Multiple myeloma (MM) is a malignant hematopoietic disease that is usually incurable. However, the ubiquitin-proteasome system (UPS) genes have not yet been established as a prognostic predictor for MM, despite their potential applications in other cancers.RNA sequencing data and corresponding clinical information were acquired from Multiple Myeloma Research Foundation (MMRF)-COMMPASS and served as a training set (n=787). Validation of the prediction signature were conducted by the Gene Expression Omnibus (GEO) databases (n=1040). To develop a prognostic signature for overall survival (OS), least absolute shrinkage and selection operator regressions, along with Cox regressions, were used.A six-gene signature, including KCTD12, SIAH1, TRIM58, TRIM47, UBE2S, and UBE2T, was established. Kaplan-Meier survival analysis of the training and validation cohorts revealed that patients with high-risk conditions had a significantly worse prognosis than those with low-risk conditions. Furthermore, UPS-related signature is associated with a positive immune response. For predicting survival, a simple to use nomogram and the corresponding web-based calculator (https://jiangyanxiamm.shinyapps.io/MMprognosis/) were built based on the UPS signature and its clinical features. Analyses of calibration plots and decision curves showed clinical utility for both training and validation datasets.As a result of these results, we established a genetic signature for MM based on UPS. This genetic signature could contribute to improving individualized survival prediction, thereby facilitating clinical decisions in patients with MM.

The anisotropic Janus nanoparticles (JNPs) provide synergistic effects by concentrating multiple properties on a single carrier. Herein, we reported a novel and simple approach to fabricate acorn-like poly(acrylic acid)-mesoporous calcium phosphate/polydopamine (PAA-mCaP/PDA) JNPs, which were selectively functionalized with methoxy-poly(ethylene glycol)thiol (PEG-SH) on PDA domains to obtain superior stability, while the other mCaP sides served as a storage space and passage for the anti-cancer drug of doxorubicin (DOX). The unique acorn-like PAA-mCaP/PDA-PEG JNPs were utilized as novel theranostic agents for photoacoustic (PA) imaging-guided synergistic cancer chemo-phototherapy. More importantly, this synthetic strategy can be applied to synthesize various mesoporous Janus nanocarriers, paving the way toward designed synthesis of acorn-like JNPs in nanomedicine, biosensing and catalysis. Statement of significance The distinct acorn-like poly(acrylic acid)-mesoporous calcium phosphate/polydopamine Janus nanoparticles (PAA-mCaP/PDA JNPs) with a spherical-shaped PAA-mCaP core and PDA half-shell were fabricated for the first time. To achieve superior stability, the acorn-like PAA-mCaP/PDA JNPs were selectively functionalized with methoxy-poly(ethylene glycol)thiol (PEG-SH) on PDA domains to obtain acorn-like PAA-mCaP/PDA-PEG JNPs. The resultant acorn-like PAA-mCaP/PDA-PEG JNPs own an excellent biocompatibility, high drug-loading contents, good photothermal conversion efficiency, photoacoustic (PA) imaging capacity and pH/NIR dual-responsive properties, enabling the acorn-like JNPs to be applied for PA imaging-guided synergistic cancer chemo-phototherapy. More importantly, the synthetic approach could be extended to prepare acorn-like mesoporous inorganic substances/PDA JNPs for specific applications.

Oxygen defects and hollow structures positively impact pseudocapacitive properties of diffusion/surface-controlled processes, a component of critical importance when building high-performance supercapacitors. Hence, we fabricated hollow nickel/cobalt molybdate rods with O-defects (D-H-NiMoO 4 @CoMoO 4 ) via a soft-template and partial reduction method, enhancing D-H-NiMoO 4 @CoMoO 4 's electrochemical performance, yielding a specific capacitance of 1329 F g -1 , and demonstrating excellent durability with 95.8% capacity retention after 3000 cycles. D-H-NiMoO 4 @CoMoO 4 was used as the positive electrode to construct an asymmetric supercapacitor, displaying an energy density of up to 34.13 W h kg -1 and demonstrating good predisposition towards practical applications. This work presents an effective approach to fabricate and use hollow nickel/cobalt molybdate rods with O-defects as pseudo-capacitor material for high-performance capacitive energy storage devices.

