6. Polyene-Resistant Mutants of Aspergillus fennelliae: Sterol Content and Genetics

Abstract

Mutants of Aspergillus fennelliae(Neosartorya fennelliae) resistant to relatively high levels of amphotericin B and low levels of nystatin were obtained by successive transfers of wild type in the presence of increasing concentrations of the polyenes. The resistance of the mutants to the polyenes was accompanied by both qualitative and quantitative changes in the sterol composition of the cells. Those resistant to amphotericin B (AF5-AB1 and p-AB1) lacked ergosterol, the major sterol of the wild type, but contained a new sterol clearly distinguished by the pattern of ultraviolet spectrophotometry and thin-layer chromatography. The mutants resistant to nystatin, however, contained both ergosterol and a new sterol, but the former was produced in a much reduced amount, as compared with the wild type. Genetic analysis indicated that the lack of ergosterol is closely associated with a reduced growth rate, poor asexual reproduction, and the loss of sexual reproduction. Growth studies revealed that the addition of ergosterol to the media did not affect the growth pattern of the mutants. Mutants resistant to amphotericin B showed an increased minimal inhibitory concentration for nystatin, pimaricin, and filipin. Mutants resistant to nystatin, however, conferred increased minimal inhibitory concentration for pimaricin and filipin but not for amphotericin B.

7. Reflectance and ellipsometric study of anodic film on platinum electrode

Abstract

The anodic films on platinum electrode in 0.05 M sulfuric acid solution were examined by means of ellipsometry. By measuring the relative phase retardation (Δ), relative amplitude reduction (tanψ) and also the intensity of the reflected polarized light, the optical constants of the films (n, k) and thickness (τ) at various potentials were obtained. The results showed that the optical properties of the film remain nearly constant (n= 2.8, k= 1.7) while the thickness increases from 2 Å to 5 Å with anodic potential. From this evidence, it is concluded that the film is made of an oxide of platinum forming a definite phase.

8. Comparison of Cardiorespiratory Changes in Surviving and Nonsurviving Shock Dogs

Abstract

Hemodynamic measurements in experimental hemorrhagic shock have been widely studied in the past.1-6 Most of the reported studies were conducted on an acute hemorrhagic shock preparation similar to that described by Wiggers.1 These studies consistently described decreased cardiac output (CO) and increased peripheral resistance, which began shortly after the onset of hemorrhage. On replacement of the shed blood, CO transiently returned toward, but usually did not reach, control values. Then CO declined again, and most of the animals died within about six hours.1 The acute hemorrhagic shock preparations such as Wiggers' technique have been used to provide a standardized experimental shock preparation to study physiological alterations.
