Abstract Background Chronic non-specific low back pain (CNSLBP) is prevalent among firefighters and may negatively affect their job performance. Alterations in the thoracolumbar fascia (TLF) structure are associated with CNSLBP. Percussive massage therapy (PT) has demonstrated potential in modifying fascial properties and alleviating pain. However, its effects on TLF structure in CNSLBP patients have not been thoroughly investigated. This study evaluates the effects of 6 weeks of PT on TLF echo intensity, thickness, pain, and disability in firefighters with CNSLBP, utilizing ultrasound imaging as a primary assessment tool. Methods This six-week randomized controlled trial involved 38 firefighters with CNSLBP. Participants were randomly assigned 1:1 to either the PT group (receiving PT and core stability exercise) or the control group (receiving only core stability exercise). TLF echo intensity and thickness were measured using ultrasound imaging, and disability was assessed using the Roland-Morris Disability Questionnaire (RMQ) at baseline and post-intervention. Pain was measured using the Visual Analog Scale (VAS) at baseline, immediately after the first intervention, three weeks post-intervention, and post-intervention. Paired t-tests were conducted to analyze within-group changes in RMQ and ultrasound data, while ANCOVA was used to assess between-group differences, and repeated measures ANOVA for VAS scores. Results Participants in the PT and control groups were aged 26.6±4.1 and 23.6±1.7 years, respectively, with BMI of 23.1±1.8 and 21.8±1.2 kg/m². The PT group showed significant reductions in bilateral TLF echo intensity post-intervention (left side: mean difference -6.4, 95% CI -9.7 to -3.1, p 0.05). Abstract Background Astroviruses (AstVs) are single-stranded RNA viruses that have been detected in a wide range of mammals and birds. They are associated with numerous interspecies transmissions and viral recombination events, posing a threat to human and animal health. Methods We collected 1,333 samples from wild animals, including bats, rodents, wild boars, and birds, from various states and cities in the Yunnan Province, China, between 2020 and 2023 to investigate the presence of AstVs. AstVs were detected using a polymerase chain reaction targeting the RdRp gene. Finally, the Molecular Evolutionary Genetics Analysis software was used to construct the phylogenetic tree. Results The overall positivity rate for AstVs was 7.12% in four species, indicating their widespread occurrence in the region. High genetic diversity among AstVs was observed in different animal species, suggesting the potential for interspecies transmission, particularly among rodents and birds. Additionally, we identified a novel AstV strain and, for the first time, provided information on the presence of bastroviruses in Yunnan, China. Conclusions The widespread distribution and high genetic diversity of AstVs, along with the observed potential for interspecies transmission, highlight the importance of further investigation and surveillance in the region. The findings emphasize the need for increased attention to AstVs and their potential impact on human and animal health in Yunnan and other regions.

The 3D object detection task is a crucial subtask of the environment perception module in Automated Driving System (ADS). The accuracy of object detection directly impacts the effectiveness of downstream autonomous driving tasks such as tracking, prediction, and planning. Existing 3D object detection networks in ADS that rely on multi-sensor fusion lack the utilization of temporal information and fail to fully consider the dynamic nature of the surrounding traffic environment. Therefore, we proposed BEVTemporal for ADS, which fuses LIDAR point clouds and surrounding multi-channel images. Its unique temporal module establishes the correlation between historical data and current data, effectively leveraging the temporal information of surrounding objects. We train and validate BEVTemporal on the nuScenes datasets. After incorporating temporal module, the Average Precision (AP) metrics of the network improved by 0.3%~1.7%, and mean Average precision (mAP) achieves 0.87% higher, nuScenes Detection Score (NDS) increased by 0.46%. The validation results on subsets of occluded objects show that the model effectively alleviates the problem of missed detection caused by sample occlusion, with significant improvements observed for heavily occluded samples. In different scenarios (sunny, rainy, daytime, night), mAP improvement ranges from 0.75% to 1.18%. Notably, in challenging scenarios such as rainy and night, AP can be improved by up to 3.6%. The experimental results show that BEVTemporal not only improves the accuracy of the 3D object detection network, but also significantly enhances the robustness of model in various scenarios and recall of objects under low visibility conditions.

