Climate change affects timing of reproduction in many bird species, but few studies have investigated its influence on annual reproductive output. Here, we assess changes in the annual production of young by female breeders in 201 populations of 104 bird species (N = 745,962 clutches) covering all continents between 1970 and 2019. Overall, average offspring production has declined in recent decades, but considerable differences were found among species and populations. A total of 56.7% of populations showed a declining trend in offspring production (significant in 17.4%), whereas 43.3% exhibited an increase (significant in 10.4%). The results show that climatic changes affect offspring production through compounded effects on ecological and life history traits of species. Migratory and larger-bodied species experienced reduced offspring production with increasing temperatures during the chick-rearing period, whereas smaller-bodied, sedentary species tended to produce more offspring. Likewise, multi-brooded species showed increased breeding success with increasing temperatures, whereas rising temperatures were unrelated to reproductive success in single-brooded species. Designed to empower researchers with access to Omics data and bioinformatic analyses, VEuPathDB projects integrate >1700 pre-analysed datasets (and associated metadata) with advanced search capabilities, visualizations, and analysis tools in a graphic interface. Diverse data types are analysed with standardized workflows including an in-house OrthoMCL algorithm for predicting orthology. Comparisons are easily made across datasets, data types and organisms in this unique data mining platform. A new site-wide search facilitates access for both experienced and novice users. Research highlighting sex-differentiated jealousy resulting from imagined scenarios has now been reaffirmed when the infidelity-revealing message is discovered on a social media platform. Participants in the current study were presented with both sexually and emotionally charged infidelity-revealing scenarios featuring a same-sex sibling, a friend and a stranger in the format of a ‘Snapchat’ message. Men indicated significantly higher jealousy to sexual as opposed to emotional messages with the reverse pattern evident in women. Sex differences were also evident in the extent of jealousy elicited by ‘third-party’ identity. Women were significantly more jealous when the imagined infidelity occurred between their sister compared to both a best friend and a stranger with males showing significantly lower jealousy directed towards their brother compared to a stranger. These findings are supportive not only of a parental investment (PI) interpretation of sex differences in jealousy but also an interpretation consistent with aspects of inclusive fitness theory.

Abstract. Anabolic Androgenic Steroids (AAS) usage has been repeatedly linked with a body image disorder called Muscle Dysmorphia (MD). However, evidence for how this relationship manifests is currently inconclusive. This study focused on the extent to which MD may precipitate or perpetuate the use of AAS. Utilizing a cross-sectional design, the sample consisted of 57 users (current and lifetime) and 51 non-using (never used AAS) bodybuilders recruited from two bodybuilding dedicated gymnasiums located in Wales, UK. Significantly higher levels of MD were found in users compared to non-users across training experience duration (0–2, 3–5, and 6+ years). MD levels irrespective of AAS usage (users vs. non-users) were consistent between 0–2 and 3–5 years of training declining however between those of 6+ years training experience. No differences were reported in symptoms of MD between users of AAS with varying lengths of AAS use exposure. This provides preliminary evidence suggesting MD may influence both initiation and maintenance of AAS use; however, neither regular gym attendance nor prolonged AAS usage may exacerbate MD symptoms.

Store-operated Ca(2+) entry (SOCE) mediated by calcium release-activated calcium (CRAC) channels contributes to calcium signaling. The resulting intracellular calcium increases activate calcineurin, which in turn activates immune transcription factor nuclear factor of activated T cells (NFAT). Microglia contain CRAC channels, but little is known whether these channels play a role in acute brain insults. We studied a novel CRAC channel inhibitor to explore the therapeutic potential of this compound in microglia-mediated injury. Cultured microglial BV2 cells were activated by Toll-like receptor agonists or IFNγ. Some cultures were treated with a novel CRAC channel inhibitor (CM-EX-137). Western blots revealed the presence of CRAC channel proteins STIM1 and Orai1 in BV2 cells. CM-EX-137 decreased nitric oxide (NO) release and inducible nitric oxide synthase (iNOS) expression in activated microglia and reduced agonist-induced intracellular calcium accumulation in microglia, while suppressing inflammatory transcription factors nuclear factor kappa B (NF-κB) and nuclear factor of activated T cells (NFAT). Male C57/BL6 mice exposed to experimental brain trauma and treated with CM-EX-137 had decreased lesion size, brain hemorrhage, and improved neurological deficits with decreased microglial activation, iNOS and Orai1 and STIM1 levels. We suggest a novel anti-inflammatory approach for managing acute brain injury. Our observations also shed light on new calcium signaling pathways not described previously in brain injury models.

