The muscle relaxant activity of cyclobenzaprine relative to chlorpromazine and diazepam in several animal models manifesting hypertonic skeletal muscle activity is described. In mice subjected to electrical and chemical induced tonic-extensor seizures, only cyclobenzaprine achieved a protective index greater than unity in all preparations. Cyclobenzaprine also appeared to be highly potent and selective in abolishing local tetanus in rabbits. Relative to the other compounds, cyclobenzaprine was consistently active in γ- and α-decerebrate as well as in ischemic spinal cord rigid cat preparations, at doses which produced no signs of motorincoordination (ataxia) or behavioural depression. Consequently, only cyclobenzaprine exhibited high protective indices and those of greater than unity for all three cat preparations. Furthermore, in contrast with chlorpromazine and diazepam, repeated administration of cyclobenzaprine to ischemic spinal cord rigid cats failed to induce tachyphylaxis. Cyclobenzaprine was also well absorbed orally in mice and cats. These observations suggest that cyclobenzaprine may be useful in the treatment of human pathological conditions manifested by excessive tonic skeletal muscle activity. The potential usefulness of cyclobenzaprine as a pharmacological tool for further studies involving somatic motor systems is also suggested.

The cardiovascular effects resulting from intraventricular injections of 600 μg picrotoxin (a brain stem noradrenaline releasing agent) were studied in vagotomized cats previously injected intraventricularly with phenoxybenzamine (2 mg) or dichloroisoproterenol (2 mg), and compared with untreated animals. Intraventricular injections of picrotoxin always induced marked pressor responses associated with electrocardiographic changes as ventricular extrasystoles, ST-T segment deviations and tachycardia. After phenoxybenzamine, picrotoxin induced smaller (but insignificantly different) pressor responses; no ventricular extrasystoles, as well as noticeably reduced degrees of ST-T segment deviations; and a significantly greater degree of cardioacceleration. Following dichloroisoproterenol, picrotoxin induced pressor responses associated with ventricular extrasystoles and ST-T segment deviations similar to those observed in untreated animals, but a significantly smaller degree of cardioacceleration. The results are discussed, and lead to the suggestion that central sympathetic brain areas have receptors similar to those postulated by ahlquist (1948) for peripheral adrenergic receptors.

Summary Experiments are presented to show that injections of small doses of picrotoxin into the lateral cerebral ventricles of rabbits induce a rise in blood pressure and cardiac arrhythmias (bradycardia, tachycardia, extrasystoles, ventricular fibrillation). The bradycardia is largely abolished by bilateral vagotomy. Both the pressor response and other cardiac arrhythmias are markedly diminished or abolished by previous injections of reserpine, phenoxybenzamine, and by spinal C2 section, but not by adrenalectomy or spinal T4-5 section. Pretreatment with LSD did not prevent these changes. The convulsant action of picrotoxin does not appear to contribute to these cardiovascular responses. It is concluded that picrotoxin can stimulate central sympathetic mechanisms leading to increased sympathetic outflow affecting the cardiovascular system, although the specific brain areas involved remain to be determined.
