Environmental changes in the Brazilian Pantanal and Cerrado facilitate the spread of parasitic diseases in wildlife, with significant implications for public health owing to their zoonotic potential. This study aimed to examine the occurrence and diversity of gastrointestinal parasites in wild felids within these regions to assess their ecological and health impacts. We collected and analyzed helminth-positive samples from 27 wild felids using specific taxonomic keys. Diverse parasitic taxa were detected, including zoonotic helminths, such as Ancylostoma braziliense , Ancylostoma caninum , Ancylostoma pluridentatum , Toxocara cati , Toxocara canis , Dipylidium caninum , Taenia spp., Echinococcus spp., and Spirometra spp. Other nematodes, such as Physaloptera praeputialis and Physaloptera anomala, were identified, along with acanthocephalans from the genus Oncicola and a trematode, Neodiplostomum spp. (potentially the first record of this parasite in wild felids in the Americas). Human encroachment into natural habitats has profound effects on wild populations, influencing parasitic infection rates and patterns. This study underscores the importance of continuous monitoring and research on parasitic infections as a means of safeguarding both wildlife and human populations and highlights the role of wild felids as bioindicators of environmental health.

Oestrus ovis larvae are obligate parasites of vertebrates and cause cavitary myiasis (oestrosis) in sheep and goats. It is also reported as a zoonosis causing ophthalmomyiasis and nasopharyngeal myiasis. Despite being relatively common in Brazil, epidemiological studies on O. ovis are scarce. Considering that the infestation is influenced by the climate and biomes of the studied region, we aimed to investigate the seasonal prevalence of O. ovis among slaughtered sheep in the northern region of the state of Mato Grosso, Brazil. The heads of sheep (n=697) slaughtered at a slaughterhouse in the municipality of Terra Nova do Norte (November 2011 to November 2013) were collected to count, catalog, and identify the larvae found in the upper respiratory tract. Overall, 45.77% (319/697) of the animals were infested with 2,412 recovered larvae, 96.89% (2,337/2,412) of which were identified at the species level as O. ovis. Seasonal variations in prevalence ranged from 41% (spring) to 56% (summer); however, no correlation was observed between prevalence and season, mean humidity, or temperature. In conclusion, parasitism by O. ovis in sheep in the studied area, occurs year-round, considering the occurrence of larvae (L1, L2, and L3) throughout the year, probably because of the area's environmental conditions.

Although Bartonella spp. have been worldwide described in rodents and bats, few studies have reported these agents in marsupials. The present work aimed to investigate the occurrence and genetic diversity of Bartonella in small mammals (rodents, marsupials, and bats) and associated ectoparasites in two ecoregions (Amazonia and Cerrado biomes) in midwestern Brazil. For this purpose, DNA samples from 378 specimens of small mammals (128 rodents, 111 marsupials, and 139 bats) and 41 fleas (Siphonaptera) were screened for the Bartonella genus employing a quantitative real-time PCR assay (qPCR) based on the nuoG (nicotinamide adenine dinucleotide dehydrogenase gamma subunit) gene. Then, positive samples in qPCR were submitted to conventional PCR (cPCR) assays targeting the gltA, ftsZ, and rpoB genes. One (0.78 %) rodent, 23 (16.54 %) bats, and 3 (7.31 %) fleas showed positive results in the qPCR for Bartonella sp. After cPCR amplification and sequencing, 13 partial Bartonella DNA sequences of the following genes were obtained only from bats´ blood samples: 9 gltA (citrate synthase), 3 ftsZ (cell division protein), and 1 rpoB (RNA polymerase beta subunit). The maximum likelihood inference based on the gltA gene positioned the obtained sequences in three different clades, closely related to Bartonella genotypes previously detected in other bat species and bat flies sampled in Brazil and other countries from Latin America. Similarly, the ftsZ sequences clustered in two different clades with sequences described in bats from Brazil, other countries from Latin America, and Georgia (eastern Europe). Finally, the Bartonella rpoB from a specimen of Lophostoma silvicolum clustered with a Bartonella sp. sequence obtained from a Noctilio albiventris (KP715475) from French Guiana. Marmosa constantiae is a species of marsupial restricted to the central portion of South America. In Brazil, it occurs in the northwestern region including five states of the Amazon, Cerrado, and Pantanal biomes. However, there is no study of the helminth fauna or helminth community structure for this marsupial. The aims of this study were to describe the species composition and to analyse the structure of the helminthic community of M. constantiae in an area of the Amazon Arc in Sinop, north of the state of Mato Grosso, Brazil. Parasites were searched in 53 specimens of this marsupial, among which 44 were infected with at least one helminth species. Parasitic helminths were counted and identified. Nine species were collected: seven nematodes, one cestode, and one acanthocephalan. The most abundant species were Gracilioxyuris agilisis , Travassostrongylus scheibelorum, Pterygodermatites sinopiensis , and Subulura eliseae. These species were the only dominant ones in the component community. No significant differences were observed in the abundance and prevalence of helminths between male and female hosts. Host body size significantly influenced helminth abundance in males. The pattern of community structure considering the infracommunities in this locality indicated more species replacement than species loss along the environmental gradient. This is the first study to report the helminth fauna and the helminth community structure of M. constantiae.

Increased interaction between wild and urban environments owing to human population growth, increased anthropization of biomes, and habitat loss for wild animals increases the spread of infectious and parasitic agents. The present study reports on the occurrence of gastrointestinal parasites in carnivorous mammals at two conservation institutions in the state of Goiás, Brazil. Fecal samples from 39 adult carnivores were collected after spontaneous defecation and analyzed by flotation and sedimentation. The structure and management data of each institution were recorded. Parasitism prevalence, binomial confidence intervals (CI) at 95%, variables associated with the presence of contact animals, size of the enclosure and type of food were recorded. The overall prevalence of gastrointestinal parasites in the samples analyzed was 71.8% (CI 55.1-83.0; 28/39). Ancylostomatidae, Toxocara spp., Toxascaris leonina, Strongyloides spp., Calodium hepaticum, and Trematoda eggs, and Cystoisospora spp. oocysts were detected. Environmental conditions were not correlated with parasitism prevalence; however, the parasites found could be managed, considering their biology, such as controlling synanthropic and domestic animals in captivity, feeding with healthy feed.

