19. REACTIVITY OF RADIOIODINATED SERUM ANTIBODY FROM BURKITT'S LYMPHOMA AND NASOPHARYNGEAL CARCINOMA PATIENTS AGAINST CULTURE LINES DERIVED FROM BURKITT'S LYMPHOMA.

Abstract

The IgG serum immunoglobulin fraction of two Burkitt's lymphoma (Mutua and Kiliopa) and one African nasopharyngeal carcinoma patient (Kipkoech) was conjugated to iodine-131 (131I) It is known from previous studies with fluorescein labelled conjugates that all three sera contain antibody against the Epstein-Barr virus (EBV)-associated membrane antigen complex, present on the surface, of lymphoblastoid cells in EBV-carrier cultures. All three radioiodinated conjugates attached to live cells of an EBV-carrying Burkitt line (Maku), but not to EBV-free Raji cells. A Swedish control serum (Berith) did not block the binding of any of the three conjugates, whereas unconjugated sera of Mutua, Kiliopa and Kipkoech showed various degrees of blocking and cross-blocking. The blocking patterns were in good agreement with previous tests, performed with the same sera against their fluorescein conjugated derivatives. Antibody release tests, involving preincubation of live cells with one of the three conjugates, followed by incubation with unlabelled serum revealed a certain 'hierarchy' between the three sera with regard to their ability to displace radioiodinated surface-coupled immunoglobulin. This ability could be related to the competitive behaviour of the same sera in the cross blocking tests. The results are believed to reflect differences in the affinity of the three antibodies, due either to differences in fit in relation to the surface antigen(s) carried by the Maku target cell, or to differences in the duration of immunization in the three patients. Abstract

A statistical observation was made on 861 cases of urolithiasis experienced during the past thirty-two years, 1928 to 1959. 92 of them showed multilocation of stones in the urinary tract. They consisted of 221 cases (25.6%) of renal calculus, 291 (33.7%) of ureteral, 252 (29.2%) of vesical, 54 (6.2%) of urethral and 43 (4.9%) of prostatic. In the early period of above 32 years, calculus in the lo w er urinary tract showed higher incidence than that of the upper, but this ratio lately reversed with greatly increasing incidence of upper urinary tract calculus. Age distribution of calculus was 223 cases (25.9%) in the third decade, 182 (21.1%) in the fourth decade. In young adults, there were many cases of calculus of the upper urinary tract ; on the other hand, there were many cases of the lower urinary tract calculus in the aged and children. 729 cas e s (84.6%) were male and 132 cases (15.3%) were female. The ratio of male to female on sites of stones was 3.2 to 1 in renal calculus, 4.2 to 1 in ureteral, 9.1 to 1 in vesical and 54 to 0 in urethral. Affected side was a little more on the left than the right. Ureteral stones were found more in the lower ureter than in the upper or mid-ureter. Male to female ratio of upper urinary calculus was 3.7 to 1, highest incidence of which was seen in the trird decade in both male and female. V e sical calculus showed highest incidence in the seventh decade and next in the fifth decade. It was interesting that 30 cases (11.8%) of vesical calculus were found in one to ten years of age. Urethral c alculus occupied 6.8% of all urinary calculus and was most frequent in one to sixten years of age, but there was no case of female. Prostatic calculus was mostly seen in man over 40 and occupied 5.09% of all urinary calculus. As far as the treatment of urolithiasis is concerned, in the early periods, those who received treatment for urinary calculus were only 44.8% of all the urolithiatic patients occupying 2.4% of outpatients. Recently, however, cases of urolithiasis have progressively increased and those who received treatment became more than 50%. Treatment of urolithiasis was, as a whole, surgical. When t h ere was a stone only in the upper urinary tract operative treatment was common, but it should be noted that cases of spontaneous discharge or chemical dissolution of stones have been recently increasing. In cases of vesical or urethral calculus, endoscopic treatment was widely used ; and in cases with prostatic calculus, treatment was all operative. Chemical composition of calculus showe d much phosphate in upper urinary and urate in lower urinary calculus.