Besides improving students' understanding of scientific concepts, chemistry teaching should also improve students' ability of applying concepts to solve problems. The research aims to explore the effects of modeling teaching on students’ proficiency in solving galvanic cell problems. This research used a quasi-experimental design, and the independent variable of the research was the teaching method. Forty-five students in the experimental class received modeling teaching, and 48 students in the control class received lecture-style teaching. The dependent variable was the performance level of the student's ability to solve the problem of the galvanic cell, which was evaluated using the galvanic cell proficiency assessment tool. The research results show that the students in the experimental class were significantly more proficient in solving galvanic cell problems than those in the control class. The results of unstructured interviews assisted in illustrating the role of modeling teaching in improving the proficiency of students in solving galvanic cell problems, and students in the experimental class had positive views on modeling teaching. Epithelial–mesenchymal transition (EMT) has been shown to be linked to a poor prognosis, particularly in patients with non-small-cell lung cancer. Nevertheless, little is known regarding the existence of EMT-related gene signatures and their prognostic values in lung adenocarcinoma (LUAD). In the current study, we systematically profiled the mRNA expression data of patients with LUAD in The Cancer Genome Atlas and Gene Expression Omnibus databases using a total of 1,184 EMT-related genes. The prognostic values of the EMT-related genes used to develop risk score models for overall survival were determined using LASSO and Cox regression analyses. A prognostic signature that consisted of nine unique EMT-related genes was generated using a training set. A nomogram, incorporating this EMT-related gene signature and clinical features of patients with LUAD, was constructed for potential clinical use. Calibration plots, decision-making curves, and receiver operating characteristic curve analysis showed that this model had a good ability to predict the survival of patients with LUAD. The EMT-associated gene signature and prognostic nomogram established in this study were reliable in predicting the survival of patients with LUAD. Thus, we first identified a novel EMT-related gene signature and developed a nomogram for predicting the prognosis of patients with LUAD.

This study aimed to investigate the clinicopathological and molecular characteristics of ELOC(TCEB1)-mutant renal cell carcinoma.Sanger sequencing was used to assess 32 cases originally diagnosed as clear cell renal cell carcinoma with CK7 positive and/or fibromyomatous stroma. Of these, 4 patients with ELOC(TCEB1) gene mutation were screened, and their clinicopathological data were collected for histomorphological observation, immunohistochemical staining, and follow-up, and relevant pieces of literature were reviewed.The 4 patients with ELOC(TCEB1) mutations were all males and aged between 57 and 64 years (median age: 59 years old). The tumor was located in the renal cortex, with a diameter of 2-3.5 cm. The cross-section was grayish-yellow and grayish brown, solid and nodular, and clearly demarcated from the surrounding tissues. Of the 4 patients, 3 harbored a thick fibrous pseudocapsule rich in smooth muscle and were separated from the surrounding normal renal tissue, and 2 of them showed focal invasion into the pseudocapsule, whereas 1 patient had no capsule but had focal invasion into the surrounding renal parenchyma. The tumor tissues mainly exhibited elongated or branched aciniform or tubular structures, commonly accompanied by interspersed small cystic and focal clustered short papillary structures. The cytoplasm of the tumor cells was rich and lightly stained, and the nuclear grading ranged from 1 to 2. All patients showed loose edema in the stroma, and 2 patients showed a small number of interspersed smooth muscle bundles. All 4 patients showed EMA, CA9, AMACR, and TCEB1 expression, and TCEB1 was mainly located in the nucleus. Vimentin, CK7, and CD10 expressions were observed in most cases; CD117, TFE3, HMB45, and melanA were not expressed in all tumors; the expression rate of Ki67 was 3%- 8%. All 4 patients had a point mutation in ELOC(TCEB1) Y79C. The patients were followed up for 24-93 months (mean 49 months), and all of them survived to date without recurrence or metastasis.ELOC(TCEB1)-mutant renal cell carcinoma is a rare type of renal cell carcinoma, which tends to occur in middle-aged and elderly men. The main characteristics of this tumor are the branching alveolar or tubular structure with clustered short papillae, presence of fibromyomatous stroma, and the expression of CK7, CA9, CD10, and AMACR. Positive TCEB1 nuclear staining may be an important marker and the Sanger sequencing method is helpful for the diagnosis of this type of RCC. Most patients harbor tumors exhibiting low nuclear grade and inert clinical behavior, and a few tumors exhibit high nuclear grade and aggressive characteristics.