Immature hard ticks from the genus Amblyomma feed on blood from a wide range of Neotropical avian hosts. They serve as vectors for pathogens of medical and veterinary importance, such as Rickettsia agents of the spotted fever group (SFG). Hence, determining ecological factors that increase encounter rates between immature ticks and their avian hosts may contribute to the understanding of tick-borne diseases transmission. Here, we used 720 individual birds from 96 species surveyed in the Brazilian Pantanal to test whether host breeding season influenced tick infestation probabilities. Additionally, collected ticks were screened for Rickettsia agents to describe new avian-tick-bacteria associations. Our models revealed that the probability of an individual bird being infested with immature ticks was similar during the breeding and pre-breeding season, but higher loads of immature tick stages were found during the breeding season. Host sex did not predict infestation probability, but Rickettsia agents recovered from ticks were more prevalent during the pre-breeding season. The new records of host usage by larvae and nymphs of Amblyomma in Pantanal and the growing body of tick surveys in Neotropical avian communities, suggest that immature ticks may benefit from avian blood sources during their annual cycle. The low number of infected ticks with Rickettsia agents on Pantanal birds suggest that this vertebrate group are likely not acting as reservoirs for these microorganisms. However, long-term surveys at the same site are imperative to determine which tick species are acting as reservoirs for Rickettsia agents in Pantanal and determine whether birds are playing a role in dispersing ticks and tick-borne pathogens., (Copyright © 2023. Gastrointestinal parasitism by helminths and protozoa poses risks to animal and human health owing to clinical changes and transmission of potentially zoonotic agents. Thus, the present study aimed to verify the occurrence of gastrointestinal parasites in dogs from the municipality of Cuiabá, Mato Grosso, Brazil, using coproparasitological tests. From June 2021 to April 2022, faecal samples were collected from domestic dogs at the Veterinary Hospital and Animal Protection Shelters in the Cuiabá municipality. A semi-structured questionnaire was applied to the owners and those responsible for the shelters to analyse the factors associated with gastrointestinal parasitism. A total of 353 faecal samples were collected and subjected to parasitological flotation and sedimentation techniques. Data were analysed using the chi-squared test and exploratory factorial analysis. The occurrence of gastrointestinal parasitism was 22.66% and the parasites found alone or in mixed infections were Ancylostoma spp., Trichuris vulpis, Toxocara spp., Dipylidium caninum, Cystoisospora spp., Giardia duodenalis, and coccidia. It was concluded that the occurrence of gastrointestinal parasites in dogs is frequent, and the variables associated with these infections were source origin, breed, age, coexistence with other animals, and dull fur.

A new species of nematode, Pterygodermatites ( Paucipectines ) sinopiensis n. sp. is described based on specimens recovered from the intestine of the white-bellied woolly mice opossum, Marmosa constantiae , trapped in the municipality of Sinop, Mato Grosso state, Brazil. The genus Pterygodermatites has 21 species described in mammals worldwide, and to date, only two species have been described for marsupials in Brazil. The new species is characterized by the presence of 23 small denticles and by the presence of 38-40 and 65 pairs of the cuticular processes in male and female species, respectively. Additionally, male species possess three ventral precloacal fans, and in female species, the cuticular processes are divided into 41 pairs of comb-like and 24 pairs of spine-like processes; the vulva opens approximately in pair 41. Ctenocephalides felis is the most common hematophagous ectoparasite of domestic cats and dogs worldwide, causing allergic dermatitis, anemia, itching, discomfort, and it is responsible for the transmission of many pathogens. In cattle, parasitism is not common but occurs due to the high adaptability of this flea species. This study aimed to report infestation of a Nellore (Bos indicus) calf from Northern Brazil by C. felis and its clinical findings. A 45 kg male calf presented diarrhea, progressive emaciation after 50 days of birth, and flea infestation. Anemia was observed (4.11 × 10 6 /μL of erythrocytes; 20.48% of hematocrit). Coproparasitological examination and molecularly detection of Anaplasma spp. and Babesia spp. indicated negatively. Fleas were collected and taxonomically identified as C. felis. Clinical manifestations of this ectoparasite in cattle were unspecific; however, they can develop severe conditions, such as restlessness and weakness, accompanied by alopecia, emaciation, anemia, and edema of the lower limbs with intense itching and self-excoriation., (Copyright © 2022 Elsevier B.V. Although the common opossum, Didelphis marsupialis (Didelphimorphia: Didelphidae) is a species widely distributed in South America, knowledge about their helminth parasites and helminth community structure is scarce. The aims of this study were to describe the species composition and analyze the structure of the helminth community of the common opossum in an area of the Amazonian Arc in northern Mato Grosso. The helminths were recovered, counted, and identified in 32 individuals. Overall, 10,198 specimens were categorized into 9 helminths taxa (seven nematodes, one cestode, and one acanthocephalan). The most abundant species were Aspidodera raillieti, Viannaia hamata, and Travassostrongylus orloffi. No statistically significant differences in helminth abundance and prevalence were observed between host sexes. However, young hosts had higher abundance and prevalence of Didelphonema longispiculata, whereas Oligacanthorhynchus microcephalus had higher abundance and prevalence in adult hosts. This was the first study to analyze the helminth fauna and helminth community structure of D. marsupialis in the Amazonian Arc. This is the first report of the presence of A. raillieti, D. longispiculata, T. orloffi, T. minuta, V. hamata, and O. microcephalus in the state of Mato Grosso, Brazil.

Background: Toxoplasma gondii and Neospora caninum infections in primates are potentially fatal and directly impact the conservation of these animals and public health., Materials and Methods: A total of 38 blood/clot samples collected from free-living and captive neotropical primates undergoing clinical care or found dead by environmental authorities in the Mato Grosso State, Brazil, were analyzed by PCR for DNA detection of T. gondii and N. caninum. Furthermore, eight animals were submitted to immunohistochemistry for the detection of T. gondii., Results: DNA of T. gondii and N. caninum was amplified in 11 (28.95%) 10 (26.32%) of samples analyzed, respectively. Coinfection was observed in three individuals. One animal returned a positive result in the immunohistochemistry for the detection of T. gondii., Conclusion: These findings reflect a concern for the conservation of these animals, as the pathogen-host interaction is unpredictable and infections by these protozoa can lead to animal mortality, which has a substantial impact on endangered species., (© 2022 John Wiley & Sons A/S. To a better insight into the epidemiology and genetic diversity of protozoan hemoparasites infections in wild mammals, this study aimed to the post mortem detection of DNA from species of the order Piroplasmida (Babesia sp., Cytauxzoon sp., and Theileria sp.) and suborder Adelorina (Hepatozoon sp.) using polymerase chain reaction based on the 18S rRNA gene followed by genetic sequencing of blood and spleen samples collected from carcasses of 164 free-ranging and captive wild mammals from Mato Grosso state. Among them, one Leopardus pardalis, three Panthera onca, two Puma concolor were positive for Cytauxzoon sp., and six Tapirus terrestris tested positive for Piroplasmida, while one L. pardalis was positive for Hepatozoon sp. Furthermore, an uncharacterized piroplasmid genetically related to Theileria sp. previously detected in cats from Brazil was described in lowland tapirs. Despite the controversy regarding the epidemiological threat of these protozoa, the detection of these tick-borne agents in wild free-living and captive mammals, even when asymptomatic, demonstrates the importance of monitoring, particularly in hotspots such as the state of Mato Grosso, to verify the circulation and genetic diversity, to anticipate the possible emergence of diseases, and even their consequences to other animals as well as humans.

