The relationship between fishers (Pekania pennanti), fire, and fuel management in the Sierra Nevada region is complex and highly controversial, and the recent listing of southern Sierra Nevada fishers as endangered has brought this issue to the forefront for many land managers. While wildfire is a natural component of the historic disturbance regime in the dry, mixed conifer forests of California, it also has the capacity to destroy large swaths of suitable habitat and to fragment tenuous populations, particularly given recent shifts in wildfire scope and severity. Therefore, balancing fuel reduction with habitat conservation requires careful consideration of spatial and temporal context, cumulative effects, and management prescriptions. There are several unresolved questions pertaining to fishers, including how and when fishers begin to reuse postfire landscapes and whether this information can be applied to postfire efforts to restore habitat connectivity and limit negative impacts of population fragmentation. To better understand fisher response to postfire landscapes, and to provide land managers with guidance regarding habitat restoration efforts, we investigated fisher recolonization behavior in and around 2 recent Sierra Nevada wildfires. We hypothesized that fisher use of postfire landscapes would be driven by at least 1 of 4 primary variables: prefire habitat quality, fire severity, topography, and time since fire. We developed 5 hypotheses regarding how fishers would begin to explore postfire landscapes and tested them using a combination of fine- and coarse-scale analyses based on data collected using scat detection dogs. At fine scales, the strongest relationship we observed was the combination of fire severity and landscape curvature on fisher scat location. At coarser scales, fisher colonization rates reflected avoidance of areas dominated by higher-severity fire, and fishers had a higher probability of being found in areas with larger and more contiguous patches of low-severity fire. Our results suggest that fisher use of postfire landscapes may center on low-severity or unburned islands (i.e., fire refugia/fire skips) and on fine-scale topographic features associated with landscape concavity, such as ravines or topographic depressions. Efforts to promote a sustainable low- to mixed-severity fire regime that creates habitat heterogeneity and forest resiliency can support fisher conservation in this region, and restoration efforts that capitalize on connecting fire refugia using microtopography as a foundation may facilitate habitat connectivity for fishers more rapidly than traditional methods.

Selection of habitat characteristics by reproductive females during neonate development can mediate the influence of adverse environmental conditions on the fitness of offspring. Previous research has suggested that cavities and burrows used for reproduction by cavity-obligate species offer thermoregulatory benefits, access to prey, and can limit predation pressure. As fishers (Pekania pennanti) are secondary cavity-obligate breeders, we hypothesized that they select particular characteristics of reproductive den cavities at discrete stages of offspring development to mediate adverse biotic and environmental effects on their neonates. To test our hypothesis, we located 406 reproductive dens and 154 cavity rest sites used by 65 individual adult female fishers during 11 reproductive seasons (2005–2016) in northwestern California. We counted 53 (27 F, 26 M) kits in 31 litters born to 19 females during six of these reproductive seasons. The weight of kits varied significantly by sex and by age, whereas the length of kits varied only by age, suggesting that adult females in this population might be preferentially investing in male kits. We found that natal and early-maternal dens buffered minimum temperatures significantly more than late-maternal dens and cavities used during the nonreproductive season. A male fisher skull was also less likely to fit through the cavity openings of natal dens than through the openings of cavities used by adult females during the nonreproductive season. Litter survival was significantly lower at natal dens than at late-maternal dens. The age of adult female fishers did not affect the probability of litter survival. Our results emphasize the vulnerability of vertebrate offspring during early developmental periods and how cavity-obligate species select cavities to mediate environmental conditions during reproduction.

Many wildlife species rely on unique features of trees during daily activities and fundamental parts of their life cycle. The fisher (Pekania pennanti) is a forest-dwelling carnivore of conservation concern in western North America that uses unique habitat elements as refuges during resting bouts and for reproduction. Prior to this study, little was known about the fine-scale habitat used during reproduction at the southernmost extent of the fisher’s range. Between 2007 and 2015, we attached radio-collars to 55 male and 72 female fishers in the southern Sierra Nevada and documented resting locations of males on 216 occasions (196 structures) and females on 824 occasions (737 structures). Beginning in 2008, we also monitored females over 8 reproductive seasons, confirming 45 females at dens and documenting 95 natal dens (83 structures) and 206 maternal dens (192 structures). We established 3 comparisons to guide our assessment of fine-scale habitat: resting males versus resting females, natal dens versus maternal dens, and resting fishers (both sexes) versus denning fishers (all dens). We expected the need for physical security and thermal protection in combination with morphology, predation risk, and aspects of reproductive ecology would influence patterns of use. Both sexes used a variety of microsites for resting, but females selected tree cavities most frequently (47%) while males used branch platforms most often (39%). For resting structures, live conifers were used most often by both sexes (males 44%, females 34%), but live hardwoods (males 16%, females 28%) and conifer snags (males 16%, females 22%) were also important. Comparing natal and maternal dens, we found that cavity microsites used early in the den season tended to be higher than those used later, and large live hardwoods comprised roughly half of all natal (46%) and maternal (51%) den structures. For resting versus denning, we found that large diameter hardwoods were an important source of cavities for both activities, live conifers used for denning were larger than those used for resting, and den structures tended to be on steeper slopes than rest structures. White fir (Abies concolor), California black oak (Quercus kelloggii), and ponderosa pine (Pinus ponderosa) were selected most often by both sexes for resting. In contrast, denning females relied on California black oak (55%), but also used white fir (24%) and incense cedar (Calocedrus decurrens; 12%). As noted in studies further north, our findings highlight the value of large trees with decay to support fisher reproduction and daily refugia.