We report on three decades of repeat surveys of beached marine debris at two locations in the Scotia Sea, in the Southwest Atlantic sector of the Southern Ocean. Between October 1989 and March 2019 10,112 items of beached debris were recovered from Main Bay, Bird Island, South Georgia in the northern Scotia Sea. The total mass of items (data from 1996 onwards) was 101 kg. Plastic was the most commonly recovered item (97.5% by number; 89% by mass) with the remainder made up of fabric, glass, metal, paper and rubber. Mean mass per item was 0.01 kg and the rate of accumulation was 100 items km−1 month−1. Analyses showed an increase in the number of debris items recovered (5.7 per year) but a decline in mean mass per item, suggesting a trend towards more, smaller items of debris at Bird Island. At Signy Island, South Orkney Islands, located in the southern Scotia Sea and within the Antarctic Treaty area, debris items were collected from three beaches, during the austral summer only, between 1991 and 2019. In total 1304 items with a mass of 268 kg were recovered. Plastic items contributed 84% by number and 80% by mass, with the remainder made up of metal (6% by number; 14% by mass), rubber (4% by number; 3% by mass), fabric, glass and paper (

As men have universally expressed attraction to younger women, awareness of this proclivity in men by women should be maximally reflected in terms of mate value in younger compared to older women. Prostitution has been recorded across all cultures and historical epochs, and one way in which mate value in women can be explored is by obtaining data from online escort advertising sites. The current study accessed an online escort site called adultwork.com to compare advertised fees between escorts at various ages and age ranges in five different cultures (UK, Ireland, Australia, the USA and Europe) offering either incall or outcall sexual services. With the exception of Irish escorts advertising for an outcall service, differences were found in all countries sampled with younger escorts charging significantly higher fees than older escorts. The current findings are another example of how by accessing real-world online data we can successfully test hypotheses relating to sexual behaviour from an evolutionary psychological perspective.

Sex differences in how and to what extent jealousy manifests have long been documented by evolutionary psychologists with males showing more pronounced responses to sexual infidelity and females to emotional infidelity. With modern technology facilitating the opportunity for extra-pair relations and the means by which inclinations towards infidelity can be monitored, social media is a fertile ground to test hypotheses derived from evolutionarily informed theories. The current study presented male (n = 21) and female (n = 23) undergraduate participants with realistic, unambiguously sexual and emotional messages both sent and received that had been discovered (imagined) on their partner’s Facebook messenger. Distress scores in response to these messages were measured on a scale of 0–10. Broad support for the evolutionary interpretation of sex differences in jealousy was found with more pronounced sexual jealousy in males, and emotional jealousy in females compared to males being evident. Similarly, salient sex differences were observed highlighting the importance of the composer of the infidelity-revealing message. For example, in females, higher distress was found resulting from the discovery of received (female rival) when compared to sent (male partner) messages, and received messages across sex (females higher). The results are discussed in relation to previous findings and in the context of growing concern relating to relationship dissolution and partner-initiated domestic violence.

Key points This work confirms previous reports that CM4620, a small molecule inhibitor of Ca2+ entry via store operated Ca2+ entry (SOCE) channels formed by stromal interaction molecule 1 (STIM1)/Orai complexes, attenuates acinar cell pathology and acute pancreatitis in mouse experimental models. Here we report that intravenous administration of CM4620 reduces the severity of acute pancreatitis in the rat, a hitherto untested species. Using CM4620, we probe further the mechanisms whereby SOCE via STIM1/Orai complexes contributes to the disease in pancreatic acinar cells, supporting a role for endoplasmic reticulum stress/cell death pathways in these cells. Using CM4620, we show that SOCE via STIM1/Orai complexes promotes neutrophil oxidative burst and inflammatory gene expression during acute pancreatitis, including in immune cells which may be either circulating or invading the pancreas. Using CM4620, we show that SOCE via STIM1/Orai complexes promotes activation and fibroinflammatory gene expression within pancreatic stellate cells. Abstract Key features of acute pancreatitis include excess cellular Ca2+ entry driven by Ca2+ depletion from the endoplasmic reticulum (ER) and subsequent activation of store-operated Ca2+ entry (SOCE) channels in the plasma membrane. In several cell types, including pancreatic acinar, stellate cells (PaSCs) and immune cells, SOCE is mediated via channels composed primarily of Orai1 and stromal interaction molecule 1 (STIM1). CM4620, a selective Orai1 inhibitor, prevents Ca2+ entry in acinar cells. This study investigates the effects of CM4620 in preventing or reducing acute pancreatitis features and severity. We tested the effects of CM4620 on SOCE, trypsinogen activation, acinar cell death, activation of NFAT and NF-κB, and inflammatory responses in ex vivo and in vivo rodent models of acute pancreatitis and human pancreatic acini. We also examined whether CM4620 inhibited cytokine release in immune cells, fibro-inflammatory responses in PaSCs, and oxidative burst in neutrophils, all cell types participating in pancreatitis. CM4620 administration to rats by i.v. infusion starting 30 min after induction of pancreatitis significantly diminished pancreatitis features including pancreatic oedema, acinar cell vacuolization, intrapancreatic trypsin activity, cell death signalling and acinar cell death. CM4620 also decreased myeloperoxidase activity and inflammatory cytokine expression in pancreas and lung tissues, fMLF peptide-induced oxidative burst in human neutrophils, and cytokine production in human peripheral blood mononuclear cells (PBMCs) and rodent PaSCs, indicating that Orai1/STIM1 channels participate in the inflammatory responses of these cell types during acute pancreatitis. These findings support pathological Ca2+ entry-mediated cell death and proinflammatory signalling as central mechanisms in acute pancreatitis pathobiology.

