Abstract Introduction The efficacy of pre-exposure prophylaxis (PrEP) is highly dependent on adherence, and adherence behavior is influenced by motivation. The aim of this study was to explore the longitudinal relationship between PrEP motivation and change in adherence among men who have sex with men (MSM) in Western China. Methods From November 2019 to June 2021, we conducted a PrEP prospective cohort study. Motivation to take medicine was measured by the PrEP Motivation Scale at baseline, and was grouped into different levels of latent categories by Latent Profile Analysis (LPA). A multinomial logistic regression model was used to explore the longitudinal relationship between change in adherence (improvement, decline, no change) and different levels of PrEP motivation. Results MSM were divided into two categories of PrEP motivation, a “high motivation group” (n = 506, 69.89%) and a “low motivation group” (n = 218, 30.11%). High PrEP motivation had no significant effect on the change in short-term adherence, however, it contributed to the improvement in long-term adherence [odds ratio (OR) = 3.028 (1.100–8.332), p = 0.031]. The predictive power of the adherence model was significantly enhanced with the addition of the PrEP motivation factor. Conclusions There was a positive correlation between high PrEP motivation at baseline and an improvement in long-term adherence. Surveillance and intervention of PrEP motivation in MSM can increase their adherence, and then promote PrEP efficacy.

Background/Aims: Hepatocellular carcinoma (HCC) represents a common neoplasm that presents a substantial worldwide health challenge. Nevertheless, the involvement of HPN-AS1 in HCC remains unknown. Materials and Methods: The quantitative reverse transcription polymerase chain reaction (qRT-PCR) assay was utilized to measure HPN-AS1 expression in HCC. The GABPA effects on the HPN-AS1 promoter were analyzed through chromatin immunoprecipitation and luciferase reporter assays. Cell proliferation potential was determined by deploying CCK-8 assay, Ki-67 immunofluorescence staining, and colony formation assay. Cell apoptosis was detected using acridine orange/ethidium bromide staining and terminal deoxynucleotidyl transferase dUTP nick end labeling staining. Western blotting was utilized to measure the protein levels of proliferation factors and apoptosis regulators. HPN-AS1 binding to eIF4A3 was accessed by RNA-binding protein immunoprecipitation assay. Results: HPN-AS1 was significantly downregulated in both HCC cells and tissues. Lower HPN-AS1 levels indicate a poorer HCC prognosis. Moreover, we found that GABPA functions as a transcription factor for HPN-AS1. Functional studies revealed that HPN-AS1 displayed inhibitory effects on HCC cell proliferation and promoted apoptosis. Mechanically, HPN-AS1 bound to and facilitated translation initiation factor eIF4A3 degradation. Loss of HPN-AS1 augmented eIF4A3 protein levels rather than eIF4A3 mRNA levels. Exogenous expression of eIF4A3 could restore eIF4A3 protein levels and reverse HPN-AS1 overexpression-induced cell proliferation inhibition and cell apoptosis. Conclusion: Our study elucidated that HPN-AS1 downregulation was mediated by GABPA. HPN-AS acted as a tumor suppressor within HCC through binding and facilitating eIF4A3 degradation. The study provides a novel insight into the biological function of HPN-AS1 in HCC, suggesting that HPN-AS1 could be a promising biomarker and a potential target for HCC diagnosis and treatment.