Obtaining bound states in the continuum (BICs) in photonic crystals gives rise to the realization of resonances with high quality factors for lasing and nonlinear applications. For BIC cavities in finite-size photonic crystals, the bulk resonance band turns into discrete modes with different mode profiles and radiation patterns. Here, photonic-crystal BIC cavities encircled by the photonic bandgap of lateral heterostructures are designed. The mirror-like photonic bandgap exhibits strong side leakage suppression to confine the mode profile in the designed cavity. Multiple bulk quantized modes are observed both in simulation and experiment. After exciting the BIC cavity at different positions, different resonance peaks are observed. The physical origin of the dependence between the resonance peak and the illuminating position is explained by analyzing the mode profile distribution and further verified by numerical simulations. Our findings have potential applications regarding the mode selectivity in BIC devices to manipulate the lasing mode in photonic-crystal surface-emitting lasers or the radiation pattern in nonlinear optics.

Sodium butyrate (SB) is a histone deacetylase inhibitor that can induce changes in gene expression and secondary metabolite titers by inhibiting histone deacetylation. Our preliminary analysis also indicated that SB significantly enhanced the biosynthesis of carotenoids in the Rhodotorula glutinis strain YM25079, although the underlying regulatory mechanisms remained unclear. Based on an integrated analysis of transcriptomics and metabolomics, this study revealed changes in cell membrane stability, DNA and protein methylation levels, amino acid metabolism, and oxidative stress in the strain YM25079 under SB exposure. Among them, the upregulation of oxidative stress may be a contributing factor for the increase in carotenoid biosynthesis, subsequently enhancing the strain resistance to oxidative stress and maintaining the membrane fluidity and function for normal cell growth. To summarize, our results showed that SB promoted carotenoid synthesis in the Rhodotorula glutinis strain YM25079 and increased the levels of the key metabolites and regulators involved in the stress response of yeast cells. Additionally, epigenetic modifiers were applied to produce fungal carotenoid, providing a novel and promising strategy for the biosynthesis of yeast-based carotenoids.

Abstract The emergence and rapid spread of methicillin‐resistant Staphylococcus aureus (MRSA) raise a critical need for alternative therapeutic options. New antibacterial drugs and targets are required to combat MRSA‐associated infections. Based on this study, celastrol, a natural product from the roots of Tripterygium wilfordii Hook. f., effectively combats MRSA in vitro and in vivo. Multi‐omics analysis suggests that the molecular mechanism of action of celastrol may be related to Δ1‐pyrroline‐5‐carboxylate dehydrogenase (P5CDH). By comparing the properties of wild‐type and rocA‐deficient MRSA strains, it is demonstrated that P5CDH, the second enzyme of the proline catabolism pathway, is a tentative new target for antibacterial agents. Using molecular docking, bio‐layer interferometry, and enzyme activity assays, it is confirmed that celastrol can affect the function of P5CDH. Furthermore, it is found through site‐directed protein mutagenesis that the Lys205 and Glu208 residues are key for celastrol binding to P5CDH. Finally, mechanistic studies show that celastrol induces oxidative stress and inhibits DNA synthesis by binding to P5CDH. The findings of this study indicate that celastrol is a promising lead compound and validate P5CDH as a potential target for the development of novel drugs against MRSA.