Background Although the fact that long non-coding RNA MCM3AP antisense RNA 1 (MCM3AP-AS1) is oncogenic in several cancers is well documented, very few researchers investigate its expression and function in prostate cancer. Methods Paired prostate cancer samples were selected, and expressions of MCM3AP-AS1, miR-876-5p and WNT5A were examined by qRT-PCR. MCM3AP-AS1 shRNA was transfected into LNCaP and PC-3 cell lines, and then the proliferative activity and apoptosis of cancer cells were detected by CCK-8 assay, EdU assay and flow cytometry analysis, respectively. qRT-PCR and Western blot were used to analyze the changes of miR-876-5p and WNT5A. Luciferase reporter gene assay was employed to determine the regulatory relationship between miR-876-5p and MCM3AP-AS1, miR-876-5p and WNT5A. Results MCM3AP-AS1 was significantly up-regulated in cancerous tissues of prostate cancer samples, positively correlated with the expression of WNT5A, while negatively related with miR-876-5p. After transfection of MCM3AP-AS1 shRNA into prostate cancer cells, the proliferative ability of cancer cells was signally inhibited, but the apoptosis of cancer cells was increased. MCM3AP-AS1 shRNA could reduce the expression of WNT5A on both mRNA and protein levels. Besides, MCM3AP-AS1 was identified as a sponge of miR-876-5p. WNT5A was validated as a target gene of miR- 876-5p. Conclusion MCM3AP-AS1 is abnormally up-regulated in prostate cancer tissues and can modulate the proliferation and apoptosis of prostate cancer cells, which has the potential to be the “ceRNA” to regulate the expression of WNT5A by targeting miR-876-5p.

Clear cell renal cell carcinoma (CCRCC) with sarcomatoid differentiation (CCRCCS) displays invasive behavior, poor prognosis, and poor therapeutic response. The present study was aimed to gain new insights into the molecular mechanisms of sarcomatoid transformation, and identify new prognostic and therapeutic targets for CCRCCS. Whole exome sequencing was performed on matched carcinomatous and sarcomatoid elements from five specimens with CCRCCS. A non‑synonymous single‑nucleotide polymorphism (SNP) of cadherin 23 (CDH23) was further studied through Sanger sequencing in expanded 40 specimens with CCRCCS and 50 specimens with CCRCC. Carcinomatous and sarcomatoid elements shared most somatic single‑nucleotide variants (SSNVs) as revealed through whole exome sequencing. Sarcomatoid element had higher overall SSNVs than carcinomatous element. A highly frequent mutation of CDH23 (rs3802711) was observed in CCRCCS that resulted in an alteration in the highly conserved calcium‑binding site in the three‑dimensional (3D) structure mediating the functions of cadherins. In the expanded 90 specimens, CDH23 SNP (rs3802711) was a highly frequent mutation in CCRCCS than that in all CCRCC samples and even high grade CCRCC. Cox multivariate analysis indicated that CDH23 (rs3802711) genotype was an independent prognostic factor affecting the overall survival of the cohort. CDH23 gene and protein were negatively or weakly expressed in most CCRCCS specimens with CDH23 mutation. The present study revealed, for the first time, that the CDH23 (rs3802711) was a highly genetic risk factor for CCRCCS. It was associated with the decreased expression of CDH23 protein, resulting in the absence of cadherin function of CDH23, indicating that the CDH23 mutation may be involved in the sarcomatoid transformation in CCRCCS. Collectively, a novel and specific SNP of CDH23 was identified in CCRCCS and a new candidate cadherin involved in EMT was revealed. Furthermore, a new prognostic evaluation factor and potential therapeutic target for CCRCCS was identified.