Purpose: To analyze the characteristics of optical coherence tomography in acute macular neuroretinopathy (AMN) following severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection and discuss the prognostic predictors. Methods: Patients with AMN following SARS-CoV-2 infection were divided into two groups according to the presence or absence of hyperreflective outer nuclear layer (ONL) lesion involving the fovea. Results: The first visit included 14 eyes in the fovea-involved group and 20 eyes in the no fovea-involved group. Ellipsoid zone (EZ) hyporeflection and interdigitation zone (IZ) interruption were detected in all eyes. Other common manifestations were myoid zone (MZ) hyperreflection (76.5%), ONL hyperreflection (73.5%), outer plexiform layer (OPL) thickening (64.7%), and EZ interruption (50%). The follow-up period was 48.4 ± 55.3 days. At the last visit, 12 eyes were in the fovea-involved group and 13 eyes in the no fovea-involved group. IZ interruption was detected in all eyes. Other common manifestations were EZ hyporeflection (92.0%), ONL atrophy (40.0%), OPL thickening (36.0%), OPL linear (32.0%), and MZ hyperreflection (32%). The improvement of visual acuity (VA) was -0.5 ± 0.5 and -0.2 ± 0.4 in the fovea-involved group and the no fovea-involved group, respectively, with a statistically significant difference between them (P = 0.045). Initial VA, initial cotton wool spot, initial ONL cyst, final ONL cyst, and final OPL linear were associated with final VA (P = 0.000, P = 0.029, P = 0.044, P = 0.049, P = 0.049, respectively). Conclusions: In the early stage of AMN following SARS-CoV-2 infection, IZ interruption and EZ hyporeflection were the most common manifestations, and pathology of IZ was more serious than that of EZ. Subsequently, OPL and ONL atrophied, and ONL atrophied faster. Regardless of whether hyperreflective ONL involved the fovea, VA improved, with a more noticeable improvement found in the fovea-involved group. The presence of initial ONL cyst and initial cotton wool spot, rapid atrophy of OPL, and poorer initial VA indicating poorer VA outcome.

Abstract A new technology for remote measurements of marine surface pressure has been proposed, employing a V‐band differential absorption radar and a radiometric temperature sounder to calculate the total column atmospheric mass. Observing System Simulation Experiments (OSSEs) are performed to evaluate the potential impact of Spaceborne Marine Surface Pressure (SMSP) on Numerical Weather Prediction. These experiments build on prior efforts (Privé, McLinden, et al., 2023, https://doi.org/10.16993/tellusa.3254), but with an updated version of the OSSE framework and with more sophisticated simulation of the SMSP observations and a longer experiment period. Several different instrument configurations are compared, including both scanning and non‐scanning orbits. SMSP impacts are calculated for analysis quality and forecast skill, and a forecast sensitivity observation impact tool is employed to place SMSP observations in context with the global observing network. The effects of rain contamination on observation quality are explored. Different magnitudes of simulated SMSP observation error are tested in the context of data assimilation to show the range of potential behaviors. Overall, SMSP observations are found to be most beneficial in the southern hemisphere extratropics, with statistically significant forecast improvements for the first 72 hr of the forecast. A constellation of four non‐scanning SMSP satellites is found to outperform a single scanning instrument with a 250 km wide swath.

Abstract Tumor immune evasion relies on the crosstalk between tumor cells and adaptive/innate immune cells. Immune checkpoints play critical roles in the crosstalk, and immune checkpoint inhibitors have achieved promising clinical effects. The long non‐coding RNA taurine‐upregulated gene 1 (TUG1) is upregulated in hepatocellular carcinoma (HCC). However, how TUG1 is upregulated and the effects on tumor immune evasion are incompletely understood. Here, METTL3‐mediated m6A modification led to TUG1 upregulation is demonstrated. Knockdown of TUG1 inhibited tumor growth and metastasis, increased the infiltration of CD8+ T cells and M1‐like macrophages in tumors, promoted the activation of CD8+ T cells through PD‐L1, and improved the phagocytosis of macrophages through CD47. Mechanistically, TUG1 regulated PD‐L1 and CD47 expressions by acting as a sponge of miR‐141 and miR‐340, respectively. Meanwhile, TUG1 interacted with YBX1 to facilitate the upregulation of PD‐L1 and CD47 transcriptionally, which ultimately regulated tumor immune evasion. Clinically, TUG1 positively correlated with PD‐L1 and CD47 in HCC tissues. Moreover, the combination of Tug1‐siRNA therapy with a Pdl1 antibody effectively suppressed tumor growth. Therefore, the mechanism of TUG1 in regulating tumor immune evasion is revealed and can inform existing strategies targeting TUG1 for enhancing HCC immune therapy and drug development.

