2. Effect of withdrawal rates on temperature field and grain competitive growth of directionally solidified DZ4125 alloy

Abstract

In order to investigate the distribution of temperature field in directionally solidified DZ4125 columnar superalloy, and to study the effects of different withdrawl rates on the changes of temperature gradient and mushy zone, the directional solidification processes with withdrawal rates of 3, 5 and 7 mm/min were simulated by ProCAST software. The results show that when the test rod is located in the heating zone, the isotherm is inclined distribution with low outside and high inside. And when the test rod is located in the cooling zone, the isotherm is inclined distribution with high outside and low inside. With the increase of withdrawal rate, the temperature gradient decreases gradually. The shape of the mushy zone depends on the withdrawal rate and the distance from the water-cooled plate. When the withdrawl rate is 5 mm/min, the horizontal state of the solidification front is maintained for a longest time. On this basis, the precision casting experiment of directional solidification test rod is carried out, and the relationship between the shape of liquidus and grain structure is found for castings withdrawn at 3 and 7 mm/min. It is found that when the liquidus is concave and convex, the grain grows towards the center and the surface of the test rod, and the horizontal liquidus can promote the grain growth along the axis of the test rod.

Abstract

In this paper we study symmetrizable intersection matrices, namely generalized intersection matrices introduced by P. Slodowy such that they are symmetrizable. Every such matrix can be naturally associated with a root basis and a Weyl root system. Using $d$-fold affinization matrices we give a classification, up to braid-equivalence, for all positive semi-definite symmetrizable intersection matrices. We also give an explicit structure of the Weyl root system for each $d$-fold affinization matrix in terms of the root system of the corresponding Cartan matrix and some special null roots.

18. Biosensing Chlorpyrifos in Environmental Water Samples by a Newly Developed Carbon Nanoparticle-Based Indirect Lateral Flow Assay

Abstract

Pesticides are used in agriculture to prevent pests. Chlorpyrifos (CHLP) is an insecticide with potentially detrimental effects on humans, bees, and the aquatic environment. Its effects have led to a total ban by the European Union (EU), but outside the EU, CHLP is still produced and used. An indirect lateral flow immunoassay (LFIA) for the detection of CHLP was developed and integrated into a cassette to create a lateral flow device (LFD). Species-specific reporter antibodies were coupled to carbon nanoparticles to create a detector conjugate. Water samples were mixed with a specific CHLP monoclonal antibody and detector conjugate and applied to the LFD. Dose-response curves elicited the detection of low concentrations of CHLP (<1 µg/L). This sensitivity was recorded through a rapid handheld digital imaging device but also visually by naked eye. The CHLP LFD was applied to a range of European surface water samples, fortified with CHLP, revealing a sensitivity in these matrices of 2 µg/L, both by digital and visual analysis. To improve the simplicity of the CHLP LFIA, the assay reagents were dried in tubes, enabling to carry out the test by simply adding water samples and inserting the LFIA strips. This CHLP LFIA is thus suited for the on-site screening of surface waters.

19. Exposure and risk assessment of acetamiprid in honey bee colonies under a real exposure scenario in Eucalyptus sp. landscapes

Abstract

Honey bee colonies have shown abnormal mortality rates over the last decades. Colonies are exposed to biotic and abiotic stressors including landscape changes caused by human pressure. Modern agriculture and even forestry, rely on pesticide inputs and these chemicals have been indicated as one of the major causes for colony losses. Neonicotinoids are a common class of pesticides used worldwide that are specific to kill insect pests, with acetamiprid being the only neonicotinoid allowed to be applied outdoors in the EU. To evaluate honeybees' exposure to acetamiprid under field conditions as well as to test the use of in-situ tools to monitor pesticide residues, two honeybee colonies were installed in five Eucalyptus sp. plantations having different area where Epik® (active substance: acetamiprid) was applied as in a common spraying event to control the eucalyptus weevil pest. Flowers, fresh nectar, honey bees and colony products samples were collected and analyzed for the presence of acetamiprid residues. Our main findings were that (1) acetamiprid residues were found in samples collected outside the spraying area, (2) the amount of residues transported into the colonies increased with the size of the sprayed area, (3) according to the calculated Exposure to Toxicity Ratio (ETR) values, spraying up to 22 % of honeybees foraging area does not harm the colonies, (4) colony products can be used as a valid tool to monitor colony accumulation of acetamiprid and (5) the use of Lateral Flow Devices (LFDs) can be a cheap, fast and easy tool to apply in the field, to evaluate the presence of acetamiprid residues in the landscape and colony products.