A three-dimensional (3D) hollow CoWO4 composite grown on Ni-foam (3D-H CoWO4 /NF) based on a flower-like metal-organic framework (MOF) is designed by utilizing a facile dipping and hydrothermal approach. The 3D-H CoWO4 /NF not only possesses large specific areas and rich active sites, but also accommodates volume expansion/contraction during charge/discharge processes. In addition, the unique structure facilitates fast electron/ion transport of 3D-H CoWO4 /NF. Meanwhile, a series of characterization measurements demonstrate the appropriate morphology and excellent electrochemical performance of the material. The 3D-H CoWO4 /NF possesses a high specific capacitance of 1395 F g-1 , an excellent cycle stability with 89% retention after 3000 cycles and superior rate property. Furthermore, the 3D-H CoWO4 /NF can be used as a cathode to configurate an asymmetric supercapacitor (ASC), and 3D-H CoWO4 /NF//AC shows a good energy density (29.0 W h kg-1 ). This work provides a facile method for the preparation of 3D-hollow electrode materials with high electrochemical capability for advanced energy storage devices.

The galvanic cell is a basic concept in electrochemistry. To assess mainland Chinese students’ proficiency levels in galvanic cells, the Galvanic Cell Proficiency Level Assessment (GCPA) was developed based on the Rasch model. The GCPA was developed through a pilot test and consists of seven multiple-choice questions and four open questions. The assessment instrument was administered to 621 high school students in the 11th grade, and the test results showed good reliability and validity. The interview results supported the validity of the data generated by the instrument.

Aims This study retrospectively investigated the morphological, immunohistochemical and molecular genetic features of papillary renal neoplasm with reverse polarity (PRNRP), a recently described renal tumor. Methods and results Eleven cases of PRNRP were collected, and 16 cases of type I and 9 cases of type II papillary renal cell carcinoma were included as a control series. Pathological features were evaluated based on HE staining and immunohistochemistry. KRAS exon 2 and BRAF V600E mutations were detected by Real-time PCR and Sanger sequencing. Fluorescence in situ hybridization was conducted for identification of chromosomal abnormalities. Hemosiderin deposition was found in a small amount of tumor cells in 6 cases. Multifocal or patchy necrosis (5/11), small focal invasion of the pseudocapsules or renal parenchyma (6/11), and breakthrough of renal capsule with nerve invasion (1/11) were revealed, inconsistent with the previous view that the tumor lacks necrosis and intercellular hemosiderin. Immunohistochemical staining (diffusely positive for CK7 and GATA3, negative for CD117 and vimentin, and negative to weakly positive for P504S) and high frequency of KRAS mutations in exon 2 (9/10) supported the identification and inclusion of our cases. Chromosome 7 trisomy (1/7), chromosome 17 trisomy (0/7) and chromosome Y deletion (0/5 male patients) were seldom detected in this tumor. All patients were alive without metastasis or recurrence at the end of the follow-up. Conclusion Our findings may highlight the possibility of a low malignant potential of this emerging entity. We suggest that the tumor be classified as a novel renal cell tumor subtype independent of papillary renal cell carcinoma.

