analysis. Their basic parameters and antimicrobial efficacy against pathogenic bacteria, Gram-positive streptococci and staphylococci, including methicillin-resistant strains, were examined. The study evaluated the effects of clays on growth and vitality using a primary dermal fibroblast line (NHDF) and a monocytic line (THP1). Studies were performed on a cell culture model to determine the effects on tissue regeneration (fibroblasts) and anti-inflammatory effects (monocytes). The study attempted to identify the mechanism of antimicrobial action, especially the textural characteristics and geochemical composition, as well as the environmental reaction (pH). SHCs were classified into the following textural classes: clay loam (D1), clay (D2), and sand (D3). The tested deposits have antimicrobial properties that reduce the bacterial population (104 CFU) compared to the control (108 CFU). The antimicrobial effect depends on the type of clay and the species or strain of bacteria used. In-house studies clearly showed that Staphylococcus aureus Pcm 2054 and Staphylococcus epidermidis MRSE ATCC 2538 cells were completely adsorbed by clay minerals from clay D3.13. Furthermore, 10% leachates also showed an antimicrobial effect, as a reduction in bacterial populations was observed ranging from 91 to 100%. The results showed stimulation of fibroblast culture proliferation and inhibition of the growth of inflammatory cells (monocytes). SHCs tested have antimicrobial potential, in particular D2.7, D2.11, and D3.13. The D3.13 deposit had a bactericidal effect against the staphylococci tested. Aqueous solutions of clays also showed bacteriostatic effect. The results obtained in cell culture model tests indicate properties that modulate the healing process – stimulation of fibroblast growth (NHDF line) and inhibition of monocyte growth (THP1 line). In order to address the challenges posed by population expansion, climate change, depletion of natural resources, malnutrition and increased prevalence of chronic diseases, disruptive innovations are needed to achieve sustainable development. The utilization of new resources and novel production methods provide opportunities for the development of novel food products (NFPs). Here, we present the potential of 3D printing with personalized design for applications in the sustainable development of NFPs, such as the development of innovative food sources, reducing the consumption of natural resources and the impact on the environment, and preventing/treating malnutrition and chronic diseases. In addition, a systematic review of optimization measures such as raw material optimization, development of enhanced evaluation techniques, improved equipment performance, and application of artificial intelligence is presented to guide the application of 3D printing of NFPs. Finally, the challenges and future opportunities for food 3D printing are presented to provide new directions for the development of NFPs. Food 3D printing offers a number of advantages such as personalized design, digital nutritional matching, streamlined supply chain, high-value use of food by-products, and broadening of food ingredient sources. Starting from the necessity and specificity of novel food development, this paper focuses on the application of 3D printing in the development of NFPs, such as innovative source foods, plant-based meat analogues, cell cultures, and foods for special medical purposes, thus facilitating the development of new possibilities for 3D printing in the food industry. In addition, a systematic overview of optimization measures such as the optimization of raw materials, the development of evaluation techniques, the improvement of equipment performance and the application of artificial intelligence is presented, thus providing theoretical guidance for the increasingly widespread application of 3D printing within the novel food industry. • Necessity and specificity of novel food products development are reviewed. • Applications of 3D printing in novel food products development are summarized. • Optimization measures are presented to guide the application of 3D food printing. • Future prospects of 3D printing in novel food products development are discussed. Piscine orthoreovirus-1 and 3 (PRV-1, PRV-3) cause highly prevalent infection in cultured salmonids and can induce heart and skeletal muscle inflammation (HSMI) resulting in economic losses in aquaculture. However, to date, PRV-1 and PRV-3 have withstood replication in continuous cell lines. In this study, we used beating heart cell cultures obtained from different developmental stages of rainbow trout (Oncorhynchus mykiss) (RTC-L and RTC-A) and tested their ability to sustain replication of PRV-1 and PRV-3. Furthermore, we compared the replication pattern of the different viruses with those in the newly developed heart fibroblast cell line (RTH-F) and the traditional established rainbow trout gonad cell line (RTG-2). Neither RTCs nor RTH-F cell lines supported replication of PRV-1 and PRV-3. Comparative experiments showed varying susceptibility of the novel cultures to viral haemorrhagic septicaemia virus (VHSV), chum salmon reovirus (CSV), infectious pancreatic necrosis virus (IPNV), piscine myocarditis virus (PMCV), salmonid alphavirus 3 (SAV-3) and tilapia lake virus (TiLV), indicating their usability for work with multiple fish viruses. Print-Light-Synthesis (PLS) combines the inkjet printing of a ruthenium precursor ink with the simultaneous photo-induced generation of ruthenium oxide films. During PLS, inkjet-printing generates on conductive as well as insulating substrates micrometer-thin reaction volumes that contain with high precision defined precursor loadings. Upon direct UV light irradiation, the Ru precursor converts to RuO 2 while all other ink components escape in the gas phase. No post PLS processes are required, and the as-obtained RuO 2 films can be immediately used as electrochemical devices. Two-dimensional RuO 2 patterns with micrometric resolution and highly-controlled ruthenium loadings (few µg/cm 2 ) are realized. Thin RuO 2 films are generated on insulating substrates, such as polyimide, as well as individual RuO 2 particles on conductive substrates, such as graphene layers. The RuO 2 films are characterized by electron microscopy and spectroscopic techniques. The sensoristic applicability of the PLS-RuO 2 electrodes is demonstrated by potentiometric pH sensing in cell cultures and amperometric detection of L-cysteine. For pH sensing the RuO 2 film electrodes show Nernstian sensitivity. The restriction in the movement of aqueous humor through the trabecular meshwork is known to play an important role in the pathogenesis of glaucomas. Among the functions of the trabecular meshwork that help to prevent this restriction and maintain the movement of aqueous humor is phagocytosis. Cells in the trabecular meshwork actively phagocytose pigment, cellular debris, and other materials in order to help keep the drainage pathway clear. A decrease in phagocytosis has thus been implicated in the development of glaucomas.