Quantifying reproductive parameters is essential for developing effective conservation plans for species of concern; however, studying reproduction in wild settings can be challenging and local data are frequently unavailable. The fisher (Pekania pennanti) is an elusive forest-dwelling carnivore of conservation concern that occurs across much of boreal North America and reaches the southernmost limit of its distribution in the southern Sierra Nevada (SSN) in California. Data on fisher reproduction in this region are limited and applicability of parameters from other areas is uncertain. To clarify how fisher populations in the SSN compare with those elsewhere in North America, we conducted a comprehensive review of available literature on reproduction throughout the species' range in North America. We then compared findings from the review with data we collected during 7 years in the SSN, focusing on the proportion of females reproducing, litter size, and the parturition date. On average across the fisher's range, the proportion of adult females that reproduced was 0.71 (range: 0.40-1.00; n = 16 studies), litter size was 2.5 (range: 1-4; n = 16 studies), and parturition occurred on 25 March (range: 3 March-17 April; n = 16 studies). In our study area, we tracked 42 adult female fishers, 35 of which reproduced and used 257 reproductive dens (74 natal, 175 maternal, 8 early failures); 0.86 of our females attempted denning (range: 0.79-1.0; n = 93 opportunities) and 0.75 were successful (range: 0.64-1.0; n = 91 opportunities monitored through the den season). Mean litter size was 1.57 (range: 1-3; n = 75 litters) and mean parturition date was 30 March (range: 17 March-12 April; n = 69 natal dens initiated). Female fishers at the southern limit of their distribution in the SSN reproduced at a rate comparable to or higher than elsewhere in North America, but average litter size was the lowest reported for the species. Female fishers in the SSN gave birth at similar or later dates than elsewhere in their range. We explore factors that might explain patterns of variation in fisher reproductive parameters and discuss conservation implications of our findings.

ObjectivesTo assess whether plasma biomarkers of oxidative stress predict diffusion-perfusion mismatch in patients with acute ischemic stroke (AIS).MethodsWe measured plasma levels of oxidative stress biomarkers such as F2-isoprostanes (F2-isoPs), total and perchloric acid Oxygen Radical Absorbance Capacity (ORACTOT and ORACPCA), urinary levels of 8-oxo-7,8-dihydro-2′-deoxyguoanosine, and inflammatory and tissue-damage biomarkers (high-sensitivity C-reactive protein, matrix metalloproteinase-2 and -9) in a prospective study of patients with AIS presenting within 9 hours of symptom onset. Diffusion-weighted (DWI) and perfusion-weighted (PWI) MRI sequences were analyzed with a semiautomated volumetric method. Mismatch was defined as baseline mean transit time volume minus DWI volume. A percent mismatch cutoff of >20% was considered clinically significant. A stricter definition of mismatch was also used. Mismatch salvage was the region free of overlap by final infarction.ResultsMismatch >20% was present in 153 of 216 (70.8%) patients (mean [±SD] age 69.2 ± 14.3 years, 41.2% women). Patients with mismatch >20% were more likely to have higher baseline plasma levels of ORACPCA (p = 0.020) and F2-isoPs (p = 0.145). Multivariate binary logistic regression demonstrated that lnF2-isoP (odds ratio [OR] 2.44, 95% confidence interval [CI] 1.19–4.98, p = 0.014) and lnORACPCA (OR 4.18, 95% CI 1.41–12.41, p = 0.010) were independent predictors of >20% PWI-DWI mismatch and the stricter mismatch definition, respectively. lnORACTOT significantly predicted mismatch salvage volume (>20% mismatch p = 0.010, stricter mismatch definition p = 0.003).ConclusionsElevated hyperacute plasma levels of F2-isoP and ORAC are associated with radiographic evidence of mismatch and mismatch salvage in patients with AIS. If validated, these findings may add to our understanding of the role of oxidative stress in cerebral tissue fate during acute ischemia.

Background and Purpose— Oxidative stress is an early response to cerebral ischemia and is likely to play an important role in the pathogenesis of cerebral ischemic injury. We sought to evaluate whether hyperacute plasma concentrations of biomarkers of oxidative stress, inflammation, and tissue damage predict infarct growth (IG). Methods— We prospectively measured plasma F2-isoprostane (F2-isoP), urinary 8-oxo-7,8-dihydro-2′-deoxyguoanosine, plasma oxygen radical absorbance capacity assay, high sensitivity C reactive protein, and matrix metalloproteinase 2 and 9 in consecutive patients with acute ischemic stroke presenting within 9 hours of symptom onset. Patients with baseline diffusion-weighted magnetic resonance imaging and follow-up diffusion-weighted imaging or computed tomographic scan were included to evaluate the final infarct volume. Baseline diffusion-weighted imaging volume and final infarct volume were analyzed using semiautomated volumetric method. IG volume was defined as the difference between final infarct volume and baseline diffusion-weighted imaging volume. Results— A total of 220 acute ischemic stroke subjects were included in the final analysis. One hundred seventy of these had IG. Baseline F2-isoP significantly correlated with IG volume (Spearman ρ=0.20; P =0.005) and final infarct volume (Spearman ρ=0.19; P =0.009). In a multivariate binary logistic regression model, baseline F2-isoP emerged as an independent predictor of the occurrence of IG (odds ratio, 2.57; 95% confidence interval, 1.37–4.83; P =0.007). In a multivariate linear regression model, baseline F2-isoP was independently associated with IG volume (B, 0.38; 95% confidence interval, 0.04–0.72; P =0.03). Conclusions— Elevated hyperacute plasma F2-isoP concentrations independently predict the occurrence of IG and IG volume in patients with acute ischemic stroke. If validated in future studies, measuring plasma F2-isoP might be helpful in the acute setting to stratify patients with acute ischemic stroke for relative severity of ischemic injury and expected progression.

