Abstract Under the background of global climate change, climate warming has led to an increase in insect herbivory, which significantly affects the growth, survival, and regeneration of forest plants in the warm temperate zone of China. Plants can adopt defense responses to adapt to insect defoliation. Therefore, field experiments were conducted on five common warm temperate species, Quercus acutissima, Quercus serrata, Quercus aliena, Quercus dentata, and Robinia pseudoacacia. We measured the leaf traits of healthy trees and insect defoliated trees, to explore the response strategies of common species in warm temperate zones to insect defoliation. Our results showed that native species stored more carbon in extreme environments for survival rather than growth, while the alien species R. pseudoacacia tended to adopt active resource acquisition strategies and were more inclined towards growth. The content of tannins and flavonoids in the alien species R. pseudoacacia did not significantly increase after leaf damage, while the content of secondary metabolites such as tannins, flavonoids, and total phenols in the native species Q. acutissima, Q. serrata, Q. aliena, Q. dentata increased significantly after leaf damage. This indicated that compared to alien species, native species invested more resources in defense, which might reduce resource allocation for growth. Thus, the native Quercus species have stronger resistance than the alien species R. pseudoacacia after insect defoliation.

ABSTRACT Research on plant functional traits have advanced our understanding of plant investment acquisition strategies. However, it is still unknown how a plant investment acquisition strategy varies or how the relative position of plants on the leaf economic spectrum changes across different habitats. Therefore, we conducted the field experiments at two long‐term ecological research sites in Shandong and Xinjiang, China, in August 2023. Twenty‐two common species in both sites and four endemics in each site were selected for leaf gas exchange traits, leaf growth traits, and leaf nutrient trait measurements. We used two different methods to quantify the leaf economic spectrum, PCA quantification and functional area quantification. We found that the 22 common species had a significantly faster investment acquisition strategy than local endemics on the leaf economic spectrum. Besides, the plasticity of the 22 common species was not coupled with the plasticity of their investment acquisition strategy. According to our results, we quantified and constructed the leaf economic spectrum of the 30 woody plant species in Shandong and Xinjiang and discussed that high plasticity and fast investment acquisition strategy may be an ecological adaptation and distribution strategy for widely distributed species.

Energy conversion technologies like fuel cells and metal-air batteries require oxygen reduction reaction (ORR) electrocatalysts with low cost and high catalytic activity. Herein, N-doped carbon spheres (N-CS) with rich micropore structure have been synthesized by a facile two-step method, which includes the polymerization of pyrrole and formaldehyde and followed by a facile pyrolysis process. During the preparation, zinc chloride (ZnCl2) was utilized as a catalyst to promote polymerization and provide a hypersaline environment. In addition, the morphology, defect content and activity area of the resultant N-CS catalysts could be regulated by controlling the content of ZnCl2. The optimum N-CS-1 catalyst demonstrated much better catalytic activity and durability towards ORR in alkaline conditions than commercial 20 wt% Pt/C catalysts, of which the half-wave potential reached 0.844 V vs. RHE. When applied in the Zn-air batteries as cathode catalysts, N-CS-1 showed a maximum power density of 175 mW cm−2 and long-term discharging stability of over 150 h at 10 mA cm−2, which outperformed 20 wt% Pt/C. The excellent performance could be due to its ultrahigh specific surface area of 1757 m2 g−1 and rich micropore channels structure. Meanwhile, this work provides an efficient method to synthesize an ultrahigh surface porous carbon material, especially for catalyst application.

IntroductionFire and nitrogen (N) deposition each impact biodiversity and ecosystem productivity. However, the effect of N deposition on ecosystem recovery after fire is still far from understood, especially in coastal wetlands.MethodsWe selected a typical coastal shrubland to simulate three N deposition levels (0, 10, and 20 g N m−2 year−1) under two different burned conditions (unburned and burned) in the Yellow River Delta of North China. Soil properties, soil microbial biodiversity, shrub growth parameters, herbaceous biodiversity, and aboveground productivity were determined after experimental treatments for 1 year.ResultsWe found that fire had a stronger influence on the ecosystem than N addition. One year after the fire, shrub growth had significantly decreased, while soil pH, soil electrical conductivity, herbaceous biodiversity, soil microbial biodiversity, and herbaceous aboveground productivity significantly increased. Conversely, a single year of N addition only slightly increased herbaceous aboveground productivity. The combined effect of fire and N addition was only significant for fungus biodiversity and otherwise had minimal influence. Interestingly, we found that herbaceous aboveground productivity was positively associated with fungal community diversity under unburned conditions but not in burned shrublands. Fire showed a great impact on soil parameters and biodiversity in the coastal wetland ecosystem even after a full year of recovery.DiscussionFire may also diminish the influence of several belowground factors on herbaceous aboveground productivity, which ultimately reduces recovery and stability. Appropriate N addition may be an effective way to improve the ecosystem productivity in a wetland dominated by shrub species.

