- Abstract
The Jumonji domain-containing protein JMJD6 is a 2-oxoglutarate-dependent dioxygenase associated with a broad range of biological functions. Cellular studies have implicated the enzyme in chromatin biology, transcription, DNA repair, mRNA splicing, and cotranscriptional processing. Although not all studies agree, JMJD6 has been reported to catalyze both hydroxylation of lysine residues and demethylation of arginine residues. However, despite extensive study and indirect evidence for JMJD6 catalysis in many cellular processes, direct assignment of JMJD6 catalytic substrates has been limited. Examination of a reported site of proline hydroxylation within a lysine-rich region of the tandem bromodomain protein BRD4 led us to conclude that hydroxylation was in fact on lysine and catalyzed by JMJD6. This prompted a wider search for JMJD6-catalyzed protein modifications deploying mass spectrometric methods designed to improve the analysis of such lysine-rich regions. Using lysine derivatization with propionic anhydride to improve the analysis of tryptic peptides and nontryptic proteolysis, we report 150 sites of JMJD6-catalyzed lysine hydroxylation on 48 protein substrates, including 19 sites of hydroxylation on BRD4. Most hydroxylations were within lysine-rich regions that are predicted to be unstructured; in some, multiple modifications were observed on adjacent lysine residues. Almost all of the JMJD6 substrates defined in these studies have been associated with membraneless organelle formation. Given the reported roles of lysine-rich regions in subcellular partitioning by liquid-liquid phase separation, our findings raise the possibility that JMJD6 may play a role in regulating such processes in response to stresses, including hypoxia.

- Abstract
The study’s objective is to evaluate the main legal difficulties and practical challenges in the cooperation within the EU in the field of the protection of vulnerable adults, and to assess the need for and the possible added value of a common legal framework for the protection of vulnerable adults in cross-border cases, either through mandatory ratification of the Hague Convention of 13 January 2000 on the International Protection of Adults or via the adoption of an EU instrument to reinforce and streamline the judicial cooperation in that area. To this end, the study examines the national laws and practices of the Member States with regard to the roles and functioning of competent authorities, the different types of measures of protection provided for in the national legal systems, the main procedural rules applying to these measures, as well as the private international law rules in place for cross-border cases. The data presented in the report is based on desk research, as well as on the results of an online survey targeted at national competent authorities, legal practitioners and other stakeholders involved in the protection of vulnerable adults (e.g. NGOs, service providers). On the basis of the data collected, and in particular the information gathered on the existing problems in judicial cooperation between Member States and disparities in the current level of protection of vulnerable adults across the EU stemming from the absence of a common legal framework, the need, added value and feasibility of further action at EU level, which may take the form of a mandatory ratification of the Convention and/or of EU legislation on the matter, was assessed.

- Abstract
Cooling is needed to protect vulnerable populations from heatwaves, keep vaccines viable and food fresh, and workforces productive. It is essential for equity and development, especially as climate change raises global temperatures. There are an estimated 3.6 billion cooling appliances in use globally today, and that number is growing by up to 10 devices every second. This growth is set to increase the sector’s greenhouse gas emissions dramatically. Air conditioners are a double burden. In most cases, they use hydrofluorocarbons (HFCs), extremely potent greenhouse gases, and require significant energy to run. Without policy intervention, direct and indirect emissions from air conditioning and refrigeration are projected to rise 90 per cent above 2017 levels by the year 2050. This report lays out ways to resolve this dilemma by delivering efficient and climate friendly cooling for all – in particular by rapidly phasing down hydrofluorocarbons in the cooling sector and delivering cooling more efficiently through more efficient equipment and more efficient buildings.

We have measured carbon nanotube quantum dots with multiple electrostatic gates and used the resulting enhanced control to investigate a nanotube double quantum dot. Transport measurements reveal honeycomb charge stability diagrams as a function of two nearly independent gate voltages. The device can be tuned from weak to strong interdot tunnel-coupling regimes, and the transparency of the leads can be controlled independently. We extract values of energy-level spacings, capacitances, and interaction energies for this system. 15. The activity of JmjC histone lysine demethylase KDM4A is highly sensitive to oxygen concentrations