Interspecific killing is common among carnivores and can have population-level effects on imperiled species. The fisher (Pekania [Martes] pennanti) is a rare forest carnivore in western North America and a candidate for listing under the United States Endangered Species Act. Interspecific killing and intraguild predation are poorly understood in fishers and potential threats to existing western populations. We studied the prevalence and patterns of interspecific killing of fishers in the southern Sierra Nevada and Coastal Range of California. We collected forensic evidence and samples from the carcasses and predation sites, conducted full necropsies when possible, and used molecular methods to determine species of predators responsible for killing fishers. We recovered 101 (59 female, 42 male) fisher carcasses; for 62 (61%) carcasses, we attributed cause of death to interspecific killing. We found that bobcats (Lynx rufus, n = 25 fisher mortalities), mountain lions (Puma concolor, n = 20), and coyotes (Canis latrans, n = 4) were predators of fishers in our study areas. Bobcats killed only female fishers, whereas mountain lions more frequently killed male than female fishers, confirming our hypothesis that female fishers would suffer lethal attacks by smaller predators than would male fishers. Coyotes rarely killed fishers. We found differences in pathologic characteristics of the predation events among the 3 predator species, which may be helpful in identifying predator species. © 2014 The Wildlife Society.

Robust scientific information is essential to proper leasing decisions in offshore waters of the United States. This information develops the necessary understanding of the environment required to protect ecosystems during sustainable energy development offshore. Collection of existing scientific information can be a laborious and time-consuming process. In an effort to collate and evaluate scientific information in a more effective and timely manner, the Bureau of Ocean Energy Management (BOEM) created a system that would improve the speed with which environmental research is performed while maintaining scientific defensibility of the resulting decisions. The EcoSpatial Information Database (ESID, pronounced “ee-sid”), available at http://esid.boem.gov , makes relevant scientific literature and ecological data for the Outer Continental Shelf (OCS) of the U.S. Atlantic Coast readily accessible via advanced location and content data searches. A unique search protocol was developed to identify and scientifically screen thousands of scientific articles and environmental reports to identify scientific datasets in the disciplines of marine geology, water quality, pelagic ecology, and benthic ecology. A prioritization process culled the results to a total of 3108 resources spanning the years of 1884 to 2010. These resources were then geospatially referenced and incorporated into the ESID. A web-based Geographic Information System (GIS), hosted in the Amazon Elastic Compute Cloud (EC2), was developed to provide broad access to the data and supporting documents. The cloud-based ESID Web Application allows users to search by content and location, view citations and abstracts, export bibliographic entries and view and download documents. The system is designed to support virtually unlimited geographic and subject matter expansion and will streamline BOEM's efforts to produce the required National Environmental Policy Act documents in a more efficient manner.

Despite many advances in symptom management, children and adolescents with cancer still have trouble maintaining adequate oral intake during routine chemotherapy treatment. The purpose of this qualitative study was to explore the eating experiences of children and adolescents receiving chemotherapy when they had problems with nausea and mucositis. Eight children and adolescents and their caregivers were interviewed to describe how and what the children and adolescents ate when they were nauseated and/or had a sore mouth. Findings reveal that these children and adolescents all experienced nausea and frequently preferred not to eat during these periods. Eating problems related to mucositis also limited oral intake in this sample. These children and adolescents and their caregivers tried a variety of foods and strategies to maintain intake, including those recommended by health care providers. Prevention and management of nausea remains a challenge for children and adolescents receiving chemotherapy. Health care providers need to offer detailed eating suggestions throughout therapy so that these patients can maintain adequate nutrition and weight for optimal treatment tolerance as well as normal growth and development. Continued research is needed to test the effectiveness of interventions focused on maintaining oral intake during cancer treatment.