Sustainable urban renewal is an important approach to achieving high-quality urban development. The elements of megacities are diverse, and their structures are complex. It is critical to carry out the scientific classification of grassroots governance units based on the concept and needs of urban renewal to promote targeted sustainability evaluation and achieve the precise application of renewal design and planning. This study takes the jurisdiction of Chengdu City as an example and constructs a hierarchical dimension composite classification. For this classification, 128 grassroots governance units are divided into nine types, according to their obvious spatial differences. Based on the properties of these types, suggestions for evaluating and implementing urban renewal are proposed: (1) high-density central areas generally face the dilemma of complex and rigid needs and administrative weaknesses, so the development of public participatory governance is an urgent issue; (2) in transitional suburban zones, areas on and between the development axes are significantly different, indicating that extra attention should be paid to the fairness of the renewal of semi-urbanized areas; (3) outer areas are generally marginalized in urban renewal processes and destructive redevelopment behaviors should be avoided.

The currently reported methods for preparing cellulose acetate hydrogels use chemical reagents as cross-linking agents, and the prepared ones are non-porous structured cellulose acetate hydrogels. Nonporous cellulose acetate hydrogels limit the range of applications, such as limiting cell attachment and nutrient delivery in tissue engineering. This research creatively proposed a facile method to prepare cellulose acetate hydrogels with porous structures. Water was added to the cellulose acetate–acetone solution as an anti-solvent to induce the phase separation of the cellulose acetate–acetone solution to obtain a physical gel with a network structure, where the cellulose acetate molecules undergo re-arrangement during the replacement of acetone by water to obtain hydrogels. The SEM and BET test results showed that the hydrogels are relatively porous. The maximum pore size of the cellulose acetate hydrogel is 380 nm, and the specific surface area reaches 62 m2/g. The porosity of the hydrogel is significantly higher than that of the cellulose acetate hydrogel reported in the previous literature. The XRD results show that the nanofibrous morphology of cellulose acetate hydrogels is caused by the deacetylation reaction of cellulose acetate.

ABSTRACT Prochlorococcus is the key primary producer in marine ecosystems, and the high-light-adapted clade II (HLII) is the most abundant ecotype. However, the genomic and ecological basis of Prochlorococcus HLII in the marine environment has remained elusive. Here, we show that the ecologically coherent subclade differentiation of HLII corresponds to genomic and ecological characteristics on the basis of analyses of 31 different strains of HLII, including 12 novel isolates. Different subclades of HLII with different core and accessory genes were identified, and their distribution in the marine environment was explored using the TARA Oceans metagenome database. Three major subclade groups were identified, viz., the surface group (HLII-SG), the transition group (HLII-TG), and the deep group (HLII-DG). These subclade groups showed different temperature ranges and optima for distribution. In regression analyses, temperature and nutrient availability were identified as key factors affecting the distribution of HLII subclades. A 35% increase in the relative abundance of HLII-SG by the end of the 21st century was predicted under the Representative Concentration Pathway 8.5 scenario. Our results show that the ubiquity and distribution of Prochlorococcus HLII in the marine environment are associated with the differentiation of diverse subclades. These findings provide insights into the large-scale shifts in the Prochlorococcus community in response to future climate change. IMPORTANCE Prochlorococcus is the most abundant oxygenic photosynthetic microorganism on Earth, and high-light-adapted clade II (HLII) is the dominant ecotype. However, the factors behind the dominance of HLII in the vast oligotrophic oceans are still unknown. Here, we identified three distinct groups of HLII subclades, viz., the surface group (HLII-SG), the transition group (HLII-TG), and the deep group (HLII-DG). We further demonstrated that the ecologically coherent subclade differentiation of HLII corresponds to genomic and ecological characteristics. Our study suggests that the differentiation of diverse subclades underlies the ubiquity and distribution of Prochlorococcus HLII in the marine environment and provides insights into the shifts in the Prochlorococcus community in response to future climate change.