Plant size influences plant responses to combined environmental factors under climate change. However, their roles in plant ecophysiological responses are not fully understood. Two rapidly growing Leguminosae species (Robinia pseudoacacia and Amorpha fruticosa) were used to examine plant responses to combined drought and defoliation treatments (two levels of both treatments). Both 1.5 month-old seedlings and 3 month-old seedlings were grown in a greenhouse, and seedling growth, leaf gas exchanges, stem hydraulics, and concentrations of non-structural carbohydrates were determined after 60 days of treatment. Our results indicated defoliation had no significant effect on plant height, basal diameter, and total biomass whatever plant sizes and species. Under the low water availability treatment, the defoliated seedlings significantly increased by 24% in stem water potential compared with non-defoliated seedlings in large R. pseudoacacia. Compared with the high water availability in large non-defoliated R. pseudoacacia seedlings, the low water availability significantly reduced by 26% in stem starch concentration to maintain the stem soluble sugar concentration stable, but not in small R. pseudoacacia seedlings. We also found a negative correlation between leaf and root soluble sugar concentration under low water availability in A. fruticosa. The results demonstrate defoliation could relieve the effect of low water availability in large seedlings. Large seedlings had more compensatory mechanisms in response to defoliation and drought treatments than small seedlings, thus species with large carbon reserves are more recommended for vegetation restoration under combined drought and defoliation conditions. Future studies with more species are crucial for obtaining more rigorous conclusions.

Objective: This study aimed to investigate the correlation of polo-like kinase 4 with clinicopathological features and survival profiles in patients with gastric cancer. Methods: This retrospective study was conducted based on the clinical data from 289 eligible patients with gastric cancer who received resection. Polo-like kinase 4 expression in adjacent tissue and tumor tissue was determined by immunohistochemical assay and semiquantified scoring method using immunohistochemical score by staining intensity score multiplying staining density score. Based on the total immunohistochemical score (ranged from 0 to 12), the polo-like kinase 4 expression was classified as low expression (immunohistochemical: 0-3) and high expression (immunohistochemical: 4-12); furthermore, high expression was divided into high+ expression (immunohistochemical: 4-6), high++ expression (immunohistochemical: 7-9), and high+++ expression (immunohistochemical: 10-12). Results: Polo-like kinase 4 expression was elevated in tumor tissue compared with adjacent tissue. Tumor polo-like kinase 4 high expression correlated with increased T stage and Tumor, Node, Metastasis (TNM) stage, while, it did not correlate with age, gender, current smoke, current drink, chronic complications, Helicobacter pylori infection, tumor location, pathological grade, or N stage. Besides, higher tumor polo-like kinase 4 expression correlated with shorter disease-free survival and overall survival. Subsequently, multivariate Cox proportional hazards regression analysis showed that higher tumor polo-like kinase 4 expression was an independent predictive factor for worse disease-free survival but not for overall survival. Conclusion: Polo-like kinase 4 possesses the clinical significance as a biomarker for aiding prognostication and facilitating postoperative tumor management in patients with gastric cancer.

Abstract Efficient energy conversion using ions as carriers necessitates membranes that sustain high permselectivity in high salinity conditions, which presents a significant challenge. This study addresses the issue by manipulating the linkages in covalent-organic-framework membranes, altering the distribution of electrostatic potentials and thereby influencing the short-range interactions between ions and membranes. We show that a charge-neutral covalent-organic-framework membrane with β-ketoenamine linkages achieves record permselectivity in high salinity environments. Additionally, the membrane retains its permselectivity under temperature gradients, providing a method for converting low-grade waste heat into electrical energy. Experiments reveal that with a 3 M KCl solution and a 50 K temperature difference, the membrane generates an output power density of 5.70 W m−2. Furthermore, guided by a short-range ionic screening mechanism, the membrane exhibits adaptable permselectivity, allowing reversible and controllable operations by finely adjusting charge polarity and magnitude on the membrane’s channel surfaces via ion adsorption. Notably, treatment with K3PO4 solutions significantly enhances permselectivity, resulting in a giant output power density of 20.22 W m−2, a 3.6-fold increase over the untreated membrane, setting a benchmark for converting low-grade heat into electrical energy.