The Deepwater Horizon (DWH) oil blowout in the Gulf of Mexico (GoM) led to the largest offshore oil spill in U.S. history. The accident resulted in oil slicks that covered between 10,000 and upward of 40,000 km2 of the Gulf between April and July 2010. Quantifying the actual spatial extent of oil over such synoptic scales on an operational basis and, in particular, estimating the oil volume (or slick thickness) of large oil slicks on the ocean surface has proven to be a challenge to researchers and responders alike. This challenge must be addressed to assess and understand impacts on marine and coastal resources and to prepare a response to future spills. We estimated surface oil volume and probability of occurrence of different oil thicknesses during the DWH blowout in the GoM by combining synoptic measurements (2330-km swath) from the satellite-borne NASA Moderate Resolution Imaging Spectroradiometer (MODIS) and near-concurrent, much narrower swath (∼5 km) hyperspectral observations from the NASA Airborne Visible/Infrared Imaging Spectrometer (AVIRIS). A histogram-matching approach was used to transfer AVIRIS-derived oil volume to MODIS pixel-scale dimensions, after masking clouds under both sun glint and nonglint conditions. Probability functions were used to apply the transformation to 19 MODIS images collected during the DWH event. This generated three types of MODIS oil maps: maps of surface oil volume, maps of relative oil thickness with four different classes (i.e., 0 μm, 8 μm), and maps of probability distributions of different thicknesses. The results were compared with satellite-based synthetic aperture radar measurements and evaluated with concurrent aerial photographs. Although the methods may not be ideal and the results may contain large uncertainties, the current attempt suggests that coarse-resolution optical remote sensing observations can provide estimates of relative oil thickness/volume for large oil slicks captured by satellites.

Increases in nitrate loading to the Mississippi River watershed during the last 50 years are considered responsible for the increase in hypoxic zone size in Louisiana-Texas shelf bottom waters. There is currently a national mandate to decrease the size of the hypoxic zone to 5000 km2 by 2015, mostly by a 30% reduction in annual nitrogen discharge into the Gulf of Mexico. We developed an ecosystem model for the Mississippi River plume to investigate the response of organic matter production and sedimentation to variable nitrate loading. The nitrogen-based model consisted of nine compartments (nitrate, ammonium, labile dissolved organic nitrogen, bacteria, small phytoplankton, diatoms, micro- and mesozooplankton, and detritus), and was developed for the spring season, when sedimentation of organic matter from plume surface waters is considered important in the development of shelf hypoxia. The model was forced by physical parameters specified along the river-ocean salinity gradient, including residence time, light attenuation by dissolved and particulate matter, mixed layer depth, and dilution. The model was developed using measurements of biological biomasses and nutrient concentrations across the salinity gradient, and model validation was performed with an independent dataset of primary production measurements for different riverine NO3 loads. Based on simulations over the range of observed springtime NO3 loads, small phytoplankton contributed on average 80% to primary production for intermediate to high salinities (>15), and the main contributors to modeled sedimentation at these salinities were diatom sinking, microzooplankton egestion, and small phytoplankton mortality. We investigated the impact of limiting factors on the relationship between NO3 loading and ecosystem rates. Model results showed that primary production was primarily limited by physical dilution of NO3, followed by abiotic light attenuation, light attenuation due to mixing, and diatom sinking. Sedimentation was mainly limited by the first three of these factors. Neither zooplankton grazing or plume residence times acted as limiting factors of ecosystem rates. Regarding nutrient reductions to the watershed, simulations showed that about half of the percent decrease in NO3 load was reflected in decreased plume sedimentation. For example, a 30% decrease in NO3 load resulted in a 19% decrease in average plume primary production and a 14% decrease in sedimentation. Finally, our model results indicated that the fraction of primary production exported from surface waters is highly variable with salinity (7–87%), a finding which has important implications for predictive models of hypoxic zone size that assume a constant value for this ratio.

We developed statistically-based, optical models to estimate tripton (sediment/detrital) and colored dissolved organic matter (CDOM) absorption coefficients (asd, ag) from physical hydrographic and atmospheric properties. The models were developed for northern Gulf of Mexico shelf waters using multi-year satellite and physical data. First, empirical algorithms for satellite-derived asd and ag were developed, based on comparison with a large data set of cruise measurements from northern Gulf shelf waters; these algorithms were then applied to a time series of ocean color (SeaWiFS) satellite imagery for 2002–2005. Unique seasonal timing was observed in satellite-derived optical properties, with asd peaking most often in fall/winter on the shelf, in contrast to summertime peaks observed in ag. Next, the satellite-derived values were coupled with the physical data to form multiple regression models. A suite of physical forcing variables were tested for inclusion in the models: discharge from the Mississippi River and Mobile Bay, Alabama; gridded fields for winds, precipitation, solar radiation, sea surface temperature and height (SST, SSH); and modeled surface salinity and currents (Navy Coastal Ocean Model, NCOM). For satellite-derived asd and ag time series (2002–2004), correlation and stepwise regression analyses revealed the most important physical forcing variables. Over our region of interest, the best predictors of tripton absorption were wind speed, river discharge, and SST, whereas dissolved absorption was best predicted by east–west wind speed, river discharge, and river discharge lagged by 1 month. These results suggest the importance of vertical mixing (as a function of winds and thermal stratification) in controlling asd distribution patterns over large regions of the shelf, in comparison to advection as the most important control on ag. The multiple linear regression models for estimating asd and ag were applied on a pixel-by-pixel basis and results were compared to monthly SeaWiFS composite imagery. The models performed well in resolving seasonal and interannual optical variability in model development years (2002–2004) (mean error of 32% for asd and 29% for ag) and in predicting shelfwide optical patterns in a year independent of model development (2005; mean error of 41% for asd and 46% for ag). The models provide insight into the dominant processes controlling optical distributions in this region, and they can be used to predict the optical fields from the physical properties at monthly timescales.

