Earlier investigations have shown that DNA synthesis in rabbit kidney cortex cells cultured directly from the animal begins after a 30- to 40-hour lag period, and that an apparently related event in the life history of these cells is a relatively abrupt doubling in the over-all rate of RNA synthesis midway through this lag period. The nature of the RNA synthesized at different times during the lag period has now been investigated by sucrose gradient sedimentation analysis.
