In this article, we study a p-Laplacian Schrödinger-Poisson system involving a parameter q≠0q\ne 0 in bounded domains. By using the Nehari manifold and the fibering method, we obtain the non-existence and multiplicity of nontrivial solutions. On one hand, there exists q*>0{q}^{* }\gt 0 such that only the trivial solution is admitted for q∈(q*,+∞).q\in \left({q}^{* },+\infty ). On the other hand, there are two positive solutions existing for q∈(0,q0*+ε)q\in \left(0,{q}_{0}^{* }+\varepsilon ), where ε>0\varepsilon \gt 0 and q0*+ε

Based on neural machine translation, this article introduced the ConvS2S system and transformer system, designed a semantic sharing combined transformer system to improve translation quality, and compared the three systems on the NIST dataset. The results showed that the operation speed of the semantic sharing combined transformer system was the highest, reaching 3934.27 words per second; the BLEU value of the ConvS2S system was the smallest, followed by the transformer system and the semantic sharing combined transformer system. Taking NIST08 as an example, the BLEU values of the designed system were 4.74 and 1.49 higher than the other two systems. The analysis of examples showed that the semantic sharing combined transformer had higher translation quality. The experimental results show that the proposed system is reliable in English content translation and can be further promoted and applied in practice.

BackgroundSingle-cell transcriptome and single-cell methylome technologies have become powerful tools to study RNA and DNA methylation profiles of single cells at a genome-wide scale. A major challenge has been to understand the direct correlation of DNA methylation and gene expression within single-cells. Due to large cell-to-cell variability and the lack of direct measurements of transcriptome and methylome of the same cell, the association is still unclear.ResultsHere, we describe a novel method (scMT-seq) that simultaneously profiles both DNA methylome and transcriptome from the same cell. In sensory neurons, we consistently identify transcriptome and methylome heterogeneity among single cells but the majority of the expression variance is not explained by proximal promoter methylation, with the exception of genes that do not contain CpG islands. By contrast, gene body methylation is positively associated with gene expression for only those genes that contain a CpG island promoter. Furthermore, using single nucleotide polymorphism patterns from our hybrid mouse model, we also find positive correlation of allelic gene body methylation with allelic expression.ConclusionsOur method can be used to detect transcriptome, methylome, and single nucleotide polymorphism information within single cells to dissect the mechanisms of epigenetic gene regulation.

Background: Mesenchymal stem cells (MSCs) are increasingly recognized for their regenerative potential. However, their clinical application is hindered by their inherent variability, which is influenced by various factors, such as the tissue source, culture conditions, and passage number. Methods: MSCs were sourced from clinically relevant tissues, including adipose tissue-derived MSCs (ADMSCs, n = 2), chorionic villi-derived MSCs (CMMSCs, n = 2), amniotic membrane-derived MSCs (AMMSCs, n = 3), and umbilical cord-derived MSCs (UCMSCs, n = 3). Passages included the umbilical cord at P0 (UCMSCP0, n = 2), P3 (UCMSCP3, n = 2), and P5 (UCMSCP5, n = 2) as well as the umbilical cord at P5 cultured under low-oxygen conditions (UCMSCP5L, n = 2). Results: We observed that MSCs from different tissue origins clustered into six distinct functional subpopulations, each with varying proportions. Notably, ADMSCs exhibited a higher proportion of subpopulations associated with vascular regeneration, suggesting that they are beneficial for applications in vascular regeneration. Additionally, CMMSCs had a high proportion of subpopulations associated with reproductive processes. UCMSCP5 and UCMSCP5L had higher proportions of subpopulations related to female reproductive function than those for earlier passages. Furthermore, UCMSCP5L, cultured under low-oxygen (hypoxic) conditions, had a high proportion of subpopulations associated with pro-angiogenic characteristics, with implications for optimizing vascular regeneration. Conclusions: This study revealed variation in the distribution of MSC subpopulations among different tissue sources, passages, and culture conditions, including differences in functions related to vascular and reproductive system regeneration. The main limitation to advances in treatment of bacterial meningitis and its complications is the incomplete knowledge of the pathogenesis and pathophysiology of this disease. The aim of this research is to detect the expression of interleukin (IL)-1β as pro-inflammatory cytokine and miRNA (miR)-146a as post transcriptional inflammation associated microRNA (miRNA) in the cerebral cortex of acute Escherichia coli (E. coli) meningitis immature rat model. Immature rats in the post natal day 11 (PN11) were used to construct a model of acute E. coli meningitis and served as controls. The expression of IL-1β and miR-146a were detected in the cerebral cortex by reverse transcription polymerase chain reaction (RT-PCR) and real-time quantitative PCR (qPCR) analysis respectively, 24 hours after bacterial inoculation. In the cerebral cortical tissue of acute E. coli meningitis immature rat model the IL-1β expression was significantly upregulated while the miR-146a expression was significantly downregulated. This study tried to add a new insight on the molecular basis of the E. coli meningitis pathogenesis at its very early stage through detecting the expression of IL-1β and miR-146a in the cerebral cortex of the infected immature rats. Consequently, modulation of the IL-1β- miR-146a axis may be a new target for treatment of acute E. coli meningitis.