Purpose: Calcium release-activated calcium (CRAC) channels activate microglia via store-operated Ca 2+ entry. We previously reported that the specific CRAC channel inhibitor (CM-EX-137) had neuroprotective effects in a model of experimental stroke due to suppression of microglial activation in male mice. However, it is well known that sex differences can influence stroke outcome as well as immune responses. Whether CRAC channel inhibition in female mice is similarly neuroprotective is unknown. The purpose of this study is to determine whether female mice respond similarly CRAC channel inhibition in a stroke model Subjects and Methods: C57/BL6 female mice, aged 2 months were subjected to distal middle cerebral artery occlusion (dMCAO) were treated with CM-EX-137 (5mg/kg IP; dMCAO-CM) or vehicle control (dMCAO-vehicle) daily for 3d. Stroke surgeries were carried out during the same phase of the estrus cycle (diestrus) to avoid potential confounds due to varying estrogen levels. These groups were also compared to sham groups. CM-EX-137 was injected immediately after occlusion and at 24 & 48h. Brains were extracted 3d post dMCAO. Neurological functions were evaluated before surgery, and 1 & 3d later. Lesion size was estimated from cresyl violet stains. Microglia and astrocyte activation were assessed using immunohistochemistry. Results: Neurological function at both 24 and 72 h post ischemia were significantly improved in dMCAO-CM group, compared with dMCAO-vehicle group (n=7/group; p Conclusion: CRAC channel inhibition reduce inflammatory responses and improve neurological outcome following experimental stroke in female animals. These results are in line with those that we observed in male mice. Thus, CRAC channel inhibition seems to be similarly effective and anti-inflammatory in both genders in a stroke model.

Gentoo penguins Pygoscelis papua breed at a number of sites at the West Antarctic Peninsula, including Goudier Island, Port Lockroy—the longest studied location for tourist-penguin interactions in the Antarctic. These penguins annually encounter some of the highest numbers of tourists in the whole of Antarctica. Using yearly count data from all ten colonies on Goudier Island, we report changes in long-term population size and breeding success over a 21 year period (1996/1997 to 2016/2017), documenting inter-annual variability in numbers of breeding pairs and chicks fledged (productivity). We found a 24.5% (1.4% per annum) decrease in breeding pairs; similar declines were evident in breeding pairs at six colonies visited by tourists as well as at four unvisited colonies. Breeding success also declined, with chick numbers declining in visited (53.7%, 3.8% per annum) and unvisited colonies (59.8%, 4.6% per annum). While gentoo penguin numbers are increasing regionally, we reveal a recent decline in the Goudier Island population occurring simultaneous with increases in tourist numbers from 262 in 1996/1997 to 19,688 in 2016/2017. Analyses suggest a complex situation with different possible drivers of change. There was a significant negative effect of increasing air temperature and shifts in sea ice variables on breeding pairs. However, similar statistical support showed a significant link existed between year and visitors, with higher numbers of visitors negatively affecting penguin numbers. Based on our results we recommend increased precaution in management at Goudier Island, and initiation of similar studies at other frequently visited penguin sites in Antarctica.

Understanding the mechanisms which determine the capacity of any species to adapt to changing environmental conditions is one of the foremost requirements in accurately predicting which populations, species and clades are likely to survive ongoing, rapid climate change. The polar oceans are amongst the most rapidly changing environments on Earth with reduced regional sea ice duration and extent, and their fauna's expected sensitivity to warming and acidification. These changes potentially pose a significant threat to a number of polar fauna. There is, therefore, a critical need to assess the vulnerability of a wide range of species to determine the tipping points or weak links in marine assemblages. Knowledge of the effect of multiple stressors on polar marine fauna has advanced over the last 40 years, but there are still many data gaps. This study applies ecological risk assessment techniques to the increasing knowledge of polar species' physiological capacities to identify their exposure to climate change and their vulnerability to this exposure. This relatively rapid, semi-quantitative assessment provides a layer of vulnerability on top of climate envelope models, until such times as more extensive physiological data sets can be produced. The risk assessment identified more species that are likely to benefit from the near-future predicted change (the winners), especially predators and deposit feeders. Fewer species were scored at risk (the losers), although animals that feed on krill scored consistently as under the highest risk.

