4. A continuum from predictive to online feedback in visuomotor interception.

Interception, essential for activities like driving and sports, can be characterized by varying degrees of predictive behaviour. We developed a visually guided task to explore how target predictability and visibility influenced interception actions. The task featured a falling dot influenced by horizontal velocity, gravity and air friction, with predictability manipulated through external forces that altered the target's trajectory. We also introduced spatial occlusion to limit visual information. Our results show that low target variability favoured predictive behaviours, while high variability led to more reactive responses relying on online feedback. Manual responses displayed increased variability with changes in target motion, whereas eye trajectories maintained constant curvature across conditions. Additionally, higher target variability delayed the onset of hand movements but did not affect eye movement onset, making gaze position a poor predictor of hand position. This distinction highlights the different adaptive patterns in hand and eye movements in response to target trajectory changes. Participants maintained stable interception behaviours within and across sessions, indicating individual preferences for either predictive or more reactive actions. Our findings reveal a dynamic interplay between target predictability and interception, illustrating how humans combine predictive and reactive behaviours to manage external variability. - Abstract
Post‐translational modifications (PTMs) of proteins are ubiquitous processes present in all life kingdoms, involved in the regulation of protein stability, subcellular location and activity. In this context, cytochrome c (Cc) is an excellent case study to analyze the structural and functional changes induced by PTMS as Cc is a small, moonlighting protein playing different roles in different cell compartments at different cell‐cycle stages. Cc is actually a key component of the mitochondrial electron transport chain (ETC) under homeostatic conditions but is translocated to the cytoplasm and even the nucleus under apoptotic conditions and/or DNA damage. Phosphorylation does specifically alter the Cc redox activity in the mitochondria and the Cc non‐redox interaction with apoptosis‐related targets in the cytoplasm. However, little is known on how phosphorylation alters the interaction of Cc with histone chaperones in the nucleus. Here, we report the effect of Cc Tyr48 phosphorylation by examining the protein interaction with SET/TAF‐Iβ in the nuclear compartment using a combination of molecular dynamics simulations, biophysical and structural approaches such as isothermal titration calorimetry (ITC) and nuclear magnetic resonance (NMR) and in cell proximity ligation assays. From these experiments, we infer that Tyr48 phosphorylation allows a fine‐tuning of the Cc‐mediated inhibition of SET/TAF‐Iβ histone chaperone activity in vitro. Our findings likewise reveal that phosphorylation impacts the nuclear, stress‐responsive functions of Cc, and provide an experimental framework to explore novel aspects of Cc post‐translational regulation in the nucleus. Intercepting moving targets is a fundamental skill in human behavior, influencing various domains such as sports, gaming, and other activities. In these contexts, precise visual processing and motor control are crucial for adapting and navigating effectively. Nevertheless, there are still some gaps in our understanding of how these elements interact while intercepting a moving target. This study explored the dynamic interplay among eye movements, pupil size, and interceptive hand movements, with visual and motion uncertainty factors. We developed a simple visuomotor task in which participants used a joystick to interact with a computer-controlled dot that moved along two-dimensional trajectories. This virtual system provided the flexibility to manipulate the target's speed and directional uncertainty during chase trials. We then conducted a geometric analysis based on optimal angles for each behavior, enabling us to distinguish between simple tracking and predictive trajectories that anticipate future positions of the moving target. Our results revealed the adoption of a strong interception strategy as participants approached the target. Notably, the onset and amount of optimal interception strategy depended on task parameters, such as the target's speed and frequency of directional changes. Furthermore, eye-tracking data showed that participants continually adjusted their gaze speed and position, continuously adapting to the target's movements. Finally, in successful trials, pupillary responses predicted the amount of optimal interception strategy while exhibiting an inverse relationship in trials without collisions. These findings reveal key interactions among visuomotor parameters that are crucial for solving complex interception tasks. [ABSTRACT FROM AUTHOR]

