National audience; A l'état solide, les fluorophores à base d'oxazole tels que le 2-(p-tert-butylphenyl)benzoxazole (Bzx) et le 2,2'-di(p-tert-butylphenyl)-6,6'-bibenzoxazole (BBzx) présentent une émission dans le bleu profond/proche UV (380 nm-460 nm) avec des rendements de luminescence pouvant atteindre 38%. Ces deux molécules, qui diffèrent par leur planéité, leur encombrement stérique et la taille du système π-conjugué, ont été intégrées dans les diodes électroluminescentes organiques (OLED). Les OLEDs à base de BBzx ont montré une émission efficace dans le bleu profond, alors qu'aucune luminescence n'a été observée sur une couche mince de Bzx. La structure moléculaire en torsion du BBzx tend à réduire l'extinction de la fluorescence par agrégation moléculaire, favorisant ainsi son émission en couche mince. Des OLEDs à base de BBzx émettant à 430 nm avec des coordonnées CIE (0,157, 0,044) et un rendement de 1.2% (pour 24 A/m 2) ont ainsi été réalisées.

International audience; Oxazole-based fluorophores, such as 2,2’-di(p-tert-butylphenyl)-6,6’-bibenzoxazole (BBzx) derivatives, show efficient emission in the UV and deep blue spectral range in the solid state. The incorporation of the bischromophore BBzx within organic light emitting diodes (OLED) as emissive layer leds to efficient emission. OLEDs displayed a deep blue emission at 430 nm arising from the BBzx layer, with CIE coordinate of (0.157, 0.044). These specific optical features were attributed to the twisted molecular structure of the bis-chromophore, leading to reduction of the aggregation induced fluorescence quenching. An EQE of 1.2% was achieved with a driving current of 24 A/m2 . The results suggest that BBzx derivatives may be useful as efficient color saturation deep-blue emitters in OLEDs.

Background: Familial nontoxic multinodular goiter (MNG) is a rare disease. One of the associated genes is Kelch-like ECH-associated protein 1 (KEAP1), which encodes the main inhibitor of nuclear factor erythroid 2-related transcription factor 2 (Nrf2), a central mediator of antioxidant responses. The association of KEAP1 with familial MNG is based on only two loss-of-function mutations identified in two families, only one of which included proper phenotyping and adequate demonstration of co-segregation of the phenotype and the mutation. There is no experimental evidence from model organisms to support that decreased Keap1 levels can, indeed, cause goiter. This study used mice hypomorphic for Keap1 to test whether decreased Keap1 expression can cause goiter, and to characterize the activation status of Nrf2 in their thyroid. Methods: C57BL/6J Keap1flox/flox (Keap1 knock-down [Keap1KD]) mice were studied at 3 and 12 months of age. Plasma and thyroid glands were harvested for evaluation of thyroid function tests and for gene and protein expression by real-time polymerase chain reaction and immunoblotting, respectively. Results: Keap1KD mice showed diffuse goiter that began to develop in early adult life and became highly prominent and penetrant with age. The goiter was characterized by a markedly increased size of thyroid follicles, most notably of the colloid compartment, and by absence of thyroid nodules or hyperplasia. Keap1KD mice also showed decreased T4 levels in early adult life that were eventually well compensated over time by increased thyrotropin (TSH) levels. Nrf2 was activated in the thyroid of Keap1KD mice. Despite a known stimulatory effect of Nrf2 on thyroglobulin (Tg) gene transcription and Tg protein abundance, the expression levels were decreased in the thyroid of Keap1KD mice. No clear patterns were observed in the expression profiles of other thyroid hormone synthesis-specific factors, with the exception of Tg-processing and Tg-degrading cathepsins, including an increase in mature forms of cathepsins D, L, and S. Conclusions: Keap1KD mice develop age-dependent diffuse goiter with elevated TSH levels. The precise mechanism accounting for the thyroidal phenotype remains to be elucidated, but it may involve enhanced Tg solubilization and excessive lysosomal Tg degradation. [ABSTRACT FROM AUTHOR]

24. Identification of the nature of trapping centers in polyspirobifluorene based diodes by using electrical characterization.