摂関政治の時代にかわって、十一世紀末に出現する院政は、軍事力編成の上からいっても中世の状況をあらわしている。このことを明らかにするために院政政権の軍事力に焦点をあててみた。軍防令に象徴されるような公民兵は、すでに八世紀において徴兵不可能となり、健児という選抜兵に軍事力を頼らざるを得なくなった。しかし、この健児もほとんど軍事的な意味をになうことなく矮少化し、ついには国家は国家の手によって軍事力の基本から組織するということをやめ、既製の軍事力を臨時的に利用するところとなる。十世紀より以後の、とくに将門の乱以後の軍事力はこうしたものであり、院政政権についても一定の官職を媒介として民間に在る武力をとりこんで自己の軍事力に編成する。その典型は、のちに院政をも打倒するところとなった平清盛のような武家棟梁である。この棟梁出現の必然性をも考え、院政政権を支えた軍事力の編成と構成について分析を加えた。, The Insei Government which had overcome the Sekkan-Regime 摂関政治 in the end of the eleventh century had shown the medieval characteristics from the viewpoint of the military organization. I will focus on the military forces under the Insei Government in order to testify it. In the eighth century, the Goverment Army had already been compelled to recruit from the elite soldiers called Kondei 健児 instead of public soldiers recruited by the Edict of Military Defense 軍防令. This system, however, collapsed without having any military significance and at last the Government, giving up organizing the army by itself, had come to use the ready-made forces temporarily. This case was predominant in the tenth century, especially after the Revolt of Masakado 将門の乱. In the Insei Period too, the Govenment organized its own army by admitting the private forces through the medium of offering public offices. This pattern was typically seen in the chief of the soldiers 武家棟梁 such as Kiyomori Taira 平清盛 who would defeat even the Insei Government. Abstract

SummaryAbsorption, tissue distribution and urinary and biliary excretion of flurbiprofen were compared after rectal and oral administration of a 14C-labelled compound to Wistar rats. Flurbiprofen was readily absorbed by both routes, plasma levels of radioactivity reaching a maximum in 20 to 30 minutes and then decreasing rapidly. Flurbiprofen was found to have a wide tissue distribution, but there was no evidence of accumulation in any particular tissue. Approximately 52 of the dose administered by either route was excreted in the urine within 96 hours. The proportion excreted in the bile within 24 hours was 45.6 after rectal administration and 38.3 after oral administration.The anti-inflammatory and analgesic effects offlurbiprofen after administration by the two routes were studied in a number of animal experiments. Flurbiprofen was shown to be more potent in the tests after rectal administration.

30. Cell agglutination mediated by concanavalin a and the dynamic state of the cell surface

Abstract

The binding of 131I-labelled concanavalin A (131I-Con A) to the cell surface has been studied in Ehrlich ascites tumour cells (EATC) and beef erythrocytes under various conditions. The binding of concanavalin A (Con A) to the cell surface was very specific and the available binding sites were saturated within a few minutes. The amount of 131I-Con A bound to EATC was 4·14 × 107 molecules/cell at 37 °C and 2·12 × 107 molecules/cell at o °C. Under these conditions, cell agglutination was observed only at 37 °C and not at o °C. However, the binding sites measured at o °C were also effective for agglutination at 37 °C. Beef erythrocytes were agglutinated by Con A only after treatment of cells with papain. The number of binding sites for Con A on the cell surface was decreased by this treatment to about half the number present on untreated cells. Various reagents such as colchicine, monoiodoacetic acid, dinitrophenol, rotenone, sodium azide and carboxyl cyanide-m-fluorophenylhydrazone (FCCP) had no effect on Con A-mediated cell agglutination. In contrast, periodate treatment produced a remarkable decrease in the agglutinability of cells. From these data, it is concluded that the cell agglutination induced by Con A was due to the topographical distribution of the surface receptors for the lectin, and not the result of energy-dependent or microtubule-dependent reaction processes. The number and the state of Con A receptors on the cell surface were in a dynamic condition, their conformation, orientation, and/or topographical distribution changing under different conditions.

Abstract

A statistical observation was made on 861 cases of urolithiasis experienced during the past thirty-two years, 1928 to 1959. 92 of them showed multilocation of stones in the urinary tract. They consisted of 221 cases (25.6%) of renal calculus, 291 (33.7%) of ureteral, 252 (29.2%) of vesical, 54 (6.2%) of urethral and 43 (4.9%) of prostatic. In the early period of above 32 years, calculus in the lo w er urinary tract showed higher incidence than that of the upper, but this ratio lately reversed with greatly increasing incidence of upper urinary tract calculus. Age distribution of calculus was 223 cases (25.9%) in the third decade, 182 (21.1%) in the fourth decade. In young adults, there were many cases of calculus of the upper urinary tract ; on the other hand, there were many cases of the lower urinary tract calculus in the aged and children. 729 cas e s (84.6%) were male and 132 cases (15.3%) were female. The ratio of male to female on sites of stones was 3.2 to 1 in renal calculus, 4.2 to 1 in ureteral, 9.1 to 1 in vesical and 54 to 0 in urethral.