Toxoplasma gondii is a widespread protozoan parasite that causes infections in animals and humans. The presence of tissue cysts in cattle musculature evidences their epidemiological significance in transmission to humans. The present study aimed to evaluate the seroprevalence of and risk factors of T. gondii seropositivity in beef cattle in the municipality of Unaí, Minas Gerais state, Brazil. Bovine blood samples (n = 612) were collected from 31 herds. Risk factors associated with the animal-level and herd-level seroprevalence were analysed using a questionnaire that investigates the epidemiological aspects of toxoplasmosis. A search for anti-T. gondii antibodies was performed by means of an indirect fluorescent antibody test (IFAT). Analysis of 612 samples showed 369 animals positive for anti-T. gondii antibodies (IFAT ≥64), with a seroprevalence of 60.29% (95% confidence interval (CI): 56.35-64.10). The herd-level seroprevalence was 96.77% (95% CI: 79.00-99.58). Applying a multivariate logistic regression analysis, a test of the association between seroprevalence of T. gondii infection and the potential predictors showed that cats (domestic or wild) with access to the drinking water of cattle were a significant (P = 0.001) predictor. The seroprevalence of T. gondii in cattle destined for human consumption is widely distributed throughout the entire study area. This study aimed to evaluate the occurrence of endoparasites in fecal samples of capybaras from anthropized areas in seven cities in the state of São Paulo, southeastern Brazil, and natural areas in two locations in the Pantanal biome. For the parasitological evaluation of samples, feces were subjected to centrifugal sedimentation in water-ether and centrifugal flotation in sucrose solution. Out of the 113 samples examined, 97.3% (110/113) were positive for the presence of helminth eggs and/or protozoan oocysts, with 96.7% (89/92) and 100% (21/21) in capybaras from anthropized and natural areas, respectively. Helminth eggs belonging to the Class Cestoda (Monoecocestus spp.), Class Digenea, and Class Nematoda (Trichostrongyloidea, Strongyloides chapini, Protozoophaga obesa, and Capillaria hydrochoeri) were detected. Coccidia oocysts of Eimeria spp. and Cryptosporidium spp. were detected. The difference found was in the occurrence of S. chapini between anthropized and natural areas, as S. chapini was not detected in natural areas. Capybaras from anthropized areas showed a greater species richness of endoparasites, including species such as Fasciola hepatica and Cryptosporidium spp., that might have zoonotic potential.

Trypanosomatids are uniflagellate protozoa belonging to the Trypanosomatidae family. The genera Trypanosoma and Leishmania are of paramount importance as they contain species that cause serious diseases, such as Chagas disease and Leishmaniasis, respectively. The objective of the present study was to identify trypanosomatids present in the whole blood of free-living and captive neotropical primates in Mato Grosso State, Midwest Brazil. Between 2017 and 2019, 38 blood samples were collected from seven different neotropical primate species in seven cities in the state. Through molecular techniques, including polymerase chain reaction (PCR) to amplify a fragment of the kinetoplast DNA (kDNA) and 18S ribosomal RNA (18S rRNA) gene, sequencing, and phylogenetic analysis, nine Leishmania spp. [seven L. infantum and two L. (Leishmania) amazonensis] and two Trypanosoma spp. (T. minasense and T. rangeli) were identified. This study contributes to understanding the occurrence and epidemiology of trypanosomatids in Mato Grosso State and the importance of neotropical primates as trypanosome hosts and possible infection sources for other animals and humans. Future identification of other blood pathogens in neotropical primates will assist in disease control and prevention strategies.

Platynosomum fastosum, which is distributed in tropical and subtropical regions, is the main parasite in the biliary system of domestic cats. Cats can be asymptomatic or show severe signs of cholangitis. Although the infection is reported throughout Brazil, only post-mortem studies are available from the Midwest region of the country. This cross-sectional study investigated the frequency of P. fastosum parasitism in domestic cats treated at the University Veterinary Hospital in Cuiabá city, Mato Grosso, Brazil, by three coproparasitological techniques (formalin-ether sedimentation, Faust, and Hoffmann), associating clinical, laboratory (haematological, biochemical) and ultrasonographic alterations. Based on the sample calculation, 171 cats were evaluated in the clinical and laboratory and a semi-structured epidemiological questionnaire was designed and used by the tutors. The prevalence observed was 26.90%, with no significant association with sex, reproductive status, age, hunting habit, access to the street, recent anthelmintic treatment, and the guardian's lack of knowledge regarding parasitism. Of the 171 cats evaluated, 55 (32.16%) showed clinical signs suggestive of cholangitis, with a significant association of this variable. Regarding the clinical and pathological variables, mild neutrophilia and elevated serum activity of alanine amino transferase and alkaline phosphatase was observed (but not significantly), while the mean values of eosinophils, leucocytes, total protein, albumin, and globulin were normal. The ultrasound findings were like those previously described. Thus, it was observed that P. fastosum occurs in moderate prevalence in cats from Cuiabá-MT, with a significant association with clinical signs and a regular agreement between the parasitological techniques employed., (Copyright © 2021 Elsevier B.V. Neospora caninum is considered to be one of the main causes of abortion among cattle. The present survey was conducted in the municipality of Rolim de Moura, Rondônia State, Brazil. A questionnaire that investigates the epidemiological aspects of neosporosis was used in the analysis of risk factors associated with the animal-level and herd-level prevalence in dairy cattle. A total of 416 bovine blood samples were collected from 30 farms, and N. caninum antibody levels were measured by Indirect Fluorescent Antibody Test (IFAT). Analysis of dairy cattle serum samples revealed the presence of anti-N. caninum antibodies to be 47.36% (n = 197). Risk factors associated with N. caninum infection were the management system and access locations of dogs. The results of the present survey indicated that infection of dairy cattle with N. caninum is widespread in the studied region of Western Amazon, which has implications for prevention and control of neosporosis in this region. Therefore, integrated control strategies and measures are recommended to prevent and control N. caninum infection in dairy cattle. In addition, direct contact between dairy cattle, dogs and wild animals, which can influence the epidemiology of neosporosis, should be investigated further.