- Abstract
Skeletal muscle is more resilient to ischemia-reperfusion injury than other organs. Tissue specific post-translational modifications of cytochrome c (Cytc) are involved in ischemia-reperfusion injury by regulating mitochondrial respiration and apoptosis. Here, we describe an acetylation site of Cytc, lysine 39 (K39), which was mapped in ischemic porcine skeletal muscle and removed by sirtuin5 in vitro. Using purified protein and cellular double knockout models, we show that K39 acetylation and acetylmimetic K39Q replacement increases cytochrome c oxidase (COX) activity and ROS scavenging while inhibiting apoptosis via decreased binding to Apaf-1, caspase cleavage and activity, and cardiolipin peroxidase activity. These results are discussed with X-ray crystallography structures of K39 acetylated (1.50 Å) and acetylmimetic K39Q Cytc (1.36 Å) and NMR dynamics. We propose that K39 acetylation is an adaptive response that controls electron transport chain flux, allowing skeletal muscle to meet heightened energy demand while simultaneously providing the tissue with robust resilience to ischemia-reperfusion injury.

Surprising violations of outcome expectancies have long been known to enhance the associability of Pavlovian cues; that is, the rate at which the cue enters into further associations. The adaptive value of such enhancements resides in promoting new learning in the face of uncertainty. However, it is unclear whether associability enhancements reflect increased associative plasticity within a particular behavior system, or whether they can facilitate learning between a cue and any arbitrary outcome, as suggested by attentional models of conditioning. Here, we show evidence consistent with the latter hypothesis. Violating the outcome expectancies generated by a cue in an appetitive setting (feeding behavior system) facilitated subsequent learning about the cue in an aversive setting (defense behavior system). In addition to shedding light on the nature of associability enhancements, our findings offer the neuroscientist a behavioral tool to dissociate their neural substrates from those of other, behavior system- or valence-specific changes. Moreover, our results present an opportunity to utilize associability enhancements to the advantage of counterconditioning procedures in therapeutic contexts.

- Abstract
Post-translational modifications frequently modulate protein functions. Lysine acetylation in particular plays a key role in interactions between respiratory cytochrome c and its metabolic partners. To date, in vivo acetylation of lysines at positions 8 and 53 has specifically been identified in mammalian cytochrome c, but little is known about the structural basis of acetylation-induced functional changes. Here, we independently replaced these two residues in recombinant human cytochrome c with glutamine to mimic lysine acetylation and then characterized the structure and function of the resulting K8Q and K53Q mutants. We found that the physicochemical features were mostly unchanged in the two acetyl-mimetic mutants, but their thermal stability was significantly altered. NMR chemical shift perturbations of the backbone amide resonances revealed local structural changes, and the thermodynamics and kinetics of electron transfer in mutants immobilized on gold electrodes showed an increase in both protein dynamics and solvent involvement in the redox process. We also observed that the K8Q (but not the K53Q) mutation slightly increased the binding affinity of cytochrome c to its physiological electron donor, cytochrome c1—which is a component of mitochondrial complex III, or cytochrome bc1—thus suggesting that Lys8 (but not Lys53) is located in the interaction area. Finally, the K8Q and K53Q mutants exhibited reduced efficiency as electron donors to complex IV, or cytochrome c oxidase