20. Neonicotinoid Microsphere Immunosensing for Profiling Applications in Honeybees and Bee-Related Matrices

Abstract

Neonicotinoids are the most commonly used insecticides due to their effectiveness. However, non-targeted insects, especially bees, are also affected by neonicotinoids. Therefore, neonicotinoid application can contribute to the declining bee populations worldwide. The presented study describes the development of novel competitive, fluorescent microsphere-based suspension immunoassays for neonicotinoid profiling and their application to bees and essential bee-related matrices, using the Multi-Analyte Profiling (xMAP) technology. For the construction of these neonicotinoid microsphere immunoassays (nMIAs), neonicotinoid–ovalbumin conjugates were coupled to unique fluorescent, paramagnetic microspheres, which competed with the free neonicotinoids that were present in test samples for interacting with the corresponding, specific antibodies. In total, five independent nMIA’s were developed for the detection of imidacloprid, acetamiprid, clothianidin, thiacloprid, thiamethoxam, dinotefuran, nitenpyram and imidaclothiz with the limits of detection being for 0.01 ng/mL, 0.01 ng/mL, 0.02 ng/mL, 0.02 ng/mL, 0.003 ng/mL, 2.95 ng/mL, 0.09 ng/mL and 0.04 ng/mL, respectively. The developed nMIAs were applied to fortified matrices including surface water, pollen, honey and honeybees. All of the neonicotinoids, except dinotefuran, could be sensitively detected in all of the tested environmental matrices and bees, with there being sensitivities of 1 ng/mL in water and 10 ng/g in solid materials. These nMIAs provide a rapid profiling method for all of the common neonicotinoids, including those that are banned by the European Union for outdoor use. The developed method can contribute to healthy and sustainable beekeeping, globally, via its application in the apiary environment.

Abstract

Abstract Background Oridonin, a tetracycline diterpenoid compound, has the potential antitumor activities. Here, we evaluate the antitumor activity and action mechanisms of oridonin in colorectal cancer. Methods Effects of oridonin on cell proliferation were determined by using a CCK-8 Kit. Cell cycle distribution was determined by flow cytometry. Apoptosis was examined by analyzing subdiploid population and terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling assay. Senescent cells were determined by senescence-associated β-galactosidase activity analysis. Semi-quantitative RT-PCR was used to examine the changes of mRNA of p16, p21, p27 and c-myc. The concomitant changes of protein expression were analyzed with Western blot. Expression of AcH3 and AcH4 were examined by immunofluorescence staining and Western blots. Effects of oridonin on colony formation of SW1116 were examined by Soft Agar assay. The in vivo efficacy of oridonin was detected using a xenograft colorectal cancer model in nude mice. Results Oridonin induced potent growth inhibition, cell cycle arrest, apoptosis, senescence and colony-forming inhibition in three colorectal cancer cell lines in a dose-dependent manner in vitro. Daily i.p. injection of oridonin (6.25, 12.5 or 25 mg/kg) for 28 days significantly inhibited the growth of SW1116 s.c. xenografts in BABL/C nude mice. With western blot and reverse transcription-PCR, we further showed that the antitumor activities of oridonin correlated with induction of histone (H3 and H4) hyperacetylation, activation of p21, p27 and p16, and suppression of c-myc expression. Conclusion Oridonin possesses potent in vitro and in vivo anti-colorectal cancer activities that correlated with induction of histone hyperacetylation and regulation of pathways critical for maintaining growth inhibition and cell cycle arrest. Therefore, oridonin may represent a novel therapeutic option in colorectal cancer treatment.

Abstract

Functionalized imidazolium-based ionic liquids (ILs) with different halides (Cl, Br, and I) were successfully immobilized on the surface of graphene oxide materials by one step through covalent condensation between alcoholic hydroxyl groups of GO and alkoxyl groups of functionalized ILs. Several characterization including TG, Raman, AFM, FT-IR, and XPS techniques have been applied to characterize the physicochemical properties of the synthesized GO-[SmIm]X materials. In the solvent-free cycloaddition reactions of CO2 to propylene oxide, GO-[SmIm]I showed remarkably catalytic activity, affording a maximum yield of propylene carbonate as ca. 96%. The heterogeneous catalyst could be reused for at least four runs without any significant loss in activity, and demonstrated versatile catalysis for a wide range of substrates. A possible catalytic mechanism has been proposed, wherein epoxides were activated by the oxygen-containing groups of GO and the halide anions of the grafted ILs acted as key active species for the catalytic cycloaddition reactions.

Abstract

This study is to investigate changes of tolerance to ischemia-reperfusion (IR) injury and protection of ischemic preconditioning (IPC) of type 1 diabetes rat heart. Type 1 diabetes rat model was made by single injection of streptozotocin and divided into 4 wk (D-4w) group and 8wk (D-8w) group randomly. The two groups were further assigned to six teams: D-4w-con, D-4w-IR, D-4w-IPC, D-8w-con, D-8w-IR and D-8w-IPC team. Normal rats were correspondingly divided into N-4w-con, N-4w-IR, N-4w-IPC, N-8w-con, N-8w-IR and N-8w-IPC team. Heart function, myocardial infarct size, release of lactate dehydrogenase (LDH) and creatine kinase (CK) were detected. And injury increment rate, injury rate and protection rate were calculated. Ultrastructure of cardiomyocyte was observed by electron microscope. The myocardial injury increment rate, infarct sizes in D-con group were all significantly higher than those in N-con rat group, and the degree of injury in D-8w rat hearts was exacerbated compared to D-4w rat hearts. The myocardial injury rate and the injury of cardiomyocyte ultrastructure in D-4w-IR team were lower compared with N-4w-IR group. Compared with D-4w-IR team, IPC decreased infarct sizes in D-4w-IPC group, but its protection rate was lower than that in N-4w-IPC team. Infarct sizes in D-8w-IPC team had no significance with those in D-8w-IR team. The fundament cardiac function of type 1 diabetes rat heart was weakened. Diabetes self could induce heart injury, which could aggravate along with time. The tolerance of 4w type 1 diabetes rat to IR injury is stronger but the protection of IPC was weaker compared to normal rat. The protection of IPC disappears in D-8w group.
