The validated hydride generation atomic absorption spectrometry (HG-AAS) method has been used to investigate total arsenic in groundwater. Under optimal experimental conditions, the concentration of arsenic in groundwater can be analysed in the range of 0.5 to 50 µg/L, with a method detection limit of 0.15 µg/L. Its recovery in the field is from 96.3 to 99.8%, with high repeatability. The method was used to observe the total arsenic pollution in groundwater collected in Phu Tho Province, Vietnam. A total of 364 groundwater samples were analysed. The results showed that arsenic pollution was significant, with 15.93% of the samples higher than the maximum permissible level of arsenic. About 20.69% of the contaminated samples had a total arsenic ten times higher (100 µg/L) than the maximum permissible level of arsenic. The pollution source was also considered by comparing the arsenic level in the groundwater with arsenic in the surface water in the same areas. Thus, the use of the high-accuracy and sensitive method, HG-AAS, supplies valuable data on groundwater pollution for water resources management and environmental protection. [ABSTRACT FROM AUTHOR]

Mycotoxins produced by Alternaria spp. act genotoxic in cell-based studies, but data on their toxicity in vivo is scarce and urgently required for risk assessment. Thus, male Sprague-Dawley rats received single doses of a complex Alternaria toxin extract (CE; 50 mg/kg bw), altertoxin II (ATX-II; 0.21 mg/kg bw) or vehicle by gavage, one of the most genotoxic metabolites in vitro and were sacrificed after 3 or 24 h, respectively. Using SDS-PAGE/Western Blot, a significant increase of histone 2a.X phosphorylation and depletion of the native protein was observed for rats that were exposed to ATX-II for 24 h. Applying RT-PCR array technology we identified genes of interest for qRT-PCR testing, which in turn confirmed an induction of Rnf8 transcription in the colon of rats treated with ATX-II for 3 h and CE for 24 h. A decrease of Cdkn1a transcription was observed in rats exposed to ATX-II for 24 h, possibly indicating tissue repair after chemical injury. In contrast to the observed response in the colon, no markers for genotoxicity were induced in the liver of treated animals. We hereby provide the first report of ATX-II as a genotoxicant in vivo. Deviating results for similar concentrations of ATX-II in a natural Alternaria toxin mixture argue for substantial mixture effects.

Australia and New Zealand have recently adopted a joint standard for the regulation of contaminants and natural toxicants in foods. The standard was developed as part of a general review of food standards in Australia and New Zealand and forms part of the new joint Food Standards Code recently adopted by both Governments. Maximum levels (MLs) for contaminants and natural toxicants were established in the new joint Standard only where they serve an effective risk management function, based on a scientific risk assessment, and only for those foods that provide a significant contribution to the total dietary exposure. To assist both enforcement agencies and industry to maintain contaminant levels at the lowest achievable levels, generally expected levels (GELs), have been established to complement the use of MLs. GELs, while not legally enforceable, provide a benchmark against which to measure contaminant levels in foods. [Copyright &y& Elsevier]