Natural basalt can be processed into rod-like basalt fibers (BFs) and flake-like basalt scales (BSs) through high-temperature melting, forming and other processes. BFs and BSs have great application prospects in the field of organic coatings. BFs possess a regular rod-like structure and excellent mechanical properties, which are mainly used to enhance the mechanical properties of organic coatings. BSs possess a relatively irregular flake-like structure, which can form “maze” effects and layered structures to enhance corrosion resistance of organic coatings. However, basalt materials have poor compatibility, adhesion, and dispersion in organic resins due to their chemical inertness, resulting in a weak basalt-resin interface. Basalt-resin interface is an important parameter that determines the service performance of basalt composite coatings. Therefore, it is necessary to improve basalt-resin interface through corresponding strategies, such as chemical etching, chemical grafting and plasma treatment. Firstly, the surface modification strategies for improving basalt-matrix interface were summarized in this paper. Secondly, we discussed the effects of BFs and BSs on corrosion resistance of organic coatings and corrosion protection mechanism of composite coatings. Finally, we reviewed the research progress of modified basalt materials reinforced organic coatings.

Aiming at the poor voltage response characteristics of the line-committed converter-based high voltage direct current (LCC-HVDC) transmission system after the fault of the AC system at the inverter side, this paper analyzes the relationship between the continuous commutation failure of the LCC-HVDC transmission system and the reactive power demand in detail from the perspective of the reactive power voltage of the inverter side converter. On this basis, an improved control method for the virtual synchronous generator (VSG) of the energy storage system is proposed to enhance the commutation ability of the LCC-HVDC transmission system, to suppress continuous commutation failure. Firstly, secondary frequency regulation is added to the active power frequency control of the traditional VSG to improve the stability of the system converter bus frequency. Secondly, the leading trigger angle negative feedback control is added to the traditional VSG control, so that the system has the advantages of rapid reactive power output and voltage recovery. At the same time, different control modes are selected according to the fault location and size to improve the recovery ability and robustness of the system. Finally, based on the CIGRE benchmark LCC-HVDC transmission system model, the improved method is proven to have a good suppression effect. The results can provide a reference for actual projects.

Sea surface air pressure observations are a significant gap in the current Earth observing systems. This study addresses retrieval algorithm development and the evaluation of the potential impact of instrumental and environmental uncertainties on sea level pressure retrievals for the measurements of O2 differential absorption radar systems operating at three spectrally evenly spaced close-frequency bands (65.5, 67.75, and 70.0 GHz). A simulated northern hemispheric summer case is used to simulate retrieval uncertainties. To avoid high attenuation and a low signal-to-noise ratio, radar measurements from weather conditions with a rain rate ≥1 mm/h are not used in the retrieval. This study finds that a retrieval algorithm combining all three channels, i.e., the 3-channel approach, can effectively mitigate major atmospheric and sea surface influences on sea surface air pressure retrieval. The major uncertainty of sea surface pressure retrieval is due to the standard deviation in radar power returns. Analysis and simulation demonstrate the potential of global sea surface pressure observations with errors of about 1∼2 mb, which is urgently needed for the improvement of numerical weather prediction models. Future work will emphasize instrument development and field experiments. It is anticipated that an O2 differential absorption radar system will be available for meteorological applications in a few years.

The effect of target tracking is not ideal when facing various complex tracking scenarios such as non-rigid deformation of target, frequent occlusion, clutter of target background and interference of similar objects. In this paper, the feature based on deep convolutional neural network is used for target tracking in moving scenes, and a sliding window target segmentation method is proposed to study the impact of data normalization and data set expansion on the final result. In order to select more distinguishing features, principal component analysis is used to process the features of Deep Convolution Neural Network (DCNN), and the features of different network layers of DCNN are compared. The feature coding algorithm is studied, and the extracted DCNN features are encoded by Fisher Vectors algorithm, and compared with the locality-constrained linear encoding technique. Experiments show that the feature based on deep convolutional neural network in this paper can obtain higher accuracy than the traditional feature fusion method. According to the result analysis, the tracking accuracy of deep convolutional neural network algorithm is improved under the condition of illumination variation. In the case of local occlusion, the tracking accuracy is also improved.

