Abstract

The Authors present the results of an immunohistochemical-chromatographic study for the evaluation of the catecholamine content in experimental microvascular anastomoses on rat femoral arteries. Immunohistochemistry showed perivascular TH immunoreactive autonomic nerve fibers and, within the vascular wall, the presence of enzymes of the catecholamine biosynthetic pathway which has been confirmed by chromatography. The data, after statistical analysis, proved a significant reduction of catecholamine levels within operated patent or thrombosed vessels compared to non operated on control vessels; these data proved also a non significant increase of catecholamine levels within operated thrombosed vessels compared to operated patent vessels. The reasons and the potential clinical repercussions of these experimental data are analyzed, underlying the importance of neurogenic factors in the pathogenesis of microvascular thrombosis.

Abstract

In this study, we measured the lymphocyte levels of proteins involved in apoptosis regulation such as Bcl-2 the peripheral benzodiazepine receptor (PBR) caspase-3, and Cu/Zn superoxide dismutase (Cu/Zn SOD) in patients with Parkinson's disease (PD), either untreated or under therapy with dopaminergic agents (L-Dopa alone or L-dopa + dopamine agonists) and in healthy volunteers. All PD groups showed increased activity of caspase-3, compared to controls, particularly those under treatment only with L-Dopa. In this latter group, the increase in caspase-3 activity was also paralleled by an increase in the concentration of Cu/Zn SOD. In addition, patients taking L-Dopa + dopamine agonists showed marked decrease in Bcl-2 levels and increased PBR expression, which seems in keeping with the hypothesis that PBR may be functionally related to Bcl-2. In conclusion, we found clear modifications in the levels of proteins involved in the control of apoptosis in lymphocytes of PD patients. These changes were disease related but also modulated by the pharmacological treatment, which confirms the potential role of apoptosis in PD pathogenesis and the modulatory influence of dopaminergic agents. Abstract

Objective To investigate the endogenous dopaminergic/adrenergic system of lymphocytes in multiple sclerosis (MS) patients during treatment with interferon (IFN)-β. Methods Patients with relapsing-remitting MS undergoing IFN-β treatment were prospectively studied during the first year of treatment. Circulating lymphocytes were obtained at baseline and after 1, 3, 6 and 12 months of treatment and assayed for catecholamine (CA) production and mRNA expression of tyrosine hydroxylase (TH, the rate-limiting enzyme in the synthesis of CA), β 2 -adrenoceptors (AR) and D2, D3 and D5 dopaminergic receptors (DR). Results In cells from patients treated with IFN-β for 12 months the production of CA hugely increased and was less sensitive to IFN-γ-induced inhibition. Expression of mRNA for TH, β 2 -AR and DRD5 was already enhanced after 1 month and further increased up to 6–12 months of treatment. On the contrary, DRD2 mRNA progressively decreased and DRD3 mRNA did not significantly change over the whole study period. Conclusions In MS patients IFN-β treatment enhances the ability of lymphocytes to produce CA, and induces extensive modifications of both β 2 -AR and DR-operated pathways. The clinical relevance of these effects deserves consideration.
