Chronic HIV infection is characterized by persistent inflammation despite antiretroviral therapy (ART). Cannabinoids may help reduce systemic inflammation in people with HIV (PWH). To assess the effects of oral cannabinoids during HIV, ten PWH on ART were randomized (n = 5/group) to increasing doses of oral Δ9-tetrahydrocannabinol (THC): cannabidiol (CBD) combination (2.5:2.5–15:15 mg/day) capsules or CBD-only (200–800 mg/day) capsules for 12 weeks. Blood specimens were collected prospectively 7–21 days prior to treatment initiation and at weeks 0 to 14. Plasma cytokine levels were determined via Luminex and ELISA. Immune cell subsets were characterized by flow cytometry. HIV DNA/RNA were measured in circulating CD4 T-cells and sperm by ultra-sensitive qPCR. Results from both arms were combined for statistical analysis. Plasma levels of IFN-γ, IL-1β, sTNFRII, and REG-3α were significantly reduced at the end of treatment (p ˂ 0.05). A significant decrease in frequencies of PD1+ memory CD4 T-cells, CD73+ regulatory CD4 T-cells, and M-DC8+ intermediate monocytes was also observed (p ˂ 0.05), along with a transient decrease in CD28–CD57+ senescent CD4 and CD8 T-cells. Ki-67+ CD4 T-cells, CCR2+ non-classical monocytes, and myeloid dendritic cells increased over time (p ˂ 0.05). There were no significant changes in other inflammatory markers or HIV DNA/RNA levels. These findings can guide future large clinical trials investigating cannabinoid anti-inflammatory properties.

Indeed, HIV remains latent and unreachable by antiretrovirals in cellular and anatomical sanctuaries, preventing its eradication. The lungs have received very little attention compared to other anatomical reservoirs despite being immunological effector sites exhibiting characteristics ideal for HIV persistence. Furthermore, PLWH suffer from a high burden of pulmonary non-opportunistic infections, suggesting impaired pulmonary immunity despite ART. Meanwhile, various immune cell populations have been proposed to be cellular reservoirs in blood, including CD4− CD8− DN T cells, a subset that may originate from CD4 downregulation by HIV proteins. The present study aims to describe DN T cells in human and humanized mice lungs in relation to intrapulmonary HIV burden. The characterization of DN T cells as cellular HIV reservoirs and the lungs as an anatomical HIV reservoir will contribute to the development of targeted HIV eradication strategies., The lungs are relatively unexplored anatomical human immunodeficiency virus (HIV) reservoirs in the antiretroviral therapy (ART) era. Double negative (DN) T cells are a subset of T cells that lack expression of CD4 and CD8 (CD4− CD8−) and may have both regulatory and effector functions during HIV infection. Notably, circulating DN T cells were previously described as cellular HIV reservoirs. Here, we undertook a thorough analysis of pulmonary versus blood DN T cells of people living with HIV (PLWH) under ART. Bronchoalveolar lavage (BAL) fluid and matched peripheral blood were collected from 35 PLWH on ART and 16 uninfected volunteers without respiratory symptoms. Both PLWH and HIV-negative (HIV−) adults displayed higher frequencies of DN T cells in BAL versus blood, and these cells mostly exhibited an effector memory phenotype. In PLWH, pulmonary mucosal DN T cells expressed higher levels of HLA-DR and several cellular markers associated with HIV persistence (CCR6, CXCR3, and PD-1) than blood. We also observed that DN T cells were less senescent (CD28− CD57+) and expressed less immunosuppressive ectonucleotidase (CD73/CD39), granzyme B, and perforin in the BAL fluid than in the blood of PLWH. Importantly, fluorescence-activated cell sorter (FACS)-sorted DN T cells from the BAL fluid of PLWH under suppressive ART harbored HIV DNA. Using the humanized bone marrow-liver-thymus (hu-BLT) mouse model of HIV infection, we observed higher infection frequencies of lung DN T cells than those of the blood and spleen in both early and late HIV infection. Overall, our findings show that HIV is seeded in pulmonary mucosal DN T cells early following infection and persists in these potential cellular HIV reservoirs even during long-term ART. People living with HIV have high burdens of chronic lung disease, lung cancers, and pulmonary infections despite antiretroviral therapy (ART). The rates of tobacco smoking by people living with HIV vastly exceed that of the general population. Furthermore, we showed that HIV can persist within the lung mucosa despite long-term ART. As CD8 T cell cytotoxicity is pivotal for controlling viral infections and eliminating defective cells, we explored the phenotypic and functional features of pulmonary versus peripheral blood CD8 T cells in ART-treated HIV+ and uninfected controls. Bronchoalveolar lavage fluid and matched blood were obtained from asymptomatic ART-treated HIV+ smokers (n = 11) and nonsmokers (n = 15) and uninfected smokers (n = 7) and nonsmokers (n = 10). CD8 T cell subsets and phenotypes were assessed by flow cytometry. Perforin/granzyme B content, degranulation (CD107a expression), and cytotoxicity against autologous Gag peptide-pulsed CD4 T cells (Annexin V+) following in vitro stimulation were assessed. In all groups, pulmonary CD8 T cells were enriched in effector memory subsets compared with blood and displayed higher levels of activation (HLA-DR+) and exhaustion (PD1+) markers. Significant reductions in proportions of senescent pulmonary CD28−CD57+ CD8 T cells were observed only in HIV+ smokers. Pulmonary CD8 T cells showed lower perforin expression ex vivo compared with blood CD8 T cells, with reduced granzyme B expression only in HIV+ nonsmokers. Bronchoalveolar lavage CD8 T cells showed significantly less in vitro degranulation and CD4 killing capacity than blood CD8 T cells. Therefore, pulmonary mucosal CD8 T cells are more differentiated, activated, and exhausted, with reduced killing capacity in vitro than blood CD8 T cells, potentially contributing to a suboptimal anti-HIV immune response within the lungs.