To understand the effect of ultraviolet (UV)-B irradiation on the antioxidant capacity and growth of lettuce (Lactuca sativa), we subjected lettuce plants to UV-B irradiation (15.55 kJ m−2 d−1) for 7 days and measured yield, photosynthetic performance, hydrogen peroxide (H2O2), reduced glutathione (GSH), and ascorbic acid (AsA) contents, and the enzyme activity and expression of genes involving AsA recycling. UV-B exposure did not significantly decrease the fresh/dry weight of the lettuce shoots. The net photosynthesis rate, internal CO2 concentration, transpiration rate, and stomatal conductance decreased during the first 4 days of irradiation and light but recovered at day 7. In UV-B-treated plants, the levels of AsA, GSH, and H2O2 increased significantly and simultaneously, with a positive correlation found between H2O2 and AsA or GSH levels. UV-B exposure upregulated the expression level of most genes encoding the enzymes involving AsA recycling but downregulated the associated enzymatic activities. The increase of AsA content in UV-B-exposed lettuce might contribute to the AsA–GSH cycle, leading to downregulation of ascorbate oxidase (AO) enzymatic activity and gene expression. UV-B irradiation had a greater impact on metabolite levels than time of UV-B treatment. These results suggest that AsA homeostasis in UV-B-treated lettuce is regulated through a feedback loop between the expression and activity of enzymes associated with AsA recycling. Short-term UV-B supplementation (24 h) could be a promising approach to enhance AsA content in lettuce.

Computer vision-based displacement measurement techniques are increasingly used for structural health monitoring. However, the vision sensors employed are easily affected by optical turbulence when capturing images of the structure, resulting in displacement measurement errors that significantly reduce the accuracy required in engineering applications. Hence, this paper develops a multi-measurement point method to address this problem by mitigating optical-turbulence errors with spatial randomness. Then, the effectiveness of the proposed method in mitigating optical-turbulence errors is verified by static target experiments, in which the RMSE correction rate can reach up to 82%. Meanwhile, the effects of target size and the number of measurement points on the proposed method are evaluated, and the optimal target design criteria are proposed to improve our method’s performance in mitigating optical-turbulence errors under different measurement conditions. Additionally, extensive dynamic target experiments reveal that the proposed method achieves an RMSE correction rate of 69% after mitigating the optical-turbulence error. The experimental results demonstrate that the proposed method improves the visual displacement measurement accuracy and retains the detailed information of the displacement measurement results.

Increased multiantibiotic-resistant bacteria means that infected bone defects remain a significant challenge to clinics. Great interest has emerged in the use of non-antibiotic antimicrobials to reduce the rate of multiantibiotic-resistant bacterial infection and facilitate bone regeneration. The cationic antimicrobial peptide LL-37 is the sole human cathelicidin and has shown nonspecific activity against a broad spectrum of microorganisms. In this study, we fabricated the poly(lactic-co-glycolic acid)/β-calcium phosphate/peptide LL-37 (PLGA/TCP/LL-37, PTL) scaffold with low-temperature 3D-printing technology for the treatment of infected segmental bone defects. The prepared scaffolds were divided into three groups: a high LL-37 concentration group (PTHL), low LL-37 concentration group (PTLL) and blank control group (PT). The cytocompatibility and antimicrobial activity of the engineered scaffolds were tested in vitro, and their osteogenesis properties were assessed in vivo in a rat infected bone defect model. We found the fabricated PTL scaffold had a well-designed porous structure that could support a steady and prolonged LL-37 release. Furthermore, the PTHL group showed strong antibacterial activity against Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) without any inhibition of the proliferation or alkaline phosphatase activity of rat bone marrow mesenchymal stem cells (BMSCs) in vitro. In addition, the infected femoral defects implanted with PTHL group displayed new bone formation in four weeks without any evidence of residual bacteria, which showed similar antibacterial outcomes to the vancomycin and cancellous bone mixture group. In conclusion, the PTHL composite scaffold is a promising non-antibiotic antimicrobial graft with good biodegradability, biocompatibility, and osteogenic capability for infected bone defects.