Nutrients from the Mississippi/Atchafalaya Rivers greatly stimulate biological production in the ‘classical’ food web on the inner shelf of the northern Gulf of Mexico. Portions of this production, especially large diatoms and zooplankton fecal pellets, sink and decompose in the bottom water, consuming oxygen and contributing to the annual development of an extensive zone of bottom water hypoxia, typically >15,000 km 2 since 1993. The microbial food web is also active in the Mississippi River plume, but consists of small organisms that sink slowly. This ‘recycling’ food web has not been considered as a significant contributor to vertical flux and hypoxia. However, gelatinous zooplankton, especially pelagic appendicularians such as Oikopleura dioica , mediate the conversion of microbial web organisms to organic particles with high sinking rates. When pelagic appendicularians are abundant in coastal regions of the northern Gulf of Mexico, they stimulate the rapid vertical transfer of microbial web productivity in the surface layer, which is only 5–15 m thick in the coastal hypoxic region, to the sub-pycnocline layer that becomes hypoxic each summer. In this paper we present results from two studies examining the significance of this pathway. In both 2002 and 2004, we observed high production rates of appendicularians in coastal waters. Discarded gelatinous houses and fecal pellets from the appendicularian populations often provided more than 1 g m −2 d −1 of organic carbon for the establishment and maintenance of hypoxia in the northern Gulf of Mexico. This source of organic matter flux is especially important in regions far from the river plumes and during periods of low river discharge. Autotrophic elements of this food web are primarily supported by recycled inorganic nutrients originating in the Mississippi and Atchafalaya Rivers. Sources of dissolved organic matter (DOM) supporting the heterotrophic components of this microbial food web may include in situ production, the Mississippi/Atchafalaya Rivers, and Louisiana's coastal wetlands. If significant, the latter source provides a possible link between Louisiana's high rates of coastal land loss and the large hypoxic zone observed along the coast during summer. Both of the latter DOM sources are independent of phytoplankton production stimulated by inputs of riverine inorganic nutrients.

In this review, we use data from field measurements of biogeochemical processes and cycles in the Mississippi River plume and in other shelf regions of the northern Gulf of Mexico to determine plume contributions to coastal hypoxia. We briefly review pertinent findings from these process studies, review recent mechanistic models that synthesize these processes to address hypoxia-related issues, and reinterpret current understanding in the context of these mechanistic models. Some of our conclusions are that both nitrogen and phosphorus are sometimes limiting to phytoplankton growth; respiration is the main fate of fixed carbon in the plume, implying that recycling is the main fate of nitrogen; decreasing the river nitrate loading results in less than a 1:1 decrease in organic matter sinking from the plume; and sedimenting organic matter from the Mississippi River plume can only fuel about 23% of observed coastal hypoxia, suggesting significant contributions from the Atchafalaya River and, possibly, coastal wetlands. We also identify gaps in our knowledge about controls on hypoxia, and indicate that some reinterpretation of our basic assumptions about this system is required. There are clear needs for improved information on the sources, rates, and locations of organic matter sedimentation; for further investigation of internal biogeochemical processes and cycling; for improved understanding of the rates of oxygen diffusion across the pycnocline; for identification and quantification of other sources of organic matter fueling hypoxia or other mechanisms by which Mississippi River derived organic matter fuels hypoxia; and for the development of a fully coupled physical-biogeochemical model.

Variability in upper ocean optical properties is often driven by changes in the particle pool. We investigated the effects of such changes by characterizing individual particles. For particles in natural assemblages, we used a combination of Mie theory and flow cytometry to determine diameter ( D), complex refractive index (n 1 in9), and optical cross-sections at 488 nm. Particles were grouped into categories of eukaryotic pico/nanophytoplankton, Synechococcus, heterotrophic prokaryotes, detritus, and minerals to interpret variability in concurrently measured bulk inherent optical properties (IOPs) in New England continental shelf waters during two seasons. The summed contributions of individual particles to phytoplankton absorption and particle scattering were close to values for these properties measured independently using bulk methods (87% and 107%, respectively). In surface waters during both seasons, eukaryotic phytoplankton were responsible for the majority of both total particle absorption and total particle scattering. Mineral particles contributed the most to backscattering ( bb) in the spring, whereas in the summer both mineral and detrital particles were important. Synechococcus and heterotrophic prokaryotes never contributed more than 14% to IOPs. Our findings emphasize that the measurement of nonliving particles, including detritus and minerals, is necessary for understanding variability in bb in the ocean, an important quantity in the interpretation of satellite ocean color. Knowledge of particle properties is important for the interpretation of variability in the inherent optical properties (IOPs; Table 1) of the upper ocean. Models of particle IOPs will be the basis for improved algorithms for light attenuation and for deriving properties such as chlorophyll concentration from satellite ocean color. Results of previous model simulations have shown how changes in particle type can give significantly different IOPs, even at the same chlorophyll concentration (Mobley and Stramski 1997; Stramski et al. 2001). These simulations were based on assumptions about the types of particles present and the distributions of diameter (D) and complex refractive index (n 1 in9) for each particle group included in the model. Certain of these particle properties are difficult to measure, especially distributions of n and n9. By definition, n governs scattering at interfaces and n9 is related to the absorption properties of a particle. Inferences of average n for a particle assemblage have been made by a variety of optical methods, in conjunction with Mie theory (e.g., Zaneveld et al. 1974; Morel and Bricaud 1986; Stramski and Morel 1990; Twardowski et al. 2001), but detailed distributions of individual particle properties (i.e., within-group variations) cannot be determined from bulk optical approaches. Micromonas pusilla (Butcher) Manton et Parke, a marine prasinophyte, was used to investigate how cell growth and division affect optical properties of phytoplankton over the light:dark cycle. Measurements were made of cell size and concentration, attenuation and absorption coefficients, flow cytometric forward and side light scattering and chl fluorescence, and chl and carbon content. The refractive index was derived from observations and Mie scattering theory. Diel variations occurred, with cells increasing in size, light scattering, and carbon content during daytime photosynthesis and decreasing during nighttime division. Cells averaged 1.6 � m in diameter and exhibited phased division, with 1.3 divisions per day. Scattering changes resulted primarily from changes in cell size and not refractive index; absorption changes were consistent with a negligible package effect. Measurements over the diel cycle suggest that in M. pusilla carbon-specific attenuation varies with cell size, and this relationship appears to extend to other phytoplankton species. Because M. pusilla is one of the smallest eukaryotic phytoplankton and belongs to a common marine genus, these results will be useful for interpreting in situ light scattering variation. The relationship between forward light scattering (FLS) and volume over the diel cycle for M. pusilla was similar to that determined for a variety of phytoplankton species over a large size range. We propose a method to estimate cellular carbon content directly from FLS, which will improve our estimates of the contribution of different phytoplankton groups to productivity and total carbon content in the oceans.

