A influência da velocidade de circulação do leite na adesão bacteriana de Pseudomonas aeruginosa foi avaliada em teste de uso simulado por meio de um modelo de circuito de processamento de leite. O circuito é composto por uma tubulação de aço inoxidável AISI 304, com 1,9 cm de diâmetro e 5,8 m de comprimento, além de um tanque de 25 L, utilizado como reservatório do produto e das soluções sanitizantes. O reservatório foi acoplado a uma bomba centrífuga de ½ HP, para impulsionar o alimento ou soluções de higienização pelo sistema equipado com cupons de prova em aço inoxidável nas formas cilíndrica, cotovelo 90º e T. As velocidades de circulação foram de 0,5, 1,0 e 1,5 m.s-1, correspondentes a fluxo turbulento com número de Reynolds de 14.000, 28.000 e 42.000, respectivamente. Quando se utilizou velocidade de 0,5 m.s-1, permaneceram aderidas à superfície 10,7% das células. Já nas velocidades e 1,0 e 1,5 m.s-1 as porcentagens de adesão foram de 5,36 e 4,9%, respectivamente, o que demonstra uma menor remoção de células aderidas à medida que o fluxo diminui, permitindo assim que mais células permaneçam aderidas na linha de produção, o que pode favorecer a formação de biofilmes. The influence of the flow milk circulation in the bacterial adhesion of Pseudomonas aeruginosa was evaluated by simulation tests through a circuit model of milk processing. The circuit is composed of a tubulation of stainless steel AISI 304, with 1.9 cm of diameter, 5.8 m of length and a tank of 25 L used as the reservoir of the product and sanitizer solutions. The reservoir was coupled to a centrifugal bomb of ½ HP to impel the food or sanitizer solutions for the system equipped with 90º and T cylindrical stainless steel specimens. The speed of circulation values were 0.5, 1.0 and 1.5 m.s-1, corresponding to turbulent flow with number of Reynolds 14.000, 28.000 and 42.000, respectively. When flow of 0.5 m.s-1 was used 10.7% the cells remained adhered, however at the speed values of 1.0 and 1.5 m.s-1 the adhesion percentages were 5.36 and 4.9%, respectively. These findings indicate a lower removal rate of adhered cells as flow decreases allowing higher number of bacteria to adhere to the production line, which can favor the biofilm formation.

A busca de matéria prima para produção de corantes naturais com propriedades adequadas ao uso pelas indústrias de alimentos processados, medicamentos, cosméticos e tintas, levou à proposta de um estudo da bertalha (Basella rubra L.) como mais uma fonte potencial de corante natural. Assim, objetivou-se com este trabalho realizar a caracterização de flavonóides e estudar a estabilidade de pigmentos. A extração foi realizada com metanol em pH 2,0, partindo-se de dois quilos de frutos, obtendo-se 13,0 gramas de pigmentos liofilizados. As substâncias presentes nos extratos liofilizados foram isoladas por CLAE preparativa, usando coluna RP 18 Luna de 5 μm, fase móvel acetonitrila:solução aquosa de ácido fórmico (9:1), fluxo de 3,5mL/mim em sistema isocrático por 30 mim. Este foi considerado um excelente método, capaz de isolar as substâncias S1, S2 e S3 das demais presentes no extrato bruto. A utilização de RMN, cromatografia em papel, métodos espectrais UV/Vis e reações químicas específicas para antocianinas foram muito úteis para a caracterização das substâncias S1, S2 e S3 como malvidina, cianidina e flavona, respectivamente. A estabilidade do extrato antociânico foi estimada determinando-se os valores de absorvância no comprimento de onda de máxima absorção, em função do tempo. A partir desses valores foram calculadas as constantes de velocidade de reação (k), assim como o tempo de meia vida (t1/2), na presença e ausência de luz, em diferentes pHs e diferentes temperaturas. O extrato de bertalha em pH 4,0, frente a luz e temperatura de 25°C apresentou tempo de meia vida de 210,0 h, enquanto que na ausência de luz apresentou tempo de meia vida de 577,5 h. Em pH 5,0 e 6,0, com luz e temperatura de 25°C apresentou tempo de meia vida de 315,0 h, enquanto que na ausência de luz apresentou tempo de meia vida de 693,0 h. Na presença de luz, observou-se o efeito drástico da temperatura sobre a degradação do pigmento. Na temperatura de 40°C o sistema mais estável foi em pH 5,0 com tempo de meia vida de 7,46h, seguido do pH 6,0 com 7,18h e o menos estável em pH 4,0 com 6,01h. Na temperatura de 60°C, o sistema mais estável foi em pH 6,0 com tempo de meia vida de 2,70h, seguido do pH 5,0 com 2,50h e o menos estável em pH 4,0 com 2,08h. A temperatura e a luz foram os agentes considerados mais destrutivos aos pigmentos. As substâncias purificadas S1 e S2 apresentaram, frente à luz e à temperatura de 25°C, tempo de meia vida de 50,90h e 40,76h respectivamente, mostrando elevada sensibilidade das antocianinas à luz, quando puras. The search for raw material for the production of natural colorants with adequate properties for use by processed food, medicine, cosmetics, and dye industries has led to the proposal of a study on Malabar nightshade (Basella rubra L.) as an additional potential source of natural colorant. Thus, the aim of this study was to carry out flavonoid characterization and to study pigment stabilization. The extraction was performed with methanol at pH 2.0, using 2 kg of fruit and obtaining 13.0 g of lyophilized pigments. The substances present in the lyophilized extracts were isolated by preparative high performance liquid chromatography (HPLC), using a 5 μm RP-18 Luna column, acetonitrile: formic acid aqueous solution (9:1) mobile phase at a flow of 3.5 mL/min in isocratic mode for 30 min. This method was considered to be excellent, capable of isolating the substances S1, S2 and S3 from the others in the raw extract. The application of the techniques NMR, paper chromatography, UV/Vis spectral methods and specific chemical reactions for anthocyanins was very useful to characterize the substances S1, S2 and S3 as maldivin, cyanidin, and flavone, respectively. The stability of the anthocyanin extract was estimated by determining the absorbance values at maximum absorption wavelength as a function of time. Based on these values, the reaction velocity constants (k), were calculated, as well as half-life time (t1/2), in the presence and absence of light, at different pHs and temperatures. Malabar nightshade extract at pH 4.0, in the presence of light and temperature of 25°C, presented half-life of 210.0 h, whereas in the absence of light it presented a half-life of 577.5 h. At pH 5.0 and 6.0 in the presence of light and temperature of 25°C, it presented a half-life of 315.0 h., but a half-life of 693.0 h. in the absence of light. In the presence of light, a drastic temperature effect was observed on pigment degradation. At temperature of 40°C the most stable system was at pH 5.0 with a half- life of 7.46 h, followed by pH 6.0 with 7.18 h while the least stable was at pH 4.0 with 6.01 h. At temperature of 60°C, the most stable system was at pH 6.0 with half-life of 2.70 h, followed by pH 5.0 with 2.50 h while the least stable was at pH 4.0 with 2.08 h. Temperature and light were considered most destructive to the pigments. The purified substances S1 and S2 in the presence of light and at temperature of 25°C, presented a half-life of 50.90h and 40.76h, respectively, showing high anthocyanin sensitivity to light, when pure.