The aim of this study was to carry out a seroepidemiological study of anti-T. gondii and anti-N. caninum antibodies in serum samples from sheep intended for human consumption in the Rondônia state, in the Western Brazilian Amazon, and identify possible risk factors associated with seropositivity. The analysis of the 616 sheep serum samples revealed animal-level seroprevalence of T. gondii in the order of 52.4 % and of N. caninum of 60.6 %. Prevalence of co-infections was 33.4 %. The herd-level seroprevalence was 100 % for both the parasites. Applying of multivariate logistic regression analysis, test of the association between seroprevalence of T. gondii infection and the potential predictors showed that feed, fetal malformation, abortion, neurological problems (lambs), presence of rodents were the significant (P < 0.05) predictors. The variables presence of rodents, cats, wild animals and main activity showed statistical association (P < 0.05) with the seroprevalence of N. caninum. Due to the high prevalence of T. gondii and N. caninum found in this study, the absence of specific slaughterhouses for sheep and raw or undercooked sheep meat for human consumption we concluded that sheep can represent an important source of infection for humans., (Copyright © 2020 Elsevier Ltd. Cutaneous lesions lead to complications in patients, since they may be recurrent and also represent risk of progression to infection and/or amputation. Therefore, effective, protective, and topical treatments of easy application and removal need to be developed to provide effective alternatives to patients. The Caryocar brasiliense Cambess (CBC) presents important pharmacological activities and proved in the healing process. This paper reports the improvement of the CBC nanostructured (LNC CBC and LNC CBC+ ) activity in dermal wounds in vivo. The oil was physico-chemically characterized and used in the development of lipid-core nanocapsules (LNCs), coated (LNC CBC+ ) or without chitosan (LNC CBC ), in concentration of 1.0 mg mL -1 . Hydrogel (HG) was tested in vivo on lesions in the back of male Wistar rats for 14 days. The oil presented appropriate physico-chemical characteristics for its use, such as moisture 0.76 %, acidity 0.85 % and oleic acid 25.90 %. The LNCs showed nanometric size (around 200 nm), monomodal distribution, slight acid pH and zeta potential of + 22.1 mV in accordance with the composition. The nanostructured oil induced dermal healing in vivo showing significantly better improvement than free oil. LNC CBC+ showed best results showing the higher increase of the production of type 1 collagen, an important protein to the healing repair. Purpose: To evaluate whether silicone foam implants have a different evolution pattern compared to conventional texture implants., Methods: Fifty-eight female patients underwent surgery. They were divided into two groups (silicone foam - Lifesil® - and microtexturized silicone - Lifesil®). The evolution was analyzed in postoperative consultations, with physical examination, photographic documentation and filling in a satisfaction questionnaire, in the postoperative period of one month, four months, one year and then annually, up to a maximum of 3 years of follow-up., Results: There were no statistically significant differences in presence of rippling, stretch marks, breast ptosis, capsular contracture and quality of scars. There was a higher rate of patients who were very satisfied with the outcome 360 days after surgery in the group receiving silicone foam implants (p = 0.036)., Conclusion: In short time, silicone foam envelope implants proved to be as reliable as textured silicone envelope implants, making them an option for augmentation mammoplasty.

Small non-volant mammals (marsupials and small rodents) were captured at three different timepoints from 23 forest fragments across three municipalities (Alta Floresta, Sinop and Cláudia) covering the Amazonian biome of the Mato Grosso State in Midwestern Brazil. The animal tissues (liver and spleen) and blood were screened using molecular tools for the detection of Babesia, Coxiella, Cytauxzoon, Hepatozoon, Theileria, and Anaplasmataceae agents. A total of 230 specimens (78 rodents and 152 marsupials) were trapped. Hepatozoon and Piroplasmorida agents were detected in the common opossums (Didelphis marsupialis). In turn, all samples (blood, liver, or spleen) collected from the small mammals were negative for the genus Coxiella and the family Anaplasmataceae, as detected by polymerase chain reaction (PCR). Phylogenetic analyses inferred from partial sequences of the 18S rRNA gene highlighted the occurrence of new Hepatozoon and Piroplasmorida haplotypes. Future studies determining the role of common opossum (D. marsupialis) in the epidemiological cycles of Hepatozoon and Babesia under natural conditions in the Amazonian biome are necessary.

Background: Fine-tuning the aeration for cultivations when oxygen-limited conditions are demanded (such as the production of vaccines, isobutanol, 2-3 butanediol, acetone, and bioethanol) is still a challenge in the area of bioreactor automation and advanced control. In this work, an innovative control strategy based on metabolic fluxes was implemented and evaluated in a case study: micro-aerated ethanol fermentation., Results: The experiments were carried out in fed-batch mode, using commercial Saccharomyces cerevisiae, defined medium, and glucose as carbon source. Simulations of a genome-scale metabolic model for Saccharomyces cerevisiae were used to identify the range of oxygen and substrate fluxes that would maximize ethanol fluxes. Oxygen supply and feed flow rate were manipulated to control oxygen and substrate fluxes, as well as the respiratory quotient (RQ). The performance of the controlled cultivation was compared to two other fermentation strategies: a conventional "Brazilian fuel-ethanol plant" fermentation and a strictly anaerobic fermentation (with ultra-pure nitrogen used as the inlet gas). The cultivation carried out under the proposed control strategy showed the best average volumetric ethanol productivity (7.0 g L -1  h -1 ), with a final ethanol concentration of 87 g L -1 and yield of 0.46 g ethanol  g substrate -1 . The other fermentation strategies showed lower yields (close to 0.40 g ethanol  g substrate -1 ) and ethanol productivity around 4.0 g L -1  h -1 ., Conclusion: The control system based on fluxes was successfully implemented. The proposed approach could also be adapted to control several bioprocesses that require restrict aeration.