Profiling floats equipped with bio-optical sensors well complement ship-based and satellite ocean color measurements by providing highly-resolved time-series data on the vertical structure of biogeochemical processes in oceanic waters. This is the first study to employ an autonomous profiling (APEX) float in the Gulf of Mexico for measuring spatiotemporal variability in bio-optics and hydrography. During the 17-month deployment (July 2011 to December 2012), the float mission collected profiles of temperature, salinity, chlorophyll fluorescence, particulate backscattering (bbp), and colored dissolved organic matter (CDOM) fluorescence from the ocean surface to a depth of 1,500 m. Biogeochemical variability was characterized by distinct depth trends and local "hot spots", including impacts from mesoscale processes associated with each of the water masses sampled, from ambient deep waters over the Florida Plain, into the Loop Current, up the Florida Canyon, and eventually into the Florida Straits. A deep chlorophyll maximum (DCM) occurred between 30 and 120 m, with the DCM depth significantly related to the unique density layer ρ = 1023.6 (R2 = 0.62). Particulate backscattering, bbp, demonstrated multiple peaks throughout the water column, including from phytoplankton, deep scattering layers, and resuspension. The bio-optical relationship developed between bbp and chlorophyll (R2 = 0.49) was compared to a global relationship and could significantly improve regional ocean-color algorithms. Photooxidation and autochthonous production contributed to CDOM distributions in the upper water column, whereas in deep water, CDOM behaved as a semi-conservative tracer of water masses, demonstrating a tight relationship with density (R2 = 0.87). In the wake of the Deepwater Horizon oil spill, this research lends support to the use of autonomous drifting profilers as a powerful tool for consideration in the design of an expanded and integrated observing network for the Gulf of Mexico.

Relationships between optical and physical properties were examined on the basis of intensive sampling at a site on the New England continental shelf during late summer 1996 and spring 1997. During both seasons, particles were found to be the primary source of temporal and vertical variability in optical properties since light absorption by dissolved material, though significant in magnitude, was relatively constant. Within the particle pool, changes in phytoplankton were responsible for much of the observed optical variability. Physical processes associated with characteristic seasonal patterns in stratification and mixing contributed to optical variability mostly through effects on phytoplankton. An exception to this generalization occurred during summer as the passage of a hurricane led to a breakdown in stratification and substantial resuspension of nonphytoplankton particulate material. Prior to the hurricane, conditions in summer were highly stratified with subsurface maxima in absorption and scattering coefficients. In spring, stratification was much weaker but increased over the sampling period, and a modest phytoplankton bloom caused surface layer maxima in absorption and scattering coefficients. These seasonal differences in the vertical distribution of inherent optical properties were evident in surface reflectance spectra, which were elevated and shifted toward blue wavelengths in the summer. Some seasonal differences in optical properties, including reflectance spectra, suggest that a significant shift toward a smaller particle size distribution occurred in summer. Shorter timescale optical variability was consistent with a variety of influences including episodic events such as the hurricane, physical processes associated with shelfbreak frontal dynamics, biological processes such as phytoplankton growth, and horizontal patchiness combined with water mass advection.

In this paper we use examples of Synthetic Aperture Radar (SAR) imagery collected during the Deepwater Horizon (DWH) oil spill and the Texture Classifier Neural Network Algorithm (TCNNA) to identify SAR image signatures that correspond to regions of emulsified (thicker) oil, which were verified by sea level observations and other remote sensing instruments. The method is sensitive to the SAR incident angles. L-band SAR was found to have the largest window of incidence angles (between 16 and 38 degrees off-nadir angle) that were able to detect Oil Emulsions (OE). C-band SAR were found to have a narrower OE detectable window (between 18 to 32 degrees off-nadir angle) than L-band. The X-band SAR had the narrowest OE detectable window (between 20 to 31 degrees off-nadir angle).

