3. Immunological and electrophoretic studies on dog alkaline phosphatase, with particular reference to serum

Abstract

An investigation was carried out to determine the tissue of origin of serum alkaline phosphatase in normal dogs and in dogs treated with the insecticide dieldrin. The work involved comparisons between tissue and serum alkaline phosphatases on the basis of immunological and electrophoretic properties. Alkaline phosphatases from dog liver, kidney, intestine and bone were separable by electrophoretic pattern on polyacrylamide gel, except for some overlapping between bone and renal bands. By electrophoretic analysis, liver appeared to be the major source of serum alkaline phosphatase in both normal and hyperphosphatasemic sera from dieldrin treated dogs. Rabbit antiserum to dog intestinal alkaline phosphatase was used to demonstrate phosphatase of intestinal origin in some dog sera. The presence of this additional serum enzyme was not related to administration of dieldrin. The work was extended to an investigation of the origin of alkaline phosphatase in dog urine. Urinary phosphatase differed electrophoretically and immunologically from that of the kidney and showed some similarity to bladder alkaline phosphatase. Alkaline phosphatase from dog intestinal mucosa was shown to differ from phosphatases extractable from the predominant bacterial flora of dog duodenum and faeces. Immunofluorescence was used to demonstrate alkaline phosphatase on the outer surface of intestinal villi.

5. Ligand bridging in the oxidation of chromium(II) at mercury electrodes

Abstract

Controlled potential electrolysis and electrochemical relaxation techniques have been used to investigate the mechanism of oxidation of chromium(II) at mercury electrodes in the presence of chloride, bromide, and iodide. It is shown that the major path for the oxidation is through a bridged activated complex of the type Hg---X--Cr. Kinetic parameters have been determined for the bromide-dependent path and the halide-independent path. Equations have been derived to predict from kinetic data the product distribution between two competing reactions. It is suggested that the activated complex is formed with halide adsorbed on the electrode surface. The limitations of chronopotentiometry at hanging mercury drop electrodes have been investigated and corrections are given for spherical diffusion effects at times greater than 0.1 sec. It is demonstrated that reverse current chronopotentiometry under these conditions is a qualitatively unreliable technique.

6. Experimental Production of Lethal Escherichia coliBacteremia of Pelvic Origin

Abstract

To reproduce the syndrome of overwhelming Escherichia colibacteremia and shock after pelvic instrumentation, a model was developed by feeding E. colivia drinking water to coliform-free rabbits, injecting nitrogen mustard intravenously, and inserting a temperature probe into the rectum. The temperature probe was inserted to mimic pelvic instrumentation of patients and to detect fever. Rabbits fed invasive serotypes of E. coliall suffered overwhelming bacteremia with high fever and fatal vascular collapse secondary to invasion of pelvic veins as the granulocyte count approached zero. In the absence of granulocytopenia, the rectal temperature probe produced an intensive inflammation with numerous polymorphonuclears and bacteremia did not develop. In the absence of rectal probing, granulocytopenic rabbits developed high fever without bacteremia. This model resembles human bacteremic shock with respect to the endogenous source of the bacteria, the high frequency of bacteremia due to E. coliand other enteric bacilli, the importance of pelvic instrumentation, and the associated immune disturbances such as granulocytopenia.