Ornithodoros fonsecai is an argasid tick that is endemic to Brazil and has been described in the municipality of Bonito, state of Mato Grosso do Sul. Some specimens of this species were found in a cave in the municipality of Nobres, state of Mato Grosso. The specific identification of this population was confirmed by means of morphology and molecular biology. The mitochondrial 16S rDNA partial sequence of this species from Nobres has been deposited in GenBank (MK158949). The objective of this study was to elucidate the biology of O. fonsecai from Nobres, and to report autogeny in this tick population. Along three laboratory generations was observed molting of first nymphal instar to the second instar without feeding, a typical behavior of species included in the subgenus Alectorobius. The first generation (F1) presented five nymphal instars (N1 to N5), and most of adults emerged through molting of N5. The last nymphal instar of second generation (F2) was N4, but most of adults emerged from N3. In the third generation (F3) the last nymphal instar was N5, with most of the adults emerging from N4. In F2, some females (n = 20) originated from N3 began laying eggs without a blood meal. It was observed that those N3 fed twice before they molted to autogenic females. However, autogenic behavior occurred in relation to third generation females (F3) with specimens originating from N4 (n = 12) that were fed only once as nymphs. This behavior has already been reported as obligatory for the genera Otobius and Antricola, while it is facultative for one species of genus Argas and for four species of genus Ornithodoros. To estimate the seroprevalence of Toxoplasma gondii and Neospora caninum, using an indirect immunofluorescent assay (IFA), and identify the risk factors associated, serum samples were collected from 1,070 pigs from 320 backyard pig farming in the of Mato Grosso state. The animal-level seroprevalence of T. gondii and N. caninum was 32.48% and 13.49%, respectively, with a herd seroprevalence of 55.63% for T. gondii and 27.81% for N. caninum. Feeding the animals with leftovers increases the probability of the presence of anti-T. gondii antibodies in pigs by 1.09-fold. Unlike to T. gondii, feeding with leftovers was found to be negatively associated with N. caninum seropositivity in farm-level analysis and in the animal-level model, so decreasing the chances of positivity. Yet, age was considered a risk factor for N. caninum seropositivity. Further studies are necessary to evaluate the impact of T. gondii infection on backyard pig farming production, and its importance as a source of toxoplasmosis infection in humans in the Mato Grosso state, as well as, the role of domestic pigs in the epidemiology of neosporosis.

Objective: to make the translation, cross-cultural adaption and content and face validation of the Hypertension Knowledge-Level Scale for use in Brazil., Methods: methodological research carried out in six stages: translation, synthesis, back-translation, expert committee's assessment, pre-test and validation. Validation was performed through the Delphi technique in two rounds. The participants were two translators and two back-translators, eight professionals in the expert committee, 40 adult participants in the pre-test, 35 experts in the first validation round and 28 in the second validation round. Data analysis included Cronbach's alpha, content validity index and one-tailed t-test., Results: the translation and cross-cultural adaptation allowed for language adjustments so that the items were comprehensible and suitable for use in Brazil. The content validity index of the Brazilian version of Hypertension Knowledge-Level Scale was 0.96 and Cronbach's alpha was 0.92., Conclusions: the scale was translated, cross-culturally adapted to Brazilian Portuguese, had its content and face validated and proved reliable to evaluate the knowledge of adults about hypertension.

Neosporosis is caused by an obligate intracellular protozoan, Neospora caninum . It is considered one of the most widespread and frequent causes of abortion in cattle worldwide. To evaluate the prevalence of anti-N. caninum antibodies and associated risk factors, serum samples were collected from 2,452 bovines at 262 farms in the northern Pantanal, state of Mato Grosso, Brazil. Each farmer was asked to fill out a questionnaire for subsequent epidemiological data analysis. Anti-N. caninum antibodies were detected by means of the indirect immunofluorescent assay (IFA), using a cut-off dilution of 1:100. The overall anti-N. caninum antibodies prevalence was 25.44% (Confidence Interval - CI 95%; 20.10%; 30.78%), and the anti-N. caninum antibodies prevalence per herd was 76.72% (CI 95%; 71.60%; 81.84%). The presence of dogs, occurrence of abortion in cows, and sale of cattle for breeding were statistically associated with seropositivity in herds, while the risk of females being seropositive for N. caninum was higher in animals ≤ 6-years-old and in the presence of dogs. A spatial analysis indicated that the relative risk of the disease is spatially constant and that the farms with the highest prevalence of anti-N. caninum antibodies are located south of the region under study.

We evaluated the distribution of piroplasmids in equids from the Mato Grosso state in Midwestern Brazil using molecular methods and the interspecific genetic diversity. For this, 1,624 blood samples of equids from 973 farms were examined by PCR, using primer pairs that amplify a fragment of the genes rap-1 and ema-1 of Babesia caballi and Theileria equi, respectively. For molecular characterization and phylogenetic studies, 13 and 60 sequences of the rap-1 and ema-1 genes, respectively, were used to build a dendogram using maximum parsimony. B. caballi and T. equi were detected in 4.11% and 28.16% of the farms, respectively, and molecular prevalence was 2.74% for B. caballi and 25.91% for T. equi. The location of the farms and animals raised in the Pantanal ecoregion influence the probability of equids testing positive for B. caballi and T. equi . Moreover, age and herd purpose were variables significantly associated with T . equi infection. The sequences of B. caballi presented 1.95% intraspecific variability, contrasting with 2.99% in T. equi. Dendrograms for both species demonstrated the presence of subgroups with high values of support of branches. However, it is not possible to associate these groups with geographic origin and/or ecoregion.

Trypanosoma (Megatrypanum) theileri is a flagellated protozoan that infects ruminants and it displays high genetic diversity. In this study, we investigated the prevalence rates of this protozoan based on hemoculture and molecular diagnosis. The isolates of T. theileri thus obtained were characterized by molecular markers SSU rDNA and gGAPDH and molecular diagnosis based on Cathepsin L-like gene (PCR-TthCATL). The PCR-TthCATL and hemoculture indicated an overall prevalence rate of 8.13%, and the CATL derived sequence named IB was identified for the first time in cattle in the western Amazon region, as well as IF in Brazil. We also describe a possible new PCR-TthCATL derived sequence in cattle, designated IL.

In the context of cellulosic ethanol production, special attention must be given to the raw material, as it affects final product yield. As observed for sugarcane, bagasse variations may derive from several elements, for instance edaphoclimatic factors, seasonality, maturation stage and harvesting techniques. Therefore, in the present work, to investigate the impact of raw material characteristics on process performance, sugarcane bagasse from four harvests from October/2010 to October/2011 was pretreated by steam explosion and had its soluble and insoluble solids contents measured, following enzymatic hydrolysis to assess glucan conversion. As confirmed by ANOVA, glucose concentration was related to the solids content in the reactor, whereas glucan conversion was related to the enzymatic load. Variations in raw material composition were indeed observed to significantly interfere in the final sugar recovery, probably due to the increase in the impurities observed as a result of the type of harvest performed in 2011., (Copyright © 2017 Elsevier Ltd. Platynosomiasis is a common feline hepatic disease caused by Platynosomum fastosum (Trematoda - Dicrocoelidae), which is also known as 'lizard poisoning'. Most reports of feline platynosomiasis show that this disease is sporadic and manifests with uncommon lesions; its pathogenicity is still not well understood. This study aimed to describe liver injuries and enzymatic changes associated with natural P. fastosum infection in 47 stray cats in an endemic area. Overall, 38.3% (18/47) of cats were parasitized, and 2,358 flukes (P. fastosum) were collected (131 - mean intensity of parasitism; 50.2 - mean abundance). The alanine transaminase (ALT) measure was significantly higher in parasitized animals, while alkaline phosphatase (ALP) showed no statistical difference between parasitized and non-parasitized animals. In infected animals, gross pathological lesions and microscopic liver injuries ranged from mild to severe, and were similar to those in previous descriptions of feline platynosomiasis. Nonetheless, the intensity of parasitism was not related to the severity of macroscopic or microscopic hepatic injuries. However, feline platynosomiasis should be considered in the differential diagnosis of feline liver disorders, as well as, in any program of helminth control, even if no clinical abnormalities are present.

