The tertiary structures of the photosynthetic cytochrome c2 of Rhodospirillum rubrum and eucaryotic mitochondrial cytochrome c are compared, together with data on their physiochemical properties and activities in physiological oxidoreduction systems. The comparison gives rise to the following observations and proposals. 1. R. rubrum cytochrome c2 does not undergo the extensive oxidoreduction-linked conformation change characteristic of eucaryotic cytochromes c, but rather is structurally constrained in a conformation in both oxidation states which is over-all most similar to that of ferrocytochrome c. 2. Upon examination and comparison of the surface structural and charge topography of cytochromes c and c2, it is apparent that they bear by far the closest similarity at their front sides (i.e. facing the heme crevice). This feature manifests itself clearly owing to the absolute invariance of many of the amino acid residues surrounding the perimeter of the heme crevice in both cytochrome c2 and 41 species of eucaryotic cytochrome c. 3. A physiological mechanism for the oxidoreduction of eucaryotic cytochrome c is proposed which is in principle similar to that previously suggested for cytochrome c2 (Sa-lemme, F.R., Freer, S.T., Xuong, Ng.H., Alden, R.A., and Kraut, J. (1973) J. Biol. Chem. 248, 3910–3921). This mechanism involves front side interaction of the cytochrome c molecule with its oxidase and reductase, and direct electron addition to and withdrawal from the heme concomitant with destabilization of the existing heme oxidation state.

The two cytochromes c for which detailed tertiary structures have been obtained recently—cytochrome c from mammalian mitochondria and cytochrome c2 from Rhodospirillum rubrum—have been compared in reactivity in the physiologic redox systems for which the former is the natural substrate. These systems are the mitochondrial DPNH-cytochrome c reductase (complex I–III) and cytochrome c oxidase (complex IV). In addition, the effects of antimycin A (a specific inhibitor in the reductase system) and L-polylysine (usually employed to inhibit the oxidase system) on the reduction and oxidation of cytochromes c and c2 have been studied. Preliminary data for these two cytochromes, as well as for five others representative of different classes of bacterial and algal cytochrome c are presented. Implications of these findings for current attempts to rationalize the structural features of cytochromes c are considered.

The structure of ferricytochrome c2 from the non-sulfur purple photosynthetic bacterium Rhodospirillum rubrum has been determined at 2 A resolution by x-ray crystallographic methods. The 112-residue polypeptide chain encloses a single covalently bound heme in a predominantly hydrophobic environment, leaving only one edge exposed to the solvent at the front of the molecule. Distributed around the exterior perimeter of the heme crevice are 11 lysine residues which form an essentially uninterrupted positively charged patch. The molecule contains four segments of α-helix, with the conformation of the remainder of the chain being primarily governed by interactions with the heme. A hypothetical scheme for folding of the molecule is presented, based primarily upon the assumption that the helical regions are formed first and are subsequently distorted by hydrophobic interactions with the heme. The heme iron is coordinated to nitrogen Ne2 of His 18 and to the sulfur atom of Met 91 in the fifth and sixth positions. A notable feature of the structure is an apparently "off axis" or bent Met 91 sulfur-iron bond. The non-axial nature of this bond is attributed to a charge-pair interaction between the Tyr 70 hydroxyl oxygen, bearing a partial negative charge, and the Met 91 sulfur atom, bearing a partial positive charge delocalized from the ferriheme iron. This interaction serves to stabilize the oxidized heme. The additional participation of Tyr 70 in a hydrogen bond network via Tyr 52 to Ser 89 on the front surface of the molecule suggests that the physiological mechanism for reduction of cytochrome c2 involves concerted protonation of Ser 89 concomitant with electron donation to the heme. The process is reversed for oxidation. This proposed mechanism has several chemical and structural implications, the most important being that the site of interaction with physiological oxido-reductants is at the front of the cytochrome c2 molecule. These conclusions appear to be substantiated by a variety of published findings, including pH dependence of the midpoint potential of cytochrome c2 and polylysine inhibition of cytochrome c2 reactivity in physiological oxidation-reduction systems.

La transición Pleistoceno/Holoceno fue un tiempo crítico ambiental en el cual se produjo la dispersión humana del último continente en ser poblado, América del Sur. Los primeros colonos, que buscaban lugares para establecerse, fueron desafiados en América por la enorme variabilidad ambiental y los profundos y repentinos cambios climáticos de los momentos de la transición. Este artículo trata de ese proceso de poblamiento en la Patagonia, donde las ocupaciones humanas más antiguas están datadas entre 13.5 y 10.5 ka AP en tres diferentes focos geográficos: (a) las cuencas andinas de la vertiente Pacífica; (b) la cuenca del Macizo del Deseado; y (c) la cuenca magallánico-fueguina. Siguiendo los datos arqueológicos y paleoambientales (polen, faunas, sedimentos) y los fechados radiocarbónicos se proponen hipótesis sobre las formas de la colonización y la movilidad humana durante el final de la última gran glaciación y el Holoceno medio. El ingreso de los primeros grupos de cazadores-recolectores podría haber ocurrido por dos rutas diferentes, el borde Pacífico y la vertiente Atlántica. Desde ambos litorales y remontando los ríos hacia el interior del territorio los grupos habrían alcanzado en forma diferida ambos flancos cordilleranos. La alta movilidad de los primeros grupos humanos habría sido la clave para enfrentar el desafío de la colonización en esas altas latitudes. A device for the rapid and accurate measurement of model molecular co-ordinates, to be used in conjunction with a Richards optical comparator, is described. The device may be operated in either a manual or automatic mode. The manual mode allows an operator to find the co-ordinates of a desired atom by optical superposition of the transmitted image of a small marker light upon the reflected image of the atom to be measured. The automatic mode allows the operator to position the marker light automatically by entering preselected co-ordinates from an electronic console. This mode of operation facilitates the rapid construction and comparison of structures the atomic co-ordinates of which are already known. The device utilizes pulsed stepping motors to position the marker light and incorporates modularized solid-state circuitry throughout. Several applications of the device are described.
