Vascular dysfunction is entwined with aging and in the pathogenesis of Alzheimer's disease (AD) and contributes to reduced cerebral blood flow (CBF) and consequently, hypoxia. Hyperbaric oxygen therapy (HBOT) is in clinical use for a wide range of medical conditions. In the current study, we exposed 5XFAD mice, a well-studied AD model that presents impaired cognitive abilities, to HBOT and then investigated the therapeutical effects using two-photon live animal imaging, behavioral tasks, and biochemical and histological analysis. HBOT increased arteriolar luminal diameter and elevated CBF, thus contributing to reduced hypoxia. Furthermore, HBOT reduced amyloid burden by reducing the volume of pre-existing plaques and attenuating the formation of new ones. This was associated with changes in amyloid precursor protein processing, elevated degradation and clearance of Aß protein and improved behavior of 5XFAD mice. Hence, our findings are consistent with the effects of HBOT being mediated partially through a persistent structural change in blood vessels that reduces brain hypoxia. Motivated by these findings, we exposed elderly patients with significant memory loss at baseline to HBOT and observed an increase in CBF and improvement in cognitive performances. This study demonstrates HBOT efficacy in hypoxia-related neurological conditions, particularly in AD and aging.

Inhibition of extracellular glutamate (Glu) release decreases proliferation and invasion, induces apoptosis, and inhibits melanoma metastatic abilities. Previous studies have shown that Blood-glutamate scavenging (BGS), a novel treatment approach, has been found to be beneficial in attenuating glioblastoma progression by reducing brain Glu levels. Therefore, in this study we evaluated the ability of BGS treatment to inhibit brain metastatic melanoma progression in-vivo. RET melanoma cells were implanted in C56BL/6J mice to induce brain melanoma tumors followed by treatment with BGS or vehicle administered for fourteen days. Bioluminescent imaging was conducted to evaluate tumor growth, and plasma/CSF Glu levels were monitored throughout. Immunofluorescence staining of Ki67 and 53BP1 was used to analyze tumor cell proliferation and DNA double-strand breaks. In addition, we analyzed CD8, CD68, CD206, p-STAT1 and iNOS expression to evaluate alterations in tumor micro-environment and anti-tumor immune response due to treatment. Our results show that BGS treatment reduces CSF Glu concentration and consequently melanoma growth in-vivo by decreasing tumor cell proliferation and increasing pro-apoptotic signaling in C56BL/6J mice. Furthermore, BGS treatment supported CD8+ cell recruitment and CD68+ macrophage invasion. These findings suggest that BGS can be of potential therapeutic relevance in the treatment of metastatic melanoma.

Loss of cognitive function with aging is a complex and poorly understood process. Recently, clinical research has linked the occurrence of cortical microinfarcts to cognitive decline. Cortical microinfarcts form following the occlusion of penetrating vessels and are considered to be restricted to the proximity of the occluded vessel. Whether and how such local events propagate and affect remote brain regions remain unknown. To this end, we combined histological analysis and longitudinal diffusion tensor imaging (DTI), following the targeted-photothrombotic occlusion of single cortical penetrating vessels. Occlusions resulted in distant tissue reorganization across the mouse brain. This remodeling co-occurred with the formation of a microglia/macrophage migratory path along subcortical white matter tracts, reaching the contralateral hemisphere through the corpus callosum and leaving a microstructural signature detected by DTI-tractography. CX3CR1-deficient mice exhibited shorter trail lengths, differential remodeling, and only ipsilateral white matter tract changes. We concluded that microinfarcts lead to brain-wide remodeling in a microglial CX3CR1-dependent manner.

