Excessive sulfur dioxide (SO₂) disturbs physiology of lysosomes causing diseases and threatening human health. A fluorescent probe has been regarded as one of the most attractive approaches, which is compatible with living cells and possesses high sensitivity. However, most of fluorescent probes' reaction sites are activated before they reach the destination. In this work, an acid‐activatable fluorescent probe PT1 was synthesized, characterized, and used for SO2 detection. The introduction of oxazolines in PT1 enables the intelligent response of probe to release the activation stie for SO2 derivatives through Michael addition upon exposure to acid. In vitro studies showed a remarkable selectivity of PT1 to SO₂ derivatives than other biothiols with a limit of detection as low as 62 nM. By using this acidic pH‐controlled fluorescence responsiveness to SO₂, precise spatiotemporal identification of lysosomal SO2 fluctuations has been successfully performed. Background: Huanshaodan (HSD) is a Chinese Herbal Compound which has a definite clinical effect on Alzheimer's disease (AD), however, the underlying mechanism remains unclear. The aim of this study is to preliminarily reveal the mechanism of HSD in the treatment of AD model of SAMP8 mice. Methods: Chemical composition of HSD and its drug-containing serum were identified by Q-Orbitrap high resolution liquid mass spectrometry. Six-month-old SAMP8 mice were treated with HSD and Donepezil hydrochloride by gavage for 2 months, and Wogonin for 28 days. Behavioral test was performed to test the learning and memory ability of mice. Immunofluorescence (IF) or Western-blot methods were used to detect the levels of pSer404-tau and β-amyloid (Aβ) in the brain of mice. Hematoxylin–eosin (H&E) staining and Transmission electron microscopy (TEM) assay was applied to observe the pathological changes of neurons. Proteomic technology was carried out to analyze and identify the protein network of HSD interventions in AD. Then the pathological process of the revealed AD-related differential proteins was investigated by IF, Q-PCR, Western-blot, Fluorescence in situ hybridization (FISH) and 16S rRNA sequencing methods. Results: The results showed that HSD and Wogonin, one of the components in its drug-containing serum, can effectively improve the cognitive impairments of SAMP8 mice, protect hippocampal neurons and synapses, and reduce the expression of pSer404-tau and Aβ. HSD and Wogonin reduced the levels of fibrinogen β chain (FGB) and γ chain (FGG), the potential therapeutic targets revealed by proteomics analysis, reduced the colocalization of FGB and FGG with Aβ, ionized calcium binding adaptor molecule 1 (Iba-1), glial fibrillary acidic protein (GFAP), increased level of and myelin basic protein (MBP). Meanwhile, HSD and Wogonin increased ZO-1 and Occludin levels, improved brain microvascular injury, and reduced levels of bacteria/bacterial DNA and lipopolysaccharide (LPS) in the brain of mice. In addition, 16S rRNA sequencing indicated that HSD regulated the structure of intestinal microbiota of mice. Conclusion: The effects of HSD on AD may be achieved by inhibiting the levels of fibrinogen and the interactions on glia cells in the brain, and by modulating the structure of intestinal microbiota and improving the blood–brain barrier function. Abnormal levels of catecholamine (CA) neurotransmitters and their metabolites in biological fluids can lead to various neurological disorders. Herein, a boric acid‐functionalized hypercrosslinked polymer was prepared and utilized as a sorbent for the dispersive solid‐phase extraction of CAs and their metabolites in rat serum. By combination with a high‐performance liquid chromatography‐fluorescence detector, the extraction parameters for the seven target analytes were optimized. Under the optimal extraction condition, the methodology for the quantitative analysis of CAs and their metabolites in rat serum samples was established. The limits of detection and limits of quantification were found to be in the ranges of 0.010–0.015 and 0.033–0.050 ng/mL, respectively. The results demonstrated satisfactory recoveries, with values ranging from 88.02% to 113.27%, accompanied by relative standard deviations within the range of 2.69%–9.59%. In addition, the method showed good anti‐interference ability (matrix effect ranged from 2.64% to 18.07%). The developed method was validated for the determination of CAs and their metabolites in normal and Alzheimer's disease model rats' serum, which proved the promising application of