Abstract

Through a simulation-based study we develop a statistical model-based quantification method for atomic resolution first moment scanning transmission electron microscopy (STEM) images. This method uses the uniformly weighted least squares estimator to determine the unknown structure parameters of the images and to isolate contributions from individual atomic columns. In this way, a quantification of the projected potential per atomic column is achieved. Since the integrated projected potential of an atomic column scales linearly with the number of atoms it contains, it can serve as a basis for atom counting. The performance of atom counting from first moment STEM imaging is compared to that from traditional HAADF STEM in the presence of noise. Through this comparison, we demonstrate the advantage of first moment STEM images to attain more precise atom counts. Finally, we compare the integrated intensities extracted from first-moment images of a wedge-shaped sample to those values from the bulk crystal. The excellent agreement found between these values proves the robustness of using bulk crystal simulations as a reference library. Abstract

Established in 2012 by members of the Food and Agriculture Organisation (FAO), the Global Soil Partnership (GSP) is a global network of stakeholders promoting sound land and soil management practices towards a sustainable world food system. However, soil survey largely remains a local or regional activity, bound to heterogeneous methods and conventions. Recognising the relevance of global and trans-national policies towards sustainable land management practices, the GSP elected data harmonisation and exchange as one of its key lines of action. Building upon international standards and previous work towards a global soil data ontology, an improved domain model was eventually developed within the GSP [54], the basis for a Global Soil Information System (GloSIS). This work also identified the Semantic Web as a possible avenue to operationalise the domain model. This article presents the GloSIS web ontology, an implementation of the GloSIS domain model with the Web Ontology Language (OWL). Thoroughly employing a host of Semantic Web standards (SOSA, SKOS, GeoSPARQL, QUDT), GloSIS lays out not only a soil data ontology but also an extensive set of ready-to-use code-lists for soil description and physio-chemical analysis. Various examples are provided on the provision and use of GloSIS-compliant linked data, showcasing the contribution of this ontology to the discovery, exploration, integration and access of soil data.

Abstract

BackgroundBone and muscle are closely linked anatomically, biochemically, and metabolically. Acute exercise affects both bone and muscle, implying a crosstalk between the two systems. However, how these two systems communicate is still largely unknown. We will explore the role of undercarboxylated osteocalcin (ucOC) in this crosstalk. ucOC is involved in glucose metabolism and has a potential role in muscle maintenance and metabolism. ObjectiveThe proposed trial will determine if circulating ucOC levels in older adults at baseline and following acute exercise are associated with parameters of muscle function and if the ucOC response to exercise varies between older adults with low muscle quality and those with normal or high muscle quality. MethodsA total of 54 men and women aged 60 years or older with no history of diabetes and warfarin and vitamin K use will be recruited. Screening tests will be performed, including those for functional, anthropometric, and clinical presentation. On the basis of muscle quality, a combined equation of lean mass (leg appendicular skeletal muscle mass in kg) and strength (leg press; one-repetition maximum), participants will be stratified into a high or low muscle function group and randomized into the controlled crossover acute intervention. Three visits will be performed approximately 7 days apart, and acute aerobic exercise, acute resistance exercise, and a control session (rest) will be completed in any order. Our primary outcome for this study is the effect of acute exercise on ucOC in older adults with low muscle function and those with high muscle function. ResultsThe trial is active and ongoing. Recruitment began in February 2018, and 38 participants have completed the study as of May 26, 2019. ConclusionsThis study will provide novel insights into bone and muscle crosstalk in older adults, potentially identifying new clinical biomarkers and mechanistic targets for drug treatments for sarcopenia and other related musculoskeletal conditions.