The mechanisms that determine patterns of species dispersal are important factors in the production and maintenance of biodiversity. Understanding these mechanisms helps to forecast the responses of species to environmental change. Here we used a comparative framework and genome-wide data obtained through RAD-seq to compare the patterns of connectivity among breeding colonies for five penguin species with shared ancestry, overlapping distributions, and differing ecological niches, allowing an examination of the intrinsic and extrinsic barriers governing dispersal patterns. Our findings show that at-sea range and oceanography underlie patterns of dispersal in these penguins. The pelagic niche of emperor (Aptenodytes forsteri), king (A. patagonicus), Adélie (Pygoscelis adeliae) and chinstrap (P. antarctica) penguins facilitates gene flow over thousands of kilometres. In contrast, the coastal niche of gentoo penguins (P. papua) limits dispersal, resulting in population divergences. Oceanographic fronts also act as dispersal barriers to some extent. We recommend that forecasts of extinction risk incorporate dispersal and that management units are defined by at-sea range and oceanography in species lacking genetic data. Aim:\ud To provide a method of analysing penguin tracking data to identify priority at-sea areas for seabird conservation (marine IBAs), based on pre-existing approaches for flying seabirds but revised according to the specific ecology of Pygoscelis penguin species.\ud Location:\ud Waters around the Antarctic Peninsula, South Shetland and South Orkney Archipelagos (FAO Subareas 48.1 and 48.2)\ud Methods:\ud We made key improvements to the pre-existing protocol for identifying marine IBAs that include refining the track interpolation method, and revision of parameters for the kernel analysis (smoothing factor and utilization distribution) using sensitivity tests. We applied the revised method to 24 datasets of tracking data on penguins (three species, seven colonies and three different breeding stages – incubation, brood and crèche).\ud Results:\ud We identified 5 new marine IBAs for seabirds in the study area, estimated to hold ca. 600,000 adult penguins.\ud Main conclusions:\ud The results demonstrate the efficacy of a new method for the designation of a network of marine IBAs in Antarctic waters for penguins based on tracking data, which can contribute to an evidence-based, precautionary, management framework for krill fisheries.

Background This study explored the direct experience of anabolic androgenic steroid (AAS) use by young men, with an emphasis on how motivations progressed from adolescent initiation to more entrenched usage. Participants and procedure Nine semi-structured interviews were conducted with individuals ranging in experience of AAS use, from novice to experienced users. Results The results indicated that the young adult men progressed through a clear transition whereby their motives for using these substances changed from a mere desire to compete with other men to more internalised body image problems. Conclusions The findings presented suggest a more complex relationship between AAS use and body image pathology than previously suggested.

The southern giant petrel (Macronectes giganteus) has a circumpolar distribution and breeds on subantarctic islands and a few continental Antarctic sites. Although this species has recently been down-listed to “Least Concern” by the World Conservation Union (IUCN), there are strong fluctuations in abundance and variable long-term trends recorded at different sites. Systematic, long-term monitoring is essential to determine drivers underlying its population dynamics. Here, we examine long-term changes in population size and productivity of southern giant petrels at Signy Island, South Orkney Islands. Comparing estimated numbers of breeding pairs over the whole island in 2000/2001, 2005/2006, 2009/2010 and 2014/2015 with historical data revealed several phases of population change: a 64 % decline (6.2 % per annum) from 1968/1969 to 1984/1985, a 162 % increase (6.2 % per annum) to 2000/2001, stability until 2005/2006, a 56 % decline (18.3 % per annum) to 2009/2010 and stability until 2014/2015. This represents a 1.8 % decline per annum between 1968/1969 and 2014/2015. Annual counts within focal study areas suggested a more rapid increase from 1996/1997 to 2006/2007, but the same downward trend from 2006/2007 to present, underlining potential pitfalls in inferring trends from part-island counts. There was also a 20 % decline in breeding success from 1996/1997 to 2014/2015. Our results indicate substantial fluctuations in southern giant petrel abundance at Signy Island over 4–5 decades and a recent decline in breeding numbers and success. As the southern giant petrels breeding at the South Orkney Islands represents ~5–10 % of the global population, continuation of these declines would be of high conservation concern.

Penguins are an important indicator of marine ecosystem health and a major contributor of nutrients to terrestrial ecosystems in Antarctica. Their stomach microbiota is influenced by both the prey consumed and their foraging environment in the sea. As penguins feed at sea and breed on land, they might be expected to transfer microbes (e.g. prey-associated and marine bacteria) as well as nutrients from their stomachs while regurgitating food or in their guano to the surrounding terrestrial environment. However, most research attention to date has focused separately on the penguin gut microbiota (via cloacal/guano samples) and the terrestrial soil microbiota, and any relationship between them has yet to be established. Here, we analysed the bacterial communities in stomach regurgitates and cloacal swabs from the same individual birds, freshly deposited guano and rookery soils of two Pygoscelis penguins that breed sympatrically on Signy Island (South Orkney Islands, maritime Antarctic) using a high-throughput DNA sequencing method. Our data do not support the hypothesis that bacteria transferred from penguin guts and/or deposited guano make a significant contribution to the communities of the surrounding terrestrial microbial ecosystem. In both penguin species, composition of bacterial communities differed between the four sample types, with Jaccard similarities ranging between 10 and 36%. Assemblages of the dominant and co-occurring bacterial communities in rookery soils were either significantly negatively correlated or not correlated with the three other sample types. Sample-specific communities were also identified in this study, contributing around 63% of the identified diversity overall.