The developmental and molecular heterogeneity of tissue macrophages is unravelling, as are their diverse contributions to physiology and pathophysiology. Moreover, also given tissues harbor macrophages in discrete anatomic locations. Functional contributions of specific cell populations can in mice be dissected using Cre recombinase-mediated mutagenesis. However, single promoter-based Cre models show limited specificity for cell types. Focusing on macrophages in the brain, we establish here a binary transgenic system involving complementation-competent NCre and CCre fragments whose expression is driven by distinct promoters: Sall1ncre: Cx3cr1ccre mice specifically target parenchymal microglia and compound transgenic Lyve1ncre: Cx3cr1ccre animals target vasculature-associated macrophages, in the brain, as well as other tissues. We imaged the respective cell populations and retrieved their specific translatomes using the RiboTag in order to define them and analyze their differential responses to a challenge. Collectively, we establish the value of binary transgenesis to dissect tissue macrophage compartments and their functions.

Seven-day cycles in numbers of COVID-19 new-cases and deaths are markedly evident in most public databases (e.g. Worldometer, ECDC), but it is unclear whether they reflect systematic artifacts of delays in information reporting/gathering, or have a more profound basis. To address this question we located 11 databases of US states that provide date- authenticated information (actual date of symptom onset and/or specimen collection, or actual hospitalization or death date) that reported more than 1,000 deaths each. Numbers of new cases showed a weekly cyclic pattern in 10 out of 11 states, commonly peaking on weekdays, 2-6 days after the weekend, corresponding with a reported median 5-day lag between infection and the manifestation of clinical symptoms. We postulate that this pattern emerges from interactions with different and/or extended social-circles during weekends, including increased inter-generational meetings, which in turn facilitate transfer of COVID-19 from younger people to older vulnerable individuals. Furthermore, we found weekly periodicity in hospitalizations in 2 out of 2 authenticated databases providing this information. Actual death date, which is more difficult to attribute to individual choice, and is expected to occur approximately 2-3 weeks following hospitalization, showed significant 7-day periodicity in 1 out of 11 states, and a trend in 2 additional states. If weekly peaks in new cases can be truncated by physical/social distancing, especially during weekends, the mortality of COVID-19 may be reduced, or at least hospitalization and mortality curves may be flattened.Significance StatementWe believe our findings are significant, as it appears that it is difficult for people to grasp an ambiguous “price” for visiting their friends and family, when they cannot be certain that they are not carrying and spreading the virus. Our findings could be used as an effective “tool” to demonstrate such a cost, clearly presented in terms of number of excessive infections. During these days of uncertainly, we believe it is fundamental to provide scientific facts that could illuminate this connection and make it tangible for the general. The amplitude of the cycles we describe here is such that many thousands of infections could be averted by carefully scrutinizing local policies, medical practice, and social norms.

Aims Autism spectrum disorder (ASD) is a wide range of neurodevelopmental disorders involving deficits in social interaction and communication. Unfortunately, autism remains a scientific and clinical challenge owing to the lack of understanding the cellular and molecular mechanisms underlying it. This study aimed to investigate the pathophysiological mechanism underlying leukocyte-endothelial adhesion in autism-related neurovascular inflammation. Methods Male BTBR T+tf/J mice were used as an autism model. The dynamic pattern of leukocyte-endothelial adhesion in mouse cerebral vessels was detected by two-photon laser scanning microscopy (TPLSM). Using FACS, RT-PCR, and Western blotting, we explored the expression of cell adhesion molecules, the mRNA expression of endothelial chemokine, the protein levels of cathepsin B, and inflammatory mediators. Results We found a significant increase in leukocyte-endothelial adhesion in BTBR mice, accompanied by elevated expression of the adhesion molecule neutrophils CD11b and endothelial ICAM-1. Our data further indicate that elevated neutrophil cathepsin B levels contribute to elevated endothelial chemokine CXCL7 levels in BTBR mice. The pharmacological inhibition of cathepsin B reverses the enhanced leukocyte-endothelial adhesion in the cerebral vessels of autistic mice. Conclusion Our results revealed the prominent role of cathepsin B in modulating leukocyte-endothelial adhesion during autism-related neurovascular inflammation and identified a promising novel approach for autism treatment.