Biphasic squamoid alveolar papillary renal cell carcinoma (BSARCC) is a recently described renal tumor, considered as a unique rare morphological pattern of PRCC in the 2022 WHO classification. We aimed to further explore the clinicopathological, immunophenotypic, and molecular genetic characteristics of BSARCC in a Chinese cohort. The clinicopathological data of 8 patients with BSARCC were collected for morphology observation and immunohistochemical staining. QRT-PCR was performed to detect BRAF gene mutation. The abnormalities of chromosomes 7, 17 and Y, and CCND1 gene were investigated by fluorescence in situ hybridization (FISH). Among the 8 cases, 5 were males and 3 were females. 6 cases were revealed by physical examination incidentally, and 2 cases were presented with low back pain and hematuria. Microscopically, the tumors were mainly composed of small cells with different proportions of large cells. The small cells were arranged in acinar, compact and long narrow tubular, solid sheet or papillary structures. Single or clustered large cells were arranged in glomerular structures in the acinar cavity formed by small cells. Among the 8 cases, 2 cases had a small amount of typical type I papillary renal cell carcinoma without large cells at the edge of the tumor. And 5 cases presented the swallowing phenomenon of lymphocytes or neutrophils in the cytoplasm of a few large cells. Immunohistochemical staining showed both large and small cells were diffusely and strongly positive for CK7, P504s, EMA, and vimentin. MET was expressed in most tumors (6/8), and large cells (70-90%) exhibited a prominent higher positive rate than small cells (20-90%). CD57 was positive in both large and small cells in 5 cases. Notably, CyclinD1 was exclusively expressed in large cells, while RCC marker was mainly expressed in small cells. Large cells showed a higher ki67 index than small cells. BRAF V600E mutation was negative in all cases. FISH showed 4 cases had chromosome 7 trisomy, 2 cases had chromosome 17 trisomy, and 4 of 5 male patients had Y chromosome deletion. CCND1 gene amplification or rearrangement was not revealed in any case. Follow-up data were available in 8 cases ranging from 3 to 66 months, and there was no recurrence or metastasis of the tumor. BSARCC is a rare unique morphological pattern of papillary renal cell carcinoma, featured by the presence of two cell populations of different morphology and immunophenotype. Distinctive CyclinD1 and RCC immunostainings are essential in diagnosing this type of renal tumor. Large and small cells may be tumor cells in different stages of proliferation and differentiation, and large cells immunoreactive to CyclinD1 and presenting higher ki67 index might be more aggressive than small cells. The anomalous expression of CyclinD1 in large cells is not due to the aberrance of CCND1 gene itself. Most BSARCC is low-stage and inert with a favorable prognosis. Our study not only validates previously described clinicopathological features, but also supplements the clinicopathological features and expands the potential genetic alterations and available clinical outcome data of BSARCC.

In this paper, the Deep Learning-Based Innovative Ideological Behavior Education Model (DL-IIBEM) has been proposed to strengthen the mechanism to promote information exchange, enhance the user’s interactive experience, and make the platform perform efficiently. Knowledge Network Mechanism Analysis is integrated with DL-IIBEM to strengthen user feedback probability, the average probability of completing social media tasks on a popular network, and the predicted utility degree for individual users. The entire platform is dramatically improved. The simulation analysis is performed based on the performance ratio based on data set 1 (98.2%) and 2 (95.3%), skill development ratio (95.3%), accuracy ratio, the teaching methods in ideological and political education, and Students Achievements ratio (98.2%) prove the proposed framework’s reliability.

Background This study aimed to establish and validate an easy-to-operate novel scoring system based on simple and readily available clinical indices for predicting the progression of chronic kidney disease (CKD). Methods We retrospectively evaluated 1045 eligible CKD patients from a publicly available database. Factors included in the model were determined by univariate and multiple Cox proportional hazard analyses based on the training set. Results Independent prognostic factors including etiology, hemoglobin level, creatinine level, proteinuria, and urinary protein/creatinine ratio were determined and contained in the model. The model showed good calibration and discrimination. The area under the curve (AUC) values generated to predict 1-, 2-, and 3-year progression-free survival in the training set were 0.947, 0.931, and 0.939, respectively. In the validation set, the model still revealed excellent calibration and discrimination, and the AUC values generated to predict 1-, 2-, and 3-year progression-free survival were 0.948, 0.933, and 0.915, respectively. In addition, decision curve analysis demonstrated that the model was clinically beneficial. Moreover, to visualize the prediction results, we established a web-based calculator (https://ncutool.shinyapps.io/CKDprogression/). Conclusion An easy-to-operate model based on five relevant factors was developed and validated as a conventional tool to assist doctors with clinical decision-making and personalized treatment.

e20547 Background: There is accumulating evidence indicating that pathogenic germline mutations may contribute to lung cancer susceptibility besides environmental factors. The prevalence and clinical significance of pathogenic germline mutations in homologous recombination repair genes on lung cancer susceptibility has not been systematically investigated. Methods: We used real-world HaploX genomic data from patients (pts) with lung cancer to determine the prevalence of pathogenic germline mutations in homologous recombination repair genes from July 2020 to January 2022. Results: Out of the 766 patients screened, 5.5% (42/766) of the patients harbored the pathogenic germline mutations：12 with ATM, 12 with FANCA, 8 with ATR, 7 with BRCA1, and 3 with BRCA2. Conclusions: A proportion of 5.5% of patients carrying germline mutations in homologous recombination repair genes may be potentially linked to increased susceptibility to lung cancer. Our study has demonstrated that pathogenic germline mutations in homologous recombination repair genes are a risk factor for lung cancer.