Since the approval of cisplatin as an antineoplastic drug, the medical and the scientific communities have been concerned about the side effects of platinum-based drugs, and this has been the dose-limiting factor that leads to reduced treatment efficiency. Another important issue is the intrinsic or acquired resistance of some patients to treatment. Identifying proper biomarkers is crucial in evaluating the efficiency of a treatment, assisting physicians in determining, at early stages, whether or not the patient presents resistance to the drug, minimizing severe side effects, and allowing them to redirect the established course of chemotherapy. A great effort is being made to identify biomarkers that can be used to predict the outcome of the treatment of cancer patients with platinum-based drugs. In this context, the metallomic approach has not yet been used to its full potential. Since the basis of these drugs is platinum, the monitoring of biomarkers containing this metal should be the natural approach to evaluate treatment progress. This review intends to show where the research in this field stands and points out some gaps that can be filled by metallomics.

Essential (Cu, Mn, Se and Zn) and non-essential (Cd and Hg) elements were analyzed in the hepatic tissue of 22 individuals of seven different species of small cetaceans (Feresa attenuata; Orcinus orca; Pontoporia blainvillei; Sotalia guianensis; Stenella frontalis; Steno bredanensis; Tursiops truncatus) accidentally caught in fishing nets or found stranded along the northern coast of the state of Rio de Janeiro, Brazil, between 2001 and 2010. Atlantic spotted dolphin (S. frontalis) showed the highest levels of Cd (20.23μgg(-1), dry weight), while rough-toothed dolphin (S. bredanensis) showed the highest levels of Hg (825.9μgg(-1)dw) and Se (221.9μgg(-1)dw). Killer whale (O. orca) presented the highest levels of Cu (64.80μgg(-1)dw) and Zn (2220μgg(-1)dw), and Guiana dolphin (S. guianensis), the highest level of Mn (13.05μgg(-1)dw). Cu, Hg, Mn and Zn in the hepatic tissue of killer whale (O. orca), Cu, Hg, Mn, Se and Zn in the hepatic tissue of rough-toothed dolphin (S. bredanensis) and Cd and Zn in the hepatic tissue of Guiana dolphin (S. guianensis) were significantly higher when compared to other studies with these species around the world. No significant correlations were observed between element accumulation and sex, sexual maturity and body length. An analysis of the interelemental relationships in the Guiana dolphin specimens showed strong positive correlations between Cd and Se, Cu and Zn, and Hg and Se. Differences were observed in the bioaccumulation of elements between the analyzed species, probably related to each species feeding habit, and differences between different element concentrations in the different dolphin species were probably due to the preference for certain preys and their bioavailability in the environment. Thus, the bioavailability of the analyzed elements in the marine environment should also be taken in consideration. Mullet (Mugil liza) were sampled in five different areas along the Guanabara Bay, southeastern Brazil, classified as non-contaminated, moderately contaminated and contaminated. Morphometric (Fulton condition factor, relative condition factor and weight to length scaling coefficient) and organosomatic (hepatosomatic index) indices of environmental stress were analysed. Fish from the differentially contaminated areas show statistically different Fulton and relative condition factors and hepatosomatic indices, but not the weight to length scaling coefficient. The Kn and the FCF followed the same trend, with fish from São Gonçalo (1.07 ± 0.04 and 0.89 ± 0.03), Itaipu (0.84 ± 0.01 and 0.86 ± 0.01) and the Rodrigo de Freitas Lagoon (1.03 ± 0.01 and 0.87 ± 0.20) showing higher FCFs than fish from Magé (0.96 ± 0.01 and 0.81 ± 0.01). Fish from Itaipu showed significantly higher HSI values than the other sampling sites (1.68 ± 0.07), with fish from Olaria and Ipiranga showing the lowest (1.56 ± 0.12 and 1.60 ± 0.07, respectively).

Total mercury (Hg) was analyzed in muscle tissue of 27 accidentally captured Guiana dolphins (Sotalia guianensis) in order to evaluate Hg contamination levels present in the Amazon Continental Shelf, in Amapá state, North Brazil. The samples showed a mean concentration of 0.4 ± 0.16 μg/g wet weight (ww), ranging from 0.07 to 0.79 μg/g ww. As observed in several other cetacean species, Hg concentrations presented positive correlations to body length, related to the capacity to bioaccumulate this element throughout life. Hg concentrations were not significantly different between males (mean = 0.38 μg/g ww; n = 15) and females (mean = 0.42 μg/g ww; n = 12). Concentrations were low when compared to results of studies carried out with small cetaceans in the Northern Hemisphere, and with some previous studies in the south-eastern region of Brazil. In contrast with high Hg concentrations normally detected in river dolphin samples from Amazon River tributaries, our results suggest that the Amazon coast contains low levels of Hg in bioavailable form.

Fish accumulate several trace elements in muscle, gills and liver, however studies also indicate that metals can be excreted through bile. Since metal contamination leads to modifications in bile composition, biliary excretion offers an alternative way to evaluate the presence of trace-elements. Bile is easier to obtain than other organs and presents a simpler matrix, making it easier for chemical pre-treatment. To verify if bile can be useful as a biomonitoring tool for metal contamination, liver and bile trace element concentrations were determined and correlated. The Artificial Neural Networks statistical technique was used to verify if liver trace-element quantification could be substituted by bile analysis. Results show that significant correlations were obtained between trace elements in bile and liver and the ANN validated the hypothesis that certain trace-elements in bile could be utilized instead of liver trace-elements. Metallothioneins (MT) are a validated biomarker for metal exposure. In fish they are usually determined in the liver, while some studies have observed MT in kidney, muscle and gills. There are, however, no reports on MT in fish bile. This study aimed to verify if these proteins are present in fish bile, if they can be used as biomarkers for metal exposure and to characterize the metal speciation present in the different species. Three independent methods to verify MT presence in fish bile were conducted: 1D/2D gel electrophoresis, spectrophotometric analyses and SEC-HPLC-ICP-MS. Results show that all methods verified the presence of these proteins in fish bile, while SEC-HPLC-ICP-MS also characterized also characterized metals responsible for the contamination of the studied sites. Quantification was successfully conducted by two methods, densitometry (1D electrophoresis) and spectrophotometry, and the data indicates that bile MTs follow the same trend as in liver. Therefore, Biliary MT show the potential to be used the same way as MT from other matrices in metal exposure studies, with the advantage of being a simpler matrix and showing the possibility of not sacrificing the animals, as chronic duct cannulation is available., (Copyright © 2012 Elsevier B.V. Total mercury (Hg) was determined in muscle tissue of 20 Guiana dolphins stranded along the coast of Rio de Janeiro state, Brazil, with a mean of 1.07 μg/g wet weight. Mercury concentrations were positively related to body length, possibly related to the capacity of the dolphins to bioaccumulate this element throughout life. The Hg concentrations were not significantly different between males and females, although females (1.08 μg/g) showed slightly higher levels than males (1.04 μg/g). Concentrations were low when compared to results of studies carried out with small cetaceans in the Northern Hemisphere, and with previous studies in the Southeastern Brazil.