Developing efficient brain imaging technologies by combining a high spatiotemporal resolution and a large penetration depth is a key step for better understanding the neurovascular interface that emerges as a main pathway to neurodegeneration in many pathologies such as dementia. This review focuses on the advances in two complementary techniques: multi-photon laser scanning microscopy (MPLSM) and functional ultrasound imaging (fUSi). MPLSM has become the gold standard for in vivo imaging of cellular dynamics and morphology, together with cerebral blood flow. fUSi is an innovative imaging modality based on Doppler ultrasound, capable of recording vascular brain activity over large scales (i.e., tens of cubic millimeters) at unprecedented spatial and temporal resolution for such volumes (up to 10 μm pixel size at 10 kHz). By merging these two technologies, researchers may have access to a more detailed view of the various processes taking place at the neurovascular interface. MPLSM and fUSi are also good candidates for addressing the major challenge of real-time delivery, monitoring, and in vivo evaluation of drugs in neuronal tissue

Recently developed methods for rapid continuous volumetric two-photon microscopy facilitate the observation of neuronal activity in hundreds of individual neurons and changes in blood flow in adjacent blood vessels across a large volume of living brain at unprecedented spatio-temporal resolution. However, the high imaging rate necessitates fully automated image analysis, whereas tissue turbidity and photo-toxicity limitations lead to extremely sparse and noisy imagery. In this work, we extend a recently proposed deep learning volumetric blood vessel segmentation network, such that it supports temporal analysis. With this technology, we are able to track changes in cerebral blood volume over time and identify spontaneous arterial dilations that propagate towards the pial surface. This new capability is a promising step towards characterizing the hemodynamic response function upon which functional magnetic resonance imaging (fMRI) is based.

Spreading edema after strokeThe brain is enveloped in a cushion of cerebrospinal fluid (CSF), which normally provides protection and helps to remove metabolic waste. CSF transport has also recently been shown to play unexpected roles in neurodegeneration and sleep. Mestreet al.used multimodal in vivo imaging in rodents and found that, after a stroke, an abnormally large volume of CSF rushes into the brain, causing swelling (see the Perspective by Moss and Williams). This influx of CSF is caused by constrictions of arteries triggered by a well-known propagating chemical reaction-diffusion wave called spreading depolarization. In Alzheimer's disease (AD), astrocytes undergo morphological changes ranging from atrophy to hypertrophy, but the effect of such changes at the functional level is still largely unknown. Here, we aimed to investigate whether alterations in astrocyte activity in AD are transient and depend on their microenvironment, or whether they are irreversible. We established and characterized a new protocol for the isolation of adult astrocytes and discovered that astrocytes isolated from old 5xFAD mice have higher GFAP expression than astrocytes derived from WT mice, as observed in vivo. We found high C1q levels in brain sections from old 5xFAD mice in close vicinity to amyloid plaques and astrocyte processes. Interestingly, while old 5xFAD astrocytes are impaired in uptake of soluble Aβ42, this effect was reversed upon an addition of exogenous C1q, suggesting a potential role for C1q in astrocyte-mediated Aβ clearance. Our results suggest that scavenger receptor B1 plays a role in C1q-facilitated Aβ uptake by astrocytes and that expression of scavenger receptor B1 is reduced in adult old 5xFAD astrocytes. Furthermore, old 5xFAD astrocytes show impairment in support of neuronal growth in co-culture and neurotoxicity concomitant with an elevation in IL-6 expression. Further understanding of the impact of astrocyte impairment on AD pathology may provide insights into the etiology of AD.

Rapid imaging of neuronal activity under multi-photon microscopy represents a photon deprived application. We show how photon counting improves signal-to-noise ratio in these experiments.

