Abstract

Magnetic gold nanoparticles (mGNP) have become a great interest of research for nano-material scientists because of their significant magnetic and plasmonic properties applicable in biomedical applications. Various synthetic approaches and surface modification techniques have been used for mGNP including the most common being the coprecipitation, thermal decomposition, and microemulsion methods in addition to the Brust Schiffrin technique, which involves the reduction of metal precursors in a two-phase system (water and toluene) in the presence of alkanethiol. The hybrid magnetic–plasmonic nanoparticles based on iron core and gold shell are being considered as potential theranostic agents. In this critical review, in addition to future works, we have summarized recent developments for synthesis and surface modification of mGNP with their applications in modern biomedical science such as drug and gene delivery, bioimaging, biosensing, and neuro-regeneration, neuro-degenerative and arthritic disorders. This review includes techniques and biological applications of mGNP majorly based on research from the previous six years.

9. Nanostructured lipid carriers based delivery of pioglitazone for treatment of type-2 diabetes

Abstract

Pioglitazone (PGZ) is utilized as a therapeutic agent in the management of (type 2) diabetes to control blood glucose levels. The existing research work was intended to make and optimize PGZ-containing NLCs (nanostructured lipid carriers). The fabricated nanostructured lipid carrier preparation was optimized by using different concentrations of the surfactants (Tween 80 and Span 80) and solid lipid (Compritol® 888 ATO) and liquid lipid (Labrasol®) while keeping the concentration of drug (PGZ), and co-surfactants (poloxamer 188) the same. The optimized NLC formulation (PGZ-NLCs) was further assessed for physical and chemical characterization, in vitro PGZ release, and stability studies. The optimized PGZ-NLCs have shown an average diameter of 150.4 nm, EE of 92.53%, PDI value of 0.076, and zeta-potential of −29.1 mV, correspondingly. The DSC thermal analysis and XRD diffractograms had not presented the spectrum of PGZ, confirming the comprehensive encapsulation of PGZ in the lipid core. PGZ-NLCs showed significantly extended release (51% in 24 h) compared to the unformulated PGZ. Our study findings confirmed that PGZ-NLCs can be a promising drug delivery system for the treatment of type 2 diabetes. 27. Notice of Retraction: Cytotoxicity of Eight Organic Solvents Towards Balb/3T3 and 293T Cells

Abstract

The aim of this study is to select organic solvents that dissolve large numbers of poor water-soluble chemicals including environmental contaminants in cell transformation assay and provide reference for solvents vehicle selection. The cytotoxicity of eight commonly used organic solvents-ethanol, methanol, acetone, isopropyl alcohol, 1,2-propanediol, glycerin, acetic ether and dimethyl sulfoxide (DMSO) towards Balb/3T3 and 293T cells has been studied by CCK-8 assay. The results indicated that the cytotoxicity increased significantly with the increasing concentration of solvent compared to the control group, and it showed a dosage-response relationship. Methanol, DMSO and ethanol exhibited much less cytotoxicity, which was slightly different between the two cell lines. The concentrations with 70% of cell viability were 3.70%, 1.40% or 1.20% (v/v) in Balb/3T3 cells and 2.20%, 1.40% or 0.80% (v/v) in 293T cells, respectively. The five other solvents displayed much greater cytotoxicity, the concentrations with 70% of cell viability were much lower. The concentrations of acetic ether and acetone with 70% of cell viability were below 0.20%. While, the difference of concentrations of isopropyl alcohol, glycerin and 1,2-propanediol with 70% of cell viability was 2-4 times between the two cell lines. The cell viability percent in solvents with little cytotoxicity should be above 90%. Methanol at 0-1.0% (v/v) and DMSO at 0 0.3% (v/v) presented much less cytotoxicity towards Balb/3T3 cells, the cell viability percent was about 95%. Ethanol at 0-0.3% (v/v) showed the least cytotoxic effect towards 293T cells, the cell viability percent was approximately 95%. Therefore, methanol/DMSO and ethanol might be compatible solvents vehicle towards Balb/3T3 and 293T cells, respectively. Methanol and ethanol were much better than DMSO towards the two cell lines. In addition, the selection of solvents vehicle might be affected by cell type.

32. Oxidative changes and apoptosis induced by 1800-MHz electromagnetic radiation in NIH/3T3 cells.

Abstract

To investigate the potential adverse effects of mobile phone radiation, we studied reactive oxygen species (ROS), DNA damage and apoptosis in mouse embryonic fibroblasts (NIH/3T3) after intermittent exposure (5 min on/10 min off, for various durations from 0.5 to 8 h) to an 1800-MHz GSM-talk mode electromagnetic radiation (EMR) at an average specific absorption rate of 2 W/kg. A 2′,7′-dichlorofluorescin diacetate fluorescence probe was used to detect intracellular ROS levels, immunofluorescence was used to detect γH2AX foci as a marker for DNA damage, and flow cytometry was used to measure apoptosis. Our results showed a significant increase in intracellular ROS levels after EMR exposure and it reached the highest level at an exposure time of 1 h ( p < 0.05) followed by a slight decrease when the exposure continued for as long as 8 h. No significant effect on the number of γH2AX was detected after EMR exposure. The percentage of late-apoptotic cells in the EMR-exposed group was significantly higher than that in the sham-exposed groups ( p < 0.05). These results indicate that an 1800-MHz EMR enhances ROS formation and promotes apoptosis in NIH/3T3 cells. [ABSTRACT FROM AUTHOR]