Abstract Membrane reactors are known for their efficiency and superior operability compared to traditional batch processes, but their limited diversity poses challenges in meeting various reaction requirements. Herein, we leverage the molecular tunability of covalent organic frameworks (COFs) to broaden their applicability in membrane reactors. Our COF membrane demonstrates an exceptional ability to achieve complete conversion in just 0.63 s at room temperature—a benchmark in efficiency for Knoevenagel condensation. This performance significantly surpasses that of the corresponding homogeneous catalyst and COF powder by factors of 176 and 375 in turnover frequency, respectively. The enhanced concentration of reactants and the rapid removal of generated water within the membrane greatly accelerate the reaction, reducing the apparent activation energy. Consequently, this membrane reactor enables reactions that are unattainable using both COF powders and homogeneous catalysts. Considering the versatility, our findings highlight the substantial promise of COF-based membrane reactors in organic transformations.

Background The prevalence of hemorrhoids in Chinese urban and rural residents is very high. However, the factors affecting the occurrence of hemorrhoids are not very clear, which is disadvantageous to the prevention of hemorrhoids. Objective: To explore the external environment and behavioral influencing factors of hemorrhoidsin the Zhuang population of Guangxi, China, and provide a basis for the prevention of hemorrhoids. Methodology: A case-control study method was used to collect hemorrhoid patients and non-hemorrhoid patients from the same hospital and department as the case group and control group. A self-made questionnaire was used to investigate the education level, family income, alcohol consumption, diet, taste, work position, and other information of the two groups of patients, and logistic regression was used to analyze the correlation between various exposure factors and hemorrhoids. Result: A total of 11 variables were analyzed in this study, and it was found that education level, constipation, chronic gastritis, and dietary taste were associated with hemorrhoids (OR=2.02, 95% CI: 1.16, 3.52; OR=2.70, 95% CI: 1.50, 4.87; OR=1.96, 95% CI: 1.14, 3.36; OR=0.50, 95% CI: 0.28, 0.89). Conclusion Education level, constipation, and chronic gastritis can increase the risk of hemorrhoids, while bland diet can reduce the risk of hemorrhoids.

Introduction: The escalating global surge in Rifampicin-resistant strains poses a formidable challenge to the worldwide campaign against tuberculosis (TB), particularly in developing countries. The frequent reports of suboptimal treatment outcomes, complications, and the absence of definitive treatment guidelines for Rifampicin-resistant spinal TB (DSTB) contribute significantly to the obstacles in its effective management. Consequently, there is an urgent need for innovative and efficacious drugs to address Rifampicin-resistant spinal tuberculosis, minimizing the duration of therapy sessions. This study aims to investigate potential targets for DSTB through comprehensive proteomic and pharmaco-transcriptomic analyses.Methods: Mass spectrometry-based proteomics analysis was employed to validate potential DSTB-related targets. PPI analysis confirmed by Immunohistochemistry (IHC) and Western blot analysis.Results: The proteomics analysis revealed 373 differentially expressed proteins (DEPs), with 137 upregulated and 236 downregulated proteins. Subsequent Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses delved into the DSTB-related pathways associated with these DEPs. In the context of network pharmacology analysis, five key targets—human leukocyte antigen A chain (HLAA), human leukocyte antigen C chain (HLA-C), HLA Class II Histocompatibility Antigen, DRB1 Beta Chain (HLA-DRB1), metalloproteinase 9 (MMP9), and Phospholipase C-like 1 (PLCL1)—were identified as pivotal players in pathways such as “Antigen processing and presentation” and “Phagosome,” which are crucially enriched in DSTB. Moreover, pharmaco-transcriptomic analysis can confirm that 58 drug compounds can regulate the expression of the key targets.Discussion: This research confirms the presence of protein alterations during the Rifampicin-resistant process in DSTB patients, offering novel insights into the molecular mechanisms underpinning DSTB. The findings suggest a promising avenue for the development of targeted drugs to enhance the management of Rifampicin-resistant spinal tuberculosis.