The trap parameters in hole-only diodes using a blue emitting polyspirobifluorene copolymer were explored by using two complementary techniques such as: charge based deep level transient spectroscopy and thermally stimulated current. The trap analyses have been further completed by the measurements of the current-voltage characteristics as a function of the temperature. The use of unipolar structures allowed us to determine hole traps in the devices and further to identify electron traps in bipolar structures. Two hole traps which are located at 0.19 and 0.37 eV from the highest occupied molecular orbital, have been identified in blue polyspirobifluorene based devices. In bipolar diodes, four electron traps are determined and are located at 0.32, 0.58, 0.74, and 0.83 eV from the lowest unoccupied molecular orbital. We also showed that the use of a palladium electrode introduced new trap states of mean activation energy of 0.4 eV. The influence of aging by electrical stress on the formation of defects has been studied in two types of polymers: blue emitting spiro copolymer and white emitting spiro blend polymer. The white emitting spiro polymer was obtained by adding green and red chromophores into the host blue copolymer. The defect states were investigated by using two complementary techniques: charge based deep level transient spectroscopy and thermally stimulated current (TSC). Six trap levels were retrieved in blue emitting diodes with a depth of 0.17–0.85 eV, and the onset of an additional trap with a depth of 0.49 eV was observed in white emitting ones. The density of these traps is of the order of 1016–1017 cm-3. The fractional TSC experiments allowed us to determine the distribution of five trap types in white emitting diodes, which could be described by Gaussian functions. The aging of devices was performed by electrical stress and not by exposure of samples to air or oxygen, in contrast to previous investigations on other organic materials. Upon aging, no new traps have been created or suppressed in both doped and undoped polymers. Furthermore, an enhancement in trap density has been observed. The analysis of the trap characteristics has highlighted that the aging would not uniformly affect the defect levels in the polymer. The density of deeper traps (above 0.3 eV) is increased, and their distribution is also modified, while the shallow traps (below 0.3 eV) remain stable. Furthermore, the investigation of the doping influence on the degradation process has shown that the chromophore addition seems to stabilize the polymer, whose degradation rate is slower than that of the undoped material. The defect states in spiro copolymer based light emitting diodes were investigated by charge based deep level transient spectroscopy (Q-DLTS). Two types of polymers have been studied: blue emitting spiro copolymer and white emitting spiro blend polymer. The white emitting spiro polymer was obtained by adding green and red chromophores into the host blue copolymer. The devices are composed of indium-tin oxide–polyethylene dioxythiophene:polystyrene sulfonate-spiro copolymer-Ba–Al. Q-DLTS measurements were performed on these diodes with various conditions of charging time, charging voltage, and temperature aiming at determining the role of chromophores in the defect formation process. Analysis of the Q-DLTS spectra obtained in both devices revealed at least five trap levels. The mean activation energies of traps are distributed in the range 0.17–0.85 eV within the band gap of the copolymers with capture cross sections of the order of 10-16–10-20 cm2. The trap densities are in the range of 1015–1016 cm-3. The results show that incorporation of dyes into the copolymer resulted in creation of an additional electron trap level and an increase in the density of the existing trap levels, indicating a more disordered state of the emitting material containing chromophores [ABSTRACT FROM AUTHOR]

- Abstract
[Background]: The thyroid gland has a special relationship with oxidative stress. While generation of oxidative substances is part of normal iodide metabolism during thyroid hormone synthesis, the gland must also defend itself against excessive oxidation in order to maintain normal function. Antioxidant and detoxification enzymes aid thyroid cells to maintain homeostasis by ameliorating oxidative insults, including during exposure to excess iodide, but the factors that coordinate their expression with the cellular redox status are not known. The antioxidant response system comprising the ubiquitously expressed NFE2-related transcription factor 2 (Nrf2) and its redox-sensitive cytoplasmic inhibitor Kelch-like ECH-associated protein 1 (Keap1) defends tissues against oxidative stress, thereby protecting against pathologies that relate to DNA, protein, and/or lipid oxidative damage. Thus, it was hypothesized that Nrf2 should also have important roles in maintaining thyroid homeostasis., [Methods]: Ubiquitous and thyroid-specific male C57BL6J Nrf2 knockout (Nrf2-KO) mice were studied. Plasma and thyroids were harvested for evaluation of thyroid function tests by radioimmunoassays and of gene and protein expression by real-time polymerase chain reaction and immunoblotting, respectively. Nrf2-KO and Keap1-KO clones of the PCCL3 rat thyroid follicular cell line were generated using CRISPR/Cas9 technology and were used for gene and protein expression studies. Software-predicted Nrf2 binding sites on the thyroglobulin enhancer were validated by site-directed in vitro mutagenesis and chromatin immunoprecipitation., [Results]: The study shows that Nrf2 mediates antioxidant transcriptional responses in thyroid cells and protects the thyroid from oxidation induced by iodide overload. Surprisingly, it was also found that Nrf2 has a dramatic impact on both the basal abundance and the thyrotropin-inducible intrathyroidal abundance of thyroglobulin (Tg), the precursor protein of thyroid hormones. This effect is mediated by cell-autonomous regulation of Tg gene expression by Nrf2 via its direct binding to two evolutionarily conserved antioxidant response elements in an upstream enhancer. Yet, despite upregulating Tg levels, Nrf2 limits Tg iodination both under basal conditions and in response to excess iodide., [Conclusions]: Nrf2 exerts pleiotropic roles in the thyroid gland to couple cell stress defense mechanisms to iodide metabolism and the thyroid hormone synthesis machinery, both under basal conditions and in response to excess iodide.