The nature of the social influence of media on social processes, the production of virtual information practices, to study these processes currently actualizes the need to use modern new tools for collecting, processing and data analysis methods. The purpose of this work is to analyze the activity of university graduates in communities, their identification through the collection of data from social networks. Assessment of the activity of graduates in social networks was carried out by “downloading” messages and news from online university communities. For each message, activity labels (“likes”, reposts, comments) were collected and graduates of these universities were identified (reconciliation with the register of graduates was carried out). The focus of the analysis is on identifying the actions of graduates - loyalty in the media space and the dissemination of information about the university community. The main methodological guideline was the approach within the framework of the microsociological paradigm, in particular, the idea of symbolic interactionism. The heuristic potential of using big data to analyze the activity of university graduates in communities allows us to expand our methodological arsenal and overcome the limitations of existing traditional methods of collection and analysis. The main research methods: interface programming, social network analysis of user interaction in social media, Web-crawling using a search engine, statistical data processing. Results: the main digital strategies of university graduates are characterized by the expansion of the audience, the promotion of content caused by the interest of users depending on the focus of the group. Four types of alumni communities have been distinguished: groups that identify with social development, with charity, with scientific research, and education. The high average value of the activity index belongs to charitable foundations, followed by the community of culture and science. The lowest average value of the activity index is recorded in educational communities.

An electrical and mechanical design process for a traction motor in a hybrid bus application is studied. Usually, the design process of an electric machine calls for close cooperation between various engineering disciplines. Compromises may be required to satisfy the boundary conditions of electrical, thermal, and mechanical performances. From the mechanical point of view, the stress values and the safety factors should be at a reasonable level and the construction lifetime predicted by a fatigue analysis. In a vehicle application, the motor has to be capable of generating high torque when accelerating, and in normal operation, the losses of the machine should be low to be able to cool the machine. Minimization of the no-load iron losses becomes a very important electrical design requirement if the traction motor and the generator are mechanically connected with an internal combustion engine when it is operating as the only source of torque. The manufacturing costs of the motor are also taken into account in this paper.

A concept has been developed for a novel lightweight wheel structure intended for rotor and stator use in direct-drive wind turbine generators. It uses a slanted spoke and rim architecture to provide maximum static structural performance with minimum weight. A unique attribute of the structure is its use of layered sheet steel elements to form the spokes and rim. Friction between layers establishes structural integrity. The interaction between layers and the resulting increase in damping normal to the stack offers improved dynamic performance. A static structural analysis of a full-scale stator wheel structure for an 8 MW permanent magnet machine demonstrates the structural effectiveness of the architecture. To understand vibration characteristics of the lightweight wheel structure, a quarter-scale prototype was built and an experimental modal analysis was carried out to measure actual radial vibration responses. This data was used to verify a numerical model, and there was good agreement between measured and predicted behaviours. Finally, a modal analysis was carried out for the full-scale stator wheel structure. The dynamic performance of the wheel is acceptable for the stator of the 8 MW direct-drive permanent magnet synchronous generator embodiment.

Offshore applications, which call for the largest and most powerful wind turbines, demand a higher standard of reliability and maintainability. Direct-drive permanent-magnet synchronous generators (DD-PMSGs) are increasingly being specified for these applications. The major shortcoming to traditional high-powered direct-drive generators is extraordinary size and mass leading to extraordinary cost. To generate higher powers at low rotational speeds, direct-drive generators must either develop greater tangential stresses or be larger in diameter. For traditional air-cooled generators, higher power generally means a much larger diameter. Dramatic cost savings can be realised with the development of a more effective stator windings cooling system that puts further the limit on current density enabling the development of high-power direct-drive generators of substantially smaller diameters. This study presents a direct liquid cooling system design for an 8 MW outer-rotor DD-PMSG. The approach is new for wind turbine generators, so its impact on the thermal behaviour and reliability for the total electrical machine has been evaluated and reported here. Testing of a stator coil prototype (1/72nd of the complete stator) with internal cooling liquid flow is also reported to demonstrate the workability of the designed cooling solution.