With the continuous scarcity of domestic zinc resources and the rapid changes in the market, the composition of zinc concentrates selected by zinc smelting companies has become more and more complex. High-silicon, high-copper, high-lead, and high-iron mineral sources have gradually become the norm. These high impurities ore seriously deteriorates the conditions of the fluidized roaster, which greatly affects the normal progress of the roasting process. In order for zinc smelting companies to achieve rapid development in the increasingly complex market competition, in addition to organizing production at the lowest cost, they also need to be able to adapt to various complex mineral sources, especially to be able to handle high value-added zinc concentrates. Therefore, this article systematically studies the behavior and influence of Cu, Pb, SiO2 and Fe, the key impurities in complex zinc concentrates, during boiling roasting, and aims to provide certain theoretical and practical guidance for zinc smelting enterprises to deal with complex zinc concentrates.

Abstract Background Osteogenesis imperfecta (OI) is a rare congenital disorder of the skeletal system, inflicting debilitating physical and psychological distress on patients and caregivers. Over the decades, much effort has been channeled towards understanding molecular mechanisms and developing new treatments. It has recently become more apparent that patient-reported outcome measurements (PROM) during treatment, healing and rehabilitation are helpful in facilitating smoother communication, refining intervention strategies and achieving higher quality of life. To date, systematic analyses of PROM in OI patients remain scarce. Results Here, utilizing a PROM Information System, we report a cross-sectional and longitudinal study in a southern Chinese cohort of 90 OI patients, covering both the child and adult age-groups. In the child group where both self and parental surveys were obtained, we identified two clusters of comparable sizes showing different outlooks in physical mobility and emotional experiences. One cluster (Cluster 1) is more negative about themselves than the other (Cluster 2). A concordance of 84.7% between self and parental assessments was recorded, suggesting the stability and validity of PROM-based stratification. Clinical subtyping, deformity, leg length discrepancy, and limited joint mobility were significantly associated with this stratification, with Cluster 1 showing higher percentages of severe phenotypes than Cluster 2. Since OI is a genetic disorder, we performed genetic testing on 72 of the 90 patients, but found no obvious association between genotypes and the PROM stratification. Analyses of longitudinal data suggested that patients tended to stay in the same psychological state, in both clusters. Adult patients also showed a continuous spectrum of self-evaluation that matches their clinical manifestations. Conclusion By systematically analyzing patient-reported outcomes, our study demonstrated the link between the sociopsychological wellbeing of OI patients, and their clinical manifestations, which may serve as the basis for evaluating clinical interventions and help achieve better patient-centric medical practices. The lack of genotype-PROM association may be due to the diverse mutational spectrum in OI, which warrants further investigation when a larger sample size is available.

Abstract Background Osteogenesis imperfecta (OI) is a group of rare skeletal dysplasia. Long bone deformity and scoliosis are often associated with progressively deforming types of OI. FKBP65 (encoded by FKBP10, OMIM *607063) plays a crucial role in the processing of type I procollagen. Autosomal recessive variants in FKBP10 result in type XI osteogenesis imperfecta. Methods Patients diagnosed with OI were recruited for a genetic test. RT‐PCR and Sanger sequencing were applied to confirm the splicing defect in FKBP10 mRNA with the splice‐site variant. The bone structure was characterized by Goldner's trichrome staining. Bioinformatic analyses of bulk RNA sequencing data were performed to examine the effect of the FKBP10 variant on gene expression. Results Here we reported three children from a consanguineous family harboured a homozygous splice‐site variant (c.918‐3C > G) in FKBP10 intron and developed long bone deformity and early onset of scoliosis. We also observed frequent long bone fractures and spinal deformity in another 3 OI patients with different FKBP10 variants. The homozygous splicing variant identified in the fifth intron of FKBP10 (c.918‐3C > G) led to abnormal RNA processing and loss of FKBP65 protein and consequently resulted in aberrant collagen alignment and porous bone morphology. Analysis of transcriptomic data indicated that genes involved in protein processing and osteoblast differentiation were significantly affected in the patient‐derived osteoblasts. Conclusion Our study characterized the clinical features of OI patients with FKBP10 variants and revealed the pathogenesis of the c.918‐3C > G variant. The molecular analyses helped to gain insight into the deleterious effects of FKBP10 variants on collagen processing and osteoblast differentiation.

