The protection against diabetes and cardiovascular disease provided by whole-grain cereal consumption has been attributed to the fiber and micronutrients present in the bran. But exactly how this occurs remains unclear due to both diversity of bran constituents and the complexity of the metabolic responses to each of these constituents. We investigated the metabolic responses of 2 groups of rats (n = 10/group) fed 2 diets, for 2 wk each, in a crossover design. One diet contained 60 g/100 g whole-grain wheat flour (WGF) and the other contained 60 g/100 g refined wheat flour (RF). Markers of oxidative stress [urinary isoprostanes and malondialdehydes (MDA), plasma ferric-reducing ability of plasma, MDA, and vitamins E and C] and lipid status (liver and plasma triglycerides and cholesterol) were measured. Urine samples collected during the feeding periods and plasma and liver samples collected at the end of experiment were analyzed by [sup.1.H] NMR spectroscopy. Metabonomic analyses showed that each group reached a new metabolic balance within 48 h of changing the diet. Urinary excretion of some tricarboxylic acid cycle intermediates, aromatic amino acids, and hippurate was significantly greater in rats fed the WGF diet. Although the diets did not affect conventional lipid and oxidative stress markers, there were decreases in some liver lipids and increases in liver reduced glutathione and betaine as shown by metabonomic analyses. Magnesium (Mg) status is currently assessed by various biochemical biomarkers, most of which, however, have some limitations. We developed an in vitro blood load test as a new Mg biomarker using a Mg stable isotope. This test is based on the hypothesis that cellular Mg uptake is increased in Mg deficiency. For this purpose, Wister male rats were fed either Mg-deficient or Mg-adequate diets for I mo and blood was sampled and incubated with the [sup.25]Mg isotope (10 mg/L) for 2 h at 37[degrees]C. Erythrocytes, lymphocytes and platelets were isolated and [sup.25]Mg concentrations were determined by inductively coupled plasma/mass spectrometry. The feasibility of this approach was then tested on human blood. [sup.25]Mg enrichments in erythrocytes, lymphocytes and platelets from Mg-deficient rats were greater than those from controls. [sup.25]Mg enrichment was low in human erythrocytes (3%) compared with rat erythrocytes (38%), whereas high [sup.25]Mg enrichments were obtained in human lymphocytes and platelets, suggesting that lymphocytes and platelets may be more appropriate cells than erythrocytes for examining Mg status in humans with this approach. Apples may have selective effects on abnormalities associated with the plurimetabolic syndrome. Therefore, the effects of 20% lyophilized apple supplementation on plasma and tissue lipids and on protection against susceptibility to oxidative stress and renal dysfunction were investigated in Zucker lean (Fa/-) or obese (fa/fa) rats. The experimental diets were equilibrated for sugar supply, contained 0.25 g/100 g cholesterol and provided only one third of the vitamin E requirement. Obese Zucker rats were hypercholesterolemic with cholesterol accumulation in LDL and HDL fractions. The apple diet lowered plasma and LDL cholesterol (-22 and -70%, respectively, P < 0.01) in obese Zucker rats and, in parallel, reduced triglyceride accumulation in heart and liver. Zucker rats fed the apple diet also had a larger intestinal pool and greater fecal excretion of bile acids. The heart concentration and urinary excretion of malondialdehyde were reduced by apple consumption in obese Zucker rats, suggesting better protection against peroxidation. Glucosuria and proteinuria in obese Zucker rats were also suppressed by the apple diet. In conclusion, despite their moderate fiber content, apples improve substantially the lipid status and peroxidative parameters in obese Zucker rats, suggesting that other plant constituents such as polyphenols are involved in these effects. More and more studies are accumulating about COVID-19. Some aspects of the pathogenesis of the disease recall events occurring in Mg deficiency, such as a drop of T cells, increased plasma concentration of inflammatory cytokines, and endothelial dysfunction. We hypothesize that a low Mg status, which is rather common, might foment the transition from mild to critical clinical manifestations of the disease. Epidemiological, clinical, and fundamental research is needed to clarify the potential role of Mg deficiency in COVID-19. [ABSTRACT FROM AUTHOR]

The article presents the magnesium global network to promote research on magnesium in diseases focusing on covid-19. Topics discussed include this prompted establishment of a worldwide collaborative network with regular virtual meetings to brainstorm the associations between magnesium and COVID-19; and hypothesize that magnesium deficiency, a common but mostly unrecognized state in modern global societies, could be an important component of the susceptibility to SARS-CoV-2 infection.