22. Estudio del crecimiento de la microalga Chlorella vulgares y su capacidad de depuración de agua residual

RESUMEN: Las microalgas son un grupo microbiano muy heterogéneo que proporciona diversas aplicaciones en la actualidad, entre las que se encuentra la depuración de aguas residuales. El tratamiento de las aguas residuales resulta muy importante, ya que estas contienen contaminantes que impiden su reutilización. En el presente estudio se ha analizado el crecimiento de la microalga Chlorella vulgaris y su capacidad de depuración en aguas residuales de secundario de diferente procedencia. Las microalgas son capaces de asimilar nitrógeno y fósforo del agua residual en forma de biomasa, pudiendo resultar un proceso más económico y sostenible frente a otros tipos de tratamientos terciarios. Para demostrar que este proceso es viable, se ha realizado un ensayo utilizando un sistema de 12 columnas de burbujeo con un volumen de cultivo de 250 ml, con cuatro diferentes medios de cultivo (ensayos por triplicado): medio control (Arnon modificado), efluente de secundario procedente de una EDAR (Estación Depuradora de Aguas Residuales), efluente de secundario procedente de un reactor de microalgas tipo raceway, y este último efluente suplementado con lixiviado procedente de la EDAR. Se ha visto que C. vulgaris es capaz de sobrevivir a las adversas condiciones presentes en las aguas residuales, eliminando eficientemente nitrógeno presente en forma de amonio y nitrato, y fósforo fundamentalmente en forma de fosfato. Los medios de cultivo que contenían aguas residuales, ya sean de la EDAR o del raceway, permitieron el crecimiento de la microalga, comprobando que esta puede crecer utilizando agua residual como único medio de cultivo, a pesar de la posible limitación de nutrientes. Se demostró que cuando el agua residual es suplementada en lixiviado (con elevada concentración de N en forma de amonio) el crecimiento es aún mayor, siempre y cuando no se alcancen los límites de inhibición para las microalgas. Así, en este trabajo se ha logrado obtener una productividad de biomasa de 0.32 g/(L·día) en ensayos en semicontinuo a una velocidad de dilución de 0.3 1/día con elevados porcentajes de depuración de nutrientes. Se demuestra, además, la importancia de determinar la cantidad óptima de lixiviado a añadir, y se proponen ensayos que añadan hasta un 25% v de lixiviado en base a un estudio bibliográfico. ABSTRACT: Currently, microalgae are a very heterogeneous microbial group that provides diverse applications including wastewater depuration. Wastewater treatment is very important as wastewater contain contaminants that prevent their re-use. The growth of Chlorella vulgaris microalgae and its sewage treatment capacity in secondary wastewater from different sources have been analyzed in this study. Microalgae are able to assimilate nitrogen and phosphorus from wastewater into biomass, possibly leading to a more economical and sustainable process against other sorts of tertiary treatments. In order to demonstrate that this depuration system is feasible, a trial has been performed using a system of 12 bubbling columns with a working volume of 250 ml, with four different culture media (trials by triplicate): medium control (modified Arnon), secondary effluent from WTP (Wastewater Treatment Plant), secondary effluent from a raceway type microalgae reactor, and the latter effluent supplemented with centrate from WTP. It has been reported that C. vulgaris is able to survive the adverse conditions present in wastewater, efficiently removing nitrogen present in the form of ammonium and nitrate, and phosphorus basically in the form of phosphate. The culture mediums containing wastewater, whether from the WTP or raceway, allowed the growth of the microalgae, proving that it can grow using wastewater as the only culture medium, despite possible nutrient limitations. It was demonstrated that when wastewater is supplemented with centrate (with high concentration of N in the form of ammonium) the growth is even greater, as long as inhibition limits for microalgae are not reached. Thus, in this work a biomass productivity of 0.32 g/(L·day) has been achieved in semi-continuous assays at a dilution rate of 0.3 1/day with high percentages of nutrient purification. The importance of determining the optimal amount of centrate to be added is also demonstrated. Studies that add up to 25% v centrate are proposed based on a bibliographic study.

Instrumental conditioning involves two different processes: Goal-directed behavior, characterized by its dependence on the causal relationship between action and outcome and the sensitivity of actions to changes in the value of the outcome; and habits, characterized for its persistence and insensitivity to changes after conditioning. It is known that the dopaminergic system is involved in both kind of learning. The present experiments analyzed two animal models of Parkinson's disease. The 6-OHDA model causes selective damage of the catecholaminergic neurons, specifically affecting the dopaminergic neurons in nigro-striatal system. This model simulates degenerative process symptomatology of Parkinson's disease. On the other hand, the LPS model generates an inflammation process in the infusion area. This model simulates the early symptoms of this disorder, including neuroinflammation and microglia activation. In order to validate both parkinsonian models, we studied if 6-OHDA and LPS models cause the same behavioral effects. The results showed that the 6-OHDA model interfered with the process involved in habit formation. In contrast, animals treated with LPS showed a goal-directed learning deficit. Differences between these models could be due to the different effects on Substantia Nigra neurons. 6-OHDA model might disrupt the nigrostriatal pathway, while LPS could interfere on efferences and afferences to Substantia Nigra.