Purpose: Inflammatory responses after ischemic stroke contribute to worsening of ischemic brain injury. Calcium release-activated calcium (CRAC) channels are known to be involved in the activation of immune responses in microglia, and have been studied indirectly through non-specific calcineurin inhibitors such as cyclosporine and FK506. Since these compounds have many off-target effects and unacceptable side effects, we studied a novel CRAC channel inhibitor which is specific to the CRAC channel and appears safe in preclinical studies in anticipation of human clinical trials for other indications. We explored whether this inhibitor might improve outcome in experimental stroke by inhibiting microglial activation. Subjects and Methods: 2-3 month old C57/BL6 male mice, were exposed to transient middle cerebral artery occlusion (tMCAO, filament model) for 2h followed by reperfusion. Some were treated with the CRAC channel inhibitor (CM-EX-137, 5mg/kg IP; tMCAO-CM) or vehicle control (tMCAO) daily beginning immediately after occlusion and the following two days. Brains were collected 3d post-stroke. Neurological function was evaluated using a Bederson score, elevated body swing test, adhesive removal test, and corner test at baseline, at 3, 24 and 72 h. Relative cerebral blood flow (rCBF) was studied in some mice using laser Doppler flowmetry. Ischemic lesion size was evaluated by 2,3,5-triphenyl tetrazolium chloride (TTC) staining. Microglial/monocyte activation was assessed by isolectin B4 histochemistry. Results: A total of 18 mice were studied (tMCAO-CM: 8; tMCAO: 8, sham:2). rCBF was no different between experimental groups, and is consistent with the notion that the compound should not alter blood flow. Neurological function at 1 & 3d after ischemia was significantly improved in tMCAO-CM group compared to tMCAO (p Conclusion: CRAC channel inhibition protects against experimental stroke by decreasing lesion size, improving neurological outcomes and decreasing microglial activation.

Reproductive timing in many taxa plays a key role in determining breeding productivity , and is often sensitive to climatic conditions . Current climate change may alter the timing of breeding at different rates across trophic levels, potentially resulting in temporal mismatch between the resource requirements of predators and their prey . This is of particular concern for highertrophic-level organisms, whose longer generation times confer a lower rate of evolutionary rescue than primary producers or consumers. However, the disconnection between studies of ecological change in marine systems makes it difficult to detect general changes in the timing of reproduction5 . Here, we use a comprehensive meta-analysis of 209 phenological time series from 145 breeding populations to show that, on average, seabird populations worldwide have not adjusted their breeding seasons over time (−0.020 days yr−1 ) or in response to sea surface temperature (SST) (−0.272 days °C−1 ) between 1952 and 2015. However, marked between-year variation in timing observed in resident species and some Pelecaniformes and Suliformes (cormorants, gannets and boobies) may imply that timing, in some cases, is affected by unmeasured environmental conditions. Knowing the spatial scales at which effective management can be implemented is fundamental for conservation planning. This is especially important for mobile species, which can be exposed to threats across large areas, but the space use requirements of different species can vary to an extent that might render some management approaches inefficient. Here the space use patterns of seabirds were examined to provide guidance on whether conservation management approaches should be tailored for taxonomic groups with different movement characteristics. Seabird tracking data were synthesised from 5419 adult breeding individuals of 52 species in ten families that were collected in the Atlantic Ocean basin between 1998 and 2017. Two key aspects of spatial distribution were quantified, namely how far seabirds ranged from their colony, and to what extent individuals from the same colony used the same areas at sea. There was evidence for substantial differences in patterns of space-use among the ten studied seabird families, indicating that several alternative conservation management approaches are needed. Several species exhibited large foraging ranges and little aggregation at sea, indicating that area-based conservation solutions would have to be extremely large to adequately protect such species. The results highlight that short-ranging and aggregating species such as cormorants, auks, some penguins, and gulls would benefit from conservation approaches at relatively small spatial scales during their breeding season. However, improved regulation of fisheries, bycatch, pollution and other threats over large spatial scales will be needed for wide-ranging and dispersed species such as albatrosses, petrels, storm petrels and frigatebirds. Penguin stomach microbiota and its variability are important as these microbes may contribute to the fitness of the host birds and their chicks, and influence the microbial ecosystem of the surrounding soils. However, there is relatively little knowledge in this area, with the majority of studies focused on their deposited faeces. Here we investigated whether similar foraging strategies in adjacent colonies of different penguin species lead to similar temporarily conserved stomach microbiota. To do this, we studied the inter- and intra-specific variations in bacterial community composition in the stomach contents of sympatrically breeding Adélie (Pygoscelis adeliae) and Chinstrap (Pygoscelis antarctica) Penguins, which consumed a diet of 100% Antarctic krill (Euphausia superba) under a similar foraging regime on Signy Island (maritime Antarctic), using a high-throughput DNA sequencing approach. Our data show that Adélie and Chinstrap Penguins shared 23–63% similarity in the stomach bacterial community composition, with no significant differences observed in the α-diversity or the assemblages of frequently encountered groups of operational taxonomic units (OTUs). The most frequently encountered OTUs that were shared between the species represented members of the phyla Fusobacteria, Firmicutes, Tenericutes and Proteobacteria. OTUs which were unique to individual birds and to single species formed approximately half of the communities identified, suggesting that stomach microbiota variability can occur in penguins that forage and breed under similar environmental conditions.

