The ever-growing need for improved in vitro models of human tissues to study both healthy and diseased states is advancing the use of techniques such as 3D Bioprinting. We here present our results on the development of a vascularized and perfusable 3D tumor mimic for studies of the early steps of Neuroblastoma metastatic spread. We used a multi-material and sacrificial bioprinting approach to fabricate vascularized 3D cell-laden structures and developed a customized perfusion system enabling maintenance of growth and viability of the constructs for up to 3 weeks. Cell phenotypes and densities in co-culture for both the bulk of the construct and the endothelialization of the vascular channels were optimized to better replicate in vivo conditions and ideally simulate tumor progression. We proved the formation of an endothelium layer lining the vascular channel after 14 days of perfused culture. Cells in the bulk of the construct reflected Neuroblastoma growth and its tendency to recruit endothelial cells contributing to neovascularization. We also collected preliminary evidence of Neuroblastoma cells migration towards the vascular compartment, recapitulating the first stages of metastatic dissemination.

BackgroundMedical simulations have emerged as a valuable tool in anatomical-medical training, allowing healthcare professionals to gain hands-on experience in a controlled and safe environment. One such simulation platform is SimLife®, which uses the Pulse for Practice (P4P) system to enable realistic restoration of airflow (“re-ventilation”) and blood flow (“revascularization”) in bodies donated to science.ObjectiveThis study aimed to evaluate the feasibility of introducing SimLife® technology in Italy. Additionally, it assessed the impact of this technology across various medical specialties, utilizing a minimal number of donated bodies.MethodsThe study utilized the existing body donation program and dissection rooms at the Anatomy Center of the University of Bologna. 62 participants from 13 medical specialties performed simulations using the SimLife® P4P platform. Post-simulation, structured interviews were used to collect data on the interventions performed, participant perceptions of the technology’s usefulness, enjoyment, and willingness to repeat the experience, as well as critical issues encountered.ResultsKey findings include that 86% of participants rated SimLife® technology as extremely useful for post-lauream training, while 84% found it highly beneficial for team-building activities. A total of 31 interventions were successfully performed across various anatomical regions, with participants reporting high satisfaction and a strong willingness to repeat the simulation experience.ConclusionThe findings support the effectiveness of SimLife® technology for body donor re-ventilation and revascularization, reinforcing its value for medical training across various specialties.

Rough surfaces known as stylolites are common geological features that are developed by pressure solution, especially in carbonate rocks, where they are used as strain markers and as stress gauges. As applications are developing in various geological settings, questions arise regarding the uncertainties associated with quantitative estimates of paleostress using stylolite roughness. This contribution reports for the first time a measurement of the temperature at which pressure solution was active by applying clumped isotopes thermometry to calcite cement found in jogs linking the tips of the stylolites. This authigenic calcite formed as a redistribution of the surrounding dissolved material by the same dissolution processes that formed the extensive stylolite network. We compare the depth derived from these temperatures to the depth calculated from the vertical stress inversion of a bedding parallel stylolite population documented on a slab of the Calcare Massiccio formation (early Jurassic) formerly collected in the Umbria-Marches Arcuate Ridge (Northern Apennines, Italy). We further validate the coevality between the jog development and the pressure solution by simulating the stress field around the stylolite tip. Calcite clumped isotopes constrain crystallization to temperatures between 35 and 40 °C from a common fluid with a δ18O signature around −1.3‰ SMOW. Additional δ18O isotopes on numerous jogs allows the range of precipitation temperature to be extended to from 25 to 53 °C, corresponding to a depth range of 650 to 1900 m. This may be directly compared to the results of stylolite roughness inversion for stress, which predict a range of vertical stress from 14 to 46 MPa, corresponding to depths from 400 to 2000 m. The overall correlation between these two independent depth estimates suggests that sedimentary stylolites can reliably be used as a depth gauge, independently of the thermal gradient. Beyond the method validation, our study also reveals some mechanisms of pressure solution and the associated p,T conditions favouring their development in carbonates. [ABSTRACT FROM AUTHOR]