Objectives: Oxidative stress occurs as part of the earliest response to ischemia and is likely to play an important role in the pathogenesis of ischemic cerebral injury. We sought to evaluate whether hyperacute plasma concentrations of F2-isoprostane (F2-isoP), a biomarker of lipid peroxidation, predict infarct growth (IG), as a mediator of brain damage. Methods: We prospectively measured plasma F2-isoP, high sensitivity-C reactive protein, matrix metalloproteinase-2 and -9, Oxygen Radical Absorbance Capacity assay, and urinary 8-hydroxydeoxyguanosine in acute ischemic stroke (AIS) patients presenting within 9 h of symptom onset. Patients with a baseline DWI MRI and a 48-h DWI or CT scan were included to evaluate the final infarct volume (FIV). Baseline DWIV and FIV were analyzed using semi-automated volumetric method. IG was defined as the difference between FIV and baseline DWIV. Results: Of 489 AIS subjects, 220 [mean (SD) age 70.1 (15.1) y; 43.4% female; 40.5% treated with IV tPA] had both baseline DWI and follow-up imaging. Of these, 170 had IG of any size, and they were more likely to be male (p=0.032), to have prior transient ischemic attack (p=0.035) or diabetes mellitus (p=0.026), higher NIHSS score (p=0.001) and blood glucose levels (p=0.003) at admission, and higher median (IQR) baseline levels of F2-isoP [56.2 (36.4-75.3) vs. 42.0 (27.0-61.8) pg/mL, p=0.009]. Baseline F2-isoP and FIV (Spearman’s rho=0.19, p=0.009) as well as absolute IG volume (IGV) (Spearman’s rho=0.20, p=0.005) were significantly correlated. In a multivariable logistic regression model including all variables associated with IG at univariate p-value Conclusions: Elevated hyperacute plasma F2-isoP concentrations independently predict IG and IGV in AIS patients. If validated in future studies, measuring plasma F2-isoP might be helpful in the acute setting to stratify AIS patients for progression and relative severity of ischemic injury.

Objectives: Identifying underlying stroke etiology plays critical role in acute management and secondary prevention strategies. Despite advanced diagnostics, up to 35% of ischemic strokes (IS) remain undetermined. We sought to evaluate the association between IS subtypes and plasma levels of biomarkers as representative of distinct etiologies underlying stroke subtypes. Methods: We prospectively measured plasma high sensitivity-C reactive protein (hs-CRP), matrix metalloproteinase (MMP)-2 and MMP-9, F2-isoprostane (F2-isoP), Oxygen Radical Absorbance Capacity assay (ORAC), homocysteine and urinary 8-hydroxydeoxyguanosine (8OHdG), in patients presenting within 9 hours from acute ischemic stroke (AIS) onset. Stroke subtype was assigned by a stroke neurologist according to the Causative Classification of Stroke System (CCSS) criteria. Results: There were 489 AIS subjects enrolled in this study [mean (SD) age 70.1 (15.1) y; 43.4% female; median (IQR) NIHSS 6 (3-13); 40.7% received IV tPA]. Among these AIS, 51% were of cardioembolic (CE), 17% of atherothrombotic, 7% of lacunar, 5% of other, and 20% of undetermined subtype. Patients with CE stroke were older and more likely to be female, non-smokers, have atrial fibrillation and congestive heart failure, but not carotid stenosis (all p Conclusions: In AIS patients, hyperacute plasma biomarkers are associated with specific stroke subtypes, reflective of the underlying pathophysiology. If validated in future studies, plasma biomarkers may play valuable role in early diagnosis and guide management of specific AIS subtypes.

We describe emerging capabilities to understand physical processes and biogeochemical cycles in coastal waters through the use of satellites, numerical models, and ship observations. Emerging capabilities provide significantly improved ability to model ecological systems and the impact of environmental management actions on them. The complex interaction of physical and biogeochemical processes responsible for hypoxic events requires an integrated approach to research, monitoring, and modeling in order to fully define the processes leading to hypoxia. Our effort characterizes the carbon cycle associated with river plumes and the export of organic matter and nutrients from coastal Louisiana wetlands and embayments in a spatially and temporally intensive manner previously not possible. Riverine nutrients clearly affect ecosystems in the northern Gulf of Mexico as evidenced in the occurrence of regional hypoxia events. Less known and largely unquantified is the export of organic matter and nutrients from the large areas of disappearing coastal wetlands and large embayments adjacent to the Louisiana Continental Shelf. This project provides new methods to track the river plume along the shelf and to estimate the rate of export of suspended inorganic and organic particulate matter and dissolved organic matter from coastal habitats of south Louisiana.