Long-term monitoring of seabird numbers around Antarctica has revealed that the chinstrap penguin (Pygoscelis antarctica) is largely declining throughout its range in the Scotia Arc. Whether archipelagos across this large area remain connected via dispersal or represent genetically isolated groups has not yet been established. The purpose of this study was to assess the level of genetic differentiation between four breeding colonies on the Western Antarctic Peninsula (WAP), South Shetland, South Orkney, and South Sandwich Islands using microsatellite-based analysis of population structure. All colonies had similar levels of genetic diversity (mean heterozygosity, H O = 0.583) but colonies from the WAP and South Orkney Island had significant inbreeding coefficients. Hierarchical and pairwise F-statistics revealed very limited population structure in the Scotia Arc, with weak differentiation between colonies from the WAP, South Shetland and South Orkney Islands relative to the South Sandwich Islands, which are situated at least 1000 km apart from these other archipelagos. Bayesian model-based clustering methods found no evidence of significant population structuring, suggesting that whilst some isolation by distance may occur, there are no strong barriers to dispersal across this wide geographic range. No evidence of sex-biased dispersal was detected. We conclude that chinstrap penguin colonies across the Scotia Arc represent one interconnected breeding population. High levels of gene flow may be important in maintaining smaller, less stable colonies, and this status should be preserved by creating dispersal corridors throughout the Scotia Arc.

An estimated 293,000 people living in the United Kingdom have used anabolic-androgenic steroids. However, there is currently no intervention to reduce usage available in practice or academic circulation throughout the United Kingdom. This study aimed to test a novel hypothesis that increased levels of intrasexual competition may play an important influential role in the use of anabolic-androgenic steroids. Significantly higher levels of intrasexual competition were evident in users compared to non-users but only in the novice group (0–2 years of experience). The research provides evidence for intrasexual competition potentially influencing anabolic-androgenic steroid use but only during the initial stages of usage.

Inflammatory responses following ischemia can worsen neurological outcome, and represent a potential target for therapeutic intervention. Store-operated Ca 2+ entry (SOCE) mediated by CRAC channels contribute to calcium signaling in immune cells. CRAC channels consist of the endoplasmic reticulum resident Ca 2+ -sensing protein stromal interaction molecule 1 (STIM1) and the calcium channel protein ORAI1 located in the plasma membrane. Prolonged Ca 2+ entry through CRAC channels activates, via calcineurin, nuclear factor of activated T cells (NFAT), involved in T cell proliferation and cytokine expression. Microglia mediate inflammation in the injured brain, but little is known about the role of CRAC channels in this process. We studied novel CRAC channel inhibitors to explore their therapeutic potential in microglia-mediated injury. A neuron cell line (Neuro-2A, N-2A) was cultured alone or with microglial BV2 cells then exposed to 2h oxygen glucose deprivation (OGD). Some cultures were treated with a novel CRAC channel inhibitor. Toll-like receptor (TLR) -3, -4 agonists or IFNγ were also used to activate microglia. Western blots revealed the presence of CRAC channel proteins STIM1 and ORAI1 in microglia. CRAC channel inhibition decreased NO release and inflammatory proteins iNOS and COX-2 expression in activated microglia, but did not affect STIM1 or ORAI1 expression. CRAC channel inhibitors also reduced agonist induced intracellular calcium accumulation in BV2 cells. Agonists also activated JNK1/2 kinase, NFAT, NF-κB, CREB & STAT1 in microglia, but only JNK1/2 kinase & NFAT were attenuated by inhibitor. International Plant Proteomics Organization (INPPO) outlined ten initiatives to promote plant proteomics in each and every country. With greater emphasis in developing countries, one of those was to "organize workshops at national and international levels to train manpower and exchange information". This third INPPO highlights covers the workshop organized for the very first time in a developing country, India, at the Department of Botany in University of Delhi on December 26-30, 2013 titled - "1(st) Plant Proteomics Workshop / Training Program" under the umbrella of INPPO India-Nepal chapter. Selected 20 participants received on-hand training mainly on gel-based proteomics approach along with manual booklet and parallel lectures on this and associated topics. In house, as well as invited experts drawn from other Universities and Institutes (national and international), delivered talks on different aspects of gel-based and gel-free proteomics. Importance of gel-free proteomics approach, translational proteomics, and INPPO roles were presented and interactively discussed by a group of three invited speakers Drs. Ganesh Kumar Agrawal (Nepal), Randeep Rakwal (Japan), and Antonio Masi (Italy). Given the output of this systematic workshop, it was proposed and thereafter decided to be organized every alternate year; the next workshop will be held in 2015. Furthermore, possibilities on providing advanced training to those students / researchers / teachers with basic knowledge in proteomics theory and experiments at national and international levels were discussed. INPPO is committed to generating next-generation trained manpower in proteomics, and it would only happen by the firm determination of scientists to come forward and do it.

