Abstract

It is increasingly recognized that Alzheimer’s disease (AD) exists before dementia is present and that shifts in amyloid beta occur long before clinical symptoms can be detected. Early detection of these molecular changes is a key aspect for the success of interventions aimed at slowing down rates of cognitive decline. Recent evidence indicates that of the two established methods for measuring amyloid, a decrease in cerebrospinal fluid (CSF) amyloid β1−42 (Aβ1−42) may be an earlier indicator of Alzheimer’s disease risk than measures of amyloid obtained from Positron Emission Tomography (PET). However, CSF collection is highly invasive and expensive. In contrast, blood collection is routinely performed, minimally invasive and cheap. In this work, we develop a blood-based signature that can provide a cheap and minimally invasive estimation of an individual’s CSF amyloid status using a machine learning approach. We show that a Random Forest model derived from plasma analytes can accurately predict subjects as having abnormal (low) CSF Aβ1−42 levels indicative of AD risk (0.84 AUC, 0.78 sensitivity, and 0.73 specificity). Refinement of the modeling indicates that only APOEε4 carrier status and four plasma analytes (CGA, Aβ1−42, Eotaxin 3, APOE) are required to achieve a high level of accuracy. Furthermore, we show across an independent validation cohort that individuals with predicted abnormal CSF Aβ1−42 levels transitioned to an AD diagnosis over 120 months significantly faster than those with predicted normal CSF Aβ1−42 levels and that the resulting model also validates reasonably across PET Aβ1−42 status (0.78 AUC). This is the first study to show that a machine learning approach, using plasma protein levels, age and APOEε4 carrier status, is able to predict CSF Aβ1−42 status, the earliest risk indicator for AD, with high accuracy.

Abstract

The Direct/Delayed Response Project (DDRP) classified more than 600 soils obtained from 145 watersheds by using surface water chemistry as basis. A study was conducted to validate the DDRP sampling classes using multivariate analysis techniques. The most important variable for grouping pedons are drainage properties, hydrologic group, total clay, depth to bedrock,depth to impermeable layer and pedon slope. The most distinct sampling classes were Entisols, Histosols and Inceptisols. The DDRP sampling classes served as effective methods for soil classification, characterization and correlation to mapped areas.

6. Competitive strategies and new technology: an empirical investigation in the UK polymer processing industry

Abstract

This paper examines how companies can achieve competitive advantage by fitting their business strategies to the way in which new technology implementation is managed. In particular, the inter-relationship between strategy, technology and performance is examined. Five strategic types and seven types of technological objectives were used to find combinations which were associated with high performance. It was found that companies pursuing a prospector strategy (a strategy based on product innovation) combined with technological objectives emphasizing marketing, image and technical development were the highest performing group of companies. The lowest performing companies were those where strategy was dominated by price competition. The general conclusion of the paper is that, in order to achieve competitive advantage, companies need to see technology objectives as an inherent part of strategy. The study was carried out on a sample of twenty companies within the polymer processing industry, but the findings should be applicable to companies in other industries, and particularly to smaller companies., Companies can enhance competitiveness by incorporating strategic planning with technology implementation. Research indicates that a strategy based on product innovation can promote improved performance when used with technological objectives focused on marketing, technical and image development. Research also reveal that business strategies emphasizing price competition realize low performance levels.

Abstract

As cities cope with new economic realities their downtown spaces are changing. Downtowns have become consumption–based places that are designed more like amusement parks for tourists than as urban places for residents. The increasing privatization of design and oversight of downtown spaces raises questions about democratic access and in whose interests public development policy is made. The case cities examined here suggest that cities are struggling to find ways to make their need for development success match their desire to make downtowns accessible. Despite citizen participation and political rhetoric about public access, cities make compromises. They are willing to give private control over formerly public space in exchange for a productive economic return.