the method for CAs neurotransmitter analysis in biological samples. In this work, a novel internal heat absorption technology using inorganic material rods is employed during the solidification process of steel ingots, aiming to control their solidification and improve the quality of the final product. The study investigates the effect of the insertion depth of inorganic materials on the solidification microstructure and macrosegregation of 2.5-ton 42CrMo ingots. The mechanical properties of samples from the product are also tested. A numerical simulation model for casting 2.5-ton ingots is established and implemented in Ansys Fluent fluid simulation software, with inorganic material rods set at different preset depths. The simulation explores the physical processes of the melting and floating of inorganic materials in molten steel, as well as their effects on the temperature and flow fields of the material. The results show that deeper insertion of inorganic materials (200 mm from the hot top) reduces the tendency for macrosegregation compared to that at the insertion depth of 100 mm. Specifically, the positive segregation area decreases by 10.35%, while the negative segregation area decreases by 15.32%. Moreover, deeper insertion results in a significant refinement of the solidification microstructure, ultimately enhancing the mechanical properties of the products machined from the ingots (i.e., the yield strength increased by 4.7%). The numerical simulation results indicate that as the placement depth of inorganic materials in the ingot mold increases, the cooling effect becomes more significant, the flow area of molten steel initiated by the inorganic materials expands, and the linear velocity of the double-circle flow increases. This further explains why the solidification quality of the ingots improves with the increasing placement depth of inorganic materials. [ABSTRACT FROM AUTHOR]

Mars Surface Composition Detector (MarSCoDe) is one of the important payloads carried by the Zhurong rover, China's first Mars exploration mission Tianwen‐1. The laser‐induced breakdown spectroscopy (LIBS) instrument of MarSCoDe is mainly used to detect major and trace elements on the surface of Mars. The quantitative analysis of alkali trace elements, namely lithium (Li), strontium (Sr), and rubidium (Rb), holds significance in unraveling the geological evolution of the Zhurong landing site. This study focuses on establishing univariate calibration models using MarSCoDe LIBS spectra from 84 samples tested in the ground laboratory. The accuracy of these models, within a few parts per million (ppm), was subsequently validated through the analysis of 12 onboard MarSCoDe Calibration Targets (MCCTs). With these models, Li, Sr, and Rb concentrations in the surface targets during the initial 300 sols (Martian days) traverse were determined. These concentrations ranged from 6 to 18, 106–628, and 22–87 ppm, respectively. Our results suggest that Li, Sr, and Rb are mainly related to the igneous rock components in the rocks and soils at the Zhurong landing site. The major secondary minerals in MarSCoDe scientific targets are likely small amounts of sulfates, which appear to have formed from the acidic weathering of recent surface brine. Clay minerals are likely either absent or very sparse in the scientific targets. The surface igneous materials at the landing site likely have originated from the most recent lava flow during the Amazonian epoch. The primary function of the laser‐induced breakdown spectroscopy (LIBS) on MarSCoDe is to measure the composition of Martian surface materials. Studying lithium (Li), strontium (Sr), and rubidium (Rb) can help us understand the history of where Zhurong is located. This study has developed precise models using MarSCoDe LIBS data from 84 ground test samples to measure these elements. The high accuracy of these models was verified through the Martian results of 12 MarSCoDe Calibration Targets (MCCTs). Using these models, we determined Li, Sr, and Rb contents in the surface targets during the first 300 Martian days of Zhurong rover exploration. Our results indicate that elevated Li, Sr, and Rb contents are primarily related to the magmatic rocks in MarSCoDe Martian targets. These targets contain small amounts of sulfates, which possibly formed due to recent acidic erosion connected to surface salty water. Clay minerals are likely either absent or very few in these targets. The surface magmatic materials where Zhurong is located likely formed from the most recent lava flow.