Background Ion homeostasis is an essential process for the survival of plants under salt stress. Na+/H+ antiporters (NHXs) are secondary ion transporters that regulate Na+ compartmentalization or efflux reduce Na+ toxicity and play a critical role during plant development and stress responses. Methods and Results To gain insight into the functional divergence of NHX genes in honeysuckle, a total of seven LjNHX genes were identified on the whole genome level and were renamed according to their chromosomal positions. All LjNHXs possessed the Na+/H+ exchanger domain and the amiloride-binding site was presented in all NHX proteins except LjNHX4. The phylogenetic analysis divided the seven NHX genes into Vac-clade (LjNHX1/2/3/4/5/7) and PM-clade (LjNHX6) based on their subcellular localization and validated by the distribution of conserved protein motifs and exon/intron organization analysis. The protein-protein interaction network showed that LjNHX4/5/6/7 shared the same putatively interactive proteins, including SOS2, SOS3, HKT1, and AVP1. Cis-acting elements and gene ontology (GO) analysis suggested that most LjNHXs involve in the response to salt stress through ion transmembrane transport. The expression profile analysis revealed that the expression levels of LjNHX3/7 were remarkably affected by salinity. These results suggested that LjNHXs play significant roles in honeysuckle development and response to salt stresses. Conclusions The theoretical foundation was established in the present study for the further functional characterization of the NHX gene family in honeysuckle.

Osteogenesis imperfecta (OI) is a rare inherited connective tissue dysplasia characterized with skeletal fragility, recurrent fractures and bone deformity, predominantly caused by mutations in the genes COL1A1 or COL1A2 that encode the chains of type I collagen. In the present study, clinical manifestations and genetic variants were analysed from 187 Chinese OI patients, majority of whom are of southern Chinese origin. By targeted sequencing, 63 and 58 OI patients were found carrying mutations in COL1A1 and COL1A2 respectively, including 8 novel COL1A1 and 7 novel COL1A2 variants. We validated a novel splicing mutation in COL1A1. A diverse mutational and phenotypic spectrum was observed, coupling with the heterogeneity observed in the transcriptomic data derived from osteoblasts of six patients from our cohort. Missense mutations were significantly associated (χ2p = 0.0096) with a cluster of patients with more severe clinical phenotypes. Additionally, the severity of OI was more correlated with the quality of bones, rather than the bone mineral density. Bone density is most responsive to bisphosphonate treatment during the juvenile stage (10–15 years old). In contrast, height is not responsive to bisphosphonate treatment. Our findings expand the mutational spectrum of type I collagen genes and the genotype-phenotype correlation in Chinese OI patients. The observation of effective bisphosphonate treatment in an age-specific manner may help to improve OI patient management.

Osteoarthritis (OA) is a major public health concern for which a reliable non-invasive treatment option has yet to be developed. In the present study, we investigated the effects of saxagliptin, a novel dipeptidyl peptidase IV (DPP-4) inhibitor, on several important aspects of the pathophysiology of OA using primary human chondrocytes. The results of real-time PCR and ELISA analyses show that saxagliptin treatment significantly decreased mRNA and protein expression of three key cartilage degrading enzymes: matrix metalloproteinase (MMP)-1, MMP-3, and MMP-13. The results of western blot confirmed that this decrease in MMP-1, -3, and -13 expression prevented degradation of type II collagen. We also found that saxagliptin significantly inhibited expression of a disintegrin and metalloproteinase with thrombospondin motif (ADAMTS)-4 and ADAMTS-5, which was reflected by markedly decreased degradation of aggrecan. Inhibition of DPP-4 by saxagliptin also reduced oxidative stress in human primary chondrocytes as evidenced by decreased production of reactive oxygen species (ROS) and increased glutathione (GSH) levels. Additionally, the results of western blot analysis show that the effects of saxagliptin are mediated through the p38/IκBα/NF-κB pathway, which is considered an important treatment target for OA. These findings suggest a potential role for saxagliptin as a novel treatment against OA.

sST2 has been shown to be a risk predictor in heart failure (HF). Our aim was to explore the characteristics and prognostic value of soluble ST2 (sST2) in hospitalized Chinese patients with HF.We consecutively enrolled 1528 hospitalized patients with HF. Receiver operating characteristic (ROC) and multivariable Cox proportional hazards analysis were used to assess the prognostic values of sST2. Adverse events were defined as all-cause death and cardiac transplantation. During a median follow-up of 19.1 months, 325 patients experienced adverse events. Compared with patients free of events, sST2 concentrations were significantly higher in patients with events (P55.6 ng/mL) independently predicted events in comparison to the lowest quartile (≤25.2 ng/mL) when adjusted by multivariable model. In ROC analysis, the area under the curve for sST2 was not different from that for NT-proBNP in short and longer term. Over time, sST2 also improved discrimination and reclassification of risk beyond NT-proBNP.sST2 is a strong independent risk predictor in Chinese patients hospitalized with HF and can significantly provide additional prognostic value to NT-proBNP in risk prediction.