BackgroundDefinitive concurrent chemoradiotherapy (CCRT) followed by maintenance therapy with immune checkpoint inhibitors offers the best chance of cure for patients with stage III non-small cell lung cancer (NSCLC). A significant challenge in this regimen is the occurrence of acute severe lymphopenia (ASL), which can compromise treatment efficacy. Currently, there are no effective strategies for preventing and treating ASL. Shenglin decoction (SLD), a traditional Chinese herbal medicine formulation, has demonstrated preliminary efficacy in mitigating ASL. However, robust evidence from clinical trials and a clear understanding of its mechanism of action are still needed. This study aims to comprehensively assess the efficacy, safety, and underlying mechanisms of SLD in the prevention of ASL.MethodsThis prospective, dual-center, open-label, randomized controlled trial will enroll 140 stage III NSCLC patients. Participants will be randomly allocated in a 1:1 ratio to a control group or an experimental group. Both groups will undergo definitive CCRT. Alongside the commencement of CCRT, the experimental group will receive an additional oral SLD intervention for a duration of three months. The primary outcome is the incidence rate of ASL, defined as the proportion of patients who experience at least one instance of a total lymphocyte count falling below 0.5 × 10^9 cells/L within 3 months of initiating CCRT treatment. Additionally, 16S rRNA gene sequencing analysis of fecal samples to assess gut microbiota, as well as metabolomic analysis of fecal/blood samples, will be conducted to explore potential mechanisms.DiscussionThis study protocol aims to rigorously evaluate the efficacy and safety of SLD, as well as elucidate its mechanism of action in preventing ASL. Successful outcomes could establish SLD as an evidence-based intervention for ASL prevention in NSCLC patients undergoing CCRT.Trial RegistrationThe trial was registered at the Chinese Clinical Trials Registry (ChiCTR2300071788, https://www.chictr.org.cn/).

Summary: We previously reported that loss of function of TYW1 led to cerebral palsy with severe intellectual disability through reduced neural proliferation. However, whether TYW1 loss affects neural differentiation is unknown. In this study, we first demonstrated that TYW1 loss blocked the formation of OHyW in tRNAphe and therefore affected the translation efficiency of UUU codon. Using the brain organoid model, we showed impaired neuron differentiation when TYW1 was depleted. Interestingly, retrotransposons were differentially regulated in TYW1−/− hESCs (human embryonic stem cells). In particular, one kind of human-specific endogenous retrovirus-K (HERVK/HML2), whose reactivation impaired human neurodevelopment, was significantly up-regulated in TYW1−/− hESCs. Consistently, a UUU codon-enriched protein, SMARCAD1, which was a key factor in controlling endogenous retroviruses, was reduced. Taken together, TYW1 loss leads to up-regulation of HERVK in hESCs by down-regulated SMARCAD1, thus impairing neuron differentiation.

The combination of water electrolysis and renewable energy to produce hydrogen is a promising way to solve the climate and energy crisis. However, the fluctuating characteristics of renewable energy not only present a significant challenge to the use of water electrolysis electrodes, but also limit the development of the hydrogen production industry. In this study, the effects of three different types of waveforms (square, step, and triangle, which were used to simulate the power input of renewable energy) on the electrochemical catalysis behavior of Ni plate cathodes for HER was investigated. During the test, the HER performance of the Ni cathode increased at first and then slightly decreased. The fluctuating power led to the degradation of the Ni cathode surface, which enhanced the catalysis effect by increasing the catalytic area and the active sites. However, prolonged operation under power fluctuations could have damaged the morphology of the electrode surface and the substances comprising this surface, potentially resulting in a decline in catalytic efficiency. In addition, the electrochemical catalysis behavior of the prepared FeNiMo-LDH@NiMo/SS cathode when subjected to square-wave potential with different fluctuation amplitudes was also extensively studied. A larger amplitude of fluctuating power led to a change in the overpotential and stability of the LDH electrode, which accelerated the degradation of the cathode. This research provides a technological basis for the coupling of water electrolysis and fluctuating renewable energy and thus offers assistance to the development of the “green hydrogen” industry.