Analytical calculation methods for all the major components of the synchronous inductance of tooth-coil permanent-magnet synchronous machines are reevaluated in this paper. The inductance estimation is different in the tooth-coil machine compared with the one in the traditional rotating field winding machine. The accuracy of the analytical torque calculation highly depends on the estimated synchronous inductance. Despite powerful finite element method (FEM) tools, an accurate and fast analytical method is required at an early design stage to find an initial machine design structure with the desired performance. The results of the analytical inductance calculation are verified and assessed in terms of accuracy with the FEM simulation results and with the prototype measurement results.

Wind is one of the most compelling forms of indirect solar energy. Available now, the conversion of wind power into electricity is and will continue to be an important element of energy self-sufficiency planning. This paper is one in a series intended to report on the development of a new type of generator for wind energy; a compact, high-power, direct-drive permanent magnet synchronous generator (DD-PMSG) that uses direct liquid cooling (LC) of the stator windings to manage Joule heating losses. The main parameters of the subject LC DD-PMSG are 8 MW, 3.3 kV, and 11 Hz. The stator winding is cooled directly by deionized water, which flows through the continuous hollow conductor of each stator tooth-coil winding. The design of the machine is to a large degree subordinate to the use of these solid-copper tooth-coils. Both steady-state and time-dependent temperature distributions for LC DD-PMSG were examined with calculations based on a lumped-parameter thermal model, which makes it possible to account for uneven heat loss distribution in the stator conductors and the conductor cooling system. Transient calculations reveal the copper winding temperature distribution for an example duty cycle during variable-speed wind turbine operation. The cooling performance of the liquid cooled tooth-coil design was predicted via finite element analysis. An instrumented cooling loop featuring a pair of LC tooth-coils embedded in a lamination stack was built and laboratory tested to verify the analytical model. Predicted and measured results were in agreement, confirming the predicted satisfactory operation of the LC DD-PMSG cooling technology approach as a whole.

The paper introduces an analytic permanent-magnet eddy-current loss calculation method for rotor surface permanent magnets. The problem has been studied widely in the literature, yet many of the suggested methods are difficult to use. A straightforward analytic solution is needed to speed up the motor preliminary design as present day computational capabilities are not fast enough to be used for 3-D time-stepped finite element analysis (FEA) of permanent-magnet eddy-current losses in every day design problems. An analytic approach is suggested to calculate different-harmonics-caused eddy-current losses. The results are compared both with 3-D FEA and measurement results. A comparison between the theoretical and experimental results is reported for an axial-flux two-stator single-rotor machine with no rotor yoke. The method has, however, been used in evaluating permanent-magnet losses also in machines with narrow slot openings where the flux density dips do not penetrate through the whole magnet or in machines having a laminated rotor yoke.

The minimum mass of the active materials needed to build an 8 MW liquid-cooled direct-drive permanent-magnet synchronous generator (LC DD-PMSG) is sought. Proper set of generator dimensions is defined based on a constrained steepest descent method with variable step size. The design variables have been defined in terms of two quantities: stator length and the ratio of induced voltage to rated voltage (EMF factor).

The use of permanent magnets offers freedom in machine design and the highest possible efficiencies for wind power machines. Although full power converters must be used with permanent magnet machines, these converters make it possible to fulfill even the strictest grid codes. If appropriate care is taken in selecting the exact magnetic material composition, Neodymium-Iron-Boron magnets are compatible with long term generator use. However, rare earth material prices have been so volatile lately that highpower applications using lower strength ferrite magnet materials are being studied. For wind power applications, the three basic types of PM generator include direct-drive generators (10–20 min−1), medium-speed generators (100–300 min−1), and high-speed generators (1000–2000 min−1). Finnish companies such as Neorem Magnets, ABB, and The Switch have been very active in the development of permanent magnet technology for wind power. Moreover, Finnish wind turbine manufacturers operating today use the most modern PM technologies in their products.

This paper presents the thermal modelling and analysis of a hybrid excitation synchronous generator with alternated current linkages (HESG-ACL) for stand-alone power supply. The HESG-ACL belongs to a class of hybrid excitation synchronous machines (HESMs), which combine the merits of electrically excited synchronous machines (EESMs) and permanent magnet synchronous machines (PMSMs). To confirm the performance and thermal behaviour of the HESG-ACL, an experimental prototype (69 kVA apparent power) equipped with thermal sensors was built and tested. A description of the HESG-ACL is given. The thermal analysis of the HESG-ACL is carried out based on the thermal resistance network model. The results obtained from the thermal model are verified by the experimental results.