- Abstract
The JmjC histone lysine demethylases (KDMs) are epigenetic regulators involved in the removal of methyl groups from post-translationally modified lysyl residues within histone tails, modulating gene transcription. These enzymes require molecular oxygen for catalytic activity and, as 2-oxoglutarate (2OG)-dependent oxygenases, are related to the cellular oxygen sensing HIF hydroxylases PHD2 and FIH. Recent studies have indicated that the activity of some KDMs, including the pseudogene-encoded KDM4E, may be sensitive to changing oxygen concentrations. Here, we report detailed analysis of the effect of oxygen availability on the activity of the KDM4 subfamily member KDM4A, importantly demonstrating a high level of O2 sensitivity both with isolated protein and in cells. Kinetic analysis of the recombinant enzyme revealed a high KMapp(O2) of 173 ± 23 μM, indicating that the activity of the enzyme is able to respond sensitively to a reduction in oxygen concentration. Furthermore, immunofluorescence experiments in U2OS cells conditionally overexpressing KDM4A showed that the cellular activity of KDM4A against its primary substrate, H3K9me3, displayed a graded response to depleting oxygen concentrations in line with the data obtained using isolated protein. These results suggest that KDM4A possesses the potential to act as an oxygen sensor in the context of chromatin modifications, with possible implications for epigenetic regulation in hypoxic disease states. Importantly, this correlation between the oxygen sensitivity of the catalytic activity of KDM4A in biochemical and cellular assays demonstrates the utility of biochemical studies in understanding the factors contributing to the diverse biological functions and varied activity of the 2OG oxygenases.

Aims: The objective of the study is to determine the demographics and the in-hospital outcome of diabetic and non-diabetic patients treated with percutaneous coronary interventions (PCI) in Europe, to report the type of equipment and technology used for PCI procedures in diabetics and to clarify whether the treatment of diabetic patients complies with current European Society of Cardiology (ESC) guidelines. Methods and results: A total of 14,458 patients treated with PCI were enrolled from 29 member countries of the ESC between June 2005 and January 2006. Data were collected on patient characteristics and treatment, using new Cardiology Audit and Registration Data standards. In total, 3,603 patients (24.9%) were diabetic. Diabetics were older, more often female and had a higher body mass index than non-diabetics. Diabetics had higher rates of hypercholesterolaemia and hypertension, while current smokers were more frequent in the non-diabetics. Diabetics also had significantly higher rates of previous cardiovascular events. Clopidogrel was administered only in 48.1% of diabetic patients before PCI, while IIb/IIIa inhibitors were 22.9% during PCI. At discharge, there was a major adjustment of treatment with increases in the use of Beta-blocker (80.4%), angiotensin converting enzyme inhibitor (ACEI, 71.3%) and statins (89.8%) compared with on admission (Beta-blocker 60.9%, ACEI 55.0%, statin 63.1%). Inhospital mortality was higher in diabetics (1.8% vs 1.2%) although the in-hospital MACCE rate was not significantly different (3.6% vs 3.0%, p=0.09). Conclusions: Diabetic patients treated with PCI were older with more comorbidity. According to ESC guideline, the under-usage of clopidogrel, GP IIb/IIIa inhibitors should be improved. PCI is now taken as a good opportunity to adjust the use of appropriate medication. © Europa Edition. All rights reserved.

18. Striking oxygen sensitivity of the peptidylglycine a-amidating monooxygenase (PAM) in neuroendocrine cells

- Abstract
Interactions between biological pathways and molecular oxygen require robust mechanisms for detecting and responding to changes in cellular oxygen availability, to support oxygen homeostasis. Peptidylglycine α-amidating monooxygenase (PAM) catalyzes a two-step reaction resulting in the C-terminal amidation of peptides, a process important for their stability and biological activity. Here we show that in human, mouse, and insect cells, peptide amidation is exquisitely sensitive to hypoxia. Different amidation events on chromogranin A, and on peptides processed from proopiomelanocortin, manifest similar striking sensitivity to hypoxia in a range of neuroendocrine cells, being progressively inhibited from mild (7% O2) to severe (1% O2) hypoxia. In developing Drosophila melanogaster larvae, FMRF amidation in thoracic ventral (Tv) neurons is strikingly suppressed by hypoxia. Our findings have thus defined a novel monooxygenase-based oxygen sensing mechanism that has the capacity to signal changes in oxygen availability to peptidergic pathways.

