Abstract

Development of vasomotor responses in fetal mesenteric arteries. Am J Physiol Heart Circ Physiol 279: H1097-H1105, 2000.--Changes in mesenteric arterial diameters were studied using intravital microscopy in chick fetuses at days 13 and 17 of incubation, corresponding to 0.6 and 0.8 fetal incubation time, both during 5 min of hypoxia followed by 5 min of reoxygenation and after topical administration of increasing concentrations, ([10.sup.-6]-[10.sup.-2] M) of norepinephrine (NE) and acetylcholine (ACh). Baseline diameters of second-order mesenteric arteries increased from 56 [micro]m at 0.6 incubation to 75 [micro]m at 0.8 incubation. Acute hypoxia induced a reduction in arterial diameter to 87 [+ or -] 4.4% of baseline at 0.6 incubation and to 44 [+ or -] 6.7% at 0.8 incubation (P [is less than] 0.01). During reoxygenation, mesenteric arteries dilated to 118 [is less than] 6.5% and 121 [is less than] 7.5% of baseline at 0.6 and 0.8 fetal incubation time, respectively. Phentolamine did not affect the vasoconstriction during hypoxia at 0.6 incubation, whereas this [Alpha]-adrenergic antagonist significantly attenuated the vasoconstrictor response at 0.8 incubation (to 93 [+ or -] 2.7% of baseline, P [is less than] 0.01). Topical NE induced maximal vasoconstriction to 71 [+ or -] 3% of baseline at 0.6 incubation and to 35 [+ or -] 3.8% at 0.8 incubation (P [is less than] 0.01). Maximal vasodilation to topical ACh was 113 [+ or -] 4.4% and 122 [+ or -] 4.8% of baseline at 0.6 and 0.8 incubation, respectively. These in vivo findings show that fetal mesenteric arteries constrict in response to acute hypoxia and that the increase in magnitude of this vasoconstrictor response from 0.6 to 0.8 of fetal development results from an increase in adrenergic constrictor capacity. cardiovascular development; hypoxia; necrotizing enterocolitis; norepinephrine; acetylcholine

Abstract

Leukocytes play an important role in the host defense as they may travel from the blood stream into the tissue in reacting to inflammatory stimuli. The leukocyte-vessel wall interactions are studied in post capillary vessels by intravital video microscopy during in vivo animal experiments. Sequences of video images are obtained and digitized with a frame grabber. A method for automatic detection and characterization of leukocytes in the video images is developed. Individual leukocytes are detected using a neural network that is trained with synthetic leukocyte images generated using a novel stochastic model. This model makes it feasible to generate images of leukocytes with different shapes and sizes under various lighting conditions. Experiments indicate that neural networks trained with the synthetic leukocyte images perform better than networks trained with images of manually detected leukocytes. The best performing neural network trained with synthetic leukocyte images resulted in an 18% larger area under the ROC curve than the best performing neural network trained with manually detected leukocytes. 25. Capillary recruitment and pain relief on leg dependency in patients with severe lower limb ischemia

Abstract

Patients suffering from severe lower limb ischemia may experience pain relief on leg dependency despite the fact that dependency normally results in arteriolar vasoconstriction. To clarify this possible paradox, skin microcirculation of the limb was investigated in 75 patients with different stages of lower limb ischemia and in 12 asymptomatic subjects. Using nailfold capillary video microscopy, red blood cell-perfused capillary density and diameter and red blood cell velocity were assessed in supine and sitting positions. Capillary density increased by changing from the supine to the sitting position, especially in patients with limb-threatening ischemia (showing a 4.5-fold increase versus a 1.5-fold increase in asymptomatic subjects). In subjects without or with mild ischemia, capillary perfusion was two to four times lower in the sitting than in the supine position. In patients with limb-threatening ischemia, perfusion was strongly reduced, being slightly higher in the sitting position. Patients with relief of pain while sitting did not always have a higher capillary perfusion but did have a higher capillary density in the sitting position. The arteriolar postural vasoconstrictive mechanism at the nutritive level is still intact in subjects without or with mild ischemia but not in patients with severe ischemia. Capillary recruitment rather than disturbed arteriolar vasoconstriction could explain why patients with severe leg ischemia prefer leg dependency

Abstract

The physical, biological and technical background for high-precision plasma surgery is prepared in a multi-disciplinary team. The aim of the research is to achieve controlled removal of diseased cells and bacteria without harming the healthy rest of the tissue. For this purpose, a small, cold, flexible and non-toxic plasma is developed (the plasma needle) and tested on cultured cells and bacterial samples. The needle is an atmospheric discharge induced by a radio-frequency voltage applied to a metal pin. This plasma operates at room temperature, in the milliwatt power regime; it poses no risk of thermal or electrical damage to living tissues. Several beneficial responses of living cells to plasma treatment have been already identified. Plasma does not cause accidental cell death (necrosis), which leads to inflammation and tissue damage. Instead, it allows to detach cells from each other and from the scaffold, and thus to remove them in a non-destructive way. Moreover, plasma is capable of bacterial inactivation. I parallel, we have determined the electrical and optical properties of the plasma and found a method of precise positioning of the plasma needle with respect to the treated tissue. © 2004 American Institute of Physics [ABSTRACT FROM AUTHOR]