Antibiotics can inhibit or kill microorganisms, while microorganisms have evolved antibiotic resistance strategies to survive antibiotics. Zymomonas mobilis is an ideal industrial microbial chassis and can tolerate multiple antibiotics. However, the mechanisms of antibiotic resistance and genes associated with antibiotic resistance have not been fully analyzed and characterized. In this study, we investigated genes associated with antibiotic resistance using bioinformatic approaches and examined genes associated with ampicillin resistance using CRISPR/Cas12a−based genome−editing technology. Six ampicillin−resistant genes (ZMO0103, ZMO0893, ZMO1094, ZMO1650, ZMO1866, and ZMO1967) were identified, and five mutant strains ZM4∆0103, ZM4∆0893, ZM4∆1094, ZM4∆1650, and ZM4∆1866 were constructed. Additionally, a four−gene mutant ZM4∆ARs was constructed by knocking out ZMO0103, ZMO0893, ZMO1094, and ZMO1650 continuously. Cell growth, morphology, and transformation efficiency of mutant strains were examined. Our results show that the cell growth of ZM4∆0103 and ZM4∆ARs was significantly inhibited with 150 μg/mL ampicillin, and cells changed to a long filament shape from a short rod shape. Moreover, the transformation efficiencies of ZM4∆0103 and ZM4∆ARs were decreased. Our results indicate that ZMO0103 is the key to ampicillin resistance in Z. mobilis, and other ampicillin−resistant genes may have a synergetic effect with it. In summary, this study identified and characterized genes related to ampicillin resistance in Z. mobilis and laid a foundation for further study of other antibiotic resistance mechanisms.

Astroviruses (AstVs) are single-stranded RNA viruses, including two main genera: Mamastroviruses (MAstVs) and Avastroviruses (AAstVs). AstVs have been detected in more than 80 different mammals and birds, with the characteristics of multiple cross-species transmission and gene recombination. All these have accelerated the process of virus mutation and posed a potential threat to human beings and animal husbandry. Yunnan province is a global hotspot with rich biodiversity and abundant animal resources and an important area with significance for public health and security because it neighbors a few Southeast Asian countries. This study collected 860 samples from 13 species of animals in Yunnan province for AstVs detection. The results showed that the positive rate of AstVs was 6.05%, and its extremely high genetic diversity was observed in different animal species. Potential cross-species transmission events were also detected from rodents to birds. Therefore, AstVs, which are widely distributed with highly diverse genes and the risk of cross-species transmission to people, deserve more attention in this region.

Staphylococcus xylosus (S. xylosus) has become an emerging opportunistic pathogen due to its strong biofilm formation ability. Simultaneously, the biofilm of bacteria plays an important role in antibiotic resistance and chronic infection. Here, we confirmed that rutin can effectively inhibit biofilm formation in S. xylosus, of which the inhibition mechanism involves its ability to interact with imidazole glycerol phosphate dehydratase (IGPD), a key enzyme in the process of biofilm formation. We designed experiments to target IGPD and inhibited its activities against S. xylosus. Our results indicated that the activity of IGPD and the amount of histidine decreased significantly under the condition of 0.8 mg/ml rutin. Moreover, the expression of IGPD mRNA (hisB) and IGPD protein was significantly down-regulated. Meanwhile, the results from molecular dynamic simulation and Bio-layer interferometry (BLI) technique showed that rutin could bind to IGPD strongly. Additionally, in vivo studies demonstrated that rutin treatment reduced inflammation and protect mice from acute mastitis caused by S. xylosus. In summary, our findings provide new insights into the treatment of biofilm mediated persistent infections and chronic bacterial infections. It could be helpful to design next generation antibiotics to against resistant bacteria.