Backgroud: This study was aimed to develop and internally validate a nomogram for risk of upgrade of ISUP (International Society of Urology Pathology) grade group from biopsy tissue to RP (radical prostatectomy) final histology.Methods: 166 patients with prostate cancer were retrospectively analyzed and divided into two groups based on ISUP upgrade status from needle biopsy to radical prostatectomy specimen, these being the 'ISUP upgrade' group and the 'no ISUP upgrade' group. Logistic regression analysis was used to predict the significant independent factors for ISUP upgrade. A nonogram was then developed based on these independent factors, which would predict risk of ISUP upgrade. The C-index, calibration plot, and decision curve analysis were used to assess the discrimination, calibration, and clinical usefulness of the predicting model. Internal validation was evaluated by using the bootstrapping validation. Results: There were 47 patients in the ISUP upgrade group and 119 patients in the no ISUP upgrade group respectively. Patients in the ISUP upgrade group tended to be of younger age, smaller PV (prostate volume), lower GS (Gleason score) of PB (prostate biopsy) tissue than the no ISUP upgrade group (p=0.043, p=0.041, p < 0.001, p =0.04, respectively). Multivariate logistic regression analysis showed that GS ≤6 (OR=14.236, P=0.001), prostate biopsy approach (TB-SB (transperineal prostate systematic biopsy) VS TR-SB (transrectal prostate systematic biopsy), OR=0.361, P=0.03) and number of positive cores < 10 (OR=0.396, P=0.04) were the independent risk factors for ISUP upgrade. A prediction nomogram model of ISUP upgrade was built based on these significant factors above, the area under the receiver operating characteristic (AUC) curve of which was 0.802. The C-index for the prediction nomogram was 0.798 (95%CI: 0.655–0.941) and the nomogram showed good calibration. High C-index value of 0.772 could still be reached in the interval validation. Decision curve analysis also demonstrated that the threshold value of RP-ISUP upgrade risk was 3% to 67%. Conclusion: A novel nomogram incorporating PSA, GS of PCa, ways of prostate biopsy and number of positive cores was built with a relatively good accuracy to assist clinicians to evaluate the risk of ISUP upgrade in the RP specimen, especially for the low-risk prostate cancer diagnosed by TR-SB.

Copious evidence reveals that long non-coding RNAs (lncRNAs) exert great regulatory functions in various human cancers. LINC01224 is a novel lncRNA, identified as a cancer regulator of HCC. However, the underlying mechanisms and clinical significance of LINC01224 in other types of cancers need further researches to explore. In this study, we aimed to elucidate the biological role of LINC01224 in NSCLC progression. Presently, LINNC01224 expression was elevated and miR-2467 expression was down-regulated in NSCLC, compared with standard control. Then we described the reciprocal correlation between LINC01224 and miR 2467. Afterward, the dual-luciferase reporter assay, RIP assay and RNA pull-down assay validated the base-pair interaction between LINC01224 and miR-2467. Moreover, our findings demonstrated that the silence of LINC01224 inhibited cell proliferation and invasion in NSCLC and enhanced cisplatin (CDDP) sensitivity in vitro. Besides, rescue assays verified that miR-2467 inhibitor could reverse the effects on cell biological activities and CDDP resistance caused by knockdown of LINC01224. Finally, in vivo experiments implicated that knockdown of LINC01224 could inhibit NSCLC tumor growth. To sum up, LINC01224 can promote tumor progression and CDDP resistance in NSCLC via sponging miR-2467, suggesting a promising therapeutic target for better diagnosis and prognosis of NSCLC patients.