The task of blood vessel segmentation in microscopy images is crucial for many diagnostic and research applications. However, vessels can look vastly different, depending on the transient imaging conditions, and collecting data for supervised training is laborious. We present a novel deep learning method for unsupervised segmentation of blood vessels. The method is inspired by the field of active contours and we introduce a new loss term, which is based on the morphological Active Contours Without Edges (ACWE) optimization method. The role of the morphological operators is played by novel pooling layers that are incorporated to the network's architecture. We demonstrate the challenges that are faced by previous supervised learning solutions, when the imaging conditions shift. Our unsupervised method is able to outperform such previous methods in both the labeled dataset, and when applied to similar but different datasets. We present PySight, an add-on hardware and software solution tailored for photon-deprived imaging conditions. PySight more than triples the median amplitude of neuronal calcium transients in awake mice, and facilitates single-trial intravital voltage imaging in fruit flies. Its unique data streaming architecture allowed us to image a fruit fly’s olfactory response over 234 ×; 600 ×; 330µm3 at 73 volumes per second, outperforming top-tier imaging setups while retaining over 200 times lower data rates. PySight requires no electronics expertise nor custom synchronization boards, and its open-source software is extensible to any imaging method based on single-pixel (bucket) detectors. PySight offers an optimal data acquisition scheme for ever increasing imaging volumes of turbid living tissue.

We review the organizational principles of the cortical vasculature and the underlying patterns of blood flow under normal conditions and in response to occlusion of single vessels. The cortex is sourced by a two-dimensional network of pial arterioles that feeds a three-dimensional network of subsurface microvessels in close proximity to neurons and glia. Blood flow within the surface and subsurface networks is largely insensitive to occlusion of a single vessel within either network. However, the penetrating arterioles that connect the pial network to the subsurface network are bottlenecks to flow; occlusion of even a single penetrating arteriole results in the death of a 500 μm diameter cylinder of cortical tissue despite the potential for collateral flow through microvessels. This pattern of flow is consistent with that calculated from a full reconstruction of the angioarchitecture. Conceptually, collateral flow is insufficient to compensate for the occlusion of a penetrating arteriole because penetrating venules act as shunts of blood that flows through collaterals. Future directions that stem from the analysis of the angioarchitecture concern cellular-level issues, in particular the regulation of blood flow within the subsurface microvascular network, and system-level issues, in particular the role of penetrating arteriole occlusions in human cognitive impairment.

The cerebral vasculature is organized to supply the brain’s metabolic needs. Sensory deprivation during the early postnatal period causes altered neural activity and lower metabolic demand. Neural activity is instructional for some aspects of vascular development, and deprivation causes changes in capillary density in the deprived brain region. However, it is not known if the pial arteriole network, which contains many leptomeningeal anastomoses (LMAs) that endow the network with redundancy against occlusions, is also affected by sensory deprivation. We quantified the effects of early-life sensory deprivation via whisker plucking on the densities of LMAs and penetrating arterioles (PAs) in anatomically-identified primary sensory regions (vibrissae cortex, forelimb/hindlimb cortex, visual cortex and auditory cortex) in mice. We found that the densities of penetrating arterioles were the same across cortical regions, though the hindlimb representation had a higher density of LMAs than other sensory regions. We found that the densities of PAs and LMAs, as well as quantitative measures of network topology, were not affected by sensory deprivation. Our results show that the postnatal development of the pial arterial network is robust to sensory deprivation.

These factors may be involved in choroid development and in the pathogenesis of the common blinding disease, age-related macular degeneration (AMD).

