BackgroundThe overdose crisis has generated innovative harm reduction and drug market monitoring strategies. In Toronto, Ontario, Canada, a multi-site drug checking service (DCS) pilot project was launched in October 2019. The project provides people who use drugs with information on the chemical composition of their substances, thereby increasing their capacity to make more informed decisions about their drug use and avoid overdose. DCS also provides real-time market monitoring to identify trends in the unregulated drug supply.MethodsSample data were obtained through analyses of drug and used drug administration equipment samples submitted anonymously and free of charge to DCS in downtown Toronto from October 10, 2019, to April 9, 2020, representing the first six months of DCS implementation. Analyses were conducted in clinical laboratories using liquid chromatography- and/or gas chromatography-mass spectrometry (LC-MS, GC-MS) techniques.ResultsOverall, 555 samples were submitted, with 49% (271) of samples that were found to contain high-potency opioids, of which 87% (235) also contained stimulants. Benzodiazepine-type drugs were found in 21% (116) of all samples, and synthetic cannabinoids in 1% (7) of all samples. Negative effects (including overdose, adverse health events, and extreme sedation) were reported for 11% (59) of samples submitted for analysis.ConclusionsToronto's DCS identified a range of high-potency opioids with stimulants, benzodiazepine-type drugs, and a synthetic cannabinoid, AMB-FUBINACA. This information can inform a range of evidence-informed overdose prevention efforts.

Research has been conducted on the technetium complex of dimethylglycyl-L-seryl-L-cys-teinyl(acm)-glycyl-L-threonyl-L-lysyl-L-prolyl-L-prolyl-L-arginine. The synthesis and characterization of this complex and its technetium and rhenium analogues have been performed and the results are reported.

Researchers continue to develop newer types of platinum-based drugs for treating cancer. The most widely use platinum cancer drug is cisplatin, but it has severe side effects and can only be used for certain tumors. Many tumors also become resistant to the drug.

The characterization and preparation of neutral, water-soluble re(V) and 99Tc(V) oxo complexes of dimethylglycyl-L-seryl-t-cysteinyl-glycinamide (SRP294) were performed using high performance liquid chromatography, X-ray crystallography and various spectroscopic techniques. Results indicate that the syn and anti isomers interconvert spontaneously in aqueous solution at room temperature. A mechanism is proposed for the interconversion of the two isomers.

The synthesis and characterization of water-soluble linear Nr4O2 amine phenols H2badd, H2Brbadd and H2Clbadd based on N,Ni-bis(3-aminopropyl)ethylendiamine are described. The deprotonation constants of the ligands and the stability constants of the metal complexes in aqueous solutions have been determined by potentiometric of the amine phenols in the presence and absence of Ga3+ and In3+. Monocationic hexacoordinated Ga and In complexes with linear amine phenols were also synthesized and characterized.

Study reports the synthesis and characterization of several potentially heptadentate N4O3 Schiff base and amine phenols, as well as a series of their mononuclear and dinuclear complexes with indium and the lanthanides. The X-ray structures of the Schiff bases H3api and H3Clapi, the mononuclear amine phenol complexes and the homodinuclear Schiff base complex have been estimated. Both of the amine phenol complexes have pentagonal bipyramidal coordination geometry around the metal ions, which are each coordinated by N4O3 donor sets. Results are discussed.

Spectroscopic studies help characterize various triethylenetetramine-based linear N4O2 amine phenols, derived from the reduction of the corresponding Schiff bases. Reaction of these phenols with Ga3+ and In3+, in the presence of 2 equiv of base, forms monocationic metal complexes. X-ray studies of the Ga complex reveal that four neutral amine nitrogens and two anionic phenolate oxygens exhibit coordination with Ga3+. NMR studies indicate the rigid solution structures of these complexes, and fluxional behavior is absent in these complexes.

The process of attenuation facilitates the synthesis of a novel sequence of N3O3 amine phenols. A novel amine phenol (H3L1) is formed when H3L1 interacts with acetone. The properties of the product are determined by crystallographic experiments to reveal the proper organization of the heteroatoms to enable metal complexation. Intra- and inter-strand H-bonds are produced to allow the prior orientation of a tripodal amine phenol ligand.