We have characterized the gamma and X-ray scintillation properties of [Ce.sup.3+] doped [Lu.sub.2][Si.sub.2][O.sub.7], a lutetium pyro-silicate (LPS) material. The rare earth ions are located in an octahedral site with [C.sub.2] symmetry. The compound exhibits chemical stability, transparency in a wide optical range and congruent melting, which allows growing of single crystals of good optical quality. The melting temperature is about 2000 [degrees] C, slightly lower than that of [Lu.sub.2]Si[O.sub.5] (LSO). Two maxima at 305 nm and 355 nm are observed in the optical absorption spectrum. The emission spectrum of [Ce.sup.3+] in LPS shows a broad band peaking at 380 nm. For a nominal [Ce.sup.3+] atomic concentration of ~0.01, under gamma-ray excitation we observe light yields in the range 13,000-23,000 photons/MeV, depending on the preparation atmosphere. The actual cerium concentration inside the crystal still needs to be determined. The scintillation decay time is around 30 ns and, in the first studies, neither a long component nor afterglow was observed.

Objective: Self-perceived health status may be helpful in identifying patients at high risk for adverse outcomes. The Euro Heart Survey on Coronary Revascularization (EHS-CR) provided an opportunity to explore whether impaired health status was a predictor of 1-year mortality in patients with coronary artery disease (CAD) undergoing angiographic procedures. Methods: Data from the EHS-CR that included 5619 patients from 31 member countries of the European Society of Cardiology were used. Inclusion criteria for the current study were completion of a self-report measure of health status, the EuroQol Questionnaire (EQ-5D) at discharge and information on 1-year follow-up, resulting in a study population of 3786 patients. Aims: Revascularization in patients with coronary artery disease changed over the last two decades, favouring the number of patients treated by means of percutaneous coronary interventions (PCI) when compared with coronary artery bypass grafting (CABG). Many randomized controlled trials (RCTs) have been performed to compare these two competing revascularization techniques. Because of the strict enrolment criteria of RCTs in which highly selected patients are recruited, the applicability of the results may be limited in clinical practice. The current study evaluates to what extent patients in clinical practice were similar to those who participated in RCTs comparing PCI with CABG. Methods and results: Clinical characteristics and 1-year outcome of 4713 patients enrolled in the Euro Heart Survey on Coronary Revascularization were compared with 8647 patients who participated in 14 major RCTs, comparing PCI with CABG. In addition, we analysed which proportion of survey patients would have disqualified for trial participation (n = 3033, 64%), aiming at identifying differences between trial-eligible and trial-ineligible survey patients. In general, important differences were observed between trial participants and survey patients. Patients in clinical practice were older, more often had comorbid conditions, single-vessel disease, and left main stem stenosis when compared with trial participants. Almost identical differences were observed between trial-eligible and trial-ineligible survey patients. In clinical practice, PCI was the treatment of choice, even in patients who were trial-ineligible (46% PCI, 26% CABG, 28% medical). PCI remained the preferred treatment option in patients with multi-vessel disease (57% in trial-eligible and 40% in trial-ineligible patients, respectively, P < 0.001); yet, the risk profile of patients treated by PCI was better than that for patients treated either by CABG or by medical therapy. In the RCTs, there was no mortality difference between PCI and CABG. In clinical practice, however, we observed 1-year unadjusted survival benefit for PCI vs. CABG (2.9 vs. 5.4%, P < 0.001). Survival benefit was only observed in trial-ineligible patients (3.3 vs. 6.2%, P < 0.001). Conclusion: Many patients in clinical practice were not represented in RCTs. Moreover, only 36% of these patients were considered eligible for participating in a trial comparing PCI with CABG. We demonstrated that RCTs included younger patients with a better cardiovascular risk profile when compared with patients in everyday clinical practice. This study highlights the disparity between patients in clinical practice and patients in whom the studies that provide the evidence for treatment guidelines are performed. © The European Society of Cardiology 2006. The overriding aim of this project is to develop guidelines for proper land use in agriculture in terms of sustainability. We analyse the bottom-up capacities for sustainable agricultural development in the Alps by specifying the conditions of sustainability in a mountain context on the basis of the points of view of both local people and scientists. The issues of sustainability are defined according to ecological, economic and social criteria, with regional adaptations to the specific conditions of the Alpine mountain regions. Five areas in the Alps are studied in this project, namely Oberes Drautal (Austria), Moyenne Tarentaise (France), Ostallgäu (Germany), Bassa Valle di Sole (Italy) and Glarner Hinterland (Switzerland).The results of SAGRI-ALP are : - a comparative analysis of the limits of sustainability, its potential and trends in the five areas, at farm level and at regional level; - regional guidelines with concrete indicators, objectives and plans of action to implement sustainable agriculture in the five research areas ; - final recommendations for the European Commission to implement a regionally adapted policy concerning sustainable agricultural land use in the Alps ; - an Alpine guideline to promote and build up local projects involving local people and targeting sustainable agriculture., L'objectif principal de ce projet de recherche européen est de proposer des recommandations en vue d'une utilisation de l'espace cohérente avec une agriculture durable. Les points de vue de scientifiques et d'acteurs locaux ont été confrontés afin de spécifier les conditions de développement d'une agriculture durable dans les Alpes. Des indicateurs écologiques, économiques et sociaux ont été élaborés en tenant compte des caractéristiques spécifiques des régions alpines. Cinq zones d'étude ont été choisies : Oberes Drautal en Autriche, Moyenne Tarentaise en France, Ostallgäu en Allemagne, Bassa Valle di Sole en Italie et Glarner Hinterland en Suisse. Les principaux résultats de ce projet portent sur : - une analyse comparative des points forts et des points faibles de l'agriculture alpine en termes de durabilité et des effets de différents scénarios de politiques agricoles, à l'échelle des cinq zones d'étude et pour les principaux types d'exploitations agricoles ; - des recommandations au plan régional avec une liste concrète d'indicateurs, d'objectifs et de plans d'actions pour développer une agriculture durable dans les cinq zones d'étude ; - des recommandations pour la Commission européenne pour mettre en oeuvre des politiques régionales adaptées en vue d'une agriculture durable ; - la proposition d'une démarche à destination des acteurs pour concevoir et mettre en oeuvre des projets locaux afin de favoriser une agriculture durable.