- Abstract
Mitochondria are the powerhouses of the cell, whilst their malfunction is related to several human pathologies, including neurodegenerative diseases, cardiovascular diseases, and various types of cancer. In mitochondrial metabolism, cytochrome c is a small soluble heme protein that acts as an essential redox carrier in the respiratory electron transport chain. However, cytochrome c is likewise an essential protein in the cytoplasm acting as an activator of programmed cell death. Such a dual role of cytochrome c in cell life and death is indeed fine-regulated by a wide variety of protein post-translational modifications. In this work, we show how these modifications can alter cytochrome c structure and functionality, thus emerging as a control mechanism of cell metabolism but also as a key element in development and prevention of pathologies.

- Abstract
Increasing the thermal stability of immobilized proteins is a motivating goal for improving the performance of electrochemical biodevices. In this work, we propose the immobilization of crosslinked plastocyanin from the thermophilic cyanobacterium Phormidium laminosum by simultaneous incubation of a mixture of plastocyanin and the coupling reagents. The thermal stability of the so built covalently immobilized protein films has been assessed by cyclic voltammetry in the 0–90 °C temperature range and has been compared to that of physisorbed films. It is shown that the protein loss along a thermal cycle is significantly reduced in the case of the crosslinked films, whose redox properties remain unaltered along a cyclic heating-cooling thermal scan, and can withstand the contact with 70 °C solutions for four hours. Comparison of thermal unfolding curves obtained by circular dichroism spectroscopy of both free and crosslinked protein confirms the improved thermic resistance of the crosslinked plastocyanin. Notably, the electron transfer thermodynamics of physisorbed and crosslinked plastocyanin films are quite similar, suggesting that the formation of intra- and inter-protein amide bonds do not affect the integrity and functionality of the copper redox centers. UV–Vis absorption and circular dichroism measurements corroborate that protein crosslinking does not alter the coordination geometry of the metal center.

Nuestra propuesta de trabajo tiene como objetivo analizar el actual aprovechamiento turístico de la antigua vía férrea Agost-Alcoy (iniciada en 1926), a través de los 2 tramos de vía verde que hay rehabilitados; La Vía Verde del Maigmó que se extiende por los términos de Agost, Petrer y Tibi con un total de 22,5 km. y está gestionada por la Diputación de Alicante y la Vía Verde de Alcoy con 10,5 km. gestionada por el Ayuntamiento de Alcoy, ambos tramos de vía suman 33 km. y pertenecían al mismo trazado ferroviario que quedó en desuso. Tanto la Vía del Maigmó como la de Alcoy se restauraron como “Vías Verdes” hace poco más de una década y actualmente quedan por acondicionar otros 33 km. de toda la parte central para completar los 66 km. de vía original y de los que forma parte los municipios de Tibi, Castalla e Ibi. La posible recuperación integral de todo el trazado vertebraría el territorio de la costa hacia el interior, siendo un eje estratégico en la provincia donde se podría desarrollar un turismo de calidad y promovería los valores históricos, culturales, paisajísticos y medioambientales, así como motivaría la participación de las comunidades locales implicadas en la reivindicación, restauración y mantenimiento de una marca “Vías Verdes” que está muy consolidada tanto en España como en Europa, cuya misión principal es la sostenibilidad y la innovación social., Our work proposal aims to analyze the current tourism development of the old railroad Agost - Alcoy (started in 1926), through the two sections of greenway that have been rehabilitated; The Greenway of Maigmó that extends to Agost, Petrer and Tibi with a total of 22.5 km. and it is managed by the Provincial Council of Alicante and the Greenway of Alcoy is 10.5 km and is managed by the town hall of Alcoy. Both railroads add up to 33 km and they belonged to the same railway line that fell into disuse. Both the Maigmó line as Alcoy were restored as “Greenways” a little more than a decade ago and currently remain to put another 33 km. to the entire center to complete the 66 km of the original line and of the ones that form part of the municipals of Tibi, Castalla and Ibi. The possible full recovery of the entire layout would structure the territory of the coast inland, being a strategic area in the province where they could develop quality tourism and promote the historical, cultural, scenic and environmental values and motivate participation of local communities involved in the claim, restoration and maintenance of the brand “Greenways” is well established both in Spain and in Europe, whose main mission is sustainability and social innovation.