The tensile and compression fatigue deformation tests of AZ31 magnesium alloy were carried out at room temperature, and the activation behavior of {10-12}-{10-12} secondary twins in different loading directions (ND normal direction, RD rolling direction) and different strain amplitude (0.5%, 1%) was studied. The results showed that {10-12}-{10-12} secondary twins were observed on the fracture surface of the fatigue samples in the ND at the strain of 1%. In contrast, no secondary twins were observed in the ND at the strain of 0.5% nor in the RD at the strain of 1% and 0.5%, and the fatigue life in the ND was greater than that in the RD. The analysis showed that the generation of {10-12}-{10-12} secondary twins was associated with the grain size and loading direction and that the fatigue deformation at 1% strain was more likely to generate {10-12}-{10-12} secondary twins due to the large lamellar thickness of the primary twins generated at relatively large strain conditions. The fatigue deformation along the ND tension was more favorable for the activation of multiple {10-12} primary twins, and the interaction of multiple twin variants was more favorable to activate {10-12}-{10-12} secondary twins.

This study aims to examine the association of breast cancer with dietary patterns among Chinese women. A population-based case-control study was conducted in Jiangsu, China. Newly diagnosed primary breast cancer patients were recruited as cases (n = 818). Controls (n = 935), selected from the general population, were frequency matched to cases. A validated food frequency questionnaire was used to assess dietary intake. Dietary patterns were identified by factor analysis and multivariable odds ratios (OR) and 95% confidence intervals (CI) were estimated. Four dietary patterns were identified: salty, vegetarian, sweet and traditional Chinese. The traditional Chinese pattern was found to be robustly associated with a lower risk of breast cancer among both pre- and post-menopausal women (4th vs. 1st quartile: OR for pre- and post-menopausal women was 0.47 and 0.68, respectively). Women with high factor scores of the sweet pattern also showed a decreased risk of breast cancer (4th vs. 1st quartile: OR for pre- and post-menopausal women was 0.47 and 0.68, respectively). No marked association was observed between a vegetarian pattern or a salty pattern and breast cancer. These findings indicate that dietary patterns of the traditional Chinese and the sweet may favorably associate with the risk of breast cancer among Chinese women.

Human umbilical cord-derived mesenchymal stem cells (UCMSCs) are particularly attractive cells for cellular and gene therapy in acute liver failure (ALF). However, the efficacy of this cell therapy in animal studies needs to be significantly improved before it can be translated into clinics. In this study, we investigated the therapeutic potential of UCMSCs that overexpress hepatocyte growth factor (HGF) in an acetaminophen-induced acute liver failure mouse model. We found that the HGF-UCMSC cell therapy protected animals from acute liver failure by reducing liver damage and prolonging animal survival. The therapeutic effect of HGF-UCMSCs was associated with the increment in serum glutathione (GSH) and hepatic enzymes that maintain redox homeostasis, including γ-glutamylcysteine synthetase (γ-GCS), superoxide dismutase (SOD), and catalase (CAT). Immunohistochemical staining confirmed that HGF-UCMSCs were mobilized to the injured areas of the liver. Additionally, HGF-UCMSCs modulated apoptosis by upregulating the antiapoptotic Bcl2 and downregulating proapoptotic genes, including Bax and TNFα. Taken together, these data suggest that ectopic expression of HGF in UCMSCs protects animals from acetaminophen-induced acute liver failure through antiapoptosis and antioxidation mechanisms.

Effective physical training is the necessary condition to achieve good physical training effect and greatly improve the level of physical competition. Therefore, it is necessary to take scientific and reasonable technical means to carry out systematic and effective supervision and management to ensure the effectiveness of training. This paper first analyzes the leg movement of swimmers, and defines the different intensity of swimming. After careful study, an image acquisition system based on light sensor is designed and developed for technical analysis and simulation of swimmers. The research uses advanced light sensor technology to design an image acquisition system that can record various movements of swimmers in the water in real time and convert them into digital data. By establishing a set of effective algorithms, these data are analyzed and simulated accurately. The experimental results show that the image acquisition system can accurately record swimmers' movements, and accurately analyze and simulate their technical performance. Compared with the traditional technical analysis method, the system in this paper is not only more accurate, but also can provide more comprehensive data support, which can effectively improve the accuracy and credibility of technical analysis, and has great significance for the improvement of swimming training and competitive level.
