Abstract

Abstract Background The β–(1 → 3),(1 → 6)-D-glucan extracellularly produced by Aureobasidium pullulans exhibits immunomodulatory activity, and is used for health supplements. To examine the effects of oral administration of the β–(1 → 3),(1 → 6)-D-glucan to domestic animals, a small scale study was conducted using Holstein cows and newborn Japanese Black calves. Findings Holstein cows of which somatic cell count was less than 3 x 105/ml were orally administered with or without the β-(1 → 3),(1 → 6)-D-glucan-enriched A. pullulans cultured fluid (AP-CF) for 3 months, and the properties of milk and serum cytokine expression were monitored. Somatic cell counts were not significantly changed by oral administration of AP-CF, whereas the concentration of solid non fat in the milk tended to increase in the AP-CF administered cows. The results of cytokine expression analysis in the serum using ELISA indicate that the expressions of tumor necrosis factor-α (TNF-α) and interleukin (IL)-6 in all cows which were orally administered with AP-CF became slightly lower than that of control cows after the two-month treatment. On the other hand, IL-8 expression tended to indicate a moderately higher level in all treated cows after the three-month administration of AP-CF in comparison with that of the control cows. Peripartum Japanese Black beef cows and their newborn calves were orally administered with AP-CF, and bacterial flora in the intestines of the calves were analyzed by T-RFLP (terminal restriction fragment length polymorphism). The results suggest that bacterial flora are tendentiously changed by oral administration of AP-CF. Conclusions Our data indicated the possibility that oral administration of the β–(1 → 3),(1 → 6)-D- glucan produced by A. pullulans affects cytokine expressions in the serum of Holstein cows, and influences bacterial flora in the intestines of Japanese Black calves. The findings may be helpful for further study on the efficacies of oral administration of β-(1 → 3),(1 → 6)-D-glucans on domestic animals.

A new terminal restriction fragment length polymorphism (T-RFLP) targeting 16S rRNA gene, designed with new primer-enzyme combinations, for distinguishing bacterial population in human feces at the genus level is described. Analysis of fecal DNAs using this method yields profiles characteristic to individual samples.

Solid bases comprising MgO covered with Al2O3 were prepared by decomposition of Al(OCH(CH3)2)3 over Mg(OH)2 in ethyl acetate. Catalysts containing more than 10 mol% Al were insoluble in acetone. Abstract

Polypeptide chain elongation factor la (EF-lα) of Saccharomyces cerevisiae is encoded by two distinct genes designated EF1 αA and EF1 αB [Nagata et al., EMBO J. 3 (1984) 1825–1830]. Both genes were cloned, and their nucleotide (nt) sequences were determined [see also Schirmaier and Philippsen, EMBO J. 3 (1984) 3311–3315, and Cottrelle et al., J. Biol. Chem. 260 (1985) 3090–3096]. They contain an open reading frame of 1374 nt coding for an identical protein of 458 amino acid residues, although their nt sequences differed at two positions. In this paper, we determined their 5' and 3'-flanking sequences which were considerably different each other. From the Sl nuclease mapping ofmRNA, both genes are found to be expressed almost to the same extent in exponentially growing cells. The transcription start points for EF1 αA and EF1 αB mRNAs were precisely located by primer extension procedure at 32 and 23 nt upstream of the start codons, respectively. The sequence which commonly exists in the 5'-flanking regions of ribosomal protein genes of S. cerevisiae was also present in the two EF1 α genes.

40. Cloning and Sequence Analysis of cDNA Coding for a Lectin from Helianthus tuberosusCallus and Its Jasmonate-induced Expression

Abstract

Two lectins (designated as HTA I and HTA II) that seemed to be isolectins were found in Helianthus tuberosuscallus. cDNA encoding HTA I was isolated from a ZAP Express expression library by immunoselection by using the anti-HTA antiserum. The sequence of this cDNA consisted of 432 bp nucleotides coding for a polypeptide of 143 amino acid residues (Mr, 15,314). When introduced into E. coli, the cDNA directed the synthesis of active HTA I as indicated by the hemagglutination activity. The deduced amino acid sequence showed homology with some lectins and jasmonate-induced proteins. When callus was cultured in the presence of methyl jasmonate (MeJA), the hemagglutination activity increased in a dose-dependent manner. The levels of expression of the HTA protein and of the corresponding mRNA also increased in the treated callus. In view of these results, HTA I is considered to be a jasmonate-induced protein.

Callus from Helianthus tuberosusexpresses a mannose-specific lectin (HTA). The level of HTA mRNA significantly increased one hour after treatment of the callus with 20 mg/l methyl jasmonate. Following this, fragmentation of the callus DNA at regular intervals was observed together with strong self-fluorescence emission in the callus cells.
