Based on the study of 40 patients in a unit of a large geriatric center, as well as on long overall experience, the problems of aged men and women with various emotional disorders are discussed. Case abstracts are presented to illustrate chronic organic brain syndrome, delirium, hallucinatory behavior, paranoid ideation, depression, and reactions triggered by physical illness. The geriatrician should be fully aware of all the interlocking factors in the patient's adjustment to his condition. Only in this way can the geriatrician and staff choose the best relevant therapy.

We have studied the properties of presumptive point mutants in the D2a region of bacteriophage T4. Dominance tests showed that the D2a mutation was recessive to the wild-type allele. The mutations were shown to map in the D2a region by complementation against rII deletions. The D2a mutations were also located between gene 52 and rIIB by two- and three-factor crosses. The mutants are located at at least two distinct loci in the D2a region. The point mutants grow normally on all hosts tested and none of the mutants makes T4 endonuclease IV. We propose the name “denB” for the D2a locus.

Levamisole was tested to determine whether the drug could reduce metastases by HSV-1-transformed cells in a model hamster system. The results presented reveal an inhibition of metastases to the lungs even when the drug is inoculated after development of subcutaneous tumors at the site of inoculation of the cells.

We have developed an in vitrocomplementation assay to demonstrate packaging and maturation of DNA of phage T7. Cells of Escherichia coliB infected with an appropriate T7 amber mutant are concentrated 200-fold and lysed by freezing and thawing. Two extracts from cells infected with different amber mutants are mixed and incubated at 30°. Positive complementation results in a 100-fold increase in phage titer.Using this assay we have demonstrated the packaging of phage DNA from an extract that contains no phage heads (gene 9-, 10-), within head structures present in an extract that contains no phage DNA (gene 5-). We have also demonstrated an activity in extracts that contain no phage DNA or heads (gene 5-, 9-, 10-), which complements gene 19--infected cells. We have proven that this activity is due to the gene-19 product by showing that the activity is temperature-sensitive if the extract is made from cells infected with a mutant having a temperature-sensitive mutation in gene 19. This assay should be useful in elucidating the mechanism of packaging and maturation of DNA of phage T7.

A practical integer programming procedure is presented which can be useful to designers of production facilities. Specifically, the version of the procedure given here can be used to determine optimal or near optimal design configurations for the unloading portion of a high speed, mixed product production line. The model is validated with actual industrial data and several examples and results are given. Tests of the design methodology indicate that least cost answers are obtained. A discussion to aid the would-be industrial user of the procedure is given.

The creep rupture behavior, and the effects of elevated temperature exposure in air and hydrogen on the subsequent room temperature properties of a 12 percent Ni-5 percent Cr-3 percent Mo maraging steel are described. Tests have been made on several heats of plate and on gas tungsten-arc, gas metal-arc, and electroslag welds. On the basis of the results obtained, maraging steels offer promise as high-strength steels for service at elevated temperatures.

An improved procedure for the purification of T4 endonuclease IV from T4+-infected Escherichia colicells is described. The enzyme preparations are highly concentrated, free of exonuclease contamination, and display about 200-fold greater activity toward single stranded DNA than toward duplex DNA. The limit product obtained by exhaustive digestion of single stranded DNA with endonuclease IV is 25% acid-soluble and consists of oligonucleotides with an average chain length of about 50 nucleotides. The enzyme may be useful in sequencing DNA.

Isolated rat liver nuclei were washed with Triton-X-100 in the presence of liver cell sap. This treatment liberated a fraction of polysomes which were isolated by differential centrifugation and were designated "outer membrane polysomes." The outer membrane polysomes synthesized protein in vivo. Shortly after injection of orotic acid-14C, the RNA of outer membrane polysomes had a higher specific activity than that of cytoplasmic polysomes. It was postulated that outer membrane polysomes may be an intermediate in the transfer of newly synthesized RNA from the nucleus to the cytoplasm. In other experiments, Triton-washed rat liver nuclei were lysed in the presence of deoxycholate and deoxyribonuclease. A ribonucleoprotein fraction was isolated from the lysate by differential centrifugation. This fraction contained "intranuclear ribosomes," which sedimented like partially degraded polysomes in sucrose gradients. This degradation could be partially prevented if intranuclear ribosomes were purified by sedimentation through heavy sucrose. The resulting pellets were termed "intranuclear polysomes" because they contained some undergraded polysomes. Intranuclear polysomes were highly radioactive after a brief pulse with orotic acid-14C, but did not appear to synthesize protein rapidly in vivo. Intranuclear polysomes may represent the initial stage of assembly of polyribosomes in the nucleus.

Rat liver nuclei were freed of cytoplasmic contamination by washing with Triton-X-100 and subsequent centrifugation through 2.2 M sucrose. Electron microscopic examination showed that the outer membranes of the nuclei had been removed, but that the nuclei otherwise resembled the nuclei of intact liver. Morphological studies, chemical estimations of DNA, RNA, and protein and the estimation of cytoplasmic "marker" enzymes suggested that contamination of nuclei by cytoplasmic components was limited. These nuclei were obtained in yields of about 70% and were suitable for the isolation of nucleoli. Nucleoli were isolated by the breaking of the nuclei by ultrasound and subsequent differential centrifugation. In ultrastructural appearance, the isolated nucleoli resembled nucleoli in intact tissue. However, at high magnifications the "granular" component of isolated nucleoli appeared to consist of tightly twisted fibers. The nucleoli could be obtained in yields of at least 30%, and the values for the chemical composition of the isolated nucleoli agreed with values previously reported.