Blood-borne brain metastases are associated with poor prognosis, but little is known about the interplay between cerebral blood flow, surgical stress responses, and the metastatic process. The intra-carotid inoculation approach, traditionally used in animal studies, involves permanent occlusion of the common carotid artery (CCA). Herein we introduced a novel intra-carotid inoculation approach that avoids CCA ligation, namely - assisted external carotid artery inoculation (aECAi) - and compared it to the traditional approach in C57/BL6 mice, assessing cerebral blood flow; particle distribution; blood-brain barrier (BBB) integrity; stress, inflammatory and immune responses; and brain tumor retention and growth. Doppler flowmetry and two-photon imaging confirmed that only in the traditional approach regional and capillary cerebral blood flux were significantly reduced. Corticosterone and plasma IL-6 levels were higher in the traditional approach, splenic numbers of NK, CD3+, granulocytes, and dendritic cells were lower, and many of these indices were more profoundly affected by surgical stress in the traditional approach. BBB integrity was unaffected. Administration of spherical beads indicated that CCA ligation significantly limited brain distribution of injected particles, and inoculation of D122-LLC syngeneic tumor cells resulted in 10-fold lower brain tumor-cell retention in the traditional approach. Last, while most of the injected tumor cells were arrested in extra-cranial head areas, our method improved targeting of brain-tissue by 7-fold. This head versus brain distribution difference, commonly overlooked, cannot be detected using in vivo bioluminescent imaging. Overall, it is crucial to maintain unperturbed cerebral blood flow while studying brain metastasis and interactions with stress and inflammatory responses.

We present a method to form an optical window in the mouse skull that spans millimeters and is stable for months without causing brain inflammation. This enabled us to repeatedly image blood flow in cortical capillaries of awake mice and determine long-range correlations in speed. We also repeatedly imaged dendritic spines, microglia and angioarchitecture, as well as used illumination to drive motor output via optogenetics and induce microstrokes via photosensitizers.

It is well known that the density of neurons varies within the adult brain. In neocortex, this includes variations in neuronal density between different lamina as well as between different regions. Yet the concomitant variation of the microvessels is largely uncharted. Here, we present automated histological, imaging, and analysis tools to simultaneously map the locations of all neuronal and non-neuronal nuclei and the centerlines and diameters of all blood vessels within thick slabs of neocortex from mice. Based on total inventory measurements of different cortical regions (∼107cells vectorized across brains), these methods revealed: (1) In three dimensions, the mean distance of the center of neuronal somata to the closest microvessel was 15 μm. (2) Volume samples within lamina of a given region show that the density of microvessels does not match the strong laminar variation in neuronal density. This holds for both agranular and granular cortex. (3) Volume samples in successive radii from the midline to the ventral-lateral edge, where each volume summed the number of cells and microvessels from the pia to the white matter, show a significant correlation between neuronal and microvessel densities. These data show that while neuronal and vascular densities do not track each other on the 100 μm scale of cortical lamina, they do track each other on the 1–10 mm scale of the cortical mantle. The absence of a disproportionate density of blood vessels in granular lamina is argued to be consistent with the initial locus of functional brain imaging signals.

Astrocytes play a pivotal role in the development and function of the central nervous system by regulating synaptic activity and supporting and guiding growing axons. It is therefore a central therapeutic and scientific challenge to develop means to control astrocytic survival and growth. We cultured primary hippocampal astrocytes on a crystalline three-dimensional (3D) aragonite biomatrix prepared from the exoskeleton of the coral Porites lutea. Such culturing led to the formation of astrocytic tissue-like 3D structures in which the cells had a higher survival rate than astrocytes grown in conventional cell culture. Within the pore void areas, multiple layers of astrocytic processes formed concave sheet structures that had no physical contact with the surface. The astrocytes attached to the crystalline perpendicular edges of the crystalline template surface extended processes in 3D and expressed glial fibrillary acidic protein. The astrocytes also expressed gap junctions and developed partly synchronized cytosolic Ca2+ oscillations. Preliminary in vivo models showed that astrocytic networks were also developed when the matrices were implanted into cortical areas of postnatal rat brains. Hence, we suggest that the biomatrix is a biocompatible supportive scaffold for astrocytes and may be exploited in applications for neuronal tissue restoration in injured or diseased central nervous system.