Manipulated prestige car ownership has been shown previously to enhance male attractiveness. In the current study the illusion of status-linked property ownership was achieved by presenting a target male and female (matched for attractiveness) adopting a casual posture standing in either a ‘high status’ (luxury apartment) or a ‘neutral status’ (standard apartment) context. A between-subjects design was used with the photographic stimuli presented amongst other masking stimuli and rated for attractiveness by opposite-sex university undergraduate participants (N = 102) on a scale of 1–10. The male model was rated significantly more attractive when presented to females in the high status compared to the neutral status context and was also adjudged to be significantly more attractive than the female target superimposed on an identical background. Prenatal stress influences the development of the fetal brain and so contributes to the risk of the development of psychiatric disorders in later life. The hippocampus is particularly sensitive to prenatal stress, and robust abnormalities have been described in the hippocampus in schizophrenia and depression. The aim of this study was to determine whether prenatal stress is associated with distinct patterns of differential protein expression in the hippocampus using a validated mouse model. We therefore performed a comparative proteomic study assessing female hippocampal samples from 8 prenatally stressed mice and 8 control mice. Differential protein expression was assessed using 2-dimensional difference in gel electrophoresis and subsequent mass spectrometry. The observed changes in a selected group of differentially expressed proteins were confirmed by Western blotting. In comparison to controls, 47 protein spots (38 individual proteins) were found to be differentially expressed in the hippocampus of prenatally stressed mice. Functional grouping of these proteins revealed that prenatal stress influenced the expression of proteins involved in brain development, cytoskeletal composition, stress response, and energy metabolism. Western blotting was utilized to validate the changes in calretinin, hippocalcin, profilin-1 and the signal-transducing adaptor molecule STAM1. Septin-5 could not be validated via Western blotting due to methodological issues. Closer investigation of the validated proteins also pointed to an interesting role for membrane trafficking deficits mediated by prenatal stress. Our findings demonstrate that prenatal stress leads to altered hippocampal protein expression, implicating numerous molecular pathways that may provide new targets for psychotropic drug development.

We report long-term changes in population size of three species of sympatrically breeding pygoscelid penguins: Adelie (Pygoscelis adeliae), chinstrap (Pygoscelis antarctica) and gentoo (Pygoscelis papua ellsworthii) over a 38 year period at Signy Island, South Orkney Islands, based on annual counts from selected colonies and decadal all-island systematic counts of occupied nests. Comparing total numbers of breeding pairs over the whole island from 1978/79 to 2015/16 revealed varying fortunes: gentoo penguin pairs increased by 255%, (3.5% per annum), chinstrap penguins declined by 68% (-3.6% per annum) and Adelie penguins declined by 42% (-1.5% per annum). The chinstrap population has declined steadily over the last four decades. In contrast, Adelie and gentoo penguins have experienced phases of population increase and decline. Annual surveys of selected chinstrap and Adelie colonies produced similar trends from those revealed by island-wide surveys, allowing total island population trends to be inferred relatively well. However, while the annual colony counts of chinstrap and Adelie penguins showed a trend consistent in direction with the results from all-island surveys, the magnitude of estimated population change was markedly different between colony wide and all island counts. Annual population patterns suggest that pair numbers in the study areas partly reflect immigration and emigration of nesting birds between different parts of the island. Breeding success for all three species remained broadly stable over time in the annually monitored colonies. Breeding success rates in gentoo and chinstrap penguins were strongly correlated, despite the differing trends in population size. This study shows the importance of effective, standardised monitoring to accurately determine long-term population trajectories. Our results indicate significant declines in the Adelie and chinstrap penguin populations at Signy Island over the last five decades, and a gradual increase in gentoo breeding pairs.