Background: Several single-nucleotide polymorphisms (SNPs) are known to increase the risk of Hashimoto's thyroiditis (HT); such SNPs reside in thyroid-specific genes or in genes related to autoimmunity, inflammation, and/or cellular defense to stress. The transcription factor Nrf2, encoded by NFE2L2, is a master regulator of the cellular antioxidant response. This study aimed to evaluate the impact of genetic variation in NFE2L2 on the risk of developing HT. Methods: In a case–control candidate gene association study, functional SNPs in the NFE2L2 promoter (rs35652124, rs6706649, and rs6721961) were examined either as independent risk factors or in combination with a previously characterized HT risk allele (rs28665122) in the gene SELENOS, encoding selenoprotein S (SelS). A total of 997 individuals from the north of Portugal (Porto) were enrolled, comprising 481 HT patients and 516 unrelated healthy controls. SELENOS and NFE2L2 SNPs were genotyped using TaqMan
® assays and Sanger sequencing, respectively. Odds ratios (ORs) were calculated using logistic regression, with adjustment for sex and age. Expression of SelS was analyzed by immunohistochemistry in thyroid tissue from HT patients and control subjects. Molecular interactions between the Nrf2 and SelS pathways were investigated in thyroid tissues from mice and in rat PCCL3 thyroid follicular cells. Results: When all three NFE2L2 SNPs were considered together, the presence of one or more minor alleles was associated with a near-significant increased risk (OR = 1.43, p = 0.072). Among subjects harboring only major NFE2L2 alleles, there was no increased HT risk associated with heterozygosity or homozygosity for the SELENOS minor allele. Conversely, in subjects heterozygous or homozygous for the SELENOS risk allele, the presence of an NFE2L2 minor allele significantly increased HT risk by 2.8-fold (p = 0.003). Immunohistochemistry showed reduced expression of SelS in thyroid follicular cells of HT patients. In Nrf2 knockout mice, there was reduced expression of SelS in thyroid follicular cells; conversely, in PCCL3 cells, reducing SelS expression caused reduced activity of Nrf2 signaling. Conclusions: The NFE2L2 promoter genotype interacts with the SELENOS promoter genotype to modulate the risk of HT in a Portuguese population. International audience; For the past thirty years, modern ground-based time-series of the solar radius have shown different apparent variations according to different instruments. The origins of these variations may result from the observer, the instrument, the atmosphere, or the Sun. Solar radius measurements have been made for a very long time and in different ways. Yet we see inconsistencies in the measurements. Numerous studies of solar radius variation appear in the literature, but with conflicting results. These measurement differences are certainly related to instrumental effects or atmospheric effects. Use of different methods (determination of the solar radius), instruments, and effects of Earth's atmosphere could explain the lack of consistency on the past measurements. A survey of the solar radius has been initiated in 1975 by Francis Laclare, at the Calern site of the Observatoire de la Côte d'Azur (OCA). Several efforts are currently made from space missions to obtain accurate solar astrometric measurements, for example, to probe the long-term variations of solar radius, their link with solar irradiance variations, and their influence on the Earth climate. Aims. The Picard program includes a ground-based observatory consisting of different instruments based at the Calern site (OCA, France). This set of instruments has been named "Picard Sol" and consists of a Ritchey-Chrétien telescope providing full-disk images of the Sun in five narrow-wavelength bandpasses (centered on 393.37, 535.7, 607.1, 782.2, and 1025.0 nm), a Sun-photometer that measures the properties of atmospheric aerosol, a pyranometer for estimating a global sky-quality index, a wide-field camera that detects the location of clouds, and a generalized daytime seeing monitor allowing us to measure the spatio-temporal parameters of the local turbulence. Picard Sol is meant to perpetuate valuable historical series of the solar radius and to initiate new time-series, in particular during solar cycle 24. Methods. We defined the solar radius by the inflection-point position of the solar-limb profiles taken at different angular positions of the image. Our results were corrected for the effects of refraction and turbulence by numerical methods. Results. From a dataset of more than 20 000 observations carried out between 2011 and 2013, we find a solar radius of 959.78 ± 0.19 arcsec (696 113 ± 138 km) at 535.7 nm after making all necessary corrections. For the other wavelengths in the solar continuum, we derive very similar results. The solar radius observed with the Solar Diameter Imager and Surface Mapper II during the period 2011-2013 shows variations shorter than 50 milli-arcsec that are out of phase with solar activity.

Developed at the Observatoire de la C\^ote d'Azur (OCA) within the framework of the PICARD space mission (Thuillier et al., 2006) and with support from the french spatial agency (CNES), MISOLFA (Moniteur d'Images Solaires Franco-Alg\'erien) is a new generation of daytime turbulence monitor. Its objective is to measure both the spatial and temporal turbulence parameters in order to quantify their effects on the solar diameter measurements that will be made from ground using the qualification model of the SODISM (SOlar Diameter Imager and Surface Mapper) instrument onboard PICARD. The comparison of simultaneous images from ground and space should allow us, with the help of the solar monitor, to find the best procedure possible to measure solar diameter variations from ground on the long term. MISOLFA is now installed at the Calern facility of OCA and PICARD is scheduled to be launched in 2010. We present here the principles of the instrument and the first results obtained on the characteristics of the turbulence observed at Calern observatory using this monitor while waiting for the launch of the space mission., Comment: 4 pages, 10 figures