A large proportion of homeless people in the UK are former members of the armed services and suffer from a mental illness. In fact, homelessness itself can be considered a symptom or manifestation of other underlying psychological difficulties. For these reasons Community Housing and Therapy (CHT) considers that providing psychological therapies to treat the homeless population is a more effective way of tackling the problem of homelessness, as it addresses the roots of the problem. This approach is one which is beginning to be recognised by leading agencies in the field. At the same time, the provision of psychological therapies for symptoms such as depression and anxiety has become accepted through the Department of Health's (DoH) Increased Access to Psychological Therapies (IAPT) initiative. Depression is the most common psychiatric disorder that homeless people suffer and it is well documented that psychological treatments for depression can be extremely effective. As well as approaching homelessness from the angle of psychological therapies, CHT in its work with the ex-service community has become increasingly aware that there are a large number of non statutory homeless that do not get the same attention as rough sleepers. Una gran proporción de personas sin hogar en el Reino Unido corresponde a ex-miembros de las fuerzas armadas, quienes sufren además de enfermedades mentales. Introduction: This study aimed to evaluate the predictive validity and discriminatory ability of clinical outcomes, inflammatory activity, oxidative and vascular damage, and metabolic mechanisms for detecting significant improve maximum heart rate after physical activity training in individuals with psychiatric disorders and obesity comorbid using a longitudinal design and transdiagnostic perspective. Methods: Patients with major depressive disorder, bipolar disorder and, schizophrenia and with comorbid obesity (n = 29) were assigned to a 12-week structured physical exercise program. Peripheral blood biomarkers of inflammation, oxidative stress, vascular mechanisms, and metabolic activity, as well as neurocognitive and functional performance were assessed twice, before and after intervention. Maximum heart rate was considered a marker of effectiveness of physical activity. Mixed one-way analysis of variance and linear regression analyses were performed. Results: Individuals with psychiatric disorders and comorbid obesity exhibited an improvement in cognition, mood symptoms and body mass index, increase anti-inflammatory activity together with enhancement of the oxidative and cardiovascular mechanisms after physical activity training (p<0.05 to 0.0001; d = 0.47 to 1.63). A better clinical outcomes along with regulation of inflammatory, oxidative, and cardiovascular mechanisms were critical for predicting significant maximum heart rate variation over time (χ
2 = 32.2 to 39.0, p < 0.0001). Conclusions: The regulation of the anti-inflammatory mechanisms may be essential for maintained of healthy physical activity across psychiatric disorders and obesity. Likewise, inflammatory activity, oxidative stress, vascular and cardio-metabolic mechanisms may be a useful to identify individuals at greater risk of multi-comorbidity. Studies have shown that COVID-19 has had a worldwide psychological impact. Confinement due to COVID-19 has had important repercussions on the mental health of the general population, with high levels of stress, anxiety, depressive symptoms, post-traumatic stress disorder, and so forth. Similarly, important labor, economic and social changes taking place are affecting people's well-being. The objective of this study was to analyze the repercussions of perceived threat from COVID-19 on the mental health of the population, and to evaluate the mediating role of perceived economic impact. The participants were 1160 adult residents of Spain aged 18 to 82, 69.9% of whom were women. A sociodemographic questionnaire, the Questionnaire on Perception of Threat from COVID-19 and the General Health Questionnaire were administered. Perceived threat had a positive direct effect on all four health dimensions analyzed. Among the indirect effects, the perceived economic impact of COVID-19 mediated in the relationship between perceived threat and each of the health dimensions. The results of this study have demonstrated the need to promote joint action promoting public mental health to minimize the psychological repercussions of new outbreaks. [ABSTRACT FROM AUTHOR]