Electroconvulsive therapy (ECT) is the most effective treatment for severe depression, yet its mechanism of action is not fully understood. Peripheral blood proteomic analyses may offer insights into the molecular mechanisms of ECT. Patients with a major depressive episode were recruited as part of the EFFECT-Dep trial (enhancing the effectiveness of electroconvulsive therapy in severe depression; ISRCTN23577151) along with healthy controls. As a discovery-phase study, patient plasma pre-/post-ECT (n=30) was analyzed using 2-dimensional difference in gel electrophoresis and mass spectrometry. Identified proteins were selected for confirmation studies using immunodetection methods. Samples from a separate group of patients (pre-/post-ECT; n=57) and matched healthy controls (n=43) were then used to validate confirmed changes. Target protein mRNA levels were also assessed in rat brain and blood following electroconvulsive stimulation (ECS), the animal model of ECT. We found that ECT significantly altered 121 protein spots with 36 proteins identified by mass spectrometry. Confirmation studies identified a post-ECT increase (PPP=0.03). PEDF levels were not associated with mood scores. Chronic, but not acute, ECS increased PEDF mRNA in rat hippocampus (P=0.02) and dentate gyrus (P=0.03). This study identified alterations in blood levels of PEDF in depressed patients and further alterations following ECT, as well as in an animal model of ECT. These findings implicate PEDF in the biological response to ECT for depression.

Regulation of protein function by reversible cysteine-targeted oxidation can be achieved by multiple mechanisms, such as S-glutathiolation, S-nitrosylation, sulfenic acid, sulfinic acid, and sulfenyl amide formation, as well as intramolecular disulfide bonding of vicinal thiols. Another cysteine oxidation state with regulatory potential involves the formation of intermolecular protein disulfides. We utilized two-dimensional sequential non-reducing/reducing SDS-PAGE (diagonal electrophoresis) to investigate intermolecular protein disulfide formation in adult cardiac myocytes subjected to a series of interventions (hydrogen peroxide, S-nitroso-N-acetylpenicillamine, doxorubicin, simulated ischemia, or metabolic inhibition) that alter the redox status of the cell. More detailed experiments were undertaken with the thiol-specific oxidant diamide (5 mm), a concentration that induces a mild non-injurious oxidative stress. This increase in cellular oxidation potential caused global intermolecular protein disulfide formation in cytosolic, membrane, and myofilament/cytoskeletal compartments. A large number of proteins that undergo these associations were identified using liquid chromatography-mass spectrometry/mass spectrometry. These associations, which involve metabolic and antioxidant enzymes, structural proteins, signaling molecules, and molecular chaperones, were confirmed by assessing "shifts" on non-reducing immunoblots. The observation of widespread protein-protein disulfides indicates that these oxidative associations are likely to be fundamental in how cells respond to redox changes.

Abnormalities of the anterior cingulate cortex have previously been described in schizophrenia, major depressive disorder and bipolar disorder. In this study 2-DE was performed followed by mass spectrometric sequencing to identify disease-specific protein changes within the anterior cingulate cortex in these psychiatric disorders. The 2-DE system comprised IPGs 4-7 and 6-9 in the first, IEF dimension and SDS-PAGE in the second dimension. Resultant protein spots were compared between control and disease groups. Statistical analysis indicated that 35 spots were differentially expressed in one or more groups. Proteins comprising 26 of these spots were identified by mass spectroscopy. These represented 19 distinct proteins; aconitate hydratase, malate dehydrogenase, fructose bisphosphate aldolase A, ATP synthase, succinyl CoA ketoacid transferase, carbonic anhydrase, alpha- and beta-tubulin, dihydropyrimidinase-related protein-1 and -2, neuronal protein 25, trypsin precursor, glutamate dehydrogenase, glutamine synthetase, sorcin, vacuolar ATPase, creatine kinase, albumin and guanine nucleotide binding protein beta subunit. All but three of these proteins have previously been associated with the major psychiatric disorders. These findings provide support for the view that cytoskeletal and mitochondrial dysfunction are important components of the neuropathology of the major psychiatric disorders.

Experimental studies have suggested that protective genes protect allografts from cardiac allograft vasculopathy (CAV), the major complication after cardiac transplantation. Here we have sought to confirm this hypothesis using long-term heart transplant recipients. Twenty-two patients that were 9 years or older after transplant were investigated; 11 of these were without angiographic evidence of CAV; 11 had developed early CAV at 1 to 3 years after transplant. To identify proteins that may act as protectors from CAV, a global proteomic approach was used comparing cardiac biopsies from 12 patients taken within the first 2 weeks after transplant and those taken after 9 years from the same patient. Proteins were separated by 2-D gel-electrophoresis, detected by silver staining, and analyzed using Progenesis software. A particular protein spot was found in 4/6 biopsies from patients without CAV, but absent from 5/6 biopsies from those with CAV ( P =0.24); however, quantitative analysis of spot intensity showed a significant difference (0.061±0.05 versus 0.003±0.01, P =0.04). This spot was identified by mass spectrometry and a combination of techniques as a diphosphorylated form of HSP27. Immunohistochemistry of further biopsies not only validated that HSP27 was more abundantly expressed on biopsies without CAV but also showed it to be localized to blood vessels. In contrast, vessels from patients with CAV did not express HSP27 ( P =0.028×10 −4 ). Immunohistochemistry of 12 further early biopsies and nontransplanted heart showed HSP27 to be present in normal blood vessels. These findings suggest that expression of a specific diphosphorylated form of HSP27 is associated with healthy blood vessels; it appears to be lost from vessels of patients with graft vasculopathy.