Fish are well-recognized bioindicators of environmental contamination. Several recent proteomic studies have demonstrated the validity and value of using fish in the search and discovery of new biomarkers. Certain analytical tools, such as comparative protein expression analyses, both in field and lab exposure studies, have been used to improve the understanding of the potential for chemical pollutants to cause harmful effects. The metallomic approach is in its early stages of development, but has already shown great potential for use in ecological and environmental monitoring contexts. Besides discovering new metalloproteins that may be used as biomarkers for environmental contamination, metallomics can be used to more comprehensively elucidate existing biomarkers, which may enhance their effectiveness. Unfortunately, metallomic profiling for fish has not been explored, because only a few fish metalloproteins have thus far been discovered and studied. Of those that have, some have shown ecological importance, and are now successfully used as biomarkers of environmental contamination. These biomarkers have been shown to respond to several types of environmental contamination, such as cyanotoxins, metals, and sewage effluents, although many do not yet possess any known function. Examples of successes include MMPs, superoxide dismutases, selenoproteins, and iron-bound proteins. Unfortunately, none of these have, as yet, been extensively studied. As data are developed for them, valuable new information on their roles in fish physiology and in inducing environmental effects should become available.

Cisplatin and carboplatin are the most common platinum-based drugs used in cancer treatment. Pharmacokinetic investigations, the evaluation of the body burden during the treatment, as well as baseline levels of platinum in humans have attracted great interest. Thus, accurate analytical methods for fast and easy Pt monitoring in clinical samples become necessary. In the present study atomic absorption spectrometric methods for the determination of platinum in the forms of cisplatin and carboplatin in human urine were investigated. Platinum, in these different forms, could be determined in urine, after simple sample dilution. Regarding electrothermal atomic absorption spectrometry, the optimum parameters were defined by a central composite design optimization. Multiplicative matrix effects were overcome by using a mixture of HCl and NaCl as modifier. The limit of detection (LOD) was 0.004 mgL(-1) of platinum in the original sample. For the analysis of more concentrated samples, high resolution continuous source flame atomic absorption spectrometry was also investigated. Flame conditions were optimized by a multivariate D-optimal design, using as response the sum of the analyte addition calibration slopes and their standard deviations. Matrix matched external calibration with PtCl(2) calibration solutions, was possible, and the LOD was 0.06 mgL(-1) in the original sample. Introduction: This study was designed aiming to determine and compare the amount of arsenic in some brands of mineral trioxide aggregate (MTA) and Portland cement., Methods: In the present study, arsenic species (As[III], As[V], and dimethylarsinic acid) were separated by high-performance liquid chromatography (HPLC) using a strong anion exchange column and converted into arsines by online HG. The instrumental coupling, HPLC-HG-AFS, was applied to 0.2 g of each cement that was prior digested in a solution of HCl, HNO(3), and HBF(4). Data were expressed as a part per million, and the preliminary analysis of the raw pooled data revealed a bell-shaped distribution. Statistical analysis was performed using one-way analysis of variance for multiple comparisons., Results: In all chromatograms obtained, only type III arsenic could be detected. The minimum amount of arsenic was detected in samples of white MTA ProRoot (3.3 x 10-4) and the maximum in the samples MTA Bio Angelus (Angelus, Londrina, PR, Brazil) (8.6 x 10-4). In the Gray MTA (Angelus), gray ProRoot MTA (Tulsa/Dentsply, Tulsa, OK) and CP Juntalider (Brasilatex Ltda, Diadema, SP, Brazil) did not detect any trace of arsenic. The values of arsenic found in CP Irajazinho (Votorantim Cimentos, Rio Branco, SP, Brazil) and white MTA Angelus were intermediaries to minimum and maximum values. The nonparametric test Kruskal-Wallis showed statistically similar results among all cements tested (p > 0.5)., Conclusions: Overall, the present study showed that all cements showed insignificant amounts of type III arsenic as well as no trace of arsenic DMA and type V could be detected.

A procedure for the direct GFAAS determination of Ni in petroleum samples using a solid sampling strategy is proposed. Palladium was used as conventional modifier. Central composite design multivariate optimization defined the optimum temperature program and the Pd mass, allowing calibration using aqueous analytical solution. The limit of detection (LOD) at the optimized conditions was 0.23 ng of Ni, for typical sample masses between of 0.10 and 0.60 mg. Linearity at least up to 11 ng of Ni and a characteristic mass of 45 pg were observed, defining a dynamic range between 0.52 and 110 microg g(-1). Typical coefficients of variation (n = 10) in the analysis of oil reference materials were 7%. Method validation was performed both by the analysis of oil certified reference materials and by comparison with an independent method (ASTM 5863-B). No statistically significant difference was observed between obtained and expected values. The total determination cycle lasted 5 min, equivalent to a sample throughput of 6 h(-1) for duplicate determinations.

A procedure for the determination of As in diesel, gasoline and naphtha at microg L(-1) levels by GFAAS is proposed. Sample stabilization was achieved by the formation of three component solutions prepared by mixing appropriate volumes of the samples propan-1-ol and nitric acid aqueous solution. This mixture resulted in a one-phase medium, which was indefinitely stable. No changes in the analyte signals were observed over several days in spiked samples, proving long-term stabilization ability. The use of conventional (Pd) and permanent (Ir) modification was investigated and the former was preferred. Central composite design multivariate optimization defined the optimum microemulsion composition as well as the temperature program. In this way, calibration using aqueous analytical solutions was possible, since the same sensitivity was observed in the investigated microemulsion media and in 0.2% v/v HNO(3). Coefficients of correlation larger than 0.999 and an As characteristic mass of 22 pg were observed. Recoveries (n=4) obtained from spiked samples were 98+/-4, 99+/-3 and 103+/-5%, and the limits of detection in the original samples were 1.8, 1.2 and 1.5 microg L(-1) for diesel, gasoline and naphtha, respectively. Validation was performed by the analysis of a set of commercial samples by independent comparative procedures. No significant difference (Student's t-test, p<0.05) was observed between comparative and proposed procedure results. The total determination cycle lasted 4 min for diesel and 3 min for gasoline and naphtha, equivalent to a sample throughput of 7 h(-1) for diesel and 10 h(-1) for gasoline and naphtha.