Brain metastasis (BrM) have poor prognosis, and prophylactic approaches are scarce. Surgical stress responses have been shown to promote metastasis in peripheral organs through their immune-suppressive impacts and through direct effects on the malignant tissue and host physiology. However, BrM have not been studied in these respects, and the unique brain immune milieu, blood supply, and BBB, may react differently to such neuroendocrine challenges. Thus, we studied the effect of laparotomy, role of NK cells, and CpG-C immune-stimulation – a TLR-9 agonist having minimal adverse effects in humans – in early stages of brain and lung metastasis. Two syngeneic animal models were used: 3LL/D122 in C57BL/J6 mice, and MADB106 mammary adenocarcinoma in F344 rats. Tumor cells were injected either through the tail vein or employing a novel internal carotid injection approach we have developed, which generates BrM with high efficacy and minimal injection-related interferences to cerebral blood flow. Employing both models and inoculation approaches in naive and NK-depleted animals, our results indicate that NK cells surprisingly have no impact on BrM, while profoundly controlling lung metastases. On the other hand, laparotomy significantly enhanced brain and lungs tumor infiltration, and CpG-C reduced it, overcoming the effects of laparotomy. Thus, surgery is a significant risk factor for BrM through yet unknown mechanisms, and CpG-C treatment may be used prophylactically in cancer patients.

This protocol describes the application of laser pulses to image and ablate neuronal tissue for the purpose of automated histology. The histology is accomplished in situ using serial two-photon imaging of labeled tissue and removal of the imaged tissue with amplified, femtosecond pulses. Together with the use of endogenous fluorescent indicators and/or deep penetration of antibody labels and organic dyes, this method may be used to automatically image, reconstruct, and vectorize structures of interest across millimeter to centimeter regions of brain with micrometer resolution.

What is the nature of the vascular architecture in the cortex that allows the brain to meet the energy demands of neuronal computations? We used high-throughput histology to reconstruct the complete angioarchitecture and the positions of all neuronal somata of multiple cubic millimeter regions of vibrissa primary sensory cortex in mouse. Vascular networks were derived from the reconstruction. In contrast with the standard model of cortical columns that are tightly linked with the vascular network, graph-theoretical analyses revealed that the subsurface microvasculature formed interconnected loops with a topology that was invariant to the position and boundary of columns. Furthermore, the calculated patterns of blood flow in the networks were unrelated to location of columns. Rather, blood sourced by penetrating arterioles was effectively drained by the penetrating venules to limit lateral perfusion. This analysis provides the underpinning to understand functional imaging and the effect of penetrating vessels strokes on brain viability.

Microinfarctions are present in the aged and injured human brain. Their clinical relevance is controversial, with postulated sequelae ranging from cognitive sparing to vascular dementia. To address the consequences of microinfarcts, we used controlled optical methods to create occlusions of individual penetrating arterioles or venules in rat cortex. Single microinfarcts, targeted to encompass all or part of a cortical column, impaired performance in a macrovibrissa-based behavioral task. Furthermore, the targeting of multiple vessels resulted in tissue damage that coalesced across cortex, even though the intervening penetrating vessels were acutely patent. Post-occlusion administration of memantine, a glutamate receptor antagonist that reduces cognitive decline in Alzheimer's disease, ameliorated tissue damage and perceptual deficits. Collectively, these data imply that microinfarcts likely contribute to cognitive decline. Strategies that have received limited success in the treatment of ischemic injury, which include therapeutics against excitotoxicity, may be successful against the progressive nature of vascular dementia.

How does the brain compute? Answering this question necessitates neuronal connectomes, annotated graphs of all synaptic connections within defined brain areas. Further, understanding the energetics of the brain's computations requires vascular graphs. The assembly of a connectome requires sensitive hardware tools to measure neuronal and neurovascular features in all three dimensions, as well as software and machine learning for data analysis and visualization. We present the state of the art on the reconstruction of circuits and vasculature that link brain anatomy and function. Analysis at the scale of tens of nanometers yields connections between identified neurons, while analysis at the micrometer scale yields probabilistic rules of connection between neurons and exact vascular connectivity.
