Phosphatidylethanolamine (PE) is a ubiquitous lipid species in higher eukaryotes. Here, we synthesized a multifunctionalized PE derivative (1) designed to identify PE-binding proteins in intact cells through photo-crosslinking and subsequent isolation and proteomic analysis of the PE–protein conjugates. We show that the tool is also useful for tracking PE translocation to mitochondria after uncaging. A trifunctional phosphatidic acid derivative was rapidly metabolized and is of more limited use. Introduction: Incisional hernias with loss of domain (IHLD) are challenging to treat. Preoperative techniques like botulinum toxin injection (BTA) and preoperative progressive pneumoperitoneum (PPP) are highly effective, potentially preventing the need for perioperative component separation in the vast majority of cases. PPP involves preoperative introduction of gas into the abdominal cavity to increase the abdominal wall volume, aiding diaphragmatic prehabilitation and hernia reintegration. This study aimed at explaining our technique in performing PPP in ambulatory setting. Description of the technique: The first insufflation and BTA injection occur during a 3-day hospitalization. Subsequently, patients are managed on an ambulatory basis with three sessions per week for at least three weeks. Each hospital visit lasts about 1 to 2 h. Patients can remain at home or in a residential center of our hospital. No preventive anticoagulation nor prophylactic antibiotics are needed. Discussion: Performing PPP in outpatient care does not compromise its efficacy. Instead, it allows for longer preparation, potentially improving efficacy. Patients maintain daily activities, possibly yielding better results than traditional physiotherapy. It reduces hospital stay costs and nosocomial infection risks. Each ambulatory hospitalization offers better patient attention. Conclusions: PPP is a valuable preoperative technique for IHLD repair, particularly in combination with botulinum toxin, offering potential benefits for selected patients. Accurate gametogenesis requires the establishment of the telomere bouquet, an evolutionarily conserved, 3D chromosomal arrangement. In this spatial configuration, telomeres temporarily aggregate at the nuclear envelope during meiotic prophase, which facilitates chromosome pairing and recombination. The mechanisms governing the assembly of the telomere bouquet remain largely unexplored, primarily due to the challenges in visualizing and manipulating the bouquet. Here, using Schizosaccharomyces pombe as a model system to elucidate telomere bouquet function, we reveal that centromeres, traditionally perceived as playing a passive role in the chromosomal reorganization necessary for bouquet assembly, play a key role in the initiation of telomere bouquet formation. We demonstrate that centromeres are capable to induce telomere mobilization, which is sufficient to trigger the first stages of bouquet assembly and the meiotic transcription program in mitotic cells. This discovery highlights the finely tuned control exerted over long-distance heterochromatic regions and underscores a pivotal step in the mechanism of eukaryotic telomere bouquet formation and meiotic transcriptional rewiring. Telomeres and centromeres are specialized chromosomal regions with distinct functions. Here, the authors uncover a long-distance molecular communication between these key regions, coordinating the formation of the telomere bouquet and the initiation of the meiotic program. Background: Undernutrition remains a global crisis and is a focus of Sustainable Development Goals. While there are multiple known, effective interventions, complex interactions between prevention and treatment and resource constraints can lead to difficulties in allocating funding. Simulation studies that use in silico simulation can help illuminate the interactions between interventions and provide insight into the cost-effectiveness of alternative packages of options. Methods: We developed an individual-based microsimulation model based on the Global Burden of Disease (GBD) 2021 study data to test a range of nutrition interventions, including antenatal interventions (iron and folic acid, multiple micronutrients, and balanced energy protein supplementation) and child interventions (treatment for severe acute malnutrition, treatment for moderate acute malnutrition, and wasting prevention with small-quantity lipid-based nutrient supplements). We also developed an analytic approach to process the results of the microsimulation and identify the optimal intervention funding allocation for a given budget size. We use Ethiopia as an example in this paper. Results: In our illustrative example of Ethiopia, the reallocation of the baseline budget to minimize disability-adjusted life years (DALYs) resulted in first funding the antenatal multiple micronutrients to their maximum coverage and then funding treatment for severe child acute malnutrition. Relative to the baseline allocation, the reallocation optimized to minimizing DALYs resulted in 592,000 fewer annual DALYs, constituting an 8.3% reduction in total DALYs in Ethiopia. For budgets larger than the baseline, our model recommended funding first targeted moderate acute malnutrition treatment, second universal moderate acute malnutrition treatment, third wasting prevention with small-quantity lipid-based nutrient supplements, and fourth balanced energy protein supplementation. Conclusions: Our simulation is a novel model for estimating optimal allocation of spending on antenatal and child health nutrition interventions which accounts for the interaction between preventive and therapeutic approaches. Our illustrative results show that an optimized