Gastrodia elata Blume, a fully mycoheterotrophic perennial plant of the family Orchidaceae, is a traditional Chinese herb with medicinal and edible value. Interestingly, G. elata requires symbiotic relationships with Mycena and Armillaria strains for seed germination and plant growth, respectively. However, there is no comprehensive summary of the symbiotic mechanism between fungi and G. elata. Here, the colonization and digestion of hyphae, the bidirectional exchange of nutrients, the adaptation of fungi and G. elata to symbiosis, and the role of microorganisms and secondary metabolites in the symbiotic relationship between fungi and G. elata are summarized. We comprehensively and deeply analyzed the mechanism of symbiosis between G. elata and fungi from three perspectives: morphology, nutrition, and molecules. The aim of this review was to enrich the understanding of the mutualistic symbiosis mechanisms between plants and fungi and lay a theoretical foundation for the ecological cultivation of G. elata. Hyperlipidemia is a major risk factor for erectile dysfunction (ED). Oxidative stress and phenotypic modulation of corpus cavernosum smooth muscle cells (CCSMCs) are the key pathological factors of ED. N-acetylcysteine (NAC) can inhibit oxidative stress; however, whether NAC can alleviate pathological variations in the corpus cavernosum and promote erectile function recovery in hyperlipidemic rats remains unclear. A hyperlipidemia model was established using 27 eight-week-old male Sprague–Dawley (SD) rats fed a high-fat and high-cholesterol diet (hyperlipidemic rats, HR). In addition, 9 male SD rats were fed a normal diet to serve as controls (NC). HR rats were divided into three groups: HR, HR+normal saline (NS), and HR+NAC (n = 9 for each group; NS or NAC intraperitoneal injections were administered daily for 16 weeks). Subsequently, the lipid profiles, erectile function, oxidative stress, phenotypic modulation markers of CCSMCs, and tissue histology were analyzed. The experimental results revealed that erectile function was significantly impaired in the HR and HR + NS groups, but enhanced in the HR + NAC group. Abnormal lipid levels, over-activated oxidative stress, and multi-organ lesions observed in the HR and HR + NS groups were improved in the HR + NAC group. Moreover, the HR group showed significant phenotypic modulation of CCSMCs, which was also inhibited by NAC treatment. This report focuses on the therapeutic effect of NAC in restoring erectile function using a hyperlipidemic rat model by preventing CCSMC phenotypic modulation and attenuating oxidative stress. ABSTRACT: It is desirable to obtain high levels of viable Lacticaseibacillus paracasei, a widely used food probiotic whose antibacterial activity and potential application in milk remain largely uninvestigated. Here, we isolated and purified the L. paracasei strain XLK 401 from food-grade blueberry ferments and found that it exhibited strong antibacterial activity against both gram-positive and gram-negative foodborne pathogens, including Staphylococcus aureus, Salmonella paratyphi B, Escherichia coli O157, and Shigella flexneri. Then, we applied alternating tangential flow (ATF) technology to produce viable L. paracasei XLK 401 cells and its cell-free supernatant (CFS). Compared with the conventional fed-batch method, 22 h of ATF-based processing markedly increased the number of viable cells of L. paracasei XLK 401 to 12.14 ± 0.13 log cfu/mL. Additionally, the CFS exhibited good thermal stability and pH tolerance, inhibiting biofilm formation in the abovementioned foodborne pathogens. According to liquid chromatography-mass spectrometry analysis, organic acids were the main antibacterial components of XLK 401 CFS, accounting for its inhibition activity. Moreover, the CFS of L. paracasei XLK 401 effectively inhibited the growth of multidrug-resistant gram-positive Staph. aureus and gram-negative E. coli O157 pathogens in milk, and caused a reduction in the pathogenic cell counts by 6 to 7 log cfu/mL compared with untreated control, thus considerably maintaining the safety of milk samples. For the first time to our knowledge, ATF-based technology was employed to obtain viable L. paracasei on a large scale, and its CFS could serve as a broad-spectrum biopreservative for potential application against foodborne pathogens in milk products.