The present paper describes an optimized C++ library for the study of electromagnetics. The implementation is based on the Finite-Difference Time-Domain method for transient analysis, and the Finite Element Method for electrostatics. Both methods share the same core and are optimized for CPU and GPU computing. To illustrate its running, FEM method is applied for solving Laplace's equation analyzing the relation between surface curvature and electrostatic potential of a long cylindrical conductor, whereas FDTD is applied for analyzing Thin Film Filters at optical wavelengths. Furthermore, a comparison of the performance of both CPU and GPU versions is analyzed as a function of the grid size simulation. This approach allows the study of a wide range of electromagnetic problems taking advantage of the benefits of each numerical method and the computing power of the modern CPUs and GPUs. Photopolymers can be used to fabricate different holographic optical elements, although maximization of the phase-shift in photopolymers has been a challenge for the last few decades. Different material compositions and irradiation conditions have been studied in order to achieve it. One of the main conclusions has been that with continuous laser exposure better results are achieved. However, our results show for the first time that higher phase-shift can be achieved using a pulsed laser. The study has been conducted with crosslinked acrylamide-based photopolymers exposed with a pulsed laser (532 nm). The increment of the phase-shift between the pulsed laser and continuous laser exposure is 17%, achieving a maximum phase-shift of 3π radians and a refractive index shift of 0.0084 at the zero spatial frequency limit, where monomer diffusion does not take place. This allows this photopolymer to be used in large-scale manufacturing. [ABSTRACT FROM AUTHOR]

García Márquez es, por derecho propio, uno de los escritores clásicos de nuestra lengua, quizás el narrador más importante después de Cervantes, y como tal, su obra presenta múltiples capas interpretativas que parecen regenerarse con cada nuevo acercamiento crítico. Es por ello que aprovechando los fastos y alegrones caribes de sus primeros ochenta años, muchos especialistas (y no pocos diletantes) se han dado a la labor de cincelar de forma obsesiva las claves formales y temáticas de su universo inextinguible, poniendo en evidencia una vez más las inalcanzables cimas estéticas a las que ha llegado el Nobel colombiano, cuyas páginas son siempre una cascada de la mejor literatura escrita nunca en lengua española. A sabiendas de que no siempre es fácil decir cosas nuevas, el libro que presentamos fue pensado como un sentido homenaje al hijo del telegrafista de Aracataca, ahora que él también parece perderse por momentos en los meandros de la memoria, para habitar definitivamente, junto a Úrsula Iguarán, ese Macondo soñado por lectores de todas las lenguas, que no es sólo un lugar mítico, sino también un estado de ánimo que anida en la conciencia de sus seguidores. Gabriel García Márquez, la modernidad de un clásico es un libro de reconocimiento sincero al gran escritor colombiano, donde tienen cabida lecturas muy originales sobre su obra, lejos de las interpretaciones más encorsetadas que se han derivado de un mal entendido macondismo.

- Abstract
The response of a H-PDLC device is improved by means of a two-step method. First, component optimization—initiator system, crosslinker, and cosolvent—enables the diffraction efficiency of the hologram to be maximized. Second, the use of N-methyl-2-pyrrolidone in combination with N-vinyl-2-pyrrolidone prevents the overmodulation in photopolymers containing ethyl eosin.