reallocation of current spending can substantially improve pregnancy-related and child health without additional funding. Purpose: Explore community-based gym exercise for non-ambulant adults with childhood-onset disability. Materials and methods: Non-ambulant adults with childhood-onset disability participated in four, weekly gym sessions co-facilitated by physiotherapists and exercise professionals. Practicalities of participating in the sessions were recorded via uptake and attrition, weekly surveys, and focus groups. Perspectives of those who designed/delivered the study were gathered via weekly debrief meetings. Quantitative data were analysed descriptively, qualitative data were analysed thematically. Results: Ten non-ambulant adults with childhood-onset disability participated; 70% completed all exercise sessions. Focus groups identified three themes. "I wouldn't be able to exercise...there's no option for a community-based setting" described the lack of opportunities for exercise in gyms. "You don't realise the benefit of coming here" highlighted benefits of exercise. "We can do better" had two sub-themes: problem solving and ingredients for community-based gym exercise. Weekly feedback and debrief meetings identified practicalities related to equipment, exercises, and collaborative working between facilitators. Conclusions: Whilst there is an interest in community-based gym exercise for non-ambulant adults with childhood-onset disability, there remains a lack of inclusive gyms. Co-design of inclusive gym guidelines and condition-specific physical activity referral scheme may enhance opportunities for participation in gym exercise for adults with childhood-onset disability. IMPLICATIONS FOR REHABILITATION: Adults with non-ambulant childhood-onset disability want to access gyms to self-manage their condition. Accessible facilities and provision of dignified toileting would reduce the barriers to participation in community-based gym exercise for non-ambulant adults with childhood-onset disability. Within aquatic ecosystems, chemosensory cues are widely used as publicly available risk assessment information, allowing prey to optimize behavioural decisions. In sexually dimorphic species, males and females often have different responses to local predation threats. According to the ‘distracted male’ hypothesis, reproductively active males should show a reduced response to risk cues compared to conspecific females due to the potentially high costs of lost courtship and mating opportunities among males. Recent evidence supports this hypothesis, showing that adult male guppies (Poecilia reticulata) show a reduced or absent response to chemosensory risk cues. Here, we further test the ‘distracted male’ hypothesis by testing non-reproductive male and female guppies, collected from the Lopinot River (high predation) or the Upper Aripo River (low predation). We found that juvenile male and female guppies exhibited similar response patterns. Consistent with our predictions, both juvenile male and female guppies collected from a high predation site exhibited a similar antipredator response (reduced number of lines crossed) when exposed to conspecific alarm cues or a novel odour (vs. water control). Those collected from a low predation site showed similar responses to conspecific alarm cues and no response to a novel odour or the water control. Combined, our results suggest that unlike adult guppies, non-reproductive male and female guppies make similar behavioural decisions in response to acute predation cues. As predicted by the ‘distracted male’ hypothesis, the previously reported absence of a response to chemosensory risk assessment cues among adult males may be due to a devaluation of risk assessment information in favour of continued mating. Introduction: The ingestion of nanomaterials (NMs) may impair the intestinal barrier, but the underlying mechanisms remain evasive, and evidence has not been systematically gathered or produced. A mechanistic-based approach would be instrumental in assessing whether relevant NMs disrupt the intestinal barrier, thereby supporting the NM risk assessment in the food sector. Methods: In this study, we developed an adverse outcome pathway (AOP) based on biological plausibility and by leveraging information from an existing NM-relevant AOP that leads to hepatic outcomes. We then extracted the current evidence from the literature for a targeted selection of NMs with high relevance to the food sector, namely, ZnO, CuO, FeO, SiO2, and Ag NMs and nanocellulose. Results: We propose a new AOP (AOP 530) that starts with endocytic lysosomal uptake, leading to lysosomal disruption inducing mitochondrial dysfunction. Mitochondrial impairments can lead to cell injury/death and disrupt the intestinal barrier. The evidence collected supports that these food-related NMs can be taken up by intestinal cells and indicates that intestinal barrier disruption may occur due to Ag, CuO, and SiO2 NMs, while only few studies support this outcome for FeO and ZnO. Lysosomal disruption and mitochondrial dysfunction are rarely evaluated. For nanocellulose, none of the studies report toxicity-related events. Conclusion: The collection of existing scientific evidence supporting our AOP linking NM uptake to intestinal barrier impairments allowed us to highlight current evidence gaps and data inconsistencies. These inconsistencies could be associated with the variety of stressors, biological systems, and key event (KE)-related assays used in different studies. Introduction Drug-related hospital admissions (DRAs) can account for 5%–40% of total hospital admissions in older adults, with a significant proportion deemed preventable. To increase the detection of