Musical instrument recognition is the process of using machine learning or audio signal processing to identify and classify different musical instruments from an audio recording. This capability enables more precise analysis of musical pieces, aiding in tasks like transcription, music recommendation, and automated composition. The challenges include (1) recognition models not being accurate enough, (2) the need to retrain the entire model when a new instrument is added, and (3) differences in audio formats that prevent direct usage. To address these challenges, this article introduces MusicTalk, a microservice based musical instrument (MI) detection system, with several key contributions. Firstly, MusicTalk introduces a novel patchout mechanism named Brightness Characteristic Based Patchout for the ViT algorithm, which enhances MI detection accuracy compared to existing solutions. Secondly, MusicTalk integrates individual MI detectors as microservices, facilitating efficient interaction with other microservices. Thirdly, MusicTalk incorporates an audio shaper that unifies diverse music open datasets such as Audioset, Openmic-2018, MedleyDB, URMP, and INSTDB. By employing Grad-CAM analysis on Mel-Spectrograms, we elucidate the characteristics of the MI detection model. This analysis allows us to optimize ensemble combinations of ViT with patchout and CNNs within MusicTalk, resulting in high accuracy rates. For instance, the system achieves precision and recall rates of 96.17% and 95.77% respectively for violin detection, which are the highest among previous approaches. An additional advantage of MusicTalk lies in its microservice-driven visualization capabilities. By integrating MI detectors as microservices, MusicTalk enables seamless visualization of songs using animated avatars. In a case study featuring “Peter and the Wolf,” we demonstrate that improved MI detection accuracy enhances the visual storytelling impact of music. The overall F1-score improvement of MusicTalk over previous approaches for this song is up to 12%.

Abstract It is long been suggested that one‐carbon metabolism (OCM) is associated with Alzheimer's disease (AD), whereas the potential mechanisms remain poorly understood. Taking advantage of chemical biology, that mitochondrial serine hydroxymethyltransferase (SHMT2) directly regulated the translation of ADAM metallopeptidase domain 10 (ADAM10), a therapeutic target for AD is reported. That the small‐molecule kenpaullone (KEN) promoted ADAM10 translation via the 5′ untranslated region (5′UTR) and improved cognitive functions in APP/PS1 mice is found. SHMT2, which is identified as a target gene of KEN and the 5′UTR‐interacting RNA binding protein (RBP), mediated KEN‐induced ADAM10 translation in vitro and in vivo. SHMT2 controls AD signaling pathways through binding to a large number of RNAs and enhances the 5′UTR activity of ADAM10 by direct interaction with GAGGG motif, whereas this motif affected ribosomal scanning of eukaryotic initiation factor 2 (eIF2) in the 5′UTR. Together, KEN exhibits therapeutic potential for AD by linking OCM with RNA processing, in which the metabolic enzyme SHMT2 “moonlighted” as RBP by binding to GAGGG motif and promoting the 5′UTR‐dependent ADAM10 translation initiation.

In this article, the authors present the design of a compact multiband monopole antenna measuring 30 × 10 × 1.6 mm3, which is aimed at optimizing performance across various communication bands, with a particular focus on Wi-Fi and sub-6G bands. These bands include the 2.4 GHz band, the 3.5 GHz band, and the 5–6 GHz band, ensuring versatility in practical applications. Another important point is that this paper demonstrates effective methods for reducing mutual coupling through two meander slits on the common ground, resembling a defected ground structure (DGS) between two antenna elements. This approach achieves mutual coupling suppression from −6.5 dB and −9 dB to −26 dB and −13 dB at 2.46 GHz and 3.47 GHz, respectively. Simulated and measured results are in good agreement, demonstrating significant improvements in isolation and overall multiple-input multiple-output (MIMO) antenna system performance. This research proposes a compact multiband monopole antenna and demonstrates a method to suppress coupling in multiband antennas, making them suitable for internet of things (IoT) sensor devices and Wi-Fi infrastructure systems.