We sought to examine trainees' choices of treatment for psychosis if they had to prescribe it for themselves, their patients, and factors influencing decision-making.Methods: Cross-sectional, semi-structured questionnaire-based study.Results: Of the 726 respondents (response rate = 66%), the majority chose second-generation antipsychotics (SGAs) if they had to prescribe it for themselves (n = 530, 93%) or for their patients (n = 546, 94%). The main factor influencing choice was perceived efficacy, 84.8% (n = 475) of trainees stating this was the most important factor for the patient, and 77.8% (n = 404) stating this was the most important factor for their own treatment. - Abstract
The HOX11 homeobox gene was first identified through studies of the t(7;10) and t(10;14) chromosomal translocations of acute T-cell leukemia. In addition, analysis of Hox11(-/-) mice has demonstrated a critical role for this gene in murine spleen development. A possible mode of in vivo function for the HOX11 protein in these two situations is regulation of target genes following DNA binding via the homeodomain, but little is known about how HOX11 regulates transcription in vivo. By performing transcriptional studies in yeast and mammalian one-hybrid systems, a modular transcriptional transactivation region at the NH 2 terminus of HOX11 has been functionally dissected from other paris of the protein. This NH 2-terminal region includes the previously identified short conserved Hep motif, which itself activates transcription in one-hybrid assays. The importance of the NH 2-terminal region for the function of HOX11 in vivo was assayed by activating a HOX11- dependent gene in NIH 3T3 cells. Activation of this gene was found to be dependent upon an intact homeodomain in HOX11, but maximal activation was obtained only when the NH 2-terminal 50 amino acids of HOX11 was present, showing that this region of HOX11 is important for in vivo transcriptional control of a chromosomal target gene.

- Abstract
Hox11 is a homeobox gene essential for spleen formation in mice, since atrophy of the anlage of a developing spleen occurs in early embryonic development in Hox11 null mice. HOX11 is also expressed in a subset of T-cell acute leukemias after specific chromosomal translocations. Since the protein has a homeodomain and can activate transcription, it probably exerts at least some of its effects in vivo by regulation of target genes. Representational difference analysis has been used to isolate cDNA clones corresponding to mRNA species activated following stable expression of HOX11 in NIH 3T3 cells. The gene encoding the retinoic acid-synthesizing enzyme aldehyde dehydrogenase 1 (Aldh1), initially called Hdg-1, was found to be ectopically activated by HOX11 in this system. Study of Aldh1 gene expression during spleen development showed that the presence of Aldh1 mRNA inversely correlated with Hox11. Hox11 null mouse embryos have elevated Aldh1 mRNA in spleen primordia prior to atrophy, while Aldh1 seems to be repressed by Hox11 during organogenesis of the spleens of wild-type mice. This result suggests that expression of Aldh1 protein is negatively regulated by Hox11 and that abnormal expression of Aldh1 in Hox11 null mice may cause loss of splenic precursor cells by aberrant retinoic acid metabolism.