The action of synthetic oxytocin on renal sodium excretion was studied in dogs under barbiturate anesthesia given a moderate saline load. Left renal artery infusion of oxytocin at a rate 5 mU/kg per hour produced comparable and significant decreases in sodium excretion by the left and by the right kidney. Since reduction of oxytocin dose to 0.5—1.5 mU/kg per hour simultaneously abolished sodium retention by the infused and the contralateral kidney, it was concluded that oxytocin's action on the kidney was indirect. The observation that postoxytocin decrease in sodium excretion occurred with no significant changes in glomerular filtration rate suggested very strongly that the cause of sodium retention was an increase in renal tubular sodium reabsorption. These findings contradict most of the previous data which indicated saluresis and inhibition of tubular sodium transport by oxytocin, and suggest that oxytocin may have differential effects, depending on dosage and mode of administration. The mechanism of antinatriuretic action of oxytocin is unknown.

The following report which explores the possibility of state action on the newsmen's privilege controversy, is from the author's 1973 Ph.D. dissertation completed at the University of Iowa. Dr. Sadowski, is on the communication faculty at Florida Atlantic University.

In this age of what many critics refer to as commercial proliferation (what the industry terms “piggybacking"), it is becoming increasingly important to advertisers and broadcasters to find out what of the plethora of commercial messages remains with the average viewer/listener. In an evening or an hour of TV watching, which messages are remembered—and why? With some answers of value to those in the industry and those aspiring to enter it, the author here compares his findings to a decade-old study on the same issue— and basically agrees with the earlier findings suggesting video is usually remembered over audio, and (perhaps surprisingly) shorter commercials are often remembered over longer ones. This research was conducted by Mr. Sadowski while he was a Ph.D. candidate in broadcasting and communication at the University of Iowa (a degree he hopes to attain by December 1972). This fall, Mr. Sadowski is to be an assistant professor of communications at Florida Atlantic University.

The utility of the thiocarbonyl group for blocking the C2hydroxyl in methyl glucosides has been demonstrated. Methyl 2-O-(benzylthio)carbonyl-α-D-glucopyranoside has been synthesized in good yield and its structure proved. Use of this 2-O-thiocarbonyl derivative in conjunction with acid-labile blocking groups has permitted the first syntheses of the following esters of methyl α-D-glucopyranoside: 3-O-benzoyl-, 3,4-di-O-benzoyl-, and 3,4,6-tri-O-benzoyl-, and the crystalline 2-O-(p-toluene)sulphonate derivative of each.

Photoluminescence from the bismuth isoelectronic trap implanted into GaP is investigated for various ion doses and annealing conditions. A thin coating of silicon oxide is found to be essential to prevent thermal decomposition of the implanted surface during annealing. In isochronal anneal studies the intensity of the Bi luminescence increases rapidly between 650 and 800°C. For 200 keV implants, the dose for maximum luminescence is ∼3.5 × 1012 ions/cm2, corresponding to an average concentration of ∼1018/cm3. However, for the optimum dose and anneal, one observes only ∼10 per cent of the light intensity expected from the estimated number of substitutional ions, and the emitting ions are situated in lattice sites with strong local strain. This strain is evident in the photoluminescence spectrum by the appearance of a strong no-phonon line which results from an otherwise forbidden transition when strain or an external field are present. The luminescence data are correlated with channeling experiments reported in a companion paper.

Bacteriophage T7 bearing amber mutations in both gene 1.3 (T7 DNA ligase) and gene 3 (T7 endonuclease I) are viable when grown in suppressor-negative, ligase-negative hosts. This is evidenced by a high plating efficiency and a large burst size compared to the single mutants. These findings may be explained by a limited destruction of cellular DNA by the double mutant.

The degradation of bacterial deoxyribonucleic acid (DNA) was studied after infection of Escherichia coliB with DNA-negative amber mutants of bacteriophage T7. Degradation occurred in three stages. (i) Release of the DNA from a rapidly sedimenting cellular structure occurred between 5 and 6 min after infection. (ii) The DNA was cleaved endonucleolytically to fragments having a molecular weight of about 2 × 106between 6 and 10 min after infection. (iii) These fragments of DNA were reduced to acid-soluble products between 7.5 and 15 min after infection. Stage 1 did not occur in the absence of the gene 1 product (ribonucleic acid polymerase sigma factor), stage 2 did not occur in the absence of the gene 3 product (phage T7-induced endonuclease), and stage 3 did not occur in the absence of the gene 6 product.

A brief review of the kinetic formation of oxides of nitrogen is presented. Distinction is made between thermal NOx and fuel nitrogen NOx. The correlation of results is based both on data from literature and field data from Riley Stoker’s Air Pollution Study Program. The emission of NOx is shown to be capable of correlation based on fuel nitrogen, excess air, fuel, burning area heat release, combustion air temperature, furnace wall deposits, and furnace type. The results correlate to a standard deviation of 60 ppm.

The biosynthesis of a phytotoxic glycopeptide of Corynebacterium insidiosuminvolves guanosine diphosphate-l-fucosyltransferase activity. This enzyme activity is most consistently associated with the cellular membranes fraction. The optimal pH for the transfer reaction is 7.5. The partially hydrolyzed toxin serves as an acceptor (primer) of l-fucose.