The rapid advancements in Artificial Intelligence of Things (AIoT) are pivotal for the healthcare sector, especially as the world approaches an aging society which will be reached by 2050. This paper presents an innovative AIoT-enabled data fusion system implemented at the CMUH Respiratory Intensive Care Unit (RICU) to address the high incidence of medical errors in ICUs, which are among the top three causes of mortality in healthcare facilities. ICU patients are particularly vulnerable to medical errors due to the complexity of their conditions and the critical nature of their care. We introduce a four-layer AIoT architecture designed to manage and deliver both real-time and non-real-time medical data within the CMUH-RICU. Our system demonstrates the capability to handle 22 TB of medical data annually with an average delay of 1.72 ms and a bandwidth of 65.66 Mbps. Additionally, we ensure the uninterrupted operation of the CMUH-RICU with a three-node streaming cluster (called Kafka), provided a failed node is repaired within 9 h, assuming a one-year node lifespan. A case study is presented where the AI application of acute respiratory distress syndrome (ARDS), leveraging our AIoT data fusion approach, significantly improved the medical diagnosis rate from 52.2% to 93.3% and reduced mortality from 56.5% to 39.5%. The results underscore the potential of AIoT in enhancing patient outcomes and operational efficiency in the ICU setting.

Porcine reproductive and respiratory syndrome virus (PRRSV) is a pathogen that causes severe abortions in sows and high piglet mortality, resulting in huge economic losses to the pig industry worldwide. The emerging and novel PRRSV isolates are clinically and biologically important, as there are likely recombination and pathogenic differences among PRRSV genomes. Furthermore, the NADC34-like strain has become a major epidemic strain in some parts of China, but the characterization and pathogenicity of the latest strain in Inner Mongolia have not been reported in detail. In this study, an NADC34-like strain (CHNMGKL1-2304) from Tongliao City, Inner Mongolia was successfully isolated and characterized, and confirmed the pathogenicity in pigs. The phylogenetic tree showed that this strain belonged to sublineage 1.5 and had high homology with the strain JS2021NADC34. There is no recombination between CHNMGKL1-2304 and any other domestic strains. Animal experiments show that the CHNMGKL1-2304 strain is moderately virulent to piglets, which show persistent fever, weight loss and high morbidity but no mortality. The presence of PRRSV nucleic acids was detected in both blood, tissues, nasal and fecal swabs. In addition, obvious pathological changes and positive signals were observed in lung, lymph node, liver and spleen tissues when subjected to hematoxylin–eosin (HE) staining and immunohistochemistry (IHC). This report can provide a basis for epidemiological investigations and subsequent studies of PRRSV.

Hyperlipidemia is a major risk factor for erectile dysfunction (ED). Oxidative stress and phenotypic modulation of corpus cavernosum smooth muscle cells (CCSMCs) are the key pathological factors of ED. N-acetylcysteine (NAC) can inhibit oxidative stress; however, whether NAC can alleviate pathological variations in the corpus cavernosum and promote erectile function recovery in hyperlipidemic rats remains unclear. A hyperlipidemia model was established using 27 eight-week-old male Sprague-Dawley (SD) rats fed a high-fat and high-cholesterol diet (hyperlipidemic rats, HR). In addition, 9 male SD rats were fed a normal diet to serve as controls (NC). HR rats were divided into three groups: HR, HR+normal saline (NS), and HR+NAC (n = 9 for each group; NS or NAC intraperitoneal injections were administered daily for 16 weeks). Subsequently, the lipid profiles, erectile function, oxidative stress, phenotypic modulation markers of CCSMCs, and tissue histology were analyzed. The experimental results revealed that erectile function was significantly impaired in the HR and HR + NS groups, but enhanced in the HR + NAC group. Abnormal lipid levels, over-activated oxidative stress, and multi-organ lesions observed in the HR and HR + NS groups were improved in the HR + NAC group. Moreover, the HR group showed significant phenotypic modulation of CCSMCs, which was also inhibited by NAC treatment.
