- Abstract
In previous work a new method of histologic grading of malignancy in squamous cell carcinomas of the larynx was described. It was based on morphologic criteria representing the tumor cell population itself and the tumor host relationship. Nuclear polymorphism was revealed to be a very important factor for determining prognosis. This method of histologic grading of malignancy was in this work further analyzed by cytophotometric DNA determinations on individual cells obtained by scraping the surface of squamous cell carcinomas of the larynx and the tongue. Increased heterogenous cellular DNA distributions were found in cases with high histological malignancy grading. Cells in the carcinoma cases with a normal epithelial morphology had the same DNA content as that of control cells of normal buccal mucosa. Cells with higher DNA values all had malignant or suspicious morphology. [ABSTRACT FROM AUTHOR]

A metric study was performed on radiographs of permanent maxillary central incisors of a normal Swedish population of 1,038 children. The normal distribution of the root lengths revealed two extreme groups: 2.4 % had short roots, formerly described as Short Root anomaly, most prevalent among girls with a boy:girl ratio of 1:2.6. In the opposite extreme group, i.e., with exceptionally long roots comprising 2.3 % of the children, the boys dominate with a boy:girl ratio of 5:1. Clinical evidence suggests that variation in root length is of genetic origin. [ABSTRACT FROM AUTHOR]

Rat liver microsomes were peroxidized in vitro and chemical, physical and morphological changes in the membrane were related to the effects on certain membrane-bound microsomal enzymes. With increasing peroxidation, microsomal phospholipids revealed decreasing concentrations of 20:4 and 22:6 fatty acids. At a high level of peroxidation, where alterations in permeability and ultrastructural changes such as breakage and deformation of microsomal vesicles were apparent, there was also a decrease in the concentration of 18:2 fatty acids. Glucose-6-phosphatase, cytochrome P-450 and uridine-5′-diphosphate glueuronyltransferase (UDP glucuronyltransferase) (three microsomal enzymes known to be highly dependent on the integrity of the membrane for their function) responded differently to lipid peroxidation. Glucose-6-phosphatase showed an initial decrease in activity as well as in K
m for glucose 6-phosphate, followed by a restoration to control values at an intermediate level of lipid peroxidation. At a high level of peroxidation, both kinetic parameters were again markedly decreased. Cytochrome P-450 concentration, on the other hand, revealed a continuous decrease with proceeding lipid peroxidation, which was only partly accounted for by a conversion to cytochrome P-420. The loss of cytochrome P-450 was paralleled by decreased rates of aminopyrine demethylation and 3,4-benzpyrene hydroxylation. Glucuronyltransferase activity, finally, was activated at low, but again normalized at high, levels of lipid peroxidation. It thus seems that although electron-microscopically observable changes in the microsomal membrane occurs only late in lipid peroxidation, chemical, physical and enzyme alterations are early phenomena during this process. The continuous loss of cytochrome P-450 and the final decrease in glucose-6-phosphatase late during peroxidation probably reflect damage to membrane lipids of vital importance for the integrity and function of these enzymes whereas the activation of UDP glucuronyltransferase early during peroxidation most likely can be ascribed to increased membrane permeability. A follow-up study was made of 125 patients with carcinoma of the gums treated from 1958 to 1969. Twenty-one of these patients with cytologically verified metastases of the neck at first visit were specially studied. The primary tumour was classified as T1 in three patients, T2 in one, T3 in nine, and T4 in eight patients. Twelve patients received radiotherapy, nine radiotherapy and surgery. The five-year determinate survival rate was 7 per cent in this group compared with 41 per cent for patients without cervical metastases. The importance of the use of fine-needle biopsy and cytology is stressed in the diagnosis of metastases of the neck for proper judgement of the prognosis and for selection of method for treatment.

The combined effects of freezing rate, frozen storage and reconstitution method on meat quality and yield were studied in multifactorial experiments using pan-fried 1.5 cm slices of longissimus muscles and deep-fat fried meat patties. On the whole, a slight improvement in sensory quality and yield was seen with increasing freezing rate, except that liquid nitrogen immersion freezing influenced flavor negatively. In contrast with earlier results of ours with raw beef, an advantage was now seen for reconstitution after previous thawing over reheating directly from the frozen state, and pan frying gave better sensory quality but lower yield than continuous microwave reheating. Tenderness of sliced, cooked beef tended to increase during frozen storage, while the opposite was earlier observed for frozen raw beef. Otherwise, a general trend was seen towards decreasing yield and sensory quality during frozen storage. A number of significant interactions between variables was obtained, suggesting that their influence on quality and yield is interrelated in a rather complex manner.

Data for basaltic rocks dredged on the Reykjanes Ridge and Jokulbanki off the southwest coast of Iceland are presented. The rocks from the Reykjanes Ridge, which are identified as recent lavas, are olivine tholeiites and quartz tholeiites very similar to those formerly described from this region. The heterogeneous Jokulbanki dredges were identified as erratics. In the light of the present and other recent data on tholeiitic rocks from Iceland and the Reykjanes Ridge, a distinct chemical gradient, together with increased scatter across Iceland is demonstrated for certain incompatible elements. This is in accordance with the mantle plume theory.

In 1969-1970 a breast mass-screening was undertaken in Gävle, with thermography and questionnaire. Mammary radiography and clinical examination of subjects with abnormal screening reports and of a group of normal subjects were performed. In spite of having achieved early detection of malignant tumours in many cases with a preponderance of carcinoma cases in the abnormal group, thermography was found to have considerable drawbacks as a screening method.

An equivalent electrical circuit has been constructed for small atrial trabecula of frog in a double sucrose gap voltage clamp apparatus. The basic strategy in constructing the circuit was to derive the distribution of membrane capacitance and extracellular resistance from the preparation's response to small voltage displacements near the resting condition, when the membrane conductance is presumably quite low. Then standard Hodgkin-Huxley channels were placed in parallel with the capacitance and the results of voltage clamp experiments were simulated. The results suggest that the membranes of the preparation cannot in fact be clamped near the control voltage nor can the ionic currents be measured directly with reasonable accuracy by axon standards. It may or may not be a realizable goal in the future to define the preparation's electrical behavior well enough to permit the ultimate quantitative description of the membrane's specific ion conductances. The result of this paper suggest that if this goal is achieved using the double sucrose gap voltage clamp, it will be by a detailed quantitative accounting for substantial irreducible errors in voltage control, rather than by experimental achievement of good voltage control.

In this paper we explore the properties of a mathematical model for the passive sodium permeability system of excitable membranes. This model is distinguished by the explicit inclusion of a rate constant which depends not on instantaneous voltage, but on rate of voltage change. Actually, the model is a rather modest modification of the Hodgkin-Huxley model, but displays some behaviors which the H-H model does not. Among these behaviors are a pronounced inactivation shift (for certain parameter values), a difference between inactivation time constant as measured by turning off a sodium current under sustained depolarization and as measured by double pulse experiments, skip runs under sustained current stimulation, and accommodation to slowing rising currents.