The present work evaluated the use of iridium (Ir) as permanent modifier for the determination of total selenium in urine and serum by graphite furnace atomic absorption spectrometry. Concerning urine, the presence of trimethylselenonium (TMSe(+)) was especially considered. Pyrolysis and atomization temperatures of 1,000 and 2,100 degrees C, respectively, were used. For nondigested urine and serum samples, 0.2% v/v HNO(3) and Triton X-100 were used as diluents, respectively, and the same initial platform Ir treatment was effective for up to 1,100 atomization cycles. Good precision [less than 5% relative standard deviation (RSD)] can be achieved with the proposed method. Low TMSe(+) recovery was observed for nondigested urine samples. Thus, if this species is to be considered in urine analysis, a previous external mineralization step was found to be necessary. Alternatively, an in situ oxidation treatment was developed. Detection limits of 8, 10, and 7 mug l(-1) were obtained after dilution, microwave-assisted digestion, and in situ oxidation procedures, respectively. The accuracy of the method was validated by the analysis of certified reference or commercial quality control materials and spiked samples.

Pituitary duplication is a rare malformation commonly associated with other major neural/craniofacial anomalies, easily shown by magnetic resonance imaging. The authors describe two girls with duplication of the pituitary gland and thickening of the hypothalamus, facial dysmorphism and precocious pubertal development. The pathogenesis of pituitary duplication and its relationship with precocious pubertal development are discussed.

Thiazolylazo p-cresol (TAC) was studied as a masking agent for the determination of Bi in rich Ni and Cu alloys by hydride generation atomic absorption spectrometry (HGAAS). In the presence of TAC, Bi/Ni and Bi/Cu ratios up to 1:160 000 and 1:16 000, respectively, were found to be tolerable. No buffering was necessary once the masking agent was effective even in very acid medium. Limits of detection at the low mug g(-1) range were reached and the accuracy of the procedure was confirmed by the good concordance between found and certified values in the analysis of certified reference materials (CRM).

This paper intends to evaluate the uptake of cadmium ions from aqueous solution by 21 hydroxyapatite samples which have been synthesized in different conditions. It has been determined thatthe variation on the hydroxyapatite sorption capacity is neither related to sample solubility nor to hydroxyapatite Ca/P molar ratio. Cd2+ sorption is controlled by sample BET surface area, which shows a direct dependence on the hydroxyapatite crystallite dimensions. The hydroxyapatite pore distribution presented modes at 1000 and 60,000 A, corresponding to intracrystallite voids and voids between the agglomerate of these crystallites, respectively. Pores belonging to the former mode immobilize the major part of Cd2+. The influence of sample thermal treatment on Cd2+ sorption efficiency has been studied using hydroxyapatite samples calcined at temperatures ranging from 500 to 1140 degrees C. Similarly to nonthermally treat samples, the Cd2+ sorption on calcined hydroxyapatite could be described by Langmuir isotherms. The results showed that the maximum sorption capacity decreased from 0.631 mmol g(-1) for the noncalcined sample to 0.150 mmol g(-1) for the one calcined at 900 degrees C. This drop in the sorption capacity could also be explained by a reduction in its specific surface area, which is induced bythe increase of the crystal size.

This study aimed to relate the reasons for interrupting intravenous therapy and the time of in-situ permanence of venous devices. The results obtained showed that in 38 of the 75 punctures performed, the reason to interrupt the venous therapy took place after the end of the therapy and that time of in-situ permanence of peripheral devices varied from 24 to 48 hours. In 15 punctures, in which infiltration was the reason for interruption, the time of permanence was shorter than 24 hours. It is believed that the data gathered in this investigation should be analyzed by nursing professionals, aiming to enhance new educational programs in services related to intravenous therapy.

The pre-concentration of mercury(II) and methylmercury by adsorption of their dithiophosphoric acid diacyl ester (DDTP) chelates on a C18 column, then detection with cold-vapor atomic-absorption spectrometry was investigated. Conditions such as sample pH, reductant and chelating agent flow and concentration, and eluent and carrier gas flow were optimized. Optimization was performed by use of evolutionary operation with a proper factorial design. At a sample flow of 5.3 mL min(-1) and a loading time of 4.5 min, column adsorption efficiency ranged from 88 to 93% for both species. Detection limits down to 10 ng L(-1) were obtained at a sample throughput of 12 h(-1). There was good agreement between found and certified values in the analysis of certified reference materials after their microwave-assisted mineralization with HNO3 and H2O2.

A procedure has been developed for vapour-phase Fourier transform infrared determination of Ziram, a dithiocarbamate pesticide. The method is based on the evolution of CS(2), after decomposition of the dithiocarbamate with diluted H(2)SO(4) at 50 degrees C. The CS(2) evolved was swept by a carrier flow of nitrogen to a laboratory-made infrared gas cell of 39 mm pathlength and 490 mul volume. The signals were registered as a function of time. The area of peaks obtained from absorbance measurement in the wavenumber range between 1600 and 1450 cm(-1) were interpolated in a calibration line established from Ziram standards treated in the same way as samples. The method provided an absolute limit of detection of 0.055 mg, a variation coefficient of the order of 6% for an analyte mass of 50 mg, and an analysis time of 3.5 min.

A flow injection Fourier transform infrared spectrometric procedure has been developed for the determination the dithiocarbamate pesticides Ziram and Thiram in solid samples. All the operations involved, such as extraction, filtration and measurement, were integrated in the experimental set-up in order to avoid excessive manipulation of samples and standards. Ultrasonic assisted and mechanical extraction were evaluated for the solubilization of the analytes and, additionally, the effect of carrier flow rate, sample loop volume and the ratio between sample mass and volume of solvent employed were studied. Quantitative extractions with chloroform were obtained for both Ziram and Thiram, after 5 and 2 min, respectively, of mechanical shaking of sample slurries. Absorbance measurement, in the wavenumber range of 1600-1460 cm-1 for Ziram and 1400-1315 cm-1 for Thiram, was carried out, and the area values of the peaks obtained, as a function of time, were interpolated in external calibration lines prepared from standard solutions of Ziram and Thiram in chloroform. Analyses of commercial formulations and spiked soil samples incubated two weeks were in a good agreement with values found by other methodologies. Absolute detection limits of 400 micrograms for Ziram and 785 micrograms for Thiram and variation coefficients of 6.4% and 2.5% were obtained by use of the aforementioned methodology.
