XD–6S cells cultured in Murashige and Skoog’s medium containing kinetin and indole–3–acetic acid (IAA) were harvested at daily intervals and fixed in 5% gtutaialdehyde. Post–fixation was done in 1% osmium tetroxide. Electron microscopic observations revealed that the microbodies were continuous to rough surfaced endoplasmic reticulum. Further electron microscopic cytochemical observations of the cells incubated in 3,3′–diaminobenzidine (DAB) and hydrogenpero–xide medium, which has been recently used to reveal the localization of catalase in microbodies in animal and plant cells, led to conclude that the microbodies were derived from rough–surfaced endoplasmic reticulum. Tobacco cultured cell clone XD-6S in medium containing kinetin and indole-3-acetic acid were fixed in 5% glutaraldehyde and post-fixed in 1%osmium tetroxide. Examinations of serial sections of crystalloids in the microbodies in XD-6S cells led to conclude that the shapes of the crystalloids were roughly cubic, and there were three principal lattice shapes. Further electron microscopic observation of the crystalloids revealed that the unit members of the crystalloid were rods, which contained 6 electron dense cores about the perimeter and additional cores in the center. The rod was approximately 0.5μ in length and had a diameter of 75Å. Generally several folds of the rods were found on each side of the mature crystalloid. [ABSTRACT FROM PUBLISHER]
