In an ecological study of the growth of two grasses, blue grama (Bouteloua gracilis) and side-oats grama (Bouteloua curtipendula), it was necessary to make a determination of the carbohydrate reserves. Both a qualitative identification and a quantitative estimation of sugars were needed to find growth and storage trends. Paper chromatography has been utilized in many instances for both qualitative and quantitative analysis of sugars (2, 9) with sufficient reliability and accuracy to be applicable to this study. Because a very large number of procedures are given in the chemical literature, with varying degrees of success reported in separation and identification, several were investigated and the methods which gave results that were reproducible and provided adequate separation were selected for the study and are described in the experimental discussion.