[1] We investigated environmental forcing mechanisms of phytoplankton absorption near the Mississippi River delta using multiyear satellite data. An algorithm for the phytoplankton absorption coefficient (aph) was developed from in situ measurements and applied to ocean color imagery. We employed a suite of chemical and physical forcing variables, including surface currents. For satellite-derived aph time series (2002–2004), correlation and stepwise regression analyses revealed the most important forcing variables of aph on the Louisiana shelf. Areally, Mississippi River discharge and nitrate concentration ([NO3]) were the two most important predictors of aph over the hypoxic zone (defined by its maximum extent). River discharge was important in a band stretching from the Mississippi River delta to the Louisiana-Texas border. Riverine [NO3] and wind magnitude best predicted aph in nearshore waters, and solar radiation and SST were most important farther offshore over the hypoxic zone, indicating upwelled nutrient sources to phytoplankton. A multiple linear regression model performed well in resolving seasonal and interannual aph variability in model development years (2002–2004) (mean error of 18%, over all pixels and months) and in predicting shelf-wide aph patterns in 2005 (mean error of 32%). Our results strongly suggest that in recent years, stratification and vertical mixing, in addition to riverine [NO3], play a primary role in regulating phytoplankton biomass over the hypoxic zone. As well, a springtime model experiment showed that aph over the hypoxic zone can differ by an average absolute 37% from its average scenario owing to changes solely in environmental variables other than NO3 flux.

[1] We used budgets of absorption (a), scattering (b), and backscattering (bb) for particles and chromophoric dissolved organic matter (CDOM) to investigate sources of seasonal variations in apparent optical properties (AOPs) of New England continental shelf surface waters. Spectral a, b, and bb budgets for particles were estimated from flow cytometric measurements of eukaryotic pico/nanophytoplankton, Synechococcus, heterotrophic prokaryotes, detritus, and minerals; AOPs were modeled with Hydrolight radiative transfer software. For late summer and spring, our modeled values of the diffuse attenuation coefficient (Kd) and remote sensing reflectance (Rrs) were on average within 15% and 9%, respectively, of independent measurements. This close agreement allowed us to examine how different seawater constituents contributed to AOP variability. Higher values of Kd in the spring, compared to summer, were due to higher absorption by eukaryotic phytoplankton (aeuk) and CDOM (aCDOM), which coincided with higher nutrient levels and less stratified conditions than in the summer. Differences in the spectral shape of Rrs between the seasons were caused by a combination of differences in aeuk, aCDOM, and bb from non-phytoplankton particles (minerals and detritus combined). For nonphytoplankton bb the major seasonal difference was a higher inverse wavelength dependence in the summer due to the effects of small organic detritus. We applied two semianalytical ocean color models to our data, in order to evaluate whether the assumptions and parameterizations inherent in these models are applicable for New England shelf waters. We show how differences between observed and modeled chlorophyll a specific phytoplankton absorption, aCDOM, and non-phytoplankton bb cause errors in chlorophyll a concentration and IOPs retrieved from reflectance inversion models. [1] From hosting key shipping lines, drilling platforms, and commercial fisheries, to sustaining mangrove swamps that shelter the coastline from the hurricanes that churn its waters, the Gulf of Mexico is important to the nations surrounding it for socio-economic, ecological, military, political, and scientific reasons. Critical to all of these sectors is the Gulf's circulation—it controls hurricane tracks and intensity, biological productivity, and larvae dispersal. Since 1982, the Environmental Studies Program (ESP) of the U.S. Bureau of Ocean Energy Management, Regulation and Enforcement (BOEMRE) has invested more than $67 million in field and numerical modeling studies to improve our understanding of the Gulf's circulation (see Figure 1). ESP-funded research has covered a broad array of topics, some applied and some basic. Studies carried out on behalf of the bureau reflect the bureau's information needs, stakeholder input, and offshore energy exploration and development trends. All ESP studies culminate in a technical report—127 technical reports on physical oceanography are publicly available (see http://www.gomr.boemre.gov/homepg/regulate/environ/techsumm/rec_pubs.html). Additionally, more than 100 peer-reviewed publications have been issued on the Gulf's physical oceanography and circulation. The AGU Geophysical MonographCirculation in the Gulf of Mexico: Observations and Models, the very first circulation overview since 1972, was funded by ESP. Additionally, data collected during fieldwork are deposited in national archives for public dissemination.

We advance a method to determine the diameter D and the complex refractive index (n + n'i) of marine particles from flow cytometric measurements of forward scattering, side scattering, and chlorophyll fluorescence combined with Mie theory. To understand better the application of Mie theory with its assumptions to flow cytometry (FCM) measurements of phytoplankton cells, we evaluate our flow cytometric-Mie (FCM-Mie) method by comparing results from a variety of phytoplankton cultures with independent estimates of cell D and with estimates of n and n' from the inversion of bulk measurements. Cell D initially estimated from the FCM-Mie method is lower than independent estimates, and n and n' are generally higher than bulk estimates. These differences reflect lower forward scattering and higher side scattering for single-cell measurements than predicted by Mie theory. The application of empirical scattering corrections improves FCM-Mie estimates of cell size, n, and n'; notably size is determined accurately for cells grown in both high- and low-light conditions, and n' is correlated with intracellular chlorophyll concentration. A comparison of results for phytoplankton and mineral particles suggests that differences in n between these particle types can be determined from FCM measurements. In application to natural mixtures of particles, eukaryotic pico/nanophytoplankton and Synechococcus have minimum mean values of n' in surface waters, and nonphytoplankton particles have higher values of n than phytoplankton at all depths.