DRAs, in 2021, a revised trigger tool listing 21 frequent causes of admissions and medications at risk was proposed. This study aimed to describe DRAs using this trigger tool in a French acute geriatric ward and to assess the performance of the tool. Methods This was a retrospective cohort study in a 20-bed geriatric unit including all patients hospitalised in 2023. During the first quarter of 2024, each patient's chart was adjudicated by using a two-step standardised review procedure to assess whether the admission was a DRA. The potentially at cause medications and reasons for admission were also assessed. Results During the study period, 483 patients were hospitalised in the acute-care geriatric ward (mean age 86.7 ± 6.15 years). After adjudication, 207 admissions (43%) were identified as DRAs; 70% were considered preventable. The main causes of DRAs were falls/fractures (33%), bleeding (23%) and delirium (14%). The drugs most frequently responsible were diuretics (21%), renin–angiotensin system inhibitors (20%) and direct oral anticoagulants (15%). The overall sensitivity and specificity of the tool for detecting DRAs was 90% (95% CI 88–93) and 72% (68–76), respectively. After adjudication, the trigger tool helped detect 83% more DRAs as compared with the attending geriatrician. Conclusion DRAs are frequent in a geriatric population and often preventable. Background: Effective evidence-based physical activity and nutrition interventions to prevent overweight and obesity and support healthy child development need to be sustained within Early Childhood Education and Care (ECEC) services. Despite this, little is known about factors that influence sustainability of these programs in ECEC settings. Therefore, the aim of this study was to describe the factors related to sustainability of physical activity and nutrition interventions in ECEC settings and examine their association with ECEC service characteristics. Methods: A cross-sectional study was undertaken with a nationally representative sample of 473 Australian ECEC services. Factors related to the sustainability of ECEC-based physical activity and nutrition interventions were assessed using the validated Integrated Measure of PRogram Element SuStainability in Childcare Settings (IMPRESS-C), measuring Outer Contextual Factors, Inner Contextual Factors, Processes and Characteristics of the Intervention domains for interventions that supervisors reported as currently implementing. Participants responded using a 5-point Likert scale, with responses ranging from 1 (completely disagree) to 5 (completely agree). Domain scores were calculated for each service by averaging item responses. Linear regression models between ECEC service characteristics and the IMPRESS-C domains were undertaken. Results: Data from 473 Australian childcare services nationally found that the domains: Processes ( x ¯ =3.78, SD = 0.64), consisting of partnership/engagement and training/support/supervision; and Outer Contextual Factors ( x ¯ =3.93, SD = 0.63), including policy and legislation, and socio-political context had the lowest mean scores indicating they may likely be barriers to sustainability. Linear regression analyses revealed no statistically significant associations between examined factors and ECEC service characteristics. There was a statistically significant association between the number of years services delivered their interventions and the Characteristics of the Intervention domain (p = 0.035) suggesting that this domain may influence sustainability of programs. Conclusions: This study suggests that factors related to the Processes and Outer Contextual Factors domains had the lowest scores and as such, strategies to support the sustainability of physical activity and nutrition interventions implemented in ECEC settings may need to consider how to best address these factors. AbstractWe examined how suicide is discussed online in chat counseling services for youth. The data included 4,340 chat conversations between young service users and adult counselors. Conversations using the word suicide (n = 558; 12.9%) were filtered, and conversations by 31 boys and 31 girls aged 15–29 years were selected for in-depth analyses; of these conversations 24 depicted previous suicide attempts and 15 suicide plans. Common reasons discussed were prolonged mental health challenges and fragmented relationships. Our results showed that young people seek help for suicidal behavior online; thus, professionalism and careful ethical consideration are needed when providing chat services. [ABSTRACT FROM AUTHOR]

The article examines the socio-educational barriers and facilitators faced by families of children with cochlear implants (CI) in Andalucía (a region in southern Spain). Through a qualitative approach based on surveys and interviews, three main categories of limitations are identified: access to and quality of services, parental stress related to caregiving and communication, and socioeconomic and individual factors. Among the facilitators, financial support, family-professional collaboration, and inclusive practices in educational settings are highlighted. The findings underscore challenges such as the lack of coordination between medical and educational services, the need for teacher training, and the emotional and financial impact on families. Conversely, effective strategies such as family support networks, specialized hospital coordinators, and inclusive educational practices are emphasized. The study concludes that improving access to comprehensive services and fostering collaboration between educational and healthcare agents is essential to promote inclusive education and the well-being of families and children with CI.