The dolostone reservoir of the Middle Permian Maokou Formation in Eastern Sichuan has good prospects for oil and gas exploration. Study of dolomitizing genesis of the Maokou Formation is essential for predicting the distribution of the dolostone reservoir. Petrography, in situ geochemistry, Sr-Mg isotopes, and fluid inclusions were carried out on samples from the Maokou Formation in Eastern Sichuan in order to discuss the dolomitizing process. Based on mineral and textural characteristics, dolomites were divided into four components: partially clouded dolomite (PCD), mosaic-like dolomite (MLD), cloudy-centered and clear-rimmed dolomite (CACD), and saddle dolomite (SDD). Results indicate that the Maokou Formation in Eastern Sichuan mainly experienced two stages of dolomitization. PCD, MLD, and cloudy-centered dolomite (CCD) were formed during the early dolomitization. They all show turbid crystal planes and bright orange-red CL and have similar trace element contents, 87Sr/86Sr ratios, and rare-earth patterns, indicating that they might be formed in the same fluid. This is a period when dolomitizing fluids mainly migrated along pores or microcracks and replaced protogenetic calcites, which occurred in the shallow burial stage of the Maokou Formation before the Late Permian. Clear-rimmed dolomite (CRD) and SDD were formed in the late stage of dolomitization. They all have clean crystal planes and darkly red CL. CRD of the ERY profile has trace element contents, 87Sr/86Sr ratios, and rare-earth patterns similar to SDD of the HLCH profile and Well TL6, inferring that both may be formed in the same fluid. Combined with high SrO contents and homogenous temperatures of fluid inclusions of CRD and SDD and Mg-isotopic compositions, they were generated by hydrothermal dolomitization. The hydrothermal fluid stage is related to the movement of the Emeishan Large Igneous Province, which was made up of basaltic magmatic fluids mixing with the surface water. The hydrothermal fluid mainly migrated upwards along structural fractures or faults and filled in structural fractures, occurring in the Late Permian to Middle-Late Triassic.

Backgroud: This study was aimed to develop and internally validate an ISUP (International Society of Urology Pathology) upgrade risk nomogram from the biopsy tissue to the specimen of radical prostatectomy. Methods: The clinical characteristics of 166 patients with prostate cancer were retrospectively analyzed, who were divided into two groups based on the upgrade of ISUP between the biopsy tissue and radical prostatectomy specimen. Logistic regression analysis was used to predict the significant independent factors of ISUP upgrade, a nomogram was established to predict ISUP upgrade of prostatectomy specimen based on the significant factors. The C-index, calibration plot, and decision curve analysis were used to assess the discrimination, calibration, and clinical usefulness of the predicting model. Internal validation was evaluated by using the bootstrapping validation. Results: There were 47 patients in the ISUP upgrade group and 119 patients in the no ISUP upgrade group respectively. Patients in the ISUP upgrade group inclined to be younger age, smaller PV, lower GS scores and PB-ISUP than the no ISUP upgrade group (p=0.043, p=0.041, p < 0.001, p =0.04, respectively), Multivariate logistic regression analysis showed that PSA≥20 ng/ml (OR=8.558, P=0.024), Gleason score of PCa≤6 (OR=9.026, P=0.004), PB-ISUP=3,4vs5 (OR=23.232, P=0.000417; OR=26.72, P=0.000241), ways of prostate biopsy (TP-SB+COG-TB (transperineal prostate biopsy + cognitive fusion targeted biopsy) VS TR-SB (transrectal prostate system biopsy), OR=033, P=0.036) and number of positive cores < 10 (OR=0.21, P=0.002) were the independent risk factors for ISUP upgrade. A prediction nomogram model of ISUP upgrade was built based on these significant factors above, the area under the receiver operating characteristic (AUC) curve of which was 0.843. The C-index for the prediction nomogram was 0.871 (95% CI: 0.817–0.925) and the nomogram showed good calibration. Decision curve analysis also demonstrated that the threshold value of RP-ISUP upgrade risk was 1% to 89%. Conclusion: A novel nomogram incorporating PSA, Gleason score of PCa, PB-ISUP, ways of prostate biopsy and number of positive cores was built with a relatively good accuracy to assist clinicians to evaluate the risk of ISUP upgrade in the radical prostatectomy specimen, especially for the low-risk prostate cancer diagnosed by TR-SB